| 1 | Analysis of characteristics and predictive factors of immune checkpoint inhibitor-related adverse events显示文摘Objective:We aimed to retrospectively analyze the toxicity profiles and predictors of immune-related adverse events(irAEs)as well as the correlation between irAEs and the clinical efficacy of multi-type immune checkpoint inhibitors(ICIs)in patients with advanced pan-cancer in a real-world setting.Methods:We retrospectively analyzed data from 105 patients with advanced pan-cancer treated with multi-type ICIs at the First Hospital of Jilin University between January 1,2016 and August 1,2020.We used logistic regression analyses to investigate the associations of irAEs with clinical baseline characteristics,blood count parameters,and biochemical indicators during treatment.Receiver operating characteristic curves were used to determine cutoff values for parameters and area under the curve values.Kaplan–Meier and Cox multivariate regression analyses were performed to estimate the relationships of baseline characteristics and irAEs with progression-free survival(PFS)and overall survival(OS).Results:A lower relative lymphocyte count(cutoff=28.5%),higher albumin level(cutoff=39.05 g/L),and higher absolute eosinophil count(AEC)(cutoff=0.175×10^(9)/L)were significantly associated with the occurrence of irAEs,among which a higher AEC(cutoff=0.205×10^(9)/L)was strongly associated with skin-related irAEs[odds ratios(ORs)=0.163,P=0.004].Moreover,a higher lactate dehydrogenase level(cutoff=237.5 U/L)was an independent predictor of irAEs of grade≥3(OR=0.083,P=0.023).In immune cell subgroup analysis,a lower absolute count of CD8+CD28−suppressor T cells(OR=0.806;95%confidence interval:0.643–1.011;P=0.062),which are regulatory T lymphocytes,was associated with the occurrence of irAEs,although the difference was not statistically significant.Furthermore,a higher percentage of CD19+B cells was associated with the occurrence of irAEs of grade≥3(P=0.02)and grade≥2(P=0.051).In addition,patients with any grade of irAE had a significantly high PFS(8.37 vs.3.77 months,hazard ratios(HR)=2.02,P=0.0038)and OS(24.77 vs.13.83 months,HR=1.84;P=0.024).Conclusions:This retrospective study reports clinical profile data for irAEs in unselected patients in a real-world setting and explored some parameters that may be potential predictive markers of the occurrence,type,or grade of irAEs in clinical practice.Evidence of a correlation between safety and efficacy may facilitate a complete assessment of the risk-benefit ratio for patients treated with ICIs. | Rilan Bai Naifei Chen Xiao Chen Lingyu Li Wei Song Wei Li Yuguang Zhao Yongfei Zhang Fujun Han Zheng Lyu Jiuwei Cui | 2021 | Cancer Biology & Medicine2021,18,4: | 4 |
| 2 | Relative dynamics estimation of non-cooperative spacecraft with unknown orbit elements and inertial tensor显示文摘The state estimation for relative motion with respect to non-cooperative spacecraft in rendezvous and docking(RVD) is a challenging problem. In this paper, a completely non-cooperative case is considered, which means that both orbit elements and inertial tensor of target spacecraft are unknown. By formulating the equations of relative translational dynamics in the orbital plane of chaser spacecraft, the issue of unknown orbit elements is solved. And for the problem for unknown inertial tensor, we propose a novel robust estimator named interaction cubature Kalman filter(In CKF) to handle it. The novel filter consists of multiple concurrent CKFs interlacing with a maximum a posteriori(MAP) estimator. The initial estimations provided by the multiple CKFs are used in a Bayesian framework to form description of posteriori probability about inertial tensor and the MAP estimator is applied to giving the optimal estimation. By exploiting special property of spherical-radial(SR) rule, a novel method with respect to approximating the likelihood probability of inertial tensor is presented. In addition, the issue about vision sensor's location inconformity with center mass of chaser spacecraft is also considered. The performance of this filter is demonstrated by the estimation problem of RVD at the final phase. | 5 | Insight into the degradation mechanism of cefixime under crystallization condition显示文摘The chemical stability of cefixime was determined by high-performance liquid chromatography(HPLC) under different conditions, including factors such as p H, solvents, initial concentration, temperature and additives.The degradation process follows the first-order kinetics. A p H-rate profile exhibits the U-shape and shows the maximum stability of cefixime at pH = 6. The stability in different pure solvents is ranked as acetone N ethanol N methanol N water, while the degradation rate of cefixime exists a maximum at the ratio of 0.6 in water + methanol mixtures. In addition, the degradation rate increases with the temperature increasing and the activation energy of degradation was found to be 27.078 k J·mol^(-1) in acetone + water mixed solvents. The addition of different additives was proven to either inhibit or accelerate the degradation. | 6 | Survival outcomes of conversion surgery for metastatic pancreatic ductal adenocarcinoma after neoadjuvant therapy显示文摘Objective:To evaluate the survival outcomes of patients who underwent conversion surgery for metastatic pancreatic ductal adenocarcinoma(mPDAC)after neoadjuvant therapy(NAT)and to identify potential candidates that may benefit from this treat-ment strategy.Background:The role and eligibility population of conversion surgery for mPDAC remains controversial in the era of NAT.Methods:A consecutive cohort of patients diagnosed with mPDAC and treated with NAT followed by conversion surgery be-tween 2019 and 2021 were confirmed from a prospective database maintained by the Department of Pancreatic Hepatobiliary Surgery of Changhai Hospital.In accordance with residual metastases and technical resectability after NAT,patients were classi-fied as the complete pathological response of metastases(ypM0)resection group,residual metastases(ypM1)resection group,and exploration group.Median overall survival(mOS)was calculated using the Kaplan-Meier method,uni-and multivariable cox regression was performed to identify clinicopathological predictors of OS.Results:A total of 244 patients with mPDAC were identified from the prospective database,with 19(7.8%)patients who un-derwent ypM0 resection,22(9.0%)underwent ypM1 resection,and 23(9.4%)underwent explorative laparotomy.The mOS was 32.6 months for ypM0 resected patients,15.1 months for ypM1 resected patients,and 13.4 months for those who underwent explorative laparotomy(P<.001).Univariable and multivariable Cox regression analyses confirmed that ypM0 resection,normal-ization of preoperative CA19-9 levels,and continued adjuvant therapy were independent prognostic factors of conversion surgery for mPDAC after NAT.Subgroup analyses revealed that oligometastases and continued adjuvant therapy were associated with improved prognosis in the ypM1 resection group.Conclusion:In patients with mPDAC who underwent NAT followed by conversion surgery,the complete pathological response of metastases,normalization of preoperative CA19-9 levels,and continued adjuvant therapy were independent risk factors for prognosis.Patients with residual oligometastases after treatment were expected to prolong survival through resection.These patients may benefit from conversion surgery and should be potential candidates for this treatment strategy. | Lingyu Zhu Suizhi Gao Xinqian Wu Bo Li Xiaohan Shi Xiaoyi Yin Huan Wang Meilong Shi Penghao Li Yikai Li Chaoliang Zhong Chuanqi Teng Jiawei Han Yiwei Ren Jian Wang Zhendong Fua Xinyu Liu Kailian Zheng Shiwei Guo Gang Jin | 2023 | Journal of Pancreatology2023,6,3: | 0 |
| 7 | Acute gastroenteritis outbreaks caused by human astrovirus,1978-2021:A systematic review显示文摘Human astrovirus(HAstV)is one of the main pathogens that cause sporadic cases of acute gastroenteritis,sometimes leading to outbreaks.This study aimed to elucidate the epidemiological and etiological character-istics of HAstV outbreaks worldwide.Literature on HAstV outbreaks published before January 2022 was retrieved from the China National Knowledge Infrastructure,WanFang,WeiPu,PubMed,and Web of Science databases.Date,region,population,settings,transmission modes,clinical symptoms,and etiological characteristics of the outbreaks were collected and analyzed.Thirty-one articles on 32 HAstV outbreaks reported between November 1978 and October 2018 were included.The outbreaks mainly occurred in autumn(14/32,43.75%),and more of them were reported in 1996,2004,and 2017.Outbreaks were primarily dis-tributed in the Northern Hemisphere and mainly occurred in nursery centers and kindergartens(9/29,31.03%),hospitals(5/29,17.24%),and schools(4/29,13.79%).Viral genotypes were identified during 19 outbreaks,and HAstV-1 was predominant(8/19,42.10%).Eleven outbreaks were caused by mixed infection,and norovirus(9/11,81.82%)and rotavirus(5/11,45.45%)were the most common mixed pathogens.The transmission routes were reported in 9 outbreaks of mixed infection,and most(7/9)were related to water-borne and foodborne transmission.Although HAstV outbreaks are infrequently reported,it is necessary to con-sider HAstV in norovirus-negative gastroenteritis outbreaks.In addition,local Centers for Disease Control and Prevention should have the capacity to handle HAstV outbreaks and identify pathogens. | Jiamei Fu Fan Yu Hanning Li Lingyu Shen Yi Tian Lei Jia Daitao Zhang Peng Yang Quanyi Wang Zhiyong Gao | 2023 | Biosafety and Health2023,5,2: | 0 |
| 8 | Prevalence of Metabolic Obese Normal Weight and Related Risk Factors in South China显示文摘[Objectives]Metabolic obese normal weight(MONW)is becoming one of the pubic problems which are threatening human health.Whereas,MONW was facing a great challenge for limited attention,especially for the female in China.The aim of this research was to estimate the prevalence of MONW and its related risk components in South China.[Methods]A community-based cross-sectional study was performed on 3349 residents aged 18-93 years in The First Affiliated Hospital of Jinan University,Guangzhou,China,in 2019.Data was collected by physical examination data which included physical measurements and laboratory examinations.[Results]In all subjects,55%were females(M/F=1509/1840).The prevalence of MONW was 16.09%(0.04%for male,16.05%for female,P<0.001).The prevalence increased significantly with increasing age in both genders(P<0.001).The binary logistic regression analysis shows that among the risk factors with MONW,age,BMI,gender,systolic pressure,hypertension[Male:ORs=2.56,95%CI(1.23,5.32);Female:ORs=2.88,95%CI(1.76,4.71)]and hypertriglyceridemia[Male:ORs=3.23,95%CI(1.67,6.19);Female:ORs=2.57,95%CI(1.64,4.03)]were found to be statistically significant.The level of ALT in MONW group was(27.88±15.85)in male and(24.33±15.75)in female,which were significantly higher than those in the non-MetS group.[Conclusions]The prevalence of MONW was pretty high.We considered MONW be significantly associated with the increase of ALT.Female gender,advanced age,and elevated ALT were independent risk factors for MONW.It was high time that the government should raise the public attention toward metabolic function in normal weight population.Effective prevention and treatment strategies for MONW and its risk factors should be developed targeting different ages and genders. | Xuehua LUO Zien LIANG Yan LI Qingbin WU Zhiqiang PAN Yuqing LIANG Lingyu LI Juanjuan MAI Huijun XIE Li HAN | 2021 | Medicinal Plant2021,12,2: | 0 |
| 9 | PTPRO represses colorectal cancer tumorigenesis and progression by reprogramming fatty acid metabolism显示文摘Background:Abnormal expression of protein tyrosine phosphatases(PTPs)has been reported to be a crucial cause of cancer.As a member of PTPs,protein tyrosine phosphatase receptor type O(PTPRO)has been revealed to play tumor suppressive roles in several cancers,while its roles in colorectal cancer(CRC)remains to be elucidated.Hence,we aimed to explore the roles and mechanisms of PTPRO in CRC initiation and progression.Methods:The influences of PTPRO on the growth and liver metastasis of CRC cells and the expression patterns of different lipid metabolism enzymes were evaluated in vitro and in vivo.Molecular and biological experiments were conducted to uncover the underpinning mechanisms of dysregulated de novo lipogenesis and fatty acidβ-oxidation.Results:PTPRO expression was notably downregulated in CRC liver metastasis compared to the primary cancer,and such a downregulation was associated with poor prognosis of patients with CRC.PTPRO silencing significantly promoted cell growth and liver metastasis.Compared with PTPRO wild-type mice,PTPROknockout mice developed more tumors and harbored larger tumor loads under treatment with azoxymethane and dextran sulfate sodium.Gene set enrichment analysis revealed that PTPRO downregulation was significantly associated with the fatty acid metabolism pathways.Blockage of fatty acid synthesis abrogated the effects of PTPRO silencing on cell growth and liver metastasis.Further experiments indicated that PTPRO silencing induced the activation of the AKT serine/threonine kinase(AKT)/mammalian target of rapamycin(mTOR)signaling axis,thus promoting de novo lipogenesis by enhancing the expression of sterol regulatory element-binding protein 1(SREBP1)and its target lipogenic enzyme acetyl-CoA carboxylase alpha(ACC1)by activating the AKT/mTOR signaling pathway.Furthermore,PTPRO attenuation decreased the fatty acid oxidation rate by repressing the expression of peroxisome proliferator-activated receptor alpha(PPARα)and its downstream enzyme peroxisomal acyl-coenzyme A oxidase 1(ACOX1)via activating the p38/extracellular signal-regulated kinase(ERK)mitogen-activated protein kinase(MAPK)signaling pathway.Conclusions:PTPRO could suppress CRC development and metastasis via modulating the AKT/mTOR/SREBP1/ACC1 and MAPK/PPARα/ACOX1 pathways and reprogramming lipid metabolism. | Weixing Dai Wenqiang Xiang Lingyu Han Zixu Yuan Renjie Wang Yanlei Ma Yongzhi Yang Sanjun Cai Ye Xu Shaobo Mo Qingguo Li Guoxiang Cai | 2022 | Cancer Communications2022,42,9: | 0 |
| 10 | The construction of pseudo-Janus silica/surfactant assembly and their application to stabilize Pickering emulsions and enhance oil recovery显示文摘Nanoparticles with high surface energy and chemical activity have drawn substantial attention in petroleum industry. Recently, Janus nanoparticles exhibited tremendous potential in enhanced oil recovery (EOR) due to their asymmetric structures and properties. In this study, a series of amphiphilic pseudo-Janus@OTAB (PJ@C18) nanoparticles with different concentrations of stearyltrimethylammoium bromide (OTAB) were successfully fabricated. The structures and properties of PJ@C18 were characterized by Fourier transform infrared spectroscopy and ζ-potential measurements. Based on the emulsification experimental results, the interaction models and the self-assembly behavior between hydrophilic nanoparticles (SiO_(2)@NH_(2)) and OTAB molecules at the oil/water interface were proposed, which was further confirmed via the measurements of the contact angle and dynamic interfacial tension. Interestingly, it was found that the change of pH value from 7.5 to 4.0 caused the type reversal of the PJ@C18-1000 stabilized Pickering emulsions. Furthermore, the PJ@C18-1000 stabilized Pickering emulsion system with excellent salt and temperature tolerances (10000 mg∙L^(–1), 90℃) significantly improved the oil recovery in the single-tube (more than 17%) and double-tube (more than 25%) sand pack model flooding tests. | 11 | Research on data pre-deployment in information service flow of digital ocean cloud computing显示文摘Data pre-deployment in the HDFS(Hadoop distributed file systems) is more complicated than that in traditional file systems. There are many key issues need to be addressed, such as determining the target location of the data prefetching, the amount of data to be prefetched, the balance between data prefetching services and normal data accesses. Aiming to solve these problems, we employ the characteristics of digital ocean information service flows and propose a deployment scheme which combines input data prefetching with output data oriented storage strategies. The method achieves the parallelism of data preparation and data processing, thereby massively reducing I/O time cost of digital ocean cloud computing platforms when processing multi-source information synergistic tasks.
