| 1 | Cancer-associated fibroblasts in breast cancer:Challenges and opportunities显示文摘The tumor microenvironment is proposed to contribute substantially to the progression of cancers,including breast cancer.Cancer-associated fibroblasts(CAFs)are the most abundant components of the tumor microenvironment.Studies have revealed that CAFs in breast cancer originate from several types of cells and promote breast cancer malignancy by secreting factors,generating exosomes,releasing nutrients,reshaping the extracellular matrix,and suppressing the function of immune cells.CAFs are also becoming therapeutic targets for breast cancer due to their specific distribution in tumors and their unique biomarkers.Agents interrupting the effect of CAFs on surrounding cells have been developed and applied in clinical trials.Here,we reviewed studies examining the heterogeneity of CAFs in breast cancer and expression patterns of CAF markers in different subtypes of breast cancer.We hope that summarizing CAFrelated studies from a historical perspective will help to accelerate the development of CAF-targeted therapeutic strategies for breast cancer. | Dengdi Hu Zhaoqing Li Bin Zheng Xixi Lin Yuehong Pan Peirong Gong Wenying Zhuo Yujie Hu Cong Chen Lini Chen Jichun Zhou Linbo Wang | 2022 | Cancer Communications2022,42,5: | 9 |
| 2 | Electroacupuncture at Tianshu (ST 25) for diarrhea-predominant irritable bowel syndrome using positron emission tomography Changes in visceral sensation center显示文摘Previous studies have demonstrated that electroacupuncture therapy is effective in the treatment of irritable bowel syndrome. However, the precise mechanism of this therapy is unknown. The present study served to investigate the effects of electroacupuncture therapy on treatment of patients with diarrhea-predominant irritable bowel syndrome (IBS). We compared brain activation maps based on the changes of cerebral glucose metabolism obtained by 18-fluorodeoxyglucose positron emission tomography scanning under three conditions: resting, rectal balloon distension and rectal balloon distension plus electroacupuncture. Under the resting condition, compared with healthy controls, IBS patients displayed an increasing regional cerebral metabolic rate of glucose over a wide range: bilateral superior temporal gyrus, right middle occipital gyrus, superior frontal gyrus and bilateral middle frontal gyrus. However, there was no significant activity in the visceral pain center. Compared with the resting condition, under the rectal balloon distension condition, patients with IBS had a greater regional cerebral metabolic rate of glucose in the prefrontal cortex, left anterior cingulate cortex, postcentral gyrus, precentral gyrus and temporal gyrus. Under the rectal balloon distension plus electroacupuncture condition, stimulation by electroacupuncture at Tianshu (ST 25) manifested a decreased regional cerebral metabolic rate of glucose in the left cingulate gyrus, right insula, right caudate nucleus, fusiform gyrus and hippocampal gyrus. | 4 | Overexpression of miR-431 attenuates hypoxia/reoxygenation-induced myocardial damage via autophagy-related 3显示文摘Myocardial injury is still a serious condition damaging the public health. Clinically, myocardial injury often leads to cardiac dysfunction and, in severe cases, death. Reperfusion of the ischemic myocardial tissues can minimize acute myocardial infarction (AMI)-induced damage. MicroRNAs are commonly recognized in diverse diseases and are often involved in the development of myocardial ischemia/reperfusion injury. However, the role of miR-431 remains unclear in myocardial injury. In this study, we investigated the underlying mechanisms of miR-431 in the cell apoptosis and autophagy of human cardiomyocytes in hypoxia/reoxygenation (H/R). H/R treatment reduced cell viability, promoted cell apoptotic rate, and down-regulated the expression of miR-431 in human cardiomyocytes. The down-regulation of miR-431 by its inhibitor reduced cell viability and induced cell apoptosis in the human cardiomyocytes. Moreover, miR-431 down-regulated the expression of autophagy-related 3 (ATG3) via targeting the 3ʹ-untranslated region of ATG3. Up-regulated expression of ATG3 by pcDNA3.1-ATG3 reversed the protective role of the overexpression of miR-431 on cell viability and cell apoptosis in H/R-treated human cardiomyocytes. More importantly, H/R treatments promoted autophagy in the human cardiomyocytes, and this effect was greatly alleviated via miR-431-mimic transfection. | 8 | Single-cell transcriptomic analysis reveals a systemic immune dysregulation in COVID-19-associated pediatric encephalopathy显示文摘Unraveling the molecular mechanisms for COVID-19-associated encephalopathy and its immunopathology is crucial for developing effective treatments.Here,we utilized single-cell transcriptomic analysis and integrated clinical observations and laboratory examination to dissect the host immune responses and reveal pathological mechanisms in COVID-19-associated pediatric encephalopathy.We found that lymphopenia was a prominent characteristic of immune perturbation in COVID-19 patients with encephalopathy,especially those with acute necrotizing encephalopathy(AE).This was characterized a marked reduction of various lymphocytes(e.g.,CD8^(+)T and CD4^(+)T cells)and significant increases in other inflammatory cells(e.g.,monocytes).Further analysis revealed activation of multiple cell apoptosis pathways(e.g.,granzyme/perforin-,FAS-and TNF-induced apoptosis)may be responsible for lymphopenia.A systemic S100A12 upregulation,primarily from classical monocytes,may have contributed to cytokine storms in patients with AE.A dysregulated type I interferon(IFN)response was observed which may have further exacerbated the S100A12-driven inflammation in patients with AE.In COVID-19 patients with AE,myeloid cells(e.g.,monocytic myeloid-derived suppressor cells)were the likely contributors to immune paralysis.Finally,the immune landscape in COVID-19 patients with encephalopathy,especially for AE,were also characterized by NK and T cells with widespread exhaustion,higher cytotoxic scores and inflammatory response as well as a dysregulated B cell-mediated humoral immune response.Taken together,this comprehensive data provides a detailed resource for elucidating immunopathogenesis and will aid development of effective COVID-19-associated pediatric encephalopathy treatments,especially for those with AE. | Yi Wang Laurence Don Wai Luu Shuang Liu Xiong Zhu Siyuan Huang Fang Li Xiaolan Huang Linying Guo Jin Zhang Haiyan Ge Yuanyuan Sun Yi Hui Yanning Qu Huicong Wang Xiaoxia Wang Weilan Na Juan Zhou Dong Qu Jun Tai | 2023 | Signal Transduction and Targeted Therapy2023,8,11: | 0 |

The present paper discusses the types of disturbances and their influence on the ecologic system in nature reserves. It is shown that nature reserves are facing a great challenge dealing with these disturbances. | 11 | PA1426 regulates Pseudomonas aeruginosa quorum sensing and virulence:an in vitro study显示文摘Objective:Pseudomonas aeruginosa(P.aeruginosa)contains a hierarchy of quorum sensing(QS)network,consisting of the las,rhl and pqs systems,which play a key role in coordinating the expression of virulence factors.PA2146 was found to be associated with P.aeruginosa pathogenicity in macrophage and host Immune response.The aim of this study was to investigate the effects of PA2146 on the virulence of P.aeruginosa and explore its mechanism.Methods:PA2146 gene knockout strain and complement strain of P.aeruginosa PAO1 were constructed.The biomass of biofilm was detected by crystal violet staining;the virulence factors were measured,including pyocyanin,rhamnolipid,LasA elastase,LasB elastase and hemolytic activity;RNA-seq and label-free relative quantitative proteomics analyses were carried out to test the influence of PA2146 on transcriptomics and proteomics.This study was approved by the Institutional Review Board of the Third Xiangya Hospital,Central South University,China(approval No.2019-S021).Results:PA2146-deficient strains showed reduced biofilm formation and increased pyocyanin,rhamnolipid,LasA elastase,LasB elastase,and hemolytic activity,as well as increased motility,compared with the wild-type strain.RNA-seq and label-free relative quantitative proteomics analyses revealed that PA2146 repressed the transcription of several genes that are integral to the pqs system and to pyocyanin biosynthesis,and increased the expression of MexEF-OprN efflux pump components at the gene and protein level.Conclusion:PA2146 gene was found to inhibit the pqs system.PA2146 may affect quorum sensing by directly inhibiting the pqs system or by enhancing the expression of MexEF-OprN efflux pump components,thereby promoting efflux of 2-heptyl-4(1H)-quinolone,a Pseudomonas quinolone signal precursor,and thus affecting P.aeruginosa virulence. | Fang Tan Pengfei She Linying Zhou Shijia Li Xianghai Zeng Lanlan Xu Yaqian Liu Zubair Hussain Yong Wu | 2021 | Journal of Bio-X Research2021,4,1: | 0 |
