1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Prediction of Primary Climate Variability Modes at the Beijing Climate Center显示文摘Climate variability modes, usually known as primary climate phenomena, are well recognized as the most important predictability sources in subseasonal–interannual climate prediction. This paper begins by reviewing the research and development carried out, and the recent progress made, at the Beijing Climate Center(BCC) in predicting some primary climate variability modes. These include the El Ni?o–Southern Oscillation(ENSO), Madden–Julian Oscillation(MJO), and Arctic Oscillation(AO), on global scales, as well as the sea surface temperature(SST) modes in the Indian Ocean and North Atlantic, western Pacific subtropical high(WPSH), and the East Asian winter and summer monsoons(EAWM and EASM, respectively), on regional scales. Based on its latest climate and statistical models, the BCC has established a climate phenomenon prediction system(CPPS) and completed a hindcast experiment for the period 1991–2014. The performance of the CPPS in predicting such climate variability modes is systematically evaluated. The results show that skillful predictions have been made for ENSO, MJO, the Indian Ocean basin mode, the WPSH, and partly for the EASM, whereas less skillful predictions were made for the Indian Ocean Dipole(IOD) and North Atlantic SST Tripole, and no clear skill at all for the AO, subtropical IOD, and EAWM. 3A direct position determination method with combined TDOA and FDOA based on particle filter显示文摘The localization of a stationary transmitter using moving receivers is considered.The original Direct Position Determination(DPD) methods, with combined Time Difference of Arrival(TDOA) and Frequency Difference of Arrival(FDOA), do not perform well under low Signal-toNoise Ratio(SNR), and worse still, the computation cost is difficult to accept when the computational capabilities are limited.To get better positioning performance, we present a new DPD algorithm that proves to be more computationally efficient and more precise for weak signals than the conventional approach.The algorithm partitions the signal received with the same receiver into multiple non-overlapping short-time signal segments, and then uses the TDOA, the FDOA and the coherency among the short-time signals to locate the target.The fast maximum likelihood estimation, one iterative method based on particle filter, is designed to solve the problem of high computation load.A secondary but important result is a derivation of closed-form expressions of the Cramer-Rao Lower Bound(CRLB).The simulation results show that the algorithm proposed in this paper outperforms the traditional DPD algorithms with more accurate results and higher computational efficiency, and especially at low SNR, it is more close to the CRLB.Zhiyu LU Bin BA Jianhui WANG Wenchao LI Daming WANG 2018Chinese Journal of Aeronautics2018,31,1:11
4Single-shot BOTDA based on an optical chirp chain probe wave for distributed ultrafast measurement显示文摘Brillouin optical time-domain analysis(BOTDA)requires frequency mapping of the Brillouin spectrum to obtain environmental information(e.g.,temperature or strain)over the length of the sensing fiber,with the finite frequencysweeping time-limiting applications to only static or slowly varying strain or temperature environments.To solve this problem,we propose the use of an optical chirp chain probe wave to remove the requirement of frequency sweeping for the Brillouin spectrum,which enables distributed ultrafast strain measurement with a single pump pulse.The optical chirp chain is generated using a frequency-agile technique via a fast-frequency-changing microwave,which covers a larger frequency range around the Stokes frequency relative to the pump wave,so that a distributed Brillouin gain spectrum along the fiber is realized.Dynamic strain measurements for periodic mechanical vibration,mechanical shock,and a switch event are demonstrated at sampling rates of 25 kHz,2.5 MHz and 6.25 MHz,respectively.To the best of our knowledge,this is the first demonstration of distributed Brillouin strain sensing with a wide-dynamic range at a sampling rate of up to the MHz level.Dengwang Zhou Yongkang Dong Benzhang Wang Chao Pang Dexin Ba Hongying Zhang Zhiwei Lu Hui Li Xiaoyi Bao 2018Light(Science & Applications)2018,7,1:11
5Involvement of the Mitochondrion-dependent and the Endoplasmic Reticulum Stress-signaling Pathways in Isoliquiritigenin-induced Apoptosis of HeLa Cell显示文摘Objective Isoliquiritigenin(ISL),a licorice chalconoid,is considered to be a bioactive agent with chemopreventive potential.This study investigates the mechanisms involved in ISL-induced apoptosis in human cervical carcinoma HeLa cells.Methods Cell viability was evaluated using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide(MTT)assay.Apoptosis was determined by flow cytometry using an Annexin V-FITC Apoptosis Detection Kit.The intracellular ROS levels were assessed using a 2,7-dichlorofluorescein probe assay.The mitochondrial membrane potential was measured with the dual-emission potential-sensitive probe 5,5',6,6'-tetra-chloro-1,1',3,3'-tetraethyl-imidacarbocyanine iodide(JC-1).The degradation of poly-ADP-ribose polymerase(PARP)protein,the phosphorylation of PKR-like ER kinase(PERK),the phosphorylation of the α-subunit of eukaryotic initiation factor 2(eIF2α),the expression of the 78 kD glucose-regulated protein(GRP 78),and the activation of caspase-12 were analyzed via western blot analysis.Results ISL significantly inhibited the proliferation,the increase in ROS levels and apoptotic rates of HeLa cells in a concentration-dependent manner.Moreover,ISL induced mitochondrial dysfunction,caspase activation,and PARP cleavage,which displayed features of mitochondria dependent on apoptotic signals.Besides,exposure of HeLa cells to ISL triggered endoplasmic reticulum(ER)stress,as indicated by the increase in p-eIF2α and GRP78 expression,ER stress-dependent apoptosis is caused by the activation of ER-specific caspase-12.Conclusion The findings from our study suggest that ISL-induced oxidative stress causes HeLa cell apoptosis via the mitochondrion-dependent and the ER stress-triggered signaling pathways.YUAN Xuan ZHANG Bo GAN Lu WANG Zhen Hua YU Ba Cui LIU Liang Liang ZHENG Qiu Sheng WANGZhi Ping 2013Biomedical and Environmental Sciences2013,26,4:8
6Irinotecan plus S-1 versus S-1 in patients with previously treated recurrent or metastatic esophageal cancer(ESWN 01):a prospective randomized,multicenter,open-labeled phase 3 trial显示文摘Background:The benefit of systemic treatments in esophageal squamous cell carcinoma(ESCC)which has pro-gressed after chemotherapy is still uncertain and optimal regimens based on randomized trials have not yet been established.We aimed to compare the efficacy of irinotecan plus S-1 with S-1 monotherapy in recurrent or metastatic ESCC patients who had resistance to platinum-or taxane-based chemotherapy.Methods:We conducted a prospective randomized,multicenter,open-label,phase 3 trial in 15 centers across China.Eligible patients were adults with histologically confirmed recurrent or metastatic ESCC,and were randomly assigned(ratio,1:1)to receive either irinotecan plus S-1(intravenous infusion of irinotecan[160 mg/m2]on day 1 and oral S-1[80-120 mg]on days 1-10,repeated every 14 days)or oral S-1 monotherapy(80-120 mg/day on days 1-14,repeated every 21 days)using a central computerized minimization procedure.The primary endpoint was progression-free survival(PFS).Results:Between December 23,2014 and July 25,2016,we screened 148 patients and randomly assigned 123 patients to receive either irinotecan plus S-1 regimen(n=61)or S-1 monotherapy(n=62).After a median follow-up of 29.2 months(95%confidence interval[CI]17.5-40.9 months),the median PFS was significantly longer in the irinotecan plus S-1 group than in the S-1 monotherapy group(3.8 months[95%CI 2.9-4.3 months]vs.1.7 months[95%CI 1.4-2.7 months],hazard ratio=0.58,95%CI 0.38-0.86,P=0.006).The objective response rates were 24.6%in the irinotecan plus S-1 group and 9.7%in the S-1 monotherapy group(P=0.002).The patients in the irinotecan plus S-1 group presented with increased rates of grade 3-4 leukopenia(16.4%vs.0%),neutropenia(14.8%vs.1.6%),and nausea(4.9%vs.0%).No significant difference in grade 3-4 diarrhea and no treatment-related deaths were observed in both groups.Conclusions: The combination of irinotecan with S-1 was similarly tolerable but significantly prolonged PFS compared to S-1 monotherapy as a second- or third-line treatment in patients with recurrent or metastatic ESCC.Jing Huang Binghe Xu Ying Liu Junxing Huang Ping Lu Yi Ba Lin Wu Yuxian Bai Shu Zhang Jifeng Feng Ying Cheng Jie Li Lu Wen Xianglin Yuan Changwu Ma Chunhong Hu Qingxia Fan Xi Wang 2019Cancer Communications2019,39,1:8
7Zircon and Monazite Ages Constraints on Devonian Magmatism and Granulite-Facies Metamorphism in the Southern Qaidam Block:Implications for Evolution of Proto-and Paleo-Tethys in East Asia显示文摘High-temperature magma generation process and granulite-facies metamorphism can provide important information about mantle-crustal interaction and tectonic evolution. The strongly peraluminous monzonite pluton, the Jinshuikou cordierite granite on the southern margin of the Qaidam Block, can provide important information about the mantle-crustal interaction and constraints on tectonic transition from Proto-Tethys to Paleo-Tethys. This pluton develops enclaves of mafic granulite, amphibolite and quartzofeldspathic rocks, and is cut by massive monzonitic leuco-granite veins. Zircon and monazite U-Pb dating for the cordierite granite, the granulite enclaves and a massive monzonitic leuco-granite vein reveal that the cordierite granitic magma was generated from Mesoproterozoic continental crust with protolith derived from a provenance that was composed of >2.8 Ga old recycled crustal materials and recorded a ~1.7 Ga magmatic event. The continental crust underwent low-pressure granulite-facies metamorphism at ~380 Ma ago, whereas the cordierite granite magmas was generated and emplaced during 380 Ma, followed by intrusion of the massive monzonitic leuco-granite vein at circa 370–330 Ma. These data suggest that after the final closure of Proto-Tethys Ocean spreading along the southern Qaidam Block at ~420 Ma, break-off of the subducted slab or delamination of the lower crustal base and upwelling of the asthenospheric mantle beneath the southern Qaidam Block occurred before the Mid-Devonian, and that the initiation of the Paleo-Tethys tectonics might initiate near the end of Early-Carboniferous in the East Kunlun-Qaidam region, East Asia.Jin Ba Lu Zhang Chuan He Neng-Song Chen Timothy M.Kusky Qinyan Wang Yusheng Wan Xiaoming Liu 2018Journal of Earth Science2018,29,5:6

11Hundred-Joule-level,nanosecond-pulse Nd:glass laser system with high spatiotemporal beam quality显示文摘A 100-J-level Nd:glass laser system in nanosecond-scale pulse width has been constructed to perform as a standard source of high-fluence-laser science experiments. The laser system, operating with typical pulse durations of 3–5 ns and beam diameter 60 mm, employs a sequence of successive rod amplifiers to achieve 100-J-level energy at 1053 nm at3 ns. The frequency conversion can provide energy of 50-J level at 351 nm. 13Investigation of influence of micro-structure on magnetic properties of amorphous powder core显示文摘The influence of micro-structure on magnetic properties of amorphous powder core was investigated.
