2A new choice of stent for transjugular intrahepatic portosystemic shunt creation: Viabahn ePTFE covered stent/bare metal stent combination显示文摘Objectives:To compare the clinical outcomes in terms of structure and function between the insertion of a transjugular intrahepatic portosystemic shunt(TIPS) created with the Viabahn ePTFE covered stent/bare metal stent(BMS) combination and the Fluency ePTFE covered stent/BMS combination.Methods:A total of 101 consecutive patients who received a TIPS from February 2016 to August 2018 in our center were retrospectively analyzed.Sixty-four subjects were enrolled in the Viabahn group and 37 were enrolled in the Fluency group.The geometry characteristics of the TIPS were calculated,and the associated occurrence of shunt dysfunction,survival,overt hepatic encephalopathy,and variceal rebleeding were evaluated.Results:The technical success rate was 100%.After the insertion of the TIPS,the rate of shunt dysfunction during the first 3 months was significantly different between the Viabahn and Fluency groups(1.6% and 13.5%,respectively;p=0.024).Multivariate analysis indicated that the angle of portal venous inflow(α) was the only independent risk factor for shunt dysfunction(hazard ratio=1.060,95% confidence interval=1.009-1.112,p=0.020).In addition,3 months after the TIPS insertion,the a angle distinctly increased from 20.9°±14.3°-26.9°±20.1°(p=0.005) in the Fluency group but did not change significantly in the Viabahn group(from 21.9°±15.1°-22.9°± 17.6°,p=0.798).Conclusions:Shunt dysfunction was related to the a angle owing to the slight effect on the a angle after the implantation of the TIPS.The Viabahn ePTFE covered stent/BMS combination was more stable in structure and promised higher short-term stent patency compared with the Fluency ePTFE covered stent/BMS combination.Jiacheng Liu Jie Meng Chen Zhou Qin Shi Chongtu Yang Jinqiang Ma Manman Chen Bin Xiong 2021Journal of Interventional Medicine2021,4,1:9
3AFF3-DNA methylation interplay in maintaining the mono-allelic expression pattern of XIST in terminally differentiated cells显示文摘X chromosome inactivation and genomic imprinting are two classic epigenetic regulatory processes that cause mono-allelic gene expression.In female mammals,mono-allelic expression of the long non-coding RNA gene X-inactive specific transcript(XIST)is essential for initiation of X chromosome inactivation upon differentiation.We have previously demonstrated that the central factor of super elongation complex-like 3(SEC-L3),AFF3,is enriched at gamete differentially methylated regions(DMRs)of the imprinted loci and regulates the imprinted gene expression.Here,we found that AFF3 can also bind to the DMR downstream of the XIST promoter.Knockdown of AFF3 leads to de-repression of the inactive allele of X IST in terminally differentiated cells.In addition,the binding of AFF3 to the XIST DMR relies on DNA methylation and also regulates DNA methylation level at DMR region.However,the KAP1-H3K9 methylation machineries,which regulate the imprinted loci,might not play major roles in maintaining the mono-aUelic expression pattern of XIST in these cells.Thus,our results suggest that the differential mechanisms involved in the X IST DMR and gDMR regulation,which both require AFF3 and DNA methylation.Yue Zhang Chao Wang Xiaoxu Liu Qian Yang Hongliang Ji Mengjun Yang Manman Xu Yunyan Zhou Wei Xie Zhuojuan Luo Chengqi Lin 2019Journal of Molecular Cell Biology2019,11,9:3

5A single-cell transcriptome of mesenchymal stromal cells to fabricate bioactive hydroxyapatite materials for bone regeneration显示文摘The osteogenic microenvironment of bone-repairing materials plays a key role in accelerating bone regeneration but remains incompletely defined,which significantly limits the application of such bioactive materials.Here,the transcriptional landscapes of different osteogenic microenvironments,including three-dimensional(3D)hydroxyapatite(HA)scaffolds and osteogenic medium(OM),for mesenchymal stromal cells(MSCs)in vitro were mapped at single-cell resolution.Our findings suggested that an osteogenic process reminiscent of endochondral ossification occurred in HA scaffolds through sequential activation of osteogenic-related signaling pathways,along with inflammation and angiogenesis,but inhibition of adipogenesis and fibrosis.Moreover,we revealed the mechanism during OM-mediated osteogenesis involves the ZBTB16 and WNT signaling pathways.Heterogeneity of MSCs was also demonstrated.In vitro ossification of LRRC75A+MSCs was shown to have better utilization of WNT-related ossification process,and PCDH10+MSCs with superiority in hydroxyapatite-related osteogenic process.These findings provided further understanding of the cellular activity modulated by OM conditions and HA scaffolds,providing new insights for the improvement of osteogenic biomaterials.This atlas provides a blueprint for research on MSC heterogeneity and the osteogenic microenvironment of HA scaffolds and a database reference for the application of bioactive materials for bone regeneration.Peng Guo Xizhe Liu Penghui Zhang Zhongyuan He Zhen Li Mauro Alini RGeoff Richards Sibylle Grad Martin J.Stoddart Guangqian Zhou Xuenong Zou Danny Chan Wei Tian Dafu Chen Manman Gao Zhiyu Zhou Shaoyu Liu 2022Bioactive Materials2022,7,3:3
6The Effect of Unannounced Inspection on Prevention of Drug Fraud显示文摘Alarge number of incidents related to fake/inferior-quality drugs have occurred in China in recent years. In order to reduce drug crimes, the China Food and Drug Administration (CFDA) has exploited the new supervision approach, i.e., unannounced inspection (UI), since September 2014. However, the effectiveness of UI in driving drug producers to put more effort into meeting the requirements of Good Manufacturing Practice (GMP) is yet to be ascertained. In this paper we apply game theory to study the effects of UI on the prevention of drug fraud, drug producers’ profits, and social welfare under scenarios of complete and incomplete information. We show that UI is not always an effective way of supervision. Specifically, if the upper bound on the punishment is not very large, UI can drive the firmto make the largest self-supervision effort only when the firm’s technical level is high and the supervision cost is low. Otherwise, UI cannot drive the firm to make the largest self-supervision effort. Under incomplete information, firms with a high technical level would adopt more self-supervision and make more profit than those with a low technical level. Moreover, we design a new mechanism under incomplete information that can incentive drug producers to make greater self-supervision effort to meet the requirements of GMP under certain conditions.Manman Zhang Juliang Zhang T.C.E. 7Detection of Ratoon Stunting Disease in Virus-free Seedcane via Real-time Fluorescence Quantitative PCR显示文摘This study was to develop the real-time fluorescence quantitative PCR technique for detecting the ratoon stunting disease (RSD) in virus-free seedcane seedlings. Healthy tissue culture seedlings were obtained from six plants of sugarcane ROC22, which had been confirmed RSD-positive by detecting the sugarcane juice, by employing the sugarcane seedlings production protocol. Real-time fluorescence quantitative PCR was used to detect RSD pathogens in tissue culture samples. The results showed that target fragment of RSD pathogens was not found in all 10 samples in real-time fluorescence quantitative PCR, with the Ct values of 37-39. 9In situ fabrication of Na3V2(PO4)3 quantum dots in hard carbon nanosheets by using lignocelluloses for sodium ion batteries显示文摘The rational assembly of quantum dots on two-dimensional(2 D) carbonaceous materials is very promising to produce materials, but remains a challenge. Here, we develop an assembly strategy of growing Na3 V2(PO4)3 quantum dots with superlattice structure(NVP-QDs-SL) for obtaining precise control of the size, distribution and crystallinity. The multifunctional lignocelluloses(LCs) used as a hard carbon source induce heterogeneous nucleation and confined growth of NVP-QDs-SL, leading to the uniform distribution of NVP-QDs-SL in H/S-doped hard carbon ultra-thin nanosheets(HCS). Detailed electrochemical analysis results from sodium-ion batteries of NVP-QDs-SL show that NVP-QDs-SL could trap the electrons inside HCS, significantly enhancing Na ion storage and transfer kinetics. Compared to the common Na3 V2(PO4)3 nanoparticle cathode, the NVP-QDs-SL/HCS cathode exhibits a high reversible capacity of 149.2 m A h g^-1 at a 0.1 C rate, which is far beyond the theoretical capacity of Na3 V2(PO4)3(117.6 m A h g^-1).At the ultrahigh current rate of 100 C, this cathode still remains a high discharge capacity of 40 m A h g-1.Even after cycling at 20 C over 3000 cycles, an ultrahigh coulombic efficiency close to 100% is still obtained,highlighting its excellent long cycling life, remarkable rate performance and energy density.Qihao Zhang Xudong Zhang Wen He Guogang Xu Manman Ren Jinhua Liu Xuena Yang Feng Wang 2019Journal of Materials Science & Technology2019,35,10:1
10The ubiquitin ligase Pelil inhibits ICOS and thereby Tfh-mediated immunity显示文摘T follicular helper(Tfh)cells are crucial for regulating autoimmune inflammation and protective immunity against viral infection.However,the molecular mechanism controlling Tfh cell differentiation is poorly understood.Here,through two mixed bone marrow chimeric experiments,we identified Peli1,a T cell-enriched E3 ubiquitin ligase,as an intrinsic regulator that inhibits Tfh cell differentiation.Peli1 deficiency significantly promoted c-Rel-mediated inducible T-cell costimulator(ICOS)expression,and PELI1 mRNA expression was negatively associated with ICOS expression on human CD4^(+)T cells.Mechanistically,increased ICOS expression on Peli1-KO CD4^(+)T cells enhanced the activation of PI3K-AKT signaling and thus suppressed the expression of Klf2,a transcription factor that inhibits Tfh differentiation.Therefore,reconstitution of Klf2 abolished the differences in Tfh differentiation and germinal center reaction between WT and Peli1-KO cells.As a consequence,Peli1-deficient CD4^(+)T cells promoted lupus-like autoimmunity but protected against H1N1 influenza virus infection in mouse models.Collectively,our findings established Peli1 as a critical negative regulator of Tfh differentiation and indicated that targeting Peli1 may have beneficial therapeutic effects in Tfhrelated autoimmunity or infectious diseases.Xinfang Huang Shumeng Hao Junli Liu Yuanyuan Huang Manman Liu Chunyuan Xiao Yan Wang Siyu Pei Tao Yu Jing Xu Haikun Wang Dongfang Dai Xiao Su Yichuan Xiao 2021Cellular & Molecular Immunology2021,18,4:1

12Experimental study on gas–liquid dispersion and mass transfer in shear-thinning system with coaxial mixer显示文摘The effects of impeller type, stirring power, gas flow rate, and liquid concentration on the gas–liquid mixing in a shear-thinning system with a coaxial mixer were investigated by experiment, and the overall gas holdup, relative power demand, and volumetric mass transfer coefficient under different conditions were compared. The results show that, the increasing stirring power or gas flow rate is beneficial in promoting the overall gas holdup and volumetric mass transfer coefficient, while the increasing system viscosity weakens the mass transfer in a shearing–thinning system. 13Effects of Different Row Ratios of Male and Female Parents and Application Amount of Nitrogen Fertilizer on Hybrid Wheat Seed Production and Its Component Factors显示文摘In order to screen suitable high hybrid wheat seed production technology,the split-plot experiment design was adopted and study was carried out about the effects of the different row ratios of male and female parents and application amount of nitrogen fertilizer on hybrid wheat seed production and its component factors. The results showed that the seed production increased with the increase in the number of female parent row. When the row ratio of male and female parents was 2 ∶ 6,the seed production was 3 683. 8 kg/ha; when the application amount of nitrogen fertilization was 50 kg/ha,the seed production was 3 649. 4 kg/ha; the interaction between the row ratio of male and female parents and the application amount of nitrogen fertilizer indicated that when the row ratio of male and female parents was 2∶ 6 and the application amount of nitrogen fertilizer was 300 kg/ha,the seed production reached the highest( 4160. 6 kg/ha). The row ratio of male and female parents and application amount of nitrogen fertilizer had significant effect on the component factors of seed production,including the number of grains per spike,spike weight and setting percentage. When the row ratio of male and female parents was 2∶ 5,the number of grains per spike,spike weight and setting percentage were the highest at 26. 7 grains,1. 12 g,and 62. 6% respectively; when application amount of nitrogen fertilizer was 450 kg/ha,the number of grains per spike,spike weight and setting percentage were the highest at 26. 0 grains,1. 08 g,and59. 2% respectively; the interaction of row ratio of male and female parents and application amount of nitrogen fertilizer had significant effect on the number of grains per spike,spike weight and setting percentage; when the row ratio of male and female parents was 2∶ 5 and the application amount of nitrogen fertilizer was 300 kg/ha,the number of grains per spike,spike weight and setting percentage were the highest at 29. 6grains,1. 24 g,and 71. 6% respectively. The number of grains per spike is the largest component factor for seed production. Increasing the number of grains per spike can increase the seed production. 14BLM helicase inhibition synergizes with PARP inhibition to improve the radiosensitivity of olaparib resistant non-small cell lung cancer cells by inhibiting homologous recombination repair显示文摘Objective:We aimed to investigate the radiosensitizing efficacy of the poly-ADP-ribose polymerase(PARP)inhibitor,olaparib,and the Bloom syndrome protein(BLM)helicase inhibitor,ML216,in non-small cell lung cancer(NSCLC)cells.Methods:Radiosensitization of NSCLC cells was assessed by colony formation and tumor growth assays.Mechanistically,the effects of ML216,olaparib,and radiation on cell and tumor proliferation,DNA damage,cell cycle,apoptosis,homologous recombination(HR)repair,and non-homologous end joining(NHEJ)repair activity were determined.Results:Both olaparib and ML216 enhanced the radiosensitivities of olaparib-sensitive H460 and H1299 cells,which was seen as decreased surviving fractions and Rad51 foci,increased total DNA damage,andγH2AX and 53BP1 foci(P<0.05).The expressions of HR repair proteins were remarkably decreased in olaparib-treated H460 and H1299 cells after irradiation(P<0.05),while olaparib combined with ML216 exerted a synergistic radiosensitization effect on olaparib-resistant A549 cells.In addition to increases of double strand break(DSB)damage and decreases of Rad51 foci,olaparib combined with ML216 also increased pDNA-PKcs(S2056)foci,abrogated G2 cell cycle arrest,and induced apoptosis in A549 lung cancer after irradiation in vitro and in vivo(P<0.05).Moreover,Western blot showed that olaparib combined with ML216 and irradiation inhibited HR repair,promoted NHEJ repair,and inactivated cell cycle checkpoint signals both in vitro and in vivo(P<0.05).Conclusions:Taken together,these results showed the efficacy of PARP and BLM helicase inhibitors for radiosensitizing NSCLC cells,and supported the model that BLM inhibition sensitizes cells to PARP inhibitor-mediated radiosensitization,as well as providing the basis for the potential clinical development of this combination for tumors intrinsically resistant to PARP inhibitors and radiotherapy.Yangyang Kong Chang Xu Xiaohui Sun Hao Sun Xiaotong Zhao Ningning He Kaihua Ji Qin Wang Liqing Du Jinhan Wang Manman Zhang Yang Liu Yan Wang Qiang Liu 2022Cancer Biology & Medicine2022,19,8:1
15Quorum sensing-based interactions among drugs,microbes,and diseases显示文摘Many diseases and health conditions are closely related to various microbes,which participate in complex interactions with diverse drugs;nonetheless,the detailed targets of such drugs remain to be elucidated.Many existing studies have reported causal associations among drugs,gut microbes,or diseases,calling for a workflow to reveal their intricate interactions.In this study,we developed a systematic workflow comprising three modules to construct a Quorum Sensing-based Drug-Microbe-Disease(QSDMD)database(http://gffzz083eea51bee54f8dh9wfk0q6unwqo66fq.ffgz.tsg.suse.edu.cn/),which includes diverse interactions for more than 8,000 drugs,163 microbes,and 42 common diseases.Potential interactions between microbes and more than 8,000 drugs have been systematically studied by targeting microbial QS receptors combined with a docking-based virtual screening technique and in vitro experimental validations.Furthermore,we have constructed a QS-based drug-receptor interaction network,proposed a systematic framework including various drug-receptor-microbe-disease connections,and mapped a paradigmatic circular interaction network based on the QS-DMD,which can provide the underlying QS-based mechanisms for the reported causal associations.The QS-DMD will promote an understanding of personalized medicine and the development of potential therapies for diverse diseases.This work contributes to a paradigm for the construction of a molecule-receptor-microbe-disease interaction network for human health that may form one of the key knowledge maps of precision medicine in the future.Shengbo Wu Shujuan Yang Manman Wang Nan Song Jie Feng Hao Wu Aidong Yang Chunjiang Liu Yanni Li Fei Guo Jianjun Qiao 2023Science China(Life Sciences)2023,66,1:0

19Ultrahigh pyroelectric effect and energy harvesting density of Pb(Lu_(1/2)Nb_(1/2))O_(3)-PbTiO_(3) crystals induced by FE-AFE phase transition显示文摘Large pyroelectric and energy harvesting properties have been attracting increasing attentions due to the practical applications in infrared detectors and energy harvesting technologies.Ferroelectricantiferroelectric(FE-AFE)phase transitions are usually accompanied by a sharp drop in polarization,which will lead to excellent pyroelectric properties and energy harvesting density.Therefore,FE-AFE phase boundary design is an effective strategy to develop new pyroelectric materials.In this paper,Pb(Lu_(1/2)Nb_(1/2))O_(3)-PbTiO_(3) (PLN-PT)single crystals with FE-AFE phase transitions were obtained by molten salt growth method.The temperature-induced FE-AFE phase transition was verified by temperaturedependent macrodomain structure,DSC curves and dielectric properties.Obviously,PLN-PT crystals display excellent peak pyroelectric coefficient(~6.8μC/(cm^(2)·K)with a maximum depolarization temperature of 118℃.Meanwhile,the pyroelectric energy harvesting density is as high as 2.62 J/cm3,which is much higher than other pyroelectric materials.The results reveal that the PLN-PT crystal is a promising candidate for infrared detectors and energy harvesting devices.Lingfei Lv Manman Liu Ying Liu Zujian Wang Rongbing Su Xiaoming Yang Chao He Xifa Long 2021Journal of Rare Earths2021,39,12:0
20Probiotics and Their Metabolites Ameliorate Inflammatory Bowel Disease:A Critical Review显示文摘Crohn disease and ulcerative colitis are the two main manifestations of inflammatory bowel disease(IBD),and both are highly morbid.However,the precise etiology of IBD is still unknown and effective therapeutics are yet to be discovered.It is becoming increasingly clear that a combination of factors,including genetic background,host immune response,and microbial reduced diversity status are related to IBD.The cardinal symptom of IBD patients is the imbalance of the intestinal microflora.According to previous studies,both probiotics and symbiotic microbiota can play a protective role through intestinal micro-ecosystem regulation,epithelial barrier integrity enhancement,and inflammation reduction.Therefore,probiotics can provide an alternative or auxiliary method to traditional IBD treatment.Here,we reviewed the possible pathogenesis of IBD,summarized the possible mechanisms of probiotics modulation of IBD,and emphasized the prevention and treatment targets of probiotics-mediated IBD,with the aim to provide theoretical support for the treatment of IBD patients by probiotics in clinical trials.Ying Liang Manman Liu Jiaqian Pu Zichun Zhu Zining Gao Qingqing Zhou Qing Gu Ping Li 2021Infectious Microbes & Diseases2021,3,1:0
