| 1 | Climate change impact and adaptation on wheat yield, water use and water use efficiency at North Nile Delta显示文摘Investigation of climate change impacts on food security has become a global hot spot.Even so,efforts to mitigate these issues in arid regions have been insufficient.Thus,in this paper,further research is discussed based on data obtained from various crop and climate models.Two DSSATcrop models(CMs)(CERESWheat and N-Wheat)were calibrated with two wheat cultivars(Gemiza9 and Misr1).A baseline simulation(1981-2010)was compared with different scenarios of simulations using three Global Climate Models(GCMs)for the 2030s,2050s and 2080s.Probable impacts of climate change were assessed using the GCMs and CMs under the high emission Representative Concentration Pathway(RCP8.5).Results predicted decreased wheat grain yields by a mean of 8.7%,11.4%and 13.2%in the 2030s,2050s and 2080s,respectively,relative to the baseline yield.Negative impacts of climatic change are probable,despite some uncertainties within the GCMs(i.e.,2.1%,5.0%and 8.0%)and CMs(i.e.,2.2%,6.0%and 9.2%).Changing the planting date with a scenario of plus or minus 5 or 10 days from the common practice was assessed as a potentially effective adaptation option,which may partially offset the negative impacts of climate change.Delaying the sowing date by 10 days(from 20 November to 30 November)proved the optimum scenario and decreased further reduction in wheat yields resulting from climate change to 5.2%,6.8%and 8.5%in the 2030s,2050s and 2080s,respectively,compared with the 20 November scenario.The planting 5-days earlier scenario showed a decreased impact on climate change adaptation.However,the 10-days early planting scenario increased yield reduction under projected climate change.The cultivar Misr1 was more resistant to rising temperature than Gemiza9.Despite the negative impacts of projected climate change on wheat production,water use efficiency would slightly increase.The ensemble of multi-model estimated impacts and adaptation uncertainties of climate change can assist decision-makers in planning climate adaptation strategies. | Marwa Gamal Mohamed Ali Mahmoud Mohamed Ibrahim Ahmed El Baroudy Michael Fullen El-Said Hamad Omar Zheli Ding Ahmed Mohammed Saad Kheir | 2020 | Frontiers of Earth Science2020，14，3： | 2 |

How end-user antibody validation facilitates insights into biology and disease显示文摘Antibodies are employed ubiquitously in biomedical sciences,including for diagnostics and therapeutics.One of the most important uses is for immunohistochemical(IHC)staining,a process that has been improving and evolving over decades.IHC is useful when properly employed,yet misuse of the method is widespread and contributes to the“reproducibility crisis”in science.We report some of the common problems encountered with IHC assays,and direct readers to a wealth of literature documenting and providing some solutions to this problem.We also describe a series of vignettes that include our approach to analytical validation of antibodies and IHC assays that have facilitated a number of biological insights into prostate cancer and the refutation of a controversial association of a viral etiology in gliomas.We postulate that a great deal of the problem with lack of accuracy in IHC assays stems from the lack of awareness by researchers for the critical necessity for end-users to validate IHC antibodies and assays in their laboratories,regardless of manufacturer claims or past publications.We suggest that one reason for the pervasive lack of end-user validation for research antibodies is that researchers fail to realize that there are two general classes of antibodies employed in IHC.First,there are antibodies that are“clinical grade”reagents used by pathologists to help render diagnoses that influence patient treatment.Such diagnostic antibodies,which tend to be highly validated prior to clinical implementation,are in the vast minority(e.g.<500).The other main class of antibodies are“research grade”antibodies(now numbering>3800000),which are often not extensively validated prior to commercialization.Given increased awareness of the problem,both the United States,National Institutes of Health and some journals are requiring investigators to provide evidence of specificity of their antibody-based assays. | Karen S.Sfanos Srinivasan Yegnasubramanian William G.Nelson Tamara L.Lotan Ibrahim Kulac Jessica L.Hicks Qizhi Zheng Charles J.Bieberich Michael C.Haffner Angelo M.De Marzo | 2019 | Asian Journal of Urology2019，6，1： | 2 |
