1Multilayer black phosphorus as saturable absorber for an Er:Lu_(2)O_(3) laser at~3 μm显示文摘Multilayer black phosphorus(BP) nanoplatelets of different thicknesses were prepared by the liquid phase exfoliation method and deposited onto yttrium aluminum garnet substrates to form saturable absorbers(SAs). These were characterized with respect to their thickness-dependent saturable absorption properties at 3 μm. The BP-SAs were employed in a passively Q-switched Er:Lu_2O_3 laser at 2.84 μm. By using BP exfoliated in different solvents,stable pulses as short as 359 ns were generated at an average output power of up to 755 m W. The repetition rate in the experiment was 107 k Hz, corresponding to a pulse energy of 7.1 μJ. 2Photocrosslinking silver nanoparticles–aloe vera–silk fibroin composite hydrogel for treatment of full-thickness cutaneous wounds显示文摘Damage to the skin causes physiological and functional issues.The most effective treatment approach is the use of wound dressings.Silk fibroin(SF)is a promising candidate biomaterial for regulating wound healing;however,its antibacterial properties and biological activity must be further improved.In this study,a photocrosslinking hydrogel was developed to treat full-thickness cutaneous wounds.The composite hydrogel(Ag-AV-SF hydrogel)was prepared by introducing the silver nanoparticles(AgNPs)and aloe vera(AV)as the modifiers.In vitro study exhibited great antibacterial ability,biocompatibility and cell-proliferation and-migration-promoting capacities.It also showed the pH-response releasing properties which release more AgNPs in a simulated chronic infection environment.The healing effect evaluation in vivo showed the healing-promoting ability of the Ag-AV-SF hydrogel was stronger than the single-modifiers groups,and the healing rate of it reached 97.02%on Day 21,higher than the commercial wound dressing,silver sulfadiazine(SS)cream on sale.Additionally,the histological and protein expression results showed that the Ag-AV-SF hydrogel has a greater effect on the pro-healing regenerative phenotype with M2 macrophages at the early stage,reconstructing the blood vessels networks and inhibiting the formation of scars.In summary,the Ag-AV-SF hydrogel developed in this study had good physical properties,overwhelming antibacterial properties,satisfactory biocompatibility and significantly promoting effect on cell proliferation,migration and wound healing.Overall,our results suggest that the Ag-AV-SF hydrogel we developed has great potential for improving the wound healing in clinical treatment.Yangkun Liu JinChuan Fan MingQi Lv Kepeng She Jiale Sun Qingqing Lu Changhao Han SongTao Ding Shuang Zhao GuiXue Wang YuChan Zhang GuangChao Zang 2021Regenerative Biomaterials2021,8,6:2

4Spectrally tunable high-power Yb:fiber chirped-pulse amplifier显示文摘Tailoring the properties of the driving laser to the need of applications often requires compromises among laser stability,high peak and average power levels,pulse duration,and spectral bandwidth.For instance,spectroscopy with optical frequency combs in the extreme/visible ultraviolet spectral region requires a high peak power of the near-IR driving laser,and therefore high average power,pulse duration of a few tens of fs,and maximal available spectral bandwidth.Contrarily,the parametric conversion efficiency is higher for pulses with a duration in the 100-fs range due to temporal walk-off and coating limitations.Here we suggest an approach to adjust the spectral characteristics of high-power chirped-pulse amplification(CPA)to the requirements of different nonlinear frequency converters while preserving the low-phase-noise(PN)properties of the system.To achieve spectral tunability,we installed a mechanical spectral shaper in a free-space section of the stretcher of an in-house-developed ytterbium-fiber-based CPA system.The CPA system delivers 100 W of average power at a repetition rate of 132.4 MHz.While gaining control over the spectral properties,we preserve the relative-intensity-noise and PN properties of the system.The high-power CPA can easily be adjusted to deliver either a spectrum ideal for mid-IR light generation(full width at half maximum of∼11 nm,compressed pulse duration of 230 fs)or a spectrum ideal for highly nonlinear processes such as high-harmonic generation(−10 dB level of>50 nm,transform-limited pulse duration of∼65 fs).VALENTINA SHUMAKOVA VITO F.PECILE JAKOB FELLINGER MICHAEL LESKOWSCHEK P.E.COLLIN ALDIA ALINE S.MAYER LUKAS W.PERNER SARPER SALMAN MINGQI FAN PRANNAY BALLA STÉPHANE SCHILT CHRISTOPH M.HEYL INGMAR HARTL GIL PORAT OLIVER H.HECKL 2022Photonics Research2022,10,10:0
5Engineered poly(A)-surrogates for translational regulation and therapeutic biocomputation in mammalian cells显示文摘Here,we present a gene regulation strategy enabling programmable control over eukaryotic translational initiation.By excising the natural poly-adenylation(poly-A)signal of target genes and replacing it with a synthetic control region harboring RNA-binding protein(RBP)-specific aptamers,cap-dependent translation is rendered exclusively dependent on synthetic translation initiation factors(STIFs)containing different RBPs engineered to conditionally associate with different eIF4F-binding proteins(eIFBPs).This modular design framework facilitates the engineering of various gene switches and intracellular sensors responding to many user-defined trigger signals of interest,demonstrating tightly controlled,rapid and reversible regulation of transgene expression in mammalian cells as well as compatibility with various clinically applicable delivery routes of in vivo gene therapy.Therapeutic efficacy was demonstrated in two animal models.To exemplify disease treatments that require on-demand drug secretion,we show that a custom-designed gene switch triggered by the FDA-approved drug grazoprevir can effectively control insulin expression and restore glucose homeostasis in diabetic mice.For diseases that require instantaneous sense-and-response treatment programs,we create highly specific sensors for various subcellularly(mis)localized protein markers(such as cancer-related fusion proteins)and show that translation-based protein sensors can be used either alone or in combination with other cell-state classification strategies to create therapeutic biocomputers driving self-sufficient elimination of tumor cells in mice.This design strategy demonstrates unprecedented flexibility for translational regulation and could form the basis for a novel class of programmable gene therapies in vivo.Jiawei Shao Shichao Li Xinyuan Qiu Jian Jiang Lihang Zhang Pengli Wang Yaqing Si Yuhang Wu Minghui He Qiqi Xiong Liuqi Zhao Yilin Li Yuxuan Fan Mirta Viviani Yu Fu Chaohua Wu Ting Gao Lingyun Zhu Martin Fussenegger Hui Wang Mingqi Xie 2024Cell Research2024,34,1:0
