| 1 | PD-1 blockade-unresponsive human tumor-infiltrating CD8^(+)T cells are marked by loss of CD28 expression and rescued by IL-15显示文摘Blockade of programmed death-1(PD-1)reinvigorates exhausted CD8^(+)T cells,resulting in tumor regression in cancer patients.Recently,reinvigoration of exhausted CD8^(+)T cells following PD-1 blockade was shown to be CD28-dependent in mouse models.Herein,we examined the role of CD28 in anti-PD-1 antibody-induced human T cell reinvigoration using tumor-infiltrating CD8^(+)T cells(CD8^(+)TILs)obtained from non-small-cell lung cancer patients.Single-cell analysis demonstrated a distinct expression pattern of CD28 between mouse and human CD8^(+)TILs.Furthermore,we found that human CD28^(+)CD8^(+)but not CD28^(–)CD8^(+)TILs responded to PD-1 blockade irrespective of B7/CD28 blockade,indicating that CD28 costimulation in human CD8^(+)TILs is dispensable for PD-1 blockade-induced reinvigoration and that loss of CD28 expression serves as a marker of anti-PD-1 antibody-unresponsive CD8^(+)TILs.Transcriptionally and phenotypically,PD-1 blockade-unresponsive human CD28^(–)PD-1^(+)CD8^(+)TILs exhibited characteristics of terminally exhausted CD8^(+)T cells with low TCF1 expression.Notably,CD28^(–)PD-1^(+)CD8^(+)TILs had preserved machinery to respond to IL-15,and IL-15 treatment enhanced the proliferation of CD28^(–)PD-1^(+)CD8^(+)TILs as well as CD28^(+)PD-1^(+)CD8^(+)TILs.Taken together,these results show that loss of CD28 expression is a marker of PD-1 blockade-unresponsive human CD8^(+)TILs with a TCF1–signature and provide mechanistic insights into combining IL-15 with anti-PD-1 antibodies. | Kyung Hwan Kim Hong Kwan Kim Hyung-Don Kim Chang Gon Kim Hoyoung Lee Ji Won Han Seong Jin Choi Seongju Jeong Minwoo Jeon Hyunglae Kim Jiae Koh Bo Mi Ku Su-Hyung Park Myung-Ju Ahn Eui-Cheol Shin | 2021 | Cellular & Molecular Immunology2021,18,2: | 6 |
| 2 | Analysis of direct operating cost of wide-body passenger aircraft: A parametric study based on Hong Kong显示文摘Analysis of the Direct Operating Cost(DOC) of aircraft is an important step towards achieving financially sustainable aviation operations. However, the value of the DOC for different aircraft types and flight scenarios is not widely available. In this study, we perform a systematic analysis of the DOC of every wide-body passenger aircraft currently in production, using the method of the Association of European Airlines(AEA). The elements of the DOC, e.g. financial costs, maintenance costs, and flight costs, are evaluated individually. Several realistic flight scenarios are considered, each with differences in route distance, fuel price, passenger number, and seating arrangement. For each flight scenario, the most cost-efficient aircraft type is identified and evaluated in the context of operations from Hong Kong International Airport. | 4 | Review on the characteristics of liver-pacifying medicinal in relation to the treatment of stroke: from scientific evidence to traditional medical theory显示文摘OBJECTIVE: To analyze the characteristics of liver pacifying medicinal in the treatment of brain disease to provide scientific evidence in clinical usage on stroke.METHODS: MEDLINE/Pub Med, Google Scholar,and China National Knowledge Infrastructure Database were used as the literature sources. The Scientific name, Latin pharmaceutical name,Chinese name of 7 kinds of liver pacifying medicinal including Gouteng(Ramulus Uncariae Rhynchophyllae cum Uncis), Tianma(Rhizoma Gastrodiae), Juemingzi(Semen Cassiae Obtusifoliae),Quanxie(Scorpio), Wugong(Scolopendra), Jiangcan(Bombyx Batryticatus), and Dilong(Pheretima Aspergillum) were used as the keywords to search the databases for relevant publications up to July2016. Their major compounds were also used as the keywords. The papers were selected based on the pharmacological activities and mechanisms of action related to brain diseases and subsequently,were analyzed and reviewed. We first described the origin, efficacy, and clinical indications of selected medicines, then brain disease specific activities focusing on stroke after the description of the general pharmacological activities.RESULTS: On the basis of the literature of scientific studies and clinical use in traditional medicine, we found and discussed the characteristics of liver pacifying medicinal in stroke treatment. First, liver-pacifying medicinal,or their components, might pass through the blood-brain barrier and act directly on neurons or on the neural network to provide protective effects against brain disease. | 12 | Design of 2D massless Dirac fermion systems and quantum spin Hall insulators based on sp-sp^(2) carbon sheets显示文摘Grapheneis a massless Dirac fermion system,featuring Dirac points in momentum space.It was also first identified as a quantum spin Hall(QSH)insulator when considering spin–orbit coupling(SOC),which opens a band gap at the Dirac points.This discovery has initiated new research efforts to study the QSH effect,towards its application for quantum computing and spintronics.Although the QSH effect has been observed in HgTe quantum wells,the SOC strength of graphene is too small(~1μeV)to induce the topological insulator phase in an experimentally achievable temperature regime.Here,we perform a systematic atomistic simulation to design two-dimensional sp–sp^(2) hybrid carbon sheets to discover new Dirac systems,hosting the QSH phase.21 out of 31 newly discovered carbon sheets are identified as Dirac fermion systems without SOC,distinct from graphene in the number,shape,and position of the Dirac cones occurring in the Brillouin zone.Moreover,we find 19 out of the 21 new Dirac fermion systems become QSH insulators with a sizable SOC gap enhanced up to an order of meV,thus allowing for the QSH effect at experimentally accessible temperatures.In addition,based on the 26 Dirac fermion systems,we make a connection between the number of Dirac points without SOC and the resultant QSH phase in the presence of SOC.Our findings present new prospects for the design of topological materials with desired properties. | Minwoo Park Youngkuk Kim Hoonkyung Lee | 2018 | npj Computational Materials2018,,1: | 1 |
