1In vitro Ruminal Gas Production Kinetics of Four Fodder Trees Ensiled With or Without Molasses and Urea显示文摘This study investigated if the addition of urea (U), molasses (M) or their 1:1 (v/v) mixture during ensiling increases the nutritional value of forage from four fodder trees (Prunus persica, Leucaena esculenta, Acacia farnesiana, and Prunus domestica). Forage samples of fodder trees were collected in triplicate (three individual samples of each species) and subjected to an in vitro gas production (GP) procedure. Fermentation at 24 h (GP 24), short-chain volatile fatty acids (SCFA), and microbial crude protein production (MCP), in vitro organic matter digestibility (OMD), metabolizable energy (ME) and dry matter degradability (DMD) were estimated. Forage samples were incubated for 72 h in an incubator at 39oC and the volume of GP was recorded at 2, 4, 6, 8, 10, 12, 24, 48, and 72 h of incubation using the reading pressure technique. The rumen fermentation profiles were highest for P. persica, which showed the highest (P<0.0001) DMD, ME, OMD, SCFA, GP 24 and MCP. On the other hand L. esculenta had the lowest (P<0.0001) DMD, SCFA, MCP; P. domestica had the lowest (P<0.0001) OMD. The addition of M to silage increased (P<0.0001) ME and OMD, as well as GP. However, the addition of U and the mixture of U and M reduced (P<0.0001) DMD, ME, OMD, SCFA, GY 24 and MCP. These results show that P. persica has the highest nutritive value and L. esculenta the lowest for ruminants. 2Liver resection versus liver transplantation for hepatocellular carcinoma within Milan criteria:a meta-analysis of 18,421 patients显示文摘Background:Outcomes after liver resection(LR)and liver transplantation(LT)for hepatocellular carcinoma(HCC)are heterogenous and may vary by region,over time periods and disease burden.We aimed to compare overall survival(OS)and disease-free survival(DFS)between LT versus LR for HCC within the Milan criteria.Methods:Two authors independently searched Medline and Embase databases for studies comparing survival after LT and LR for patients with HCC meeting the Milan criteria.Meta-analyses and meta-regression were conducted using random-effects models.Results:We screened 2,278 studies and included 35 studies with 18,421 patients.LR was associated with poorer OS[hazard ratio(HR)=1.44;95%confidence interval(CI):1.14-1.81;P<0.01]and DFS(HR=2.71;95%CI:2.23-3.28;P<0.01)compared to LT,with similar findings among intention-to-treat(ITT)studies.In uninodular disease,OS in LR was comparable to LT(P=0.13)but DFS remained poorer(HR=2.95;95%CI:2.30-3.79;P<0.01).By region,LR had poorer OS versus LT in North America and Europe(P≤0.01),but not Asia(P=0.25).LR had inferior survival versus LT in studies completed before 2010(P=0.01),but not after 2010(P=0.12).Cohorts that underwent enhanced surveillance had comparable OS after LT and LR(P=0.33),but cohorts undergoing usual surveillance had worse OS after LR(HR=1.95;95%CI:1.24-3.07;P<0.01).Conclusions:Mortality after LR for HCC is nearly 50%higher compared to LT.Survival between LR and LT were similar in uninodular disease.The risk of recurrence after LR is threefold that of LT.Jin Hean Koh Darren Jun Hao Tan Yuki Ong Wen Hui Lim Cheng Han Ng Phoebe Wen Lin Tay Jie Ning Yong Mark D.Muthiah Eunice X.Tan Ning Qi Pang Beom Kyung Kim Nicholas Syn Alfred Kow Brian K.P.Goh Daniel Q.Huang 2022Hepatobiliary Surgery and Nutrition2022,11,1:3

6Role of regenerating islet-derived proteins in inflammatory bowel disease显示文摘Inflammatory bowel disease(IBD)is an inflammatory disorder of the gastrointestinal tract that affects millions of patients worldwide.It has a complex and multifactorial etiology leading to excessive exposure of intestinal epithelium to microbial antigens,inappropriate activation of the immune system and ultimately to the damage of intestinal tissues.Although numerous efforts have been made to improve the disease management,IBD remains persistently recurring and beyond cure.This is due largely to the gaps in our understanding of the pathogenesis of IBD that hamper the development of timely diagnoses and effective treatment.However,some recent discoveries,including the beneficial effects of interleukin-22(IL-22)on the inflamed intestine,have shed light on a self-protective mechanism in IBD.Regenerating islet-derived(REG/Reg)proteins are small secretory proteins which function as IL-22’s downstream effectors.Mounting studies have demonstrated that IBD patients have significantly increased REG expressions in the injured intestine,but with undefined mechanisms and roles.The reported functions of REG/Reg proteins in intestinal homeostasis,such as those of antibacterial,anti-inflammatory and tissue repair,lead us to discuss their potential mechanisms and clinical relevance in IBD in order to advance IBD research and management.Jodi-Ann Edwards Nicholas Tan Nadlie Toussaint Peiqi Ou Cathy Mueller Albert Stanek Vladimir Zinsou Sean Roudnitsky Michelle Sagal Lisa Dresner Alexander Schwartzman Chongmin Huan 2020World Journal of Gastroenterology2020,26,21:1

9Geospatial Analytics for COVID-19 Active Case Detection显示文摘Ever since the COVID-19 pandemic started in Wuhan,China,much research work has been focusing on the clinical aspect of SARS-CoV-2.Researchers have been leveraging on various Artificial Intelligence techniques as an alternative to medical approach in understanding the virus.Limited studies have,however,reported on COVID-19 transmission pattern analysis,and using geography features for prediction of potential outbreak sites.Predicting the next most probable outbreak site is crucial,particularly for optimizing the planning of medical personnel and supply resources.To tackle the challenge,this work proposed distance-based similarity measures to predict the next most probable outbreak site together with its magnitude,when would the outbreak likely to happen and the duration of the outbreak.The work began with preprocessing of 1365 patient records from six districts in the most populated state named Selangor in Malaysia.The dataset was then aggregated with population density information and human elicited geography features that might promote the transmission of COVID-19.Empirical findings indicated that the proposed unified decision-making approach outperformed individual distance metric in predicting the total cases,next outbreak location,and the time interval between start dates of two similar sites.Such findings provided valuable insights for policymakers to perform Active Case Detection.Choo-Yee Ting Helmi Zakariah Fadzilah Kamaludin Darryl Lin-Wei Cheng Nicholas Yu-Zhe Tan Hui-Jia Yee 2021Computers, Materials & Continua2021,,4:1

13The impact of urological resection and reconstruction on patients undergoing cytoreductive surgery (CRS) and hyperthermic intraperitoneal chemotherapy (HIPEC)显示文摘Objective:Cytoreductive surgery(CRS)and hyperthermic intraperitoneal chemotherapy(HIPEC)are increasingly being used to treat peritoneal malignancies.Urological resections and reconstruction(URR)are occasionally performed during the surgery.We aim to evaluate the impact of these procedures on peri-operative outcomes of CRS and HIPEC patients.Methods:A retrospective review of a prospectively maintained database of all patients who underwent CRS-HIPEC from April 2001 to February 2016 was performed.Outcomes between patients who had surgery involving,and not involving URR were compared.Primary outcomes were the rate of major complications and the duration of stay in the intensive care unit(ICU)and hospital.Secondary outcomes were that of overall survival(OS)and prognostic factors that would indicate a need for URR.Results:A total of 214 CRS-HIPEC were performed,21 of which involved a URR.Baseline clinical characteristics did not vary between the groups(URR vs.No URR).Urological resections comprised of 52%bladder resections,24%ureteric resections,and 24%involving both bladder and ureteric resections.All bladder defects were closed primarily while ureteric reconstructions consisted of two end-to-end anastomoses,one ureto-uretostomy,five direct implantations into the bladder and three boari flaps.URR were more frequently required in patients with colorectal peritoneal disease(p Z 0.029),but was not associated with previous pelvic surgery(76%vs.54%,p Z 0.065).Patients with URR did not suffer more serious complications(14%vs.24%,p Z 0.42).ICU(2.2 days vs.1.4 days,p Z 0.51)and hospital stays(18 days vs.25 days,p Z 0.094)were not significantly affected.Undergoing a URR did not affect OS(p Z 0.99),but was associated with increased operation time(570 min vs.490 min,p Z 0.046).Conclusion:While concomitant URR were associated with an increase in operation time,there were no significant differences in postoperative complications or OS.Patients with colorectal peritoneal metastases are more likely to require a URR compared to other primary tumours,and needs to be considered during pre-operative planning.Grace Hwei Ching Tan Nicholas B.Shannon Claramae Shulyn Chia Lui Shiong Lee Khee Chee Soo Melissa Ching Ching Teo 2018Asian Journal of Urology2018,5,3:1

15通过分布式人工智能实现CT结肠成像的计算机辅助慢性息肉辨识显示文摘目的通过计算机辅助诊断技术(CAD),利用分布式人工智能进行CT结肠成像的病理诊断。材料与方法本研究经人体实验保护局(OfficeofHumanSubjectsResearch)批准。研究内容中涉及健康保险流通与责任法案(HIPAA)的患者知情同意要求并未报批。CT数据采集于24名患有至少一块6mm息肉的病人,相关图样的分析通过计算机辅助诊断(CAD)软件完成,共鉴别268处疑似病灶。每处疑似病灶均由20名来自同一众包平台的知识工作者(KnowledgeWorkers,KWs)判断是否是息肉。为了检验本方法的可重复性,共228名知识工作者参与了两轮病灶鉴别工作。本文中,通过比较知识工作者和CAD软件分析结果的受试者工作特征曲线覆盖面积(AUC)来评价本方法的效果。结果知识工作者的检测水平AUC结果为:第1轮0.845±0.045(标准差),第2轮0.855±0.044。计算机辅助诊断软件(CAD)分析结果的AUC为0.859±0.043,两种方法之间并没有显著区别。根据病灶判断难度分析,容易的诊断中知识工作者表现更佳:第1轮AUC为0.951±0.032,第2轮0.966±0.027(P=0.039),CAD的AUC为0.877±0.048。对于两轮检验都参加的知识工作者而言,第2轮的诊断结果有显著的提升:第1轮AUC为0.759±0.052,第2轮为0.839±0.046(P=0.041)。结论在慢性息肉的诊断方面,分布式人工智能和计算机辅助诊断(CAD)技术的结果之间并无显著差异。Tan B. 16A Class Effect Network Meta-analysis of Lipid Modulation in Non-alcoholic Steatohepatitis for Dyslipidemia显示文摘Background and Aims:Pharmaceutical therapy for NASH is associated with lipid modulation,but the consensus on drug treatment is limited and lacks comparative analysis of effectiveness.A network meta-analysis was conducted to compare NASH drug classes in lipid modulation.Methods:Online databases were searched for randomized controlled trails(RCTs)evaluating NASH treatments in biopsy-proven NASH patients.Treatments were classified into four groups:(1)inflammation,(2)energy,(3)bile acids,and(4)fibro-sis based on the mechanism of action.A Bayesian network analysis was conducted with outcome measured by mean difference(MD)with credible intervals(Crl)and surface un-der the cumulative ranking curve(SUCRA).Results:Forty-four RCTs were included in the analysis.Bile acid modulat-ing treatments(MD:0.05,Crl:0.03-0.07)were the best treatment for improvement in high-density lipid(HDL)cho-lesterol,followed by treatments modulating energy(MD:0.03,Crl:0.02-0.04)and fibrosis(MD:0.01,Crl:−0.12 to 0.14)compared with placebo.The top three treatments for reduction in triglycerides were treatments modulating energy(MD:−0.46,Crl:−0.49 to−0.43),bile acids(MD:−0.22,Crl:−0.35 to−0.09),and inflammation(MD:−0.08,Crl:−0.13 to−0.03)compared with placebo.SUCRA found treatment modulating fibrosis(MD:−1.27,Crl:−1.76 to−0.79)was the best treatment for reduction in low-density lipid(LDL)cholesterol followed by treatment modulating in-flammation(MD:−1.03,Crl:−1.09 to−0.97)and energy(MD:−0.37,Crl:−0.39 to−0.34)compared with placebo,but LDL cholesterol was worsened by treatments modulat-ing bile acids.Conclusions:Network analysis comparing the class effects of dyslipidemia modulation in NASH found that treatment targets can include optimization of athero-genic dyslipidemia.Future studies are required to evaluate the cardiovascular outcomes.Jieling Xiao Cheng-Han Ng Yip-Han Chin Darren Jun Hao Tan Wen-Hui Lim Grace Lim Jingxuan Quek Ansel Shao Pin Tang Kai-En Chan Rou-Yi Soong Nicholas Chew Benjamin Tay Daniel Q.Huang Nobuharu Tamaki Roger Foo Mark Y.Chan Mazen Noureddin Mohammad Shadab Siddiqui Arun J.Sanyaland Mark D.Muthiah 2022Journal of Clinical and Translational Hepatology2022,10,6:0
17Assessing the mechanism of citywide test-trace-isolate Zero-COVID policy and exit strategy of COVID-19 pandemic显示文摘Background:Countries that aimed for eliminating the cases of COVID-19 with test-trace-isolate policy are found to have lower infections,deaths,and better economic performance,compared with those that opted for other mitigation strategies.However,the continuous evolution of new strains has raised the question of whether COVID-19 eradication is still possible given the limited public health response capacity and fatigue of the epidemic.We aim to investigate the mechanism of the Zero-COVID policy on outbreak containment,and to explore the possibility of eradication of Omicron transmission using the citywide test-trace-isolate(CTTI)strategy.Methods:We develop a compartmental model incorporating the CTTI Zero-COVID policy to understand how it contributes to the SARS-CoV-2 elimination.We employ our model to mimic the Delta outbreak in Fujian Province,China,from September 10 to October 9,2021,and the Omicron outbreak in Jilin Province,China for the period from March 1 to April 1,2022.Projections and sensitivity analyses were conducted using dynamical system and Latin Hypercube Sampling/Partial Rank Correlation Coefcient(PRCC).Results:Calibration results of the model estimate the Fujian Delta outbreak can end in 30(95%confdence interval CI:28–33)days,after 10(95%CI:9–11)rounds of citywide testing.The emerging Jilin Omicron outbreak may achieve zero COVID cases in 50(95%CI:41–57)days if supported with sufcient public health resources and population compliance,which shows the efectiveness of the CTTI Zero-COVID policy.Conclusions:The CTTI policy shows the capacity for the eradication of the Delta outbreaks and also the Omicron outbreaks.Nonetheless,the implementation of radical CTTI is challenging,which requires routine monitoring for early detection,adequate testing capacity,efcient contact tracing,and high isolation compliance,which constrain its benefts in regions with limited resources.Moreover,these challenges become even more acute in the face of more contagious variants with a high proportion of asymptomatic cases.Hence,in regions where CTTI is not possible,personal protection,public health control measures,and vaccination are indispensable for mitigating and exiting the COVID-19 pandemic.Pei Yuan Yi Tan Liu Yang Elena Arufo Nicholas H.Ogden Guojing Yang Haixia Lu Zhigui Lin Weichuan Lin Wenjun Ma Meng Fan Kaifa Wang Jianhe Shen Tianmu Chen Huaiping Zhu 2022Infectious Diseases of Poverty2022,11,5:0

19Ultra-precision Machining of Micro-step Pillar Array Using a Straight-Edge Milling Tool显示文摘Ultra-precision machining has been well known as a very precise and effective way to prototype and fabricate different types of components with microstructures.In this paper,an ultra-precision micro-milling method has been presented to produce a micro-step pillar array as an embossing mold for semiconductor application.Theoretical analysis on machining mechanics indicates that work material property,machining strategy,and cutting tool geometry largely afFect machining distortion on the machined microstructure.Experimental investigation has been conducted on micromachining of the micro-step pillar array using different types of cutting tools and work materials.Experimental results demonstrate that the material property,cutting tool edge radius,and cutting force play a significant role in the machined micro-structure accuracy and surface finish,while grain boundary of brass has no significant effect on the micro-milling performance.The best outcome among the tests done is achieved when using brass as work material and cutting with a straight-edge single crystalline diamond tool.The main reasons to achieve this outcome are based on:the brass material having a higher elastic modulus and the diamond tool having a smaller cutting edge radius,which contributes to better machinability of brass.One micro-step pillar array has been successfully obtained with very precise feature and dimensional accuracy using brass work material and single-crystal diamond tool.It is believed that the outcomes of this study would largely benefit the research community and end-users.Kui Liu Hu Wu Rui Huang Nicholas Yew Jin Tan 2020Nanomanufacturing and Metrology2020,3,4:0
20Tumor heterogeneity in VHL drives metastasis in clear cell renal cell carcinoma显示文摘Loss of function of the von Hippel-Lindau(VHL)tumor suppressor gene is a hallmark of clear cell renal cell carcinoma(ccRCC).The importance of heterogeneity in the loss of this tumor suppressor has been under reported.To study the impact of intratumoral VHL heterogeneity observed in human ccRCC,we engineered VHL gene deletion in four RCC models,including a new primary tumor cell line derived from an aggressive metastatic case.The VHL gene-deleted(VHL-KO)cells underwent epithelial-to-mesenchymal transition(EMT)and exhibited increased motility but diminished proliferation and tumorigenicity compared to the parental VHL-expressing(VHL+)cells.Renal tumors with either VHL+or VHL-KO cells alone exhibit minimal metastatic potential.Combined tumors displayed rampant lung metastases,highlighting a novel cooperative metastatic mechanism.The poorly proliferative VHL-KO cells stimulated the proliferation,EMT,and motility of neighboring VHL+cells.Periostin(POSTN),a soluble protein overexpressed and secreted by VHL non-expressing(VHL−)cells,promoted metastasis by enhancing the motility of VHL-WT cells and facilitating tumor cell vascular escape.Genetic deletion or antibody blockade of POSTN dramatically suppressed lung metastases in our preclinical models.This work supports a new strategy to halt the progression of ccRCC by disrupting the critical metastatic crosstalk between heterogeneous cell populations within a tumor.Junhui Hu Ping Tan Moe Ishihara Nicholas ABayley Shiruyeh Schokrpur Jeremy GReynoso Yangjun Zhang Raymond JLim Camelia Dumitras Lu Yang Steven MDubinett Parmjit SJat Jacques Van Snick Jiaoti Huang Arnold IChin Robert MPrins Thomas GGraeber Hua Xu Lily Wu 2023Signal Transduction and Targeted Therapy2023,8,5:0
