| 1 | Efficacy and safety of Shaoyang Xibi decoction in patients with knee osteoarthritis:a multi-center,single-blind,randomized controlled trial显示文摘PURPOSE: To observe the efficacy and safety of Shaoyang Xibi decoction(SYXBD) in patients with knee osteoarthritis(KOA), and to verify that the theory of 'Shaoyang dominating bone' in Traditional Chinese Medicine(TCM) can be applied to KOA treatment.METHODS: Participants were randomly allocated to two groups: SYXBD(treatment group, n = 66)and Meloxicam(control group, n = 66). Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC) and 36-Item Short Form Health Survey(SF-36) were used to assess efficacy before the treatment and 8 weeks after the treatment.RESULTS: Baseline data before the treatment between the two groups were similar. The WOMAC scores significantly decreased and the SF-36 scores significantly increased after 8-week treatment in both groups compared with before the treatment(P < 0.05). SYXBD significantly decreased pain scores(P < 0.001), physical function scores(P <0.001) and the total scores(P < 0.001) in WOMAC compared to Meloxicam. SYXBD significantly improved physical function(P = 0.021), bodily pain(P = 0.002) and general health(P = 0.014), with no significant difference in role emotional(P = 0.053),role physical(P = 0.517), vitality(P = 0.241), social function(P = 0.712) and mental health(P = 0.800)in SF-36 compared to Meloxicam. No adverse events were reported in the treatment group while13 adverse events happened in the control group during the study.CONCLUSION: SYXBD, prepared based on the theory of 'Shaoyang dominating bone', has a better curative efficay and safety in patients with KOA compared with Meloxicam. | 2 | Potential chronic liver toxicity in rats orally administered an ethanol extract of Huangqin(Radix Scutellariae Baicalensis)显示文摘OBJECTIVE: To investigate the potential chronic liver toxicity of oral administration of ethanol extract of Huangqin(Radix Scutellariae Baicalensis)(SBE) in Wistar rats.METHODS: SBE was administered to rats by gavage for 26 weeks, at doses of 300, 1250, or 2500 mg·kg-1·d-1 respectively. The rats were euthanized at the end of 13 and 26 weeks daily oral dosing and following 4 weeks of recovery time. The changes of hematology, urinary, blood biochemistry and histomorphology were examined at each time point and focus on liver function and histological changes.RESULTS: When SBE at a dose of up to 2500 mgkg-1 d-1 was fed to male and female rats for 2··6 weeks, the liver tissue showed some inflammatory change that predominated by leukocyte infiltrationbut returned to normal after withdrawal. In addition, high-dose SBE treatment of 26 weeks in rats,glucose, electrolyte and lipid levels also have some changes. In addition, there are no other functional or organic lesions related to SBE treatment.CONCLUSIONS: Long-term and high-dose SBE may cause liver damage, however, the structural damage of the liver can be restored after the ethanol extract stopping. | 3 | Peptide binding specificities of HLA-B*5701 and B*5801显示文摘Recently, genome wide association studies showed that there is a strong association between abacavir-induced serious, idio-syncratic, adverse drug reactions (ADRs) and human leukocyte antigen-B*5701 (HLA-B*5701). Studies also found that ab-acavir-induced ADRs were seldom observed in patients carrying the HLA-B*5801 subtype. HLA-B*5801 of the same sero type (B17) as B*5701 differs by only 4 amino acids from B*5701. It is believed that because of these sequence differences, HLA-B*5801 cannot bind the specific peptides which are required for HLA-B*5701 to stimulate the T cell immune response. Thus, the difference in peptide binding profiles between HLA-B*5701 and B*5801 is an important clue for exploring the mechanisms of abacavir-induced ADRs. VHSE (principal component score vector of hydrophobic, steric, and electronic properties), a set of amino acid structural descriptors, was employed to establish QSAR models of peptide-binding affinities of HLA-B*5701 and B*5801. Optimal linear SVM (support vector machine) models with high predictive capabilities were obtained for both B*5701 and B*5801. The R 2 (coefficient of determination), Q 2 (cross-validated R 2 ), and R PRE 2 (R 2 of test set) of two optimal models were 0.7530, 0.7037, 0.6153 (B*5701) and 0.6074, 0.5966, 0.5762 (B*5801), respectively. For B*5701 and B*5801, the mutations in positions 45 (MET-THR) and 46 (ALA-GLU) have little influence on the selection specificity of the P2 position of the bound peptide. However, the mutation in position 97 (VAL-ARG) greatly influences the selection specificity of the P7 position. HLA-B*5701 prefers the bulky and positively charged amino acids at the P7 position. | 6 | Relationship between groundwater cadmium and vicinity resident urine cadmium levels in the non-ferrous metal smelting area,China显示文摘Cadmium(Cd)has received widespread attention owing to its persistent toxicity and non-degradability.Cd in the human body is mainly absorbed from the external environment and is usually assessed using urinary Cd.Hunan Province is the heartland of the Chinese non-ferrous mining area,where several serious Cd pollution events have occurred,including high levels of Cd in the urine of residents.However,the environmental factors influencing high urinary Cd levels(UCLs)in nearby residents remain unclear.Therefore,211 nearby residents’UCLs and the corresponding sociological characteristics from nine groundwater samples in this area were analyzed using statistical analysis models.Groundwater Cd concentration ranged from 0.02 to 1.15μg/L,aligning with class III of the national standard;the range of UCL of nearby residents was 0.37–36.60μg/L,exceeding the national guideline of 0–2.5μg/L.Groundwater Cd levels were positively correlated with the UCL(P<0.001,correlation coefficient 95%CI=9.68,R^(2)=0.06).In addition,sociological characteristics,such as smoking status and education level,also affect UCL.All results indicate that local governments should strengthen the prevention and abatement of groundwater Cd pollution.This study is the first to systematically evaluate the relationship between groundwater Cd and UCL using internal and external environmental exposure data.These findings provide essential bases for relevant departments to reduce Cd exposure in regions where the heavy metal industry is globally prevalent. | Yujie Pan Yalan Li Hongxia Peng Yiping Yang Min Zeng Yang Xie Yao Lu Hong Yuan | 2023 | Frontiers of Environmental Science & Engineering2023,17,5: | 0 |

| 9 | Toxicity study of ethanol extract from Oroxylumindicum(L.) Vent in rats显示文摘OBJECTIVE: To evaluate the safety of Oroxylumindicum(L.) Vent extract administered for 26 weeks in Wistar rats.METHODS: Oroxylumindicum(L.) Vent extract was administrated to male and female rats by gavage once daily at doses of 54, 225, and450 mg·kg-1·d-1.The rats were sacrificed after administration for13 weeks and 26 weeks. Part of the rats in each group were allowed to recover for 4 weeks after 26-week administration. Systematic examinations including haematology, urology, blood biochemistry and histomorphology were performed at the end of13, 26 weeks of administration and 4 weeks of recovery.RESULTS: No treatment related adverse effect shappened on rats' general status, body weight, food consumption, urinary index and histomorphologyexamination. Although during the administration,in some rats of extract's groups, the value of Red blood cell count(RBC), white blood cell counts(WBC), hemoglobin(HGB), haematocrit(HCT) and K iron were decreased, and biochemistry index,such as glucose(GLU), triglyceride(TG), alanine transaminase(ALT) and blood urea nitrogen(BUN)were increased, the above parameters were within the normal ranges and all returned to baseline after the drug stopping for 4 weeks.CONCLUSION: The administration of Oroxylumindicum(L.)
