| 1 | Recent advances in electrogenerated chemiluminescence biosensing methods for pharmaceuticals显示文摘Electrogenerated chemiluminescence(electrochemiluminescence, ECL) generates species at electrode surfaces, which undergoes electron-transfer reactions and forms excited states to emit light. It has become a very powerful analytical technique and has been widely used in such as clinical testing, biowarfare agent detection, and pharmaceutical analysis. This review focuses on the current trends of molecular recognition-based biosensing methods for pharmaceutical analysis since 2010. It introduces a background of ECL and presents the recent ECL developments in ECL immunoassay(ECLIA), immunosensors, enzyme-based biosensors, aptamer-based biosensors, and molecularly imprinted polymers(MIP)-based sensors. | 7 | Selective DNA detection at Zeptomole level based on coulometric measurement of gold nanoparticle-mediated electron transfer across a self-assembled monolayer显示文摘A selective DNA sensing with zeptomole detection level is developed based on coulometric measurement of gold nanoparticle (AuNPs)-mediated electron transfer (ET) across a self-assembled monolayer on the gold electrode. After immobilization of a thiolated hairpin-structured DNA probe, an alkanethiol monolayer was self-assembled on the resultant electrode to block [Fe(CN)6 ]-3-/4in a solution from accessing the electrode. In the presence of DNA target, hybridization between the DNA probe and the DNA target breaks the stem duplex of DNA probe. Consequently, stem moiety at the 3′-end of the DNA probes was removed from the electrode surface and made available for hybridization with the reporter DNA-AuNPs conjugates (reporter DNA-AuNPs). The thiolated reporter DNA matches the stem moiety at the 3′-end of the DNA probe. AuNPs were then enlarged by immersing the electrode in a growth solution containing HAuCl 4 and H2O2 after the reporter DNA-AuNPs bound onto the electrode surface. The enlarged AuNPs on the electrode restored the ET between the electrode and the [Fe(CN)6]3 -/4- , as a result, amplified signals were achieved for DNA target detection using the coulometric measurement of Fe(CN)6 3- electro-reduction by prolonging the electrolysis time. The quantities of ET on the DNA sensor increased with the increase in DNA target concentration through a linear range of 3.0 fM to 1.0 pM when electrolysis time was set to 300 s, and the detection limit was 1.0 fM. Correspondingly, thousands of DNA (zeptomole) copies were detected in 10L samples. | 10 | Monitoring casein kinase Ⅱ at subcellular level via bio-bar-code-based electrochemiluminescence biosensing method显示文摘A highly sensitive electrochemiluminescence(ECL) biosensing method was developed for monitoring casein kinase Ⅱ(CK2) at subcellular level via bio-bar-code assay.A bio-bar-code probe(h-DNA/AuNPs/pDNA) prepared by conjugating phosphorylated DNA(p-DNA) and hairpin DNA(h-DNA) onto gold nanoparticles(AuNPs) was used as a carrier for ECL signal reagent(Ru(phen)32+) while a specific peptide was used as a recognition substance.A gold ultramicroelectrode with a diameter of 400 nm was fabricated and then modified with the specific peptide via self-assembly technique to obtain peptide modified gold ultramicroelectrode.The peptide on gold ultramicroelectrode was phosphorylated in the presence of CK2 and adenosine 5’-triphosphate,and then the phosphorylated peptide was integrated with the h-DNA/AuNPs/p-DNA through a process mediated by zirconium cations(Zr4+),and finally Ru(phen)32+ was intercalated into h-DNA.A 'signal on' ECL method was developed for the detection of CK2 in the range of 0.005-0.2 U/mL with a detection limit of 0.001 U/mL.Additionally,combined efficient subcellular phosphorylation in vivo with bio-bar-code-based ECL biosensing method,the ECL method was further applied to monitor CK2 at subcellular level without tedious subcellular fractionation.It was found that the concentration of CK2 by inserting the peptide modified gold ultramicroelectrode into the nucleus was higher than that into cytoplasm of HeLa cells.A distinct heterogeneity among CK2 concentrations in single cells was observed for cellular heterogeneity assessment. | Lifen Wang Jiajia Song Xiaofei Wang Honglan Qi Qiang Gao Chengxiao Zhang | 2020 | Chinese Chemical Letters2020,31,9: | 1 |

| 14 | Highly selective electrochemical method for the detection of serotonin at carbon fiber microelectrode modified with gold nanoflowers and overoxidized polypyrrole显示文摘A highly selective and sensitive electrochemical method was developed for the detection of serotonin (5-hydroxytryptamine, 5-HT) at gold nanoflowers (Au NFs) and overoxidized polypyrrole (OPPy) modified carbon fiber microelectrode (CFME). Carbon fiber was firstly modified with gold nanoflowers using electroless deposition method, and then modified with overoxidized polypyrrole using electrochemical polymerization and overoxidization to obtain OPPy/Au NFs/CFME. The obtained OPPy/Au NFs/CFME was characterized by field emission scanning electron microscopy and electrochemical techniques. It was found that the OPPy/Au NFs/CFME showed good sensitivity for the detection of 5-HT in the range of 10 nmol/L-7.0μmol/L with detection limit of 2.3 nmol/L, and negligible interferences from ascorbic acid, 5-hydroxyindole acetic acid and uric acid. The OPPy/Au NFs/CFME was successfully applied to the detection of 5-HT in human serum samples with good recovery. | 15 | Electrogenerated chemiluminescence aptasensor for thrombin incorporating poly(pyrrole-co-pyrrole propylic acid) nanoparticles loaded with aptamer and ruthenium complex显示文摘A novel electrogenerated chemiluminescence (ECL) aptasensor for highly sensitive detection of thrombin was developed on the basis of poly(pyrrole-co-pyrrole propylic acid) nanoparticles loaded with aptamer and ruthenium complex. Thrombin binding aptamers served as the molecular recognition elements and ruthenium bis(2,2′-bipyridine) (2,2′-bipyridine-4,4′-dicarboxylic acid)-ethylenediamine (Ru1) was used as an ECL signal complex. Novel electroactive polymers poly(pyrrole-co-pyrrole propylic acid) nanoparticles (Ppy-pa NPs) were synthesized by a simple alcohol-assisted microemulsion polymerization. Ru1-Ppy-pa NPs were synthesized by covalently coupling Ru1 with the Ppy-pa NPs. Ppy-pa NPs and Ru1-Ppy-pa NPs were characterized using a fourier transform infrared spectrometer, super-conducting fourier digital NMR spectrometer, and transmission electron microscope. One ECL chemical sensor fabricated by immobilizing the Ru1-Ppy-pa NPs on PIGE was developed for the determination of TprA with a high sensitivity and stability. The ECL aptasensor was fabricated by covalently coupling the thrombin binding aptamer-I (TBA-I) onto the surface of the paraffin-impregnated graphite electrode, which had been covalently modified with a monolayer of 4-aminobenzene sulfonic acid via electrochemical oxidations, for capturing thrombin onto the electrode and then the TBA-II labeled with Ru1-Ppy-pa NPs was bound with epitope of thrombin. The ECL aptasensor showed an extremely low detection limit of 3.0×l0?16 mol/L for thrombin and a good selectivity. | 16 | pH-responsive aldehyde-bearing cyclometalated iridium(Ⅲ) complex for tracking intracellular pH fluctuations under external stimulation显示文摘Intracellular p H undertakes critical functions in various biological and pathological processes. It is important to monitor intracellular p H fluctuations for understanding physiological and pathological processes.Here, one aldehyde-bearing cyclometalated iridium(Ⅲ) complex([(4-pba)_(2)Ir(dcphen)]PF_(6), 4-pba = 4-(2-pyridyl) benzaldehyde, dcphen = 4,7-dichloro-1,10-phenanthroline, probe 1) was synthesized and used to track intracellular p H fluctuations. Probe 1 displayed p H-dependent luminescence property in p H range of 1.81–6.81 with an evaluated p Kavalue of 4.30 in BR buffer-DMSO(v:v = 99:1). An intramolecular hydrogen bonds assisted p H-responsive mechanism was proposed for the p H-responsive behavior of probe1. Probe 1 was successfully applied for imaging and tracking p H fluctuations in He La cells under external stimulation with fast response time, good photostability as well as low cytotoxicity and high cell permeability.
