1Tissue-specific transcription reprogramming promotes liver metastasis of colorectal cancer显示文摘Metastasis,the development of secondary malignant growths at a distance from a primary tumor,is the cause of death for 90%of cancer patients,but little is known about how metastatic cancer cells adapt to and colonize new tissue environments.Here,using clinical samples,patient-derived xenograft(PDX)samples,PDX cells,and primary/metastatic cell lines,we discovered that liver metastatic colorectal cancer(CRC)cells lose their colon-specific gene transcription program yet gain a liver-specific gene transcription program.We showed that this transcription reprogramming is driven by a reshaped epigenetic landscape of both typical enhancers and super-enhancers.Further,we identified that the liver-specific transcription factors FOXA2 and HNF1A can bind to the gained enhancers and activate the liver-specific gene transcription,thereby driving CRC liver metastasis.Importantly,similar transcription reprogramming can be observed in multiple cancer types.Our data suggest that reprogrammed tissue-specific transcription promotes metastasis and should be targeted therapeutically.Shuaishuai Teng Yang Eric Li Ming Yang Rui Qi Yiming Huang Qianyu Wang Yanmei Zhang Shanwen Chen Shasha Li Kequan Lin Yang Cao Qunsheng Ji Qingyang Gu Yujing Cheng Zai Chang Wei Guo Pengyuan Wang Ivan Garcia-Bassets Zhi John Lu Dong Wang 2020Cell Research2020,30,1:11

3A rice XANTHINE DEHYDROGENASE gene regulates leaf senescence and response to abiotic stresses显示文摘Xanthine dehydrogenase, a member of the molybdenum enzyme family, participates in purine metabolism and catalyzes the generation of ureides from xanthine and hypoxanthine. However, the mechanisms by which xanthine dehydrogenase affects rice growth and development are poorly understood. In the present study, we identified a mutant with early leaf senescence and reduced tillering that we named early senescence and less-tillering 1(esl1). Map-based cloning revealed that ESL1 encodes a xanthine dehydrogenase, and it was expressed in all tissues. Chlorophyll content was reduced and chloroplast maldevelopment was severe in the esl1 mutant. Mutation of ESL1 led to decreases in allantoin, allantoate, and ABA contents. Further analysis revealed that the accumulation of reactive oxygen species in esl1 resulted in decreased photosynthesis and impaired chloroplast development, along with increased sensitivity to abscisic acid and abiotic stresses. 5The Thermogel Chronicle——From Rational Design of Thermogelling Copolymers to Advanced Thermogel Applications显示文摘CONSPECTUS:Temperature responsive supramolecular hydrogels,also known as thermogels,are produced by the self-assembly of amphiphilic copolymers in solution following temperature stimulus.They are a class of high caliber novel materials possessing highly attractive properties such as injectability,ability to undergo temperature controlled reversible sol−gel phase transitions,high biocompatibility,and tunable biodegradability.Much research vigor has been dedicated to designing advanced amphiphilic copolymers and investigating their molecular interactions in order to enhance the properties of the thermogelling systems and expand the scope of their applications.As such,thermogelling systems have since become well established in the field of sustained localized drug or protein delivery and have also been demonstrated as high potential materials for niche applications such as threedimensional cell culture,wound healing patches,and vitreous endotamponades.Recent developments saw the advent of thermogelling systems with advanced biomedical applications ranging from enhanced cancer therapy to radiology imaging to tissue engineering,and these are usually achieved by the conjugating of biologically relevant molecules such as drugs and peptides to the thermogel copolymer or by incorporating nanoparticles into the thermogel systems.New developments in this field see a shift away from employing traditional synthetic polymers such as polypropylene glycol(PPG)and poly(lactic-co-glycolic acid)(PLGA)to utilizing more advanced nature-derived bioactive molecules and also introducing chiral moieties in the thermogelling copolymer backbone.Qianyu Lin Cally Owh Jason Y.C.Lim Pei Lin Chee Michelle P.Y.Yew Eilis T.Y.Hor Xian Jun Loh 2021Accounts of Materials Research2021,2,10:1
6Research Progress and Ideas on the Anti-liver Fibrosis Effect of Ethnic Medicine Plumbagin Based on microRNAs/TLR4/NF-κB and NLRP3 Inflammasome Activation显示文摘The core of hepatic fibrosis is the activation of hepatic stellate cells.Through the lipopolysaccharide/TLR4/MyD88/NF-κB signal transduction pathway,the inflammatory response in the liver is directly enhanced,and then returns to promote the activation of hepatic stellate cells.And TLR4/MyD88/NF-κB signaling pathway can directly regulate the activation of NLRP3 inflammasome and is an important pathway for activating hepatic stellate cells.TLR4/MyD88/NF-κB/NLRP3 inflammasome pathway is regulated by upstream microRNAs.These miRNAs can significantly regulate the inflammatory response of the liver and the activation behavior of hepatic stellate cells,affecting the formation of liver fibrosis.Previous studies have found that the active ingredient of Guangxi specialty ethnic medicine,plumbagin,has a definite anti liver fibrosis effect,but its mechanism of action is not clear.This paper provides a review of the research progress on the above issues,and further research ideas have been derived from this,stating that'the anti liver fibrosis effect of plumbagin is achieved by regulating miRNA/TLR4/MyD88/NF-κB inflammatory pathway and activating downstream NLRP3 inflammasome'.Mingzhe LU Qianyu LIU Yue PENG Jiang LIN Weiqian GUO Miao YANG 2023Medicinal Plant2023,14,5:0

8Highly sensitive vector magnetic fiber sensor based on hyperbolic metamaterials显示文摘Hyperbolic metamaterials(HMMs) are novel artificial materials that excite the surface plasmon resonance(SPR) because of their unique hyperbolic dispersion properties. Herein, to the best of our knowledge, we propose the first HMM-based fiber SPR(HMM-SPR) sensor for vector magnetic detection. By selecting the composite materials and structural parameters of the HMM dispersion management, HMM-SPR sensors can achieve a high refractive index sensitivity of 14.43 μm/RIU. Vector magnetic field detection was performed with the HMM-SPR sensor encapsulated with a magnetic fluid. Compared with other ferrofluidbased magnetic field fiber sensors, the proposed sensor shows pronounced advantages in intensity and direction sensitivity of 1.307 nm/Oe and 7.116 nm/°, respectively. 9A rare cause of fever of unknown origin - cervical spinal cord lesion显示文摘To the editor:A female patient,aged 49,visited the emergency room at Beijing Union Medical College Hospital on August 21,2013.She had had 'malaise for five years,aggravated for more than a month with fever.' Since early 2008,the patient had whole body fatigue without a cause,but it was not affecting her daily work.As the symptoms gradually worsened,she became incapable of daily chores and had difficulty climbing stairs to the second floor.Two years ago,she resigned from work to rest at home,but the symptoms of fatigue aggravatedand she was reluctant to get out of bed.One month ago,the patient accidentally fell from her bed due to fatigue and she was only able to get up with help from family.Since then,the patient started to have fever (self-measured temperatures up to 38-39℃) and dizziness.Self-prescribed antipyretic drugs did not have an effect on reducing the fever.She did not sweat,but her body temperature decreased slightly after a warm sponge bath.She could not walk,comb her hair,or go to the bathroom by herself.She had intermittent palpitations,dizziness,and loss of appetite,with normal urine and bowel movements,and no other discomfort.On August 20,2013,the patient's temperature increased to 41.6℃,she then came to our hospital emergency room for further treatment.Ge Ying Li Taisheng Liu Zhengyin Liu Mingsheng Guan Hongzhi Chert Lin Tian Ye Zhuang Qianyu 2014Chinese Medical Journal2014,,19:0
10Identification of an immune classifier for predicting the prognosis and therapeutic response in triple-negative breast cancer显示文摘Triple-negative breast cancer(TNBC)poses a significant challenge due to the lack of reliable prognostic gene signatures and an understanding of its immune behavior.Methods:We analyzed clinical information and mRNA expression data from 162 TNBC patients in TCGA-BRCA and 320 patients in METABRIC-BRCA.Utilizing weighted gene coexpression network analysis,we pinpointed 34 TNBC immune genes linked to survival.The least absolute shrinkage and selection operator Cox regression method identified key TNBC immune candidates for prognosis prediction.We calculated chemotherapy sensitivity scores using the“pRRophetic”package in R software and assessed immunotherapy response using the Tumor Immune Dysfunction and Exclusion algorithm.Results:In this study,34 survival-related TNBC immune gene expression profiles were identified.A least absolute shrinkage and selection operator-Cox regression model was used and 15 candidates were prioritized,with a concomitant establishment of a robust risk immune classifier.The high-risk TNBC immune groups showed increased sensitivity to therapeutic agents like RO-3306,Tamoxifen,Sunitinib,JNK Inhibitor VIII,XMD11-85h,BX-912,and Tivozanib.An analysis of the Search Tool for Interaction of Chemicals database revealed the associations between the high-risk group and signaling pathways,such as those involving Rap1,Ras,and PI3K-Akt.The low-risk group showed a higher immunotherapy response rate,as observed through the tumor immune dysfunction and exclusion analysis in the TCGA-TNBC and METABRIC-TNBC cohorts.Conclusion:This study provides insights into the immune complexities of TNBC,paving the way for novel diagnostic approaches and precision treatment methods that exploit its immunological intricacies,thus offering hope for improved management and outcomes of this challenging disease.KUAILU LIN QIANYU GU XIXI LAI 2023BIOCELL2023,47,12:0
11Yunpi Qushi Jiangzhuo Recipe Alleviates Lipid Deposition and Reduces Liver Damage in Mice with Non-alcoholic Fatty Liver Disease显示文摘Background:Non-alcoholic fatty liver disease(NAFLD)is a multifactorial liver metabolic disease,which af-fects nearly a quarter of the world’s population.Yunpi Qushi Jiangzhuo Recipe(QSJZR)is a traditional Chi-nese medicine compound,which is composed of Amomum kravanh Pierre ex Gagnep,Coix lacryma-jobi L.var.ma-yuen(Roman.)Stapf,Prunus armeniac a L.var.ansu Maxim,Salvia miltiorrhiza Bge,Nelumbo nucifera Gaertn,Alisma plantago-aquatica Linn,Polyporus umbellatus(Pers.)Fries,Poria cocos(Schw.)Wolf,and Artemisia capillaris Thunb.QSJZR has a certain therapeutic effect on NAFLD patients,but its specific mechanism is not clear.Objective:To investigate the effect of QSJZR on high-fat diet(HFD)-fed NAFLD mice and its mechanism.Methods:Twenty-four SPF female C57BL/6 J mice(21.0±0.5 g)were randomly divided into normal diet(ND)group,HFD group and QSJZR group.ND group was given basal diet,while the other two groups were given HFD.Meanwhile,each mouse in QSJZR group was given 0.2 mL/day(containing 2.27 g crude drug per mL)QSJZR,ND group and HFD group were given the same amount of normal saline for 13 consecutive weeks.Then,the serum was collected for biochemical assay,and the liver was removed for pathological examination and biochemical analysis.Results:Body weight and white fat weight of the HFD-induced NAFLD mice significantly decreased after min-istration with QSJZR,while liver weight had no significant change.QSJZR also significantly reduced liver and serum triglyceride levels,and alleviated hepatocyte lipid deposition by regulating genes and proteins expression related to lipid metabolism,including AMPK,SREBP1C,CPT1A and ACC.In addition,compared with HFD group,liver malondialdehyde(MDA)content was lower in QSJZR group,while glutathione peroxidase(GPx)content was higher.Serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels were lower in QSJZR group than those in HFD group.Treatment with QSJZR significantly alleviated liver injury by increasing BCL2/BAX protein ratio and down-regulating ASK1/JNK pathway.Conclusion:Administration of QSJZR to NAFLD mice once daily for 13 weeks can decrease lipid levels,and alleviate liver damage.These results suggest that QSJZR might be used to treat NAFLD,although more studies need to be conducted for further verification.Xiangyao Wu Linwensi Zhu Mengmeng You Yanli Chen Hui Chai Lin Chen Qianyu Qian Zuifei Shangguan Fusheng Jiang Songtao Li Xiaobing Dou 2023Clinical Complementary Medicine and Pharmacology2023,3,1:0
12Recent Advances in Asymmetric Transformations of Unactivated Alkanes and Cycloalkanes through Direct C—H Functionalization显示文摘The direct conversion of unactivated alkanes and cycloalkanes into structurally diverse molecules through aliphatic C—H functionalization is a useful process, which has attracted intense interest from academia and industry. Methods to control chemo- and site-selectivity, combined with asymmetric catalysis, provide appealing access to high value-added enantiomer-enriched compounds but are far less developed. This review focuses on recent progress in (i) asymmetric reactions of alkanes or cycloalkanes with prochiral substrates which generate a stereocenter adjacent to the cleaved C(sp3)–H bond, and (ii) C(sp3)–H enantiodiscriminatory reactions creating a new stereogenic center on the carbon of a cleaved C(sp3)–H bond. Elegant strategies are discussed, including (a) metal carbene-induced C—H insertions by chiral rhodium catalysts, (b) metal-oxo-mediated C—H oxidation by biomimetic manganese catalysts, (c) enzyme catalysis by cytochromes P450 variants, and (d) dual catalysis by a photocatalyst and a chiral Lewis acid (CLA) or a chiral phosphoric acid (CPA). These catalytic systems can not only precisely recognize primary, secondary and tertiary C—H bonds at specific positions in alkanes and cycloalkanes, but also support a high level of stereoselectivity in the reactions. 13Research Progress and Prospects on the Anti-liver Fibrosis,Blood Circulation-promoting and Stasis-resolving Effects of Curcuma kwangsiensis Based on W-P Bodies显示文摘Liver fibrosis is a necessary stage in the progression of chronic liver disease to cirrhosis.So far,no satisfactory drugs have been found to intervene in liver fibrosis.Liver microcirculation disorders are one of the important pathogenesis of chronic liver disease,and hepatic sinusoidal endothelial cells(HSECs)are the main cells that constitute the liver microcirculation barrier.In clinical practice,W-P bodies have been detected in HSECs of most patients with liver fibrosis.W-P bodies serve as a site for the synthesis and storage of vW factors,ET-1 and other cytokines that promote liver fibrosis.They can disrupt the structure and function of HSECs,cause liver microcirculation disorders,and exacerbate the progression of liver fibrosis.Previous studies have found that the Guangxi specialty ethnic medicine,C.kwangsiensis S.G.Lee et C.F.Liang,has definite effects in promoting blood circulation,resolving blood stasis,and resisting liver fibrosis.Based on this,a further research idea has been derived,stating that the blood circulation-promoting,blood stasis-resolving,and anti-liver fibrosis effects of C.kwangsiensis are produced by affecting the formation of W-P bodies,the synthesis and storage of contents in W-P bodies,and intervening in their exocytosis capacity.Weiqian GUO Mingzhe LU Yue PENG Jiang LIN Qianyu LIU Miao YANG 2023Agricultural Biotechnology2023,12,4:0
