1Clinically relevant factors associated with quantitative optical coherence tomography angiography metrics in deep capillary plexus in patients with diabetes显示文摘Background:To test clinically relevant factors associated with quantitative artifact-free deep capillary plexus(DCP)metrics in patients with diabetes mellitus(DM).Methods:563 eligible eyes(221 with no diabetic retinopathy[DR],135 with mild DR,130 with moderate DR,and 77 with severe DR)from 334 subjects underwent optical coherence tomography-angiography(OCT-A)with a swept-source OCT(Triton DRI-OCT,Topcon,Inc.,Tokyo,Japan).Strict criteria were applied to exclude from analysis those DCP images with artifacts and of poor quality,including projection artifacts,motion artifacts,blurriness,signal loss,B-scan segmentation error,or low-quality score.A customized MATLAB program was then used to quantify DCP morphology from the artifact-free DCP images by calculating three metrics:foveal avascular zone(FAZ),vessel density(VD),and fractal dimension(FD).Results:166(29.5%)eyes were excluded after quality control,leaving in the analysis 397 eyes(170 with no DR,101 with mild DR,90 with moderate DR,36 with severe DR)from 250 subjects.In the multiple regression models,larger FAZ area was associated with more severe DR(β=0.687;p=0.037),shorter axial length(AL)(β=−0.171;p=0.003),thinner subfoveal choroid thickness(β=−0.122;p=0.031),and lower body mass index(BMI)(β=−0.090;p=0.047).Lower VD was associated with more severe DR(β=−0.842;p=0.001),shorter AL(β=0.107;p=0.039),and poorer visual acuity(VA)(β=−0.133;p=0.021).Lower FD was associated with more severe DR(β=−0.891;p<0.001)and with older age(β=−0.142;p=0.004).Conclusions:Quantitative artifact-free DCP metrics are associated with VA,DR severity,AL,subfoveal choroidal thickness,age,and BMI in diabetic patients.The effects of ocular and systemic factors should be considered for meaningful interpretations of DCP changes in DM patients.Fang Yao Tang Erica O.Chan Zihan Sun Raymond Wong Jerry Lok Simon Szeto Jason C.Chan Alexander Lam Clement C.Tham Danny S.Ng Carol Y.Cheung 2020Eye and Vision2020,7,1:7

3Drug development in targeting ion channels for brain edema显示文摘Cerebral edema is a pathological hallmark of various central nervous system(CNS)insults,including traumatic brain injury(TBI)and excitotoxic injury such as stroke.Due to the rigidity of the skull,edema-induced increase of intracranial fluid significantly complicates severe CNS injuries by raising intracranial pressure and compromising perfusion.Mortality due to cerebral edema is high.With mortality rates up to 80%in severe cases of stroke,it is the leading cause of death within the first week.Similarly,cerebral edema is devastating for patients of TBI,accounting for up to 50%mortality.Currently,the available treatments for cerebral edema include hypothermia,osmotherapy,and surgery.However,these treatments only address the symptoms and often elicit adverse side effects,potentially in part due to non-specificity.There is an urgent need to identify effective pharmacological treatments for cerebral edema.Currently,ion channels represent the third-largest target class for drug development,but their roles in cerebral edema remain ill-defined.The present review aims to provide an overview of the proposed roles of ion channels and transporters(including aquaporins,SUR1-TRPM4,chloride channels,glucose transporters,and proton-sensitive channels)in mediating cerebral edema in acute ischemic stroke and TBI.We also focus on the pharmacological inhibitors for each target and potential therapeutic strategies that may be further pursued for the treatment of cerebral edema.Zheng-wei Luo Andrea Ovcjak Raymond Wong Bao-xue Yang Zhong-ping Feng Hong-shuo Sun 2020Acta Pharmacologica Sinica2020,41,10:3

9Hollow silica reinforced magnesium nanocomposites with enhanced mechanical and biological properties with computational modeling analysis for mandibular reconstruction显示文摘The present study investigates Mg-SiO2 nanocomposites as biodegradable implants for orthopedic and maxillofacial applications.The effect of presence and progressive addition of hollow silica nanoparticles(0.5,1,and 1.5)vol.%on the microstructural,mechanical,degradation,and biocompatibility response of pure Mg were investigated.Results suggest that the increased addition of hollow silica nanoparticles resulted in a progressive increase in yield strength and ultimate compressive strength with Mg-1.5 vol.%SiO2 exhibiting superior enhancement.The response of Mg-SiO2 nanocomposites under the influence of Hanks’balanced salt solution revealed that the synthesized composites revealed lower corrosion rates,indicating rapid dynamic passivation when compared with pure Mg.Furthermore,cell adhesion and proliferation of osteoblast cells were noticeably higher than pure Mg with the addition of 1 vol.%SiO2 nanoparticle.The biocompatibility and the in vitro biodegradation of the Mg-SiO2 nanocomposites were influenced by the SiO2 content in pure Mg with Mg-0.5 vol.%SiO2 nanocomposite exhibiting the best corrosion resistance and biocompatibility when compared with other nanocomposites.Enhancement in mechanical,corrosion,and biocompatibility characteristics of Mg-SiO2 nanocomposites developed in this study are also compared with properties of other metallic biomaterials used in alloplastic mandibular reconstruction in a computational model.Somasundaram Prasadh Vyasaraj Manakari Gururaj Parande Raymond Chung Wen Wong Manoj Gupta 2020International Journal of Oral Science2020,12,4:2

13Molecular Advances in Severe Acute Respiratory Syndrome-associated Coronavirus (SARS-CoV)显示文摘The sudden outbreak of severe acute respiratory syndrome (SARS) in 2002 prompted the establishment of a global scientific network subsuming most of the traditional rivalries in the competitive field of virology. Within months of the SARS outbreak, collaborative work revealed the identity of the disastrous pathogen as SARS-associated coronavirus (SARS-CoV). However, although the rapid identification of the agent represented an important breakthrough, our understanding of the deadly virus remains limited. Detailed biological knowledge is crucial for the development of effective countermeasures, diagnostic tests, vaccines and antiviral drugs against the SARS-CoV.
