2Chemical compound cinobufotalin potently induces FOXO1-stimulated cisplatin sensitivity by antagonizing its binding partner MYH9显示文摘In this study,we present novel molecular mechanisms by which FOXO1 functions as a tumor suppressor to prevent the pathogenesis of nasopharyngeal carcinoma(NPC).First,we observed that FOXO1 not only controlled tumor stemness and metastasis,but also sensitized NPC cells to cisplatin(DDP)in vitro and in vivo.Mechanistic studies demonstrated that FOXO1-induced miR-200b expression through the GSK3β/β-catenin/TCF4 network-mediated stimulation of ZEB1,which reduced tumor stemness and the epithelial–mesenchymal transition(EMT)signal.Furthermore,we observed FOXO1 interaction with MYH9 and suppression of MYH9 expression by modulating the PI3K/AKT/c-Myc/P53/miR-133a-3p pathway.Decreased MYH9 expression not only reduced its interactions with GSK3β,but also attenuated TRAF6 expression,which then decreased the ubiquitin-mediated degradation of GSK3βprotein.Increased GSK3βexpression stimulated theβ-catenin/TCF4/ZEB1/miR-200b network,which increased the downstream tumor stemness and EMT signals.Subsequently,we observed that chemically synthesized cinobufotalin(CB)strongly increased FOXO1-induced DDP chemosensitivity by reducing MYH9 expression,and the reduction in MYH9 modulated GSK3β/β-catenin and its downstream tumor stemness and EMT signal in NPC.In clinical samples,the combination of low FOXO1 expression and high MYH9 expression indicated the worst overall survival rates.Our studies demonstrated that CB potently induced FOXO1-mediated DDP sensitivity by antagonizing its binding partner MYH9 to modulate tumor stemness in NPC.YongHao Li Xiong Liu Xian Lin Menyang Zhao Yanyi Xiao Chen Liu Zixi Liang Zelong Lin Renhui Yi Zibo Tang Jiahao Liu Xin Li Qingping Jiang Libo Li Yinyin Xie Zhen Liu Weiyi Fang 2019Signal Transduction and Targeted Therapy2019,4,1:6
3Responses of Synechocystis sp. PCC 6803 (cyanobacterium) photosystem Ⅱ to pyrene stress显示文摘In order to explore the mechanism of acute toxicity for pyrene to cyanobacterial organisms,the responses of Synechocystis sp.PCC 6803 photosystem II(PS II) under pyrene stress were studied.The results showed there was no significant difference about the oxygen evolution under 0.125 mg/L pyrene stress when compared with control,but it was significantly lower than control at 0.625 mg/L pyrene.Polyphasic chlorophyll-a fluorescence transients in cells of Synechocystis sp.PCC 6803 exhibited a typical increase including O,J,I,and P phases.Fluorescence yield at phases J,I and P declined slightly at 0.125 and 0.625 mg/L pyrene,and significantly lower than control at 3.125 mg/L.According to the parameters deviated from JIP-test,no modification was induced by pyrene both at the donor side and at the acceptor side of PS II,and the reaction centre of PS II is the primary damaging target.Based on the expressing of four key genes(psbA,psbB,psbC and psbO) of PS II,only psbA showed significant difference at 3.125 mg/L pyrene when compared with control.Jihai Shao Gongliang Yu Zhongxing Wu Xin Peng Renhui Li 2010Journal of Environmental Sciences2010,22,7:2
4First report of microcystin production in Microcystis smithii Komrek and Anagnostidis(Cyanobacteria) from a water bloom in Eastern China显示文摘A water bloom sample collected from Lake Dishui in Shanghai was characterized.The morphological identification showed that Micorcystis wesenbergii and Micorcystis smithii were the main component of the bloom.Five strains of M.smithii were successfully isolated.Their 16S rRNA gene sequences based phylogenetic tree showed that the five strains of M.smithii intermixed with strains of other morphospecies in Microcystis.A fragment of mcy gene encoding for microcystin synthetase was detected in one of the five M.smithii strains(CHAB 2183),indicating its potential of microcystin production.High performance liquid chromatography analysis confirmed M.smithii CHAB 2183 to produce microcystin-RR as 1550 μg per gram dry weight cells.The present investigation,for the first time,reported the isolated strains of M.smithii and microcystin production from M.smithii.Yang Liu Wenhua Tan Xingqiang Wu Zhongxing Wu Gongliang Yu Renhui Li 2011Journal of Environmental Sciences2011,23,1:2
5Molecular characterization and toxin quantification of Microcystis panniformis: A microcystin producer in Lake Taihu,China显示文摘Microcystis panniformis is a bloom forming species with flat panniform-like colonies. This species was recently found in Lake Taihu, China. To specifically characterize M. panniformis based on isolated strains, morphological examination on colonial transition and genetic examination are needed. Three M. panniformis strains isolated from a water bloom sample in Lake Taihu were characterized by molecular analysis and toxin quantification. Phylogenetic analysis based on both 16S rRNA gene and internal transcribed spacer(ITS) between 16S and 23S rRNA genes were performed and compared to facilitate easy identification of the species.Relatively high similarities(98%–99%) were shown in 16S rDNA sequences between the strains of M. panniformis and those of other Microcystis species, whereas the similarities for ITS sequences were 88%–95%. In the phylogenetic tree based on the 16S rDNA sequences, the M. panniformis and M. aeruginosa strains were intermixed together with no clear division,whereas all of the M. panniformis strains were clustered together in a single clade based on the ITS sequences based phylogenyetic tree. The mcyE gene was detected in all three strains, and microcystin was determined by high-performance liquid chromatography. 11Structural evolution of low-dimensional metal oxide semiconductors under external stress显示文摘Metal oxide semiconductors(MOSs) are attractive candidates as functional parts and connections in nanodevices.Upon spatial dimensionality reduction, the ubiquitous strain encountered in physical reality may result in structural instability and thus degrade the performance of MOS. Hence, the basic insight into the structural evolutions of low-dimensional MOS is a prerequisite for extensive applications, which unfortunately remains largely unexplored. Herein, we review the recent progress regarding the mechanical deformation mechanisms in MOSs, such as CuO and ZnO nanowires(NWs). We report the phase transformation of CuO NWs resulting from oxygen vacancy migration under compressive stress and the tensile strain-induced phase transition in ZnO NWs. 12Harmful algal bloom-forming dinofl agellate Prorocentrum donghaiense inhibits the growth and photosynthesis of seaweed Sargassum fusiformis embryos显示文摘Harmful algal bloom(HAB)is an ecological disaster to local mariculture.At present,its impact on macrophytes has not been well studied.In this study,we cultivated sexually propagated embryos of S argassum fusiformis-an edible seaweed-in Prorocentrum donghaiense suspensions at different cell densities(0,0.50×10^(5),0.75×10^(5),1.00×10^(5),and 1.50×10^(5) cells/mL)for 10 days,during which growth and photosynthetic activities of the embryos were determined,and a monocultivation was set up for comparison.Results show that the relative growth rate and photosynthetic activities of the embryos co-cultivated with P.donghaiense were inhibited mostly and significantly in the cell densities of 0.75×10^(5),1.00×10^(5),and 1.50×10^(5) cells/mL,and the inhibitory effects increased in overall with increased cell densities.The maximum relative electron transport rates(rETR max)and apparent photosynthetic efficiency(a)of co-cultivated embryos were all significantly lower than monocultivation ones on the 10 th day.Furthermore,the photosynthetic activity detected by chlorophyll-a fluorescence transient(i.e.,OJIP),the electron transport among electron transfer accepters of PSII(photosystem II)and that from PSII to PSI(photosystem I)was restricted,which is probably responsible for the decreases of rETR max andain the co-cultivated embryos.In addition,parts of the photosynthetic reaction centers of PSII in the co-cultivated embryos were inactivated.Therefore,P.donghaiense bloom could restrain the development and photosynthetic activities of S.fusiformis embryos,reduce the seedlings stock,and eventually hinder the development of S.fusiformis production industry.Caixia WANG Min WANG Binbin CHEN Wenli QIN Lidong LIN Chuanjun DAI Hengguo YU Renhui LI Min ZHAO Zengling MA 2021Journal of Oceanology and Limnology2021,39,6:1

14Competitiveness of alga Microcystis aeruginosa co-cultivated with cyanobacterium Raphidiopsis raciborskii confirms its dominating position显示文摘Microcystis aeruginosa has always been regarded as the main culprit of cyanobacterial blooms in freshwater.However,in recent years,Raphidiopsis raciborskii has gradually replaced M.aeruginosa as the culprit of cyanobacterial blooms in some tropical and subtropical shallow lakes.To reveal which one plays a more dominant role,interactions between cylindrospermospin(CYN)-producing R.raciborskii and microcystins(MCs)-producing or non-MCs-producing M.aeruginosa strains were studied using bialgal cultures at different initial ratios of biomasses of the two species at 25℃.During the co-cultivation,the M.aeruginosa strains inhibited the growth and heterocyst formation of R.raciborskii filaments,and thus occupied a dominant position during the co-cultivation regardless of the initial biomass ratios in the cultures.In addition,the MCs-producing M aeruginosa strain contributed to a higher portion of the total biomass and exerted a stronger inhibitory effect on R.raciborskii compared with the non-MCs-producing strain.However,the growth of both MCs-producing and non-MCs-producing M.aeruginosa strains was stimulated by R.raciborskii in the co-cultures compared with M.aeruginosa monoculture,indicating that M.aeruginosa could outcompete R.raciborskii if given enough time,enabling it to develop into the dominant species even in very low initial concentration.To our best knowledge,this is the first report on the loss of heterocyst formation by a species of cyanobacteria that resulted from interactions between two different species of cyanobacteria.These findings indicate that it is difficult for R.raciborskii to replace the dominant position of M.aeruginosa under the experimental environmental condition,and the allelopathic effects of M.aeruginosa on R.raciborskii could significantly contribute to the success of M.aeruginosa.Zengling MA Xiaoqiao ZHANG Renhui LI Min WANG Wenli QIN He ZHANG Gang LI Henguo YU Chuanjun DAI Min ZHAO 2022Journal of Oceanology and Limnology2022,40,5:1

19Theoretical study of the charge carrier mobilities of the molecular materials tetrathiafulvalene (TTF) and 2,5-bis(1,3-dithiolan-2-ylidene)-1,3,4,6-tetrathiapentalene (BDH-TTP)显示文摘Tetrathiafulvalene (TTF) is a kind of fused ring aromatic compound containing four sulfur atoms in one molecule, which is well known as a charge transport material. In order to calculate the charge mobility of this semiconductor, Marcus electron transfer theory and the embedded model, which can give small intramolecular reorganization energies, were employed. The calculated results were in good agreement with the experimental values, so the above computing model is appropriate to assess the electrical property of TTF. On this basis, we predicted the charge mobility of 2,5-bis(1,3-dithiolan-2-ylidene)-1,3,4,6-tetrathiapentalene (BDH-TTP) crystals, for which the molecular structure is similar to TTF. The calculated results indicated that BDH-TTP is a p-type material, which has a better performance than TTF in hole transfer due to larger hole coupling and the smaller hole injection barrier. In addition, the direct coupling (DC) and the site energy correction (SEC) methods were used to calculate the charge transfer integrals. Although the results were slightly different, the qualitative trends were the same. Furthermore we took into account the anisotropic transfer properties of TTF and BDH-TTF, since obviously the mobilities along one dimension are larger than those along three dimensions.
