1External use of Ruyanneixiao cream efficiently blocks precancerous mammary lesions by interfering with glycolysis induced by inhibition of hypoxia inducible factor-1α, hexokinase 2, phosphofructokinase, and pyruvate kinase M2 expression显示文摘OBJECTIVE:To investigate the effect of Ruyanneixiao cream(RYNX) on the expression of hypoxia inducible factor-1α(HIF-1α), hexokinase 2(HK2),phosphofructokinase(PFK), and pyruvate kinase M2(PKM2) mRNA and protein in MCF-10 AT cells and in an animal model of precancerous mammary lesions.METHODS:Following treatment of MCF-10 AT cells with RYNX, tamoxifen(TAM) and YC-1 for 48 h,HIF-1α, HK2, PFK, PKM2 mRNA and protein expression was analyzed.Fifty female SD rats were randomly divided into control, model, TAM, and highand low-dose RYNX groups, with 10 rats in each group.A precancerous mammary lesion model was established for all groups except the control group.High-and low-dose RYNX cream containing TAM was coated on the breasts of animals in the corresponding groups.The rat mammary tissue was removed in the 10 th week and HIF-1α, HK2, PFK,PKM2 mRNA and protein was analyzed.RESULTS:In vitro analyses demonstrated that, compared with the matrix group, HIF-1α, HK2, PFK,PKM2 mRNA and protein expression was significantly decreased in the RYNX group(P < 0.05).Compared with the YC-1 + RYNX group, HK2, PFK,and PKM2 protein expression was significantly reduced in the RYNX group.HIF-1α, HK2, PFK, and PKM2 protein expression was increased significantly in the model group(P < 0.05) compared with the control group, while HIF-1α, HK2, PFK, and PKM2 mRNA and protein expression was significantly decreased in both the high-and low-dose RYNX groups(P < 0.05), with the effect being greater in the high-dose group.CONCLUSION:RYNX can block precancerous breast lesions by decreasing the expression of HK2,PFK, and PKM2 mRNA and protein via inhibition of HIF-1α mRNA and protein overexpression in a dose-dependent manner.Li Xiaobo Ma Min Zhang Guijuan Ma Yi Liao Rui Chen Ruixue Yan Xianxin Bie Fengjie Huang Maojie Liang Shijie 2017Journal of Traditional Chinese Medicine2017,37,2:5
2Metal Particle Contamination in Gas-insulated Switchgears/Gas-insulated Transmission Lines显示文摘Metal particle contamination in a gas-insulated switchgear(GIS)or a gas-insulated transmission line(GIL)is an important factor leading to the decline of insulation performance.Exploring the deterioration mechanism and suppression measures of metal particles on insulation is a key technical problem in enhancing the dielectric strength of GIS/GIL equipment.In this paper,the charge and motion characteristics of metal particles are first introduced.The gas gap breakdown caused by free metal particles and the surface flashover caused by metal particles near or adsorbed on the insulator are then analyzed according to different particle motion patterns and spatial locations.Subsequently in terms of operation managements,the existing methods of particle detection are analyzed.In addition,the main inhibition methods of metal particles are introduced from three aspects:particle trap,insulator surface treatment and electrode coating.Finally,the prospects in the future research on particle pollution in GIS/GIL are also pointed out.Jian Wang Qi Hu Yanan Chang Jingrui Wang Ruixue Liang Youping Tu Chuanyang Li Qingmin Li 2021CSEE Journal of Power and Energy Systems2021,7,5:1
3A 1.8-V 240-MHz 2.19-mW Four-Stage CMOS OTA with a Segmenting Frequency Compensation Technique显示文摘A four-stage Operational transconductance amplifier(OTA)used in an infrared temperature sensor adopting the proposed Feed-forward Gm-stage and segmenting nested Miller compensation technique is presented.The purpose of the proposed segment compensation is primarily to make more amplifier stages concatenated.The circuit linked several transconductance stages to form a segment,and linked several segments to form a large multistage amplifier.For example,a twostage amplifier linked with a three-stage amplifier can realize a five-stage amplifier,two three-stage amplifiers linked can realize a six-stage amplifier.A four-stage amplifier in the form of 2+2(two stages and two stages)was used as an example to verify this compensation method.The proposed OTA is designed in the 180 nm complementary metal oxide semiconductor process.It consists of two parts which ensure the stability and improve the bandwidth performance.The first part is a feedforward transconductance stage,and the second part is the two-segment transconductance stages with the Miller compensation techniques employed within and between the two segments.Based on the small-signal model,stability analysis and theoretical derivation are performed in theory.On condition of a 2-pF load capacitance,a direct current gain of 109 dB and a gain-bandwidth of 240 MHz with a phase margin of 50°can be achieved.The proposed design consumes 2.19 mW in a 1.8-V supply voltage.The transient simulation indicates that the settling time of the output is 19 ns with the settling error being 1%,and the slew rate is 114 V/μs.LIANG Yuhua ZHENG Zirui LIU Shubin LI Dengquan DING Ruixue ZHU Zhangming 2021Chinese Journal of Electronics2021,30,5:0
4Identification of oleic acid as an endogenous ligand of GPR3显示文摘Although GPR3 plays pivotal roles in both the nervous system and metabolic processes,such as cold-induced thermogenesis,its endogenous ligand remains elusive.Here,by combining structural approach(including cryo-electron microscopy),mass spectrometry analysis,and functional studies,we identify oleic acid(OA)as an endogenous ligand of GPR3.Our study reveals a hydrophobic tunnel within GPR3 that connects the extracellular side of the receptor to the middle of plasma membrane,enabling fatty acids to readily engage the receptor.Functional studies demonstrate that OA triggers downstream Gs signaling,whereas lysophospholipids fail to activate the receptor.Moreover,our research reveals that cold stimulation induces the secretion of OA in mice,subsequently activating Gs/cAMP/PKA signaling in brown adipose tissue.Notably,brown adipose tissues from Gpr3 knockout mice do not respond to OA during cold stimulation,reinforcing the significance of GPR3 in this process.Finally,we propose a“born to be activated and cold to enhance”model for GPR3 activation.Our study provides a starting framework for the understanding of GPR3 signaling in cold-stimulated thermogenesis.Yangjie Xiong Zhenmei Xu Xinzhi Li Yuqin Wang Jing Zhao Na Wang Yaning Duan Ruixue Xia Zhengbin Han Yu Qian Jiale Liang Anqi Zhang Changyou Guo Asuka Inoue Yu Xia Zheng Chen Yuanzheng He 2024Cell Research2024,34,3:0
5Research on discharge phenomenon caused by cross‐adsorption of linear insulating fibre and metal dust under DC voltage显示文摘A large amount of metal dust is often generated during production and operation of Gas Insulated Switchgear/Gas‐Insulated Transmission Line,and insulating fibres are very easy to mix in during assembly.The discharge phenomenon induced by these two impurities remains unknown.By setting up a test rig with parallel electrodes,the discharge characteristics of metal dust and linear insulating fibres under the negative DC voltage were studied separately and jointly.The obtained research results indicate some interesting phenomena that the metal dust may render‘sand storm’motion characteristics under the action of the electric field,and the linear insulating fibres appear to show‘standing’,‘jumping’,‘flying fire’and other phenomena.A small amount of dust and fibres alone present little effect on the breakdown voltage of the air gap.However,while the insulating fibres and metal dust coexist,the metal dust will be adsorbed on the surface of the insulating fibres,then the surface charge of the fibres is reallocated,and the discharge phenomenon at both ends of the fibres is enhanced,so that the insulating fibre exhibits similar movement and discharge characteristics to the linear metal particles.At the same time,the fibres with dust absorbed on will result in an air gap breakdown voltage drop of approximately 25%-40%.What's more,if the discharge of fibres and the‘sand storm’movement of the dust coexist,it may directly lead to‘explosion’of the moving dust,causing severely ablation of the plate surfaces,and thereby the degree of destruction increases dramatically.Ruixue Liang Qi Hu Heng Liu Jian Wang Qingmin Li 2022High Voltage2022,7,2:0

8Study on ethylene/1-hexene copolymerization catalyzed by α-diimine nickel catalysts with different ligands--Dedicated to Professor Xiuwen Han on the occasion of her 80th birthday显示文摘The structure of polyolefin has an important influence on its performance and application.Ethylene/1-hexene copolymerization is one of the important ways to control the structure of the polyolefin.However,research on the ethylene/1-hexene copolymerization catalyzed by nickel complexes with different steric ligands remains to be refined.Here,three α-dimine nickel catalysts are used to study the ligand effect on catalytic performance in the ethylene/1-hexene copolymerization.Reaction activity,molecular weight,phase-transition temperature and branching density of the resultant copolymer are measured to evaluate the catalytic performance.The results indicate that the steric ligands could exert great effect on the copolymerization.As for the chemical valence of Ni species,detailed EPR demonstrate that the presence of excess xo-catalyst can reduce Ni(Ⅱ)to the lower valence and affect the catalytic performance.Yusheng Xing Haojie Yu Li Wang Nan Wang Lei Zhu Ruixue Liang Md Alim Uddin 2021Magnetic Resonance Letters2021,1,2:0
