| 1 | The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells.However,there are limited studies on the relationship between the variation and molecular basis of stone cells,lignin and cellulose content among different pear varieties.Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation.In our results,we measured the content of stone cells,lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell,lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell,lignin and cellulose content were detected,and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits,indicating that lignin is the main component of stone cell in pears.Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families,which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content.This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation. | Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu | 2021 | Horticultural Plant Journal2021,7,2: | 3 |

| 4 | Genomic selection of eight fruit traits in pear显示文摘Genomic selection (GS) has the potential to improve selection efficiency and shorten the breeding cycle in fruit tree breeding. In this study,we evaluated the effect of prediction methods, marker density and the training population (TP) size on pear GS for improving its performance and reducing cost. We evaluated GS under two scenarios:(1) five-fold cross-validation in an interspecific pear family;(2) independent validation. Based on the cross-validation scheme, the prediction accuracy (PA) of eight fruit traits varied between 0.33 (fruit core vertical diameter)and 0.65 (stone cell content). Except for single fruit weight, a slightly better prediction accuracy (PA) was observed for the five parametrical methods compared with the two non-parametrical methods. In our TP of 310 individuals, 2 000 single nucleotide polymorphism (SNP) markers were sufficient to make reasonably accurate predictions. PAs for different traits increased by 18.21%-46.98%when the TP size increased from 50to 100, but the increment was smaller (-4.13%-33.91%) when the TP size increased from 200 to 250. For independent validation, the PAs ranged from 0.11 to 0.45 using rrBLUP method. In summary, our results showed that the TP size and SNP numbers had a greater impact on the PA than prediction methods. | 6 | Phosphate-dependent regulation of vacuolar trafficking of OsSPX-MFSs is critical for maintaining intracellular phosphate homeostasis in rice显示文摘Vacuolar storage of inorganic phosphate(Pi)is essential for Pi homeostasis in plants.The SPX-MFS family proteins have been demonstrated to be vacuolar Pi transporters in many plant species.Transcriptional regulation of the predominant transporter among rice SPX-MFSs,OsSPX-MFS3,was only moderately suppressed by Pi starvation.Thus,post-transcriptional mechanisms were hypothesized to regulate the activity of OsSPX-MFS3.In this study,we found that the tonoplast localization of OsSPX-MFSs is inhibited under Pi-depleted conditions,resulting in their retention in the pre-vacuolar compartments(PVCs).A yeast two-hybrid screen identified that two SNARE proteins,OsSYP21 and OsSYP22,interact with the MFS domain of OsSPX-MFS3.Further genetic and cytological analyses indicate that OsSYP21 and OsSYP22 facilitate trafficking of OsSPX-MFS3 from PVCs to the tonoplast.Although a homozygous frameshift mutation in OsSYP22 appeared to be lethal,tonoplast localization of OsSPX-MFS3 was significantly inhibited in transgenic plants expressing a negative-dominant form of OsSYP22(OsSYP22-ND),resulting in reduced vacuolar Pi concentrations in OsSYP22-ND plants.Under Pi-depleted conditions,the interaction between OsSYP22 and OsSPX-MFS3 was disrupted,and this process depended on the presence of the SPX domain.Deleting the SPX domains of OsSPX-MFSs resulted in their tonoplast localization under both Pi-depleted and Pi-replete conditions.Complementation of the osspx-mfs1/2/3 triple mutants with the MFS domain or the SPX domain of OsSPX-MFS3 confirmed that the MFS and SPX domains are responsive to Pi transport activity and Pi-dependent regulation,respectively.These data indicated that the SPX domains of OsSPX-MFSs sense cellular Pi(InsP)levels and,under Pi-depleted conditions,inhibit the interaction between OsSPX-MFSs and OsSYP21/22 and subsequent trafficking of OsSPX-MFSs from PVCs to the tonoplast. | Runze Guo Qi Zhang Kun Qian Yinghui Ying Wenying Liao Lening Gan Chuanzao Mao Yong Wang James Whelan Huixia Shou | 2023 | Molecular Plant2023,16,8: | 0 |

| 9 | Protective performance test and safety risk evaluation of a powered air-purifying suit显示文摘The positive pressure biological protective suit is the highest degree of personal protective equipment, and its performance directly impacts the health and safety of the wearer. Protection factor is the most critical measure of the performance of a positive pressure bio-protective suit. This study establishes a dynamic detection system for the positive pressure bio-protective suit, to evaluate its safety risks, and explores methods to reduce these risks. A manikin was used to perform human actions, such as raising the arms, sitting, and walking, that were simulated at different levels. Integrity breaches of different shapes and sizes were made to different parts of the suit for evaluation. The aerosol concentration and the pressure of the suit were detected. Positive-pressure suits provide exceptionally good protection even in accident scenarios and provide a greater flexibility and more ergonomic benefits. However, sufficiently large (>3 cm) breaches caused a negative-pressure and an inward airflow during vigorous activities. The location of the breach site on the suit also had a significant effect on bio-aerosol leakage. Studies have shown that effective methods to avoid the risk of damage include increasing the air supply flow and performing gentle movements while wearing the suit. | 10 | Narrowband EUV Sc/Si Multilayer for the Solar Upper Transition Region Imager at 46.5 nm显示文摘The transition region is the key region between the lower solar atmosphere and the corona, which has been limitedly understood by human beings. Therefore, the Solar Upper Transition Region Imager(SUTRI) was proposed by Chinese scientists and launched in 2022 July. Right now, the first imaging observation of the upper transition region around 46.5 nm has been carried out by SUTRI. To ensure the spectral and temporal resolution of the SUTRI telescope, we have developed a narrowband Sc/Si multilayer. Based on the extreme ultraviolet(EUV)reflectivity measurements, the multilayer structure has been modified for ensuring the peak position of reflectivity was at 46.5 nm. Finally, the narrowband Sc/Si multilayer was successfully deposited on the hyperboloid primary mirror and secondary mirrors. The deviation of multilayer thickness uniformity was below than 1%, and the average EUV reflectivity at 46.1 nm was 27.8% with a near-normal incident angle of 5°.
