Erianin,a novel dibenzyl compound in Dendrobium extract, inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases, including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment. Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl, exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells, which was accompanied by ROS accumulation, lipid peroxidation, and GSH depletion. The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death, indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together, our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration, and erianin will hopefully serve as a prospective compound for lung cancer treatment. | Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 42
Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently, the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however, the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown. In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo, accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment. The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death, indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together, our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis, which will hopefully provide a prospective compound for bladder cancer treatment. | Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 23
Transcriptional activation and phosphorylation of OsCNGC9 confer enhanced chilling tolerance in rice显示文摘Low temperature is a major environmental factor that limits plant growth and productivity.Although transient elevation of cytoplasmic calcium has long been recognized as a critical signal for plant cold tolerance, the calcium channels responsible for this process have remained largely elusive. Here we report that OsCNGC9,a cyclic nucleotide-gated channel, positively regulates chilling tolerance by mediating cytoplasmic calcium elevation in rice(Oryza sativa). We showed that the loss-of-function mutant of OsCNGC9 is defective in cold-induced calcium influx and more sensitive to prolonged cold treatment, whereas OsCNGC9 overexpression confers enhanced cold tolerance.Mechanistically,we demonstrated that in response to chilling stress,OsSAPK8,a homolog of Arabidopsis thaliana OST1,phosphorylates and activates OsCNGC9 to trigger Ca2+influx.Moreover,we found that the transcription of OsCNGC9 is activated by a rice dehydration-responsive element-binding transcription factor,OsDREB1A.Taken together, our results suggest that OsCNGC9 enhances chilling tolerance in rice through regulating cold-induced calcium influx and cytoplasmic calcium elevation. | Jiachang Wang Yulong Ren Xi Liu Sheng Luo Xiao Zhang Xin Liu Qibing Lin Shanshan Zhu Hua Wan Yang Yang Yu Zhang Bin Lei Chunlei Zhou Tian Pan Yongfei Wang Mingming Wu Ruonan jing Yang Xu Meng Han Fuqing Wu Cailin Lei Xiuping Guo Zhijun Cheng Xiaoming Zheng Yihua Wang Zhigang Zhao Ling Jiang Xin Zhang Yong-Fei Wang Haiyang Wang Jianmin Wan | 2021 | Molecular Plant2021,14,2: | 10
Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin, has been reported that exerted its antitumor potential in a few cancer types. However, the effect and molecular mechanism of curcumenol in lung cancer are largely unknown. Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently, using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol, when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol, while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target, ferritin heavy chain 1(FTH1),a marker of ferroptosis. In conclusion, our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis, and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death. Therefore, our findings will hopefully provide a valuable drug for treating lung cancer patients. | Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui | 2022 | Bioactive Materials2022,7,7: | 8
An ATF24 peptide-functionalized β-elemene-nanostructured lipid carrier combined with cisplatin for bladder cancer treatment显示文摘Objective:In this study,we aimed to develop an amino-terminal fragment(ATF)peptide-targeted liposome carryingβ-elemene(ATF24-PEG-Lipo-β-E)for targeted delivery into urokinase plasminogen activator receptor-overexpressing bladder cancer cells combined with cisplatin(DDP)for bladder cancer treatment. Methods:The liposomes were prepared by ethanol injection and high-pressure microjet homogenization. The liposomes were characterized, and the drug content, entrapment efficiency, andin vitro release were studied. The targeting efficiency was investigated using confocal microscopy,ultra-fast liquid chromatography, and an orthotopic bladder cancer model. The effects of ATF24-PEG-Lipo-β-E combined with DDP on cell viability and proliferation were evaluated by a Cell Counting Kit-8(CCK-8)assay,a colony formation assay, and cell apoptosis and cell cycle analyses. The anticancer effects were evaluated in a KU-19-19 bladder cancer xenograft model.Results:ATF24-PEG-Lipo-β-E had small and uniform sizes(~79 nm),high drug loading capacity(~5.24 mg/mL),high entrapment efficiency(98.37±0.95%),and exhibited sustained drug release behavior.ATF24-PEG-Lipo-β-E had better targeting efficiency and higher cytotoxicity than polyethylene glycol(PEG)ylatedβ-elemene liposomes(PEG-Lipo-β-E).DDP,combined with ATF24-PEG-Lipo-β-E,exerted a synergistic effect on cellular apoptosis and cell arrest at the G2/M phase, and these effects were dependent on the caspase-dependent pathway and Cdc25C/Cdc2/cyclin B1 pathways.Furthermore, thein vivo antitumor activity showed that the targeted liposomes effectively inhibited the growth of tumors, using the combined strategy. Conclusions:The present study provided an effective strategy for the targeted delivery ofβ-elemene(β-E)to bladder cancer, and a combined strategy for bladder cancer treatment. | Bingtao Zhai Peng Chen Wengang Wang Shuiping Liu Jiao Feng Ting Duan Yu Xiang Ruonan Zhang Mingming Zhang Xuemeng Han Xiaying Chen Qiujie Li Guohua Li Ying Liu Xingxing Huang Wenzheng Zhang Ting Pan Lili Yan Ting Jin Tian Xie Xinbing Sui | 2020 | Cancer Biology & Medicine2020,17,3: | 5
PCDH17 increases the sensitivity of colorectal cancer to 5-fluorouracil treatment by inducing apoptosis and autophagic cell death显示文摘5-Fluorouracil(5-FU)is known as a first-line chemotherapeutic agent against colorectal cancer(CRC),but drug resistance occurs frequently and significantly limits its clinical success. Our previous study showed that the protocadherin 17(PCDH17)gene was frequently methylated and functioned as a tumor suppressor in CRC.However, the relationship between PCDH17 and 5-FU resistance in CRC remains unclear. Here,we revealed that PCDH17 was more highly expressed in 5-FU-sensitive CRC tissues than in 5-FU-resistant CRC tissues, and high expression of PCDH17 was correlated with high BECN1 expression. Moreover, this expression profile contributed to superior prognosis and increased survival in CRC patients.Restoring PCDH17 expression augmented the 5-FU sensitivity of CRC in vitro and in vivo by promoting apoptosis and autophagic cell death.Furthermore, autophagy played a dominant role in PCDH17-induced cell death,as an autophagy inhibitor blocked cell death to a greater extent than the pancaspase inhibitor Z-VAD-FMK.PCDH17 inhibition by siRNA decreased the autophagy response and 5-FU sensitivity.Mechanistically,we showed that c-Jun NH2-terminal kinase(JNK)activation was a key determinant in PCDH17-induced autophagy. The compound SP600125,an inhibitor of JNK,suppressed autophagy and 5-FU-induced cell death in PCDH17-reexpressing CRC cells.Taken together, our findings suggest for the first time that PCDH17 increases the sensitivity of CRC to 5-FU treatment by inducing apoptosis and JNK-dependent autophagic cell death.PCDH17 may be a potential prognostic marker for predicting 5-FU sensitivity in CRC patients. | Shuiping Liu Haoming Lin Da Wang Qiang Li Hong Luo Guoxiong Li Xiaohui Chen Yongqiang Li Peng Chen Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Mingming Zhang Xuemeng Han Qiujie Li Liuxi Chen Ying Liu Xiaying Chen Guohua Li Yu Xiang Ting Duan Jiao Feng Jianshu Lou Xingxing Huang Qin Zhang Ting Pan Lili Yan Ting Jin Wenzheng Zhang Lvjia Zhuo Yitian Sun Tian Xie Xinbing Sui | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 4
Asymmetric Ridge–Furrow and Film Cover Improves Plant Morphological Traits and Light Utilization in Rain-Fed Maize显示文摘Light is one of the most important natural resources for plant growth. Light interception(LI) and use efficiency(LUE) are often affected by the structure of canopy caused by growing pattern and agronomy managements. Agronomy practices, such as the ridge–furrow system and plastic film cover, might affect the leaf morphology and then light transmission within the canopy, thus change light extinction coefficient(k), and LI and LUE. The objective of this study is to quantify LI and LUE in rain-fed maize(Zea Mays L.), a major cropping system in Northeast China, under different combinations of ridge–furrow and film covering ratios. The tested ridge–furrow system(DRF:'double ridges and furrows') was asymmetric and alternated with wide ridge(0.70 m in width and 0.15 m in height),narrow furrow(0.10 m), narrow ridge(0.40 m in width and 0.20 m in height), and narrow furrow(0.10 m). Field experiments were conducted in 2013 and 2014 in Jilin Province, Northeast China. Four treatments were tested: no ridges and plastic film cover(control, NRF), ridges without film cover(DRF_0), ridges with 58% film cover(DRF_(58)),and ridges with 100% film cover(DRF_(100)). DRF_0 significantly increased LI by 9% compared with NRF, while film cover showed a marginal improvement. Specific leaf area in DRF experiments with film cover was significantly lower than in NRF, and leaf angle was 16% higher than in NRF, resulting in a 4% reduction in k. LUE of maize was not increased by DRF_0, but was significantly enhanced by covering film in other DRF experiments, especially by22% in DRF_(100). The increase of LUE by film cover was due to a greater biomass production and a lower assimilation portioning to vegetative organs, which caused a higher harvest index. The results could help farmers to optimize maize managements, especially in the region with decreased solar radiation under climate change. | Wanlin DONG Hang YU Lizhen ZHANG Ruonan WANG Qi WANG Qingwu XUE Zhihua PAN Zhigang SUN Xuebiao PAN | 2018 | Journal of Meteorological Research2018,32,5: | 3
Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants, are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. These results suggested that FLO18 is involved in 5′-end processing of nad5 messenger RNA and plays an important role in mitochondrial function and endosperm development. | Mingzhou Yu Mingming Wu Yulong Ren Yihua Wang Jingfang Li Cailin Lei Yinglun Sun Xiuhao Bao Hongming Wu Hang Yang Tian Pan Yongfei Wang Ruonan Jing Mengyuan Yan Houda Zhang Lei Zhao Zhichao Zhao Xin Zhang Xiuping Guo Zhijun Cheng Bing Yang Ling Jiang Jianmin Wan | 2021 | Journal of Integrative Plant Biology2021,63,5: | 2
From borophosphate to fluoroborophosphate:a rational design of fluorine-induced birefringence enhancement显示文摘The introduction of oxofluoride anion into anionic group assists to tune optical properties owing to the change of coordination, electronegativity, and according anionic framework. Here,we proposed a rational design of new compounds by fluorine-driven structure and optical property evolution.A new borophosphate Ba_(2)BP_(3)O_(11)with the monoclinic space group P2_(1/c) has been synthesized in the sealed system.Ba_(2)BP_(3)O_(11)exhibits a rare P-O-P bridge formation, which is the first example in alkaline-earth metal borophosphates.By further substituting[BO_(4)]^(3-)with[BO_(3)F]^(4-),the first alkaline-earth metal/lead fluoroborophosphates M_(2)BP_(2)O_(8)F(M=Ba and Pb)with the same space group were designed.Since the scissors effect of fluorine,in M_(2)BP_(2)O_(8)F(M=Ba and Pb),a BO_(3)F tetrahedron corner-sharing with three PO_(4)tetrahedra forms 1D chains along the b-axial direction, which are filled by MO_(n)(M=Ba/Pb,n=5,6,8)distorted polyhedra. The first principles calculation shows that the borophosphate Ba_(2)BP_(3)O_(11)has a birefringence about 0.013@1,064 nm,while the fluoroborophosphates M_(2)BP_(2)O_(8)F(M=Ba and Pb)have the values of 0.035 and 0.043@1,064 nm,respectively.Such an apparent enhancement in birefringence is derived from synergies of the oxyfluoride and cation. The introduction of fluorine-containing heteroanionic groups provides a feasible strategy to design novel promising optical materials. | Wenyao Zhang Zhizhong Zhang Wenqi Jin Ruonan Zhang Meng Cheng Zhihua Yang Shilie Pan | 2021 | Science China Chemistry2021,64,9: | 1

The inflammatory microenvironment and the urinary microbiome in the initiation and progression of bladder cancer显示文摘Accumulating evidence suggests that chronic inflammation may play a critical role in various malignancies, including bladder cancer. This hypothesis stems in part from inflammatory cells observed in the urethral microenvironment.Chronic inflammation may drive neoplastic transformation and the progression of bladder cancer by activating a series of in-flammatory molecules and signals.Recently,it has been shown that the microbiome also plays an important role in the development and progression of bladder cancer, which can be mediated through the stimulation of chronic inflammation. In effect, the urinary microbiome can play a role in establishing the inflammatory urethral microenvironment that may facilitate the development and progression of bladder cancer. In other words, chronic inflammation caused by the urinary microbiome may promote the initiation and progression of bladder cancer. Here,we provide a detailed and comprehensive account of the link between chronic inflammation, the microbiome and bladder cancer. Finally,we highlight that targeting the urinary microbiome might enable the development of strategies for bladder cancer prevention and personalized treatment. | Xingxing Huang Ting Pan Lili Yan Ting Jin Ruonan Zhang Bi Chen Jiao Feng Ting Duan Yu Xiang Mingming Zhang Xiaying Chen Zuyi Yang Wenzheng Zhang Xia Ding Tian Xie Xinbing Sui | 2021 | Genes & Diseases2021,8,6: | 1
TBBPA rather than its main derivatives enhanced growth of endometrial cancer via p53 ubiquitination显示文摘Tetrabromobisphenol A(TBBPA)and its derivatives widely exist in various environments and biota.Although the available data indicate that TBBPA exposure is highly associated with the increased incidence of endometrial cancer(EC),the effects of TBBPA and its main derivatives on EC proliferation and the involved crucial mechanism remain unclear. The present study aimed to investigate the effects of TBBPA and its derivatives under environmental concentrations on the proliferation of EC,and the crucial mechanism on the progression of EC caused by bromine fame retardants exposure. In this research,TBBPA and two of the most common TBBPA derivatives including TBBPA bis(2-hydroxyethyl ether)(TBBPABHEE)and TBBPA bis(dibromopropyl ether)(TBBPA-BDBPE)were screened for their capacities in induced EC proliferation and explored the related mechanism by in vitro cell culture model and in vivo mice model.Under environmental concentrations,TBBPA promoted the proliferation of EC,the main derivatives of TBBPA(TBBPA-BHEE and TBBPA-BDBPE)did not present the similar facilitation effects. The ubiquitination degradation of p53 was crucial in TBBPA induced EC proliferation, which resulted in the increase of downstream cell cycle and decrease of apoptosis. The further molecular docking result suggested the high affinity between TBBPA and ubiquitinated proteasome. This finding revealed the effects of TBBPA and its derivatives on EC proliferation, thus providing novel insights into the underlying mechanisms of TBBPA-caused EC. | Hongyan Yu Jingxu Zhang Jing Liu Ruonan Pan Yu Wang Xiaoting Jin Rifat Zubair Ahmed Yuxin Zheng | 2024 | Journal of Environmental Sciences2024,,3: | 0
