3Single-port transperitoneal robotic-assisted laparoscopic radical prostatectomy (spRALP): Initial experience显示文摘Objective:To assess the feasibility of single-port transperitoneal robotic-assisted laparoscopic radical prostatectomy(spRALP)and discuss its surgical technique.Methods:A 60-year-old male was admitted with an elevated prostate-specific antigen(PSA)level of 13.89 ng/mL and confirmed with prostate cancer on biopsy showing three of 22 positive cores with a Gleason score of 3þ4=7.Multiparametric magnetic resonance(MR)and bone scintigraphy showed organ-confined disease.spRALP was performed using da Vinci Si HD surgical system,with access of a quadri-channel laparoscopic port placed supraumbilically.Two drainage tubes were placed before wound closure.The surgical procedure was largely in consistence with a conventional robotic-assisted laparoscopic radical prostatectomy.Results:The surgery was successfully carried out with a duration of 152 min and an estimated blood loss of 100 mL.The patient was discharged on postoperative Day 4 after removal of both pelvic drainage tubes.Foley catheter was removed on postoperative Day 14.No major complications were encountered.Postoperative pathology showed a Gleason score of 3þ4=7 with no extraprostatic extension and negative surgical margins.Conclusion:Single-port robotic prostatectomy is feasible using the currently available robotic instruments in most Chinese robotic urological centers.Meticulous preoperative planning and careful patient selection are mandatory.Further studies concerning perioperative complications and pentafecta outcome compared with the conventional multi-port robotic prostatectomy is required.Yifan Chang Xiaojun Lu Qingliang Zhu Chuanliang Xu Yinghao Sun Shancheng Ren 2019Asian Journal of Urology2019,6,3:9
4Percent free prostate-specific antigen for prostate cancer diagnosis in Chinese men with a PSA of 4.0-10.0 ng/mL:Results from the Chinese Prostate Cancer Consortium显示文摘Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years.Rui Chen Liping Xie Xiaobing Cai Yiran Huang Liqun Zhou Lulin Ma Xu Gao Chuanliang Xu Shancheng Ren Pengfei Shao Danfeng Xu Kexin Xu Zhangqun Ye Chunxiao Liu Dingwei Ye Li Lu Qiang Fu Jianquan Hou Jianlin Yuan Dalin He Tie Zhou Fubo Wang Biming He Yinghao Sun 2015Asian Journal of Urology2015,2,2:8

7Genome-wide association studies on prostate cancer:the end or the beginning?显示文摘Prostate cancer(PCa)is the second most frequently di-agnosed malignancy in men.Ge nome-wide association st udies(GWAS)has been highly successful in discover-ing susceptibility loci for prostate cancer.Currently,more than twenty GWAS have identified more than fifty com-mon variants associated with susceptibility with PCa.Yet with the increase in loci,voices from the scientifi c society are calling for more.In this review,we summarize current fi ndings,discuss the common problems troubling current studies and shed light upon possible breakthroughs in the future.GWAS is the beginning of something wonder-ful.Although we are quite near the end of the beginning,post-GWAS studies are just taking off and future studies are needed extensively.It is believed that in the future GWAS information will be helpful to build a comprehen-sive system intergraded with PCa prevention,diagnosis,molecular classifi cation,personalized therapy.Rui Chen Shancheng Ren Yinghao Sun 2013Protein & Cell2013,4,9:2
8Targeting signaling pathways in prostate cancer:mechanisms and clinical trials显示文摘Prostate cancer(PCa)affects millions of men globally.Due to advances in understanding genomic landscapes and biological functions,the treatment of PCa continues to improve.Recently,various new classes of agents,which include next-generation androgen receptor(AR)signaling inhibitors(abiraterone,enzalutamide,apalutamide,and darolutamide),bone-targeting agents(radium-223 chloride,zoledronic acid),and poly(ADP-ribose)polymerase(PARP)inhibitors(olaparib,rucaparib,and talazoparib)have been developed to treat PCa.Agents targeting other signaling pathways,including cyclin-dependent kinase(CDK)4/6,Ak strain transforming(AKT),wingless-type protein(WNT),and epigenetic marks,have successively entered clinical trials.Furthermore,prostate-specific membrane antigen(PSMA)targeting agents such as 177Lu-PSMA-617 are promising theranostics that could improve both diagnostic accuracy and therapeutic efficacy.Advanced clinical studies with immune checkpoint inhibitors(ICIs)have shown limited benefits in PCa,whereas subgroups of PCa with mismatch repair(MMR)or CDK12 inactivation may benefit from ICIs treatment.In this review,we summarized the targeted agents of PCa in clinical trials and their underlying mechanisms,and further discussed their limitations and future directions.Yundong He Weidong Xu Yu-Tian Xiao Haojie Huang Di Gu Shancheng Ren 2022Signal Transduction and Targeted Therapy2022,7,7:2
9Developments in stability and passivation strategies for black phosphorus显示文摘Black phosphorus(BP),a promising two-dimensional layer material,has attracted increasing attention due to its high carrier mobility,thickness-dependent tunable bandgap,in-plane anisotropy,and other advantageous characteristics.Because of these excellent characteristics,BP has been considered for applications in optics,electronics,optoelectronics,sensors,and energy storage.However,early studies found that BP has high chemical activity due to the lone pair electrons of P atoms on the surface and edges,resulting in rapid degradation under ambient conditions and limiting many applications.Recently,these thorny issues have been alleviated through superior physical and chemical passivation techniques,and passivated BP can be used in various devices under ambient and water conditions with excellent performance over a long period.This review,highlights the critical problems addressed in solving the serious instability of BP in a harsh environment by effective passivation technology.These unique strategies can provide more researchers with a fundamental study of the fascinating properties of BP.Finally,we found that passivated BP not only showed good stability under ambient conditions but also exhibited excellent performance compared with the original BP.Therefore,it is anticipated that this overview can contribute to the application of BP.Haizeng Song Han Wu Tianqi Ren Shancheng Yan Tianhong Chen Yi Shi 2021Nano Research2021,14,12:1

11Glycolytic potential enhanced by blockade of pyruvate influx into mitochondria sensitizes prostate cancer to detection and radiotherapy显示文摘Objective:This study aimed to evaluate the effects of mitochondrial pyruvate carrier(MPC)blockade on the sensitivity of detection and radiotherapy of prostate cancer(PCa).Methods:We investigated glycolysis reprogramming and MPC changes in patients with PCa by using metabolic profiling,RNASeq,and tissue microarrays.Transient blockade of pyruvate influx into mitochondria was observed in cellular studies to detect its different effects on prostate carcinoma cells and benign prostate cells.Xenograft mouse models were injected with an MPC inhibitor to evaluate the sensitivity of 18F-fluorodeoxyglucose positron emission tomography with computed tomography and radiotherapy of PCa.Furthermore,the molecular mechanism of this different effect of transient blockage towards benign prostate cells and prostate cancer cells was studied in vitro.Results:MPC was elevated in PCa tissue compared with benign prostate tissue,but decreased during cancer progression.The transient blockade increased PCa cell proliferation while decreasing benign prostate cell proliferation,thus increasing the sensitivity of PCa cells to 18F-PET/CT(SUVavg,P=0.016;SUVmax,P=0.03)and radiotherapy(P<0.01).This differential effect of MPC on PCa and benign prostate cells was dependent on regulation by a VDAC1-MPC-mitochondrial homeostasis-glycolysis pathway.Conclusions:Blockade of pyruvate influx into mitochondria increased glycolysis levels in PCa but not in non-carcinoma prostate tissue.This transient blockage sensitized PCa to both detection and radiotherapy,thus indicating that glycolytic potential is a novel mechanism underlying PCa progression.The change in the mitochondrial pyruvate influx caused by transient MPC blockade provides a critical target for PCa diagnosis and treatment.Huan Xu Junyi Chen Zhi Cao Xi Chen Caihong Huang Jin Ji Yalong Xu Junfeng Jiang Yue Wang Guowang Xu Lina Zhou Jingyi He Xuedong Wei Jason Boyang Wu Zhong Wang Shancheng Ren Fubo Wang 2022Cancer Biology & Medicine2022,19,9:1
12Comparative risk of acute kidney injury among cancer patients treated with immune checkpoint inhibitors显示文摘With the development and introduction of immune checkpoint inhibitors(ICIs)in cancer patients,immune-related side effects have increasingly attracted attention.However,the risks of immune-related renal toxicity are poorly characterized.In this study,we performed a network meta-analysis(NMA)of ICI-related randomized clinical trials(RCTs)to elucidate the comparative risk of acute kidney injury(AKI)in cancer patients receiving different ICIs.We also sought to identify other factors potentially affecting the risk of AKI.PubMed and EMBASE were searched for peer-reviewed trial reports published between January 2000 and May 2021.Eligible studies were RCTs studying ICIs in cancer patients and reporting AKI data.We performed a frequentist NMA to evaluate the risk ratios for grade 1-5 and grade 3-5 AKI between the treatment groups.We also assessed the absolute incidence of AKI in the ICI-containing arm using traditional direct meta-analysis.Once significant heterogeneity was detected in a traditional direct meta-analysis,multivariable meta-regression analysis was applied to identify factors that significantly affected the absolute incidence of AKI.A total of 85 RCTs were included in this study.In the NMA for the risk of grade 1-5 and 3-5 AKI,ipilimumab showed a significantly higher risk than avelumab and durvalumab,whereas 1 mg/kg nivolumab plus 3 mg/kg ipilimumab(N1I3)showed a significantly higher risk than other groups.In terms of treatment ranking,durvalumab±low-dose tremelimumab and avelumab were consistently among the top three safest treatments for grade 1-5 or 3-5 AKI,whereas N1I3,ipilimumab and tremelimumab were consistently among the top three treatments with the highest risk for grade 1-5 or 3-5 AKI.Compared with other cancers,renal cell carcinoma and urothelial carcinoma showed a significantly higher risk of AKI.The incidence of AKI was significantly higher with ICI+chemotherapy than with ICI monotherapy.In this NMA involving largescale up-to-date ICI trials,we demonstrated the comparative safety of existing ICI drugs for grade 1-5 and grade 3-5 AKI.Based on data from the ICI arms of these trials,we also revealed several potential risk factors for immune-related AKI,including tumor type and treatment paradigm.Fei Liu ZixianWang Xiaofan Li Zhen Zhang Yue Yang Junquan Chen Dinghua Chen Lingling Wu Xiangyu Liu Sujun Han Fangming Wang Wasilijiang Wahafu Yibo Gao Shancheng Ren Nianzeng Xing Guangyan Cai Xiangmei Chen 2023Cancer Communications2023,43,2:1
13Neoadjuvant radiohormonal therapy for oligo-metastatic prostate cancer:safety and efficacy outcomes from an open-label,dose-escalation,single-center,phase Ⅰ/Ⅱ clinical trial显示文摘To evaluate the safety and efficacy of neoadjuvant radiohormonal therapy for oligometastatic prostate cancer(OMPC),we conducted a 3+3 dose escalation,prospective,phase Ⅰ/Ⅱ,single-arm clinical trial(CHiCTR1900025743),in which long-term neoadjuvant androgen deprivation was adopted 1 month before radiotherapy,comprising intensity modulated radiotherapy to the pelvis,and stereotactic body radiation therapy to all extra-pelvic bone metastases for 4‒7 weeks,at 39.6,45,50.4,and 54 Gy.Robotic-assisted radical prostatectomy was performed after 5‒14 weeks.The primary outcome was treatment-related toxicities and adverse events;secondary outcomes were radiological treatment response,positive surgical margin(pSM),postoperative prostate-specific antigen(PSA),pathological down-grading and tumor regression grade,and survival parameters.Twelve patients were recruited from March 2019 to February 2020,aging 66.2 years in average(range,52‒80).Median baseline PSA was 62.0 ng/mL.All underwent RARP successfully without open conversions.Ten patients recorded pathological tumor down-staging(83.3%),and 5(41.7%)with cN1 recorded negative regional lymph nodes on final pathology.66.7%(8/12)recorded tumor regression grading(TRG)–Ⅰ and 25%(3/12)recorded TRG-Ⅱ.Median follow-up was 16.5 months.Mean radiological progression-free survival(RPFS)was 21.3 months,with 2-year RPFS of 83.3%.In all,neoadjuvant radiohormonal therapy is well tolerated for oligometastatic prostate cancer.Yifan Chang Xianzhi Zhao Yutian Xiao Shi Yan Weidong Xu Ye Wang Huojun Zhang Shancheng Ren 2023Frontiers of Medicine2023,17,2:0
14Cell-free DNA in the management of prostate cancer:Current status and future prospective显示文摘Objective:With the escalating prevalence of prostate cancer(PCa)in China,there is an urgent demand for novel diagnostic and therapeutic approaches.Extensive investigations have been conducted on the clinical implementation of circulating free DNA(cfDNA)in PCa.This review aims to provide a comprehensive overview of the present state of cfDNA as a biomarker for PCa and to examine its merits and obstacles for future clinical utilization.Methods:Relevant peer-reviewed manuscripts on cfDNA as a PCa marker were evaluated by PubMed search(2010-2022)to evaluate the roles of cfDNA in PCa diagnosis,prognosis,and prediction,respectively.Results:cfDNA is primarily released from cells undergoing necrosis and apoptosis,allowing for non-invasive insight into the genomic,transcriptomic,and epigenomic alterations within various PCa disease states.Next-generation sequencing,among other detection methods,enables the assessment of cfDNA abundance,mutation status,fragment characteristics,and epigenetic modifications.Multidimensional analysis based on cfDNA can facilitate early detection of PCa,risk stratification,and treatment monitoring.However,standardization of cfDNA detection methods is still required to expedite its clinical application.Conclusion:cfDNA provides a non-invasive,rapid,and repeatable means of acquiring multidimensional information from PCa patients,which can aid in guiding clinical decisions and enhancing patient management.Overcoming the application barriers of cfDNA necessitates increased data sharing and international collaboration.Wei He Yutian Xiao Shi Yan asheng Zhu Shancheng Ren 2023Asian Journal of Urology2023,10,3:0
15Exploring the potential mechanisms of impairment on genitourinary system associated with coronavirus disease 2019 infection:Bioinformatics and molecular simulation analyses显示文摘Objective:The novel coronavirus(severe acute respiratory syndrome coronavirus 2)has been spreading worldwide since December 2019,posing a serious danger to human health and socioeconomic development.A large number of clinical trials have revealed that coronavirus disease 2019(COVID-19)results in multi-organ damage including the urogenital system.This study aimed to explore the potential mechanisms of genitourinary damage associated with COVID-19 infection through bioinformatics and molecular simulation analysis.Methods:We used multiple publicly available databases to explore the expression patterns of angiotensin-converting enzyme 2(ACE2),transmembrane serine protease 2(TMPRSS2),and CD147 in major organs in the healthy and disease-specific populations,particularly the genitourinary organs.Single-cell RNA sequencing was used to analyze the cell-specific expression patterns of ACE2,TMPRSS2,CD147,cytokine receptors,and cytokine interacting proteins in genitourinary organs,such as the bladder,kidney,prostate,and testis.Additionally,gene set enrichmentanalysis was used to investigate the relationship between testosterone levels and COVID-19 vulnerability in patients with prostate cancer.Results:The results revealed that ACE2,TMPRSS2,and CD147 were highly expressed in normal urogenital organs.Then,they were also highly expressed in multiple tumors and chronic kidney diseases.Additionally,ACE2,TMPRSS2,and CD147 were significantly expressed in a range of cells in urogenital organs according to single-cell RNA sequencing.Cytokine receptors and cytokine interacting proteins,especially CCL2,JUN,and TIMP1,were commonly highly expressed in urogenital organs.Finally,gene set enrichment analysis results showed that high testosterone levels in prostate cancer patients were significantly related to the JAK-STAT signaling pathway and the Toll-like receptor signaling pathway which were associated with COVID-19.Conclusion:Our study provides new insights into the potential mechanisms of severe acute respiratory syndrome coronavirus 2 damage to urogenital organs from multiple perspectives,which may draw the attention of urologists to COVID-19 and contribute to the development of targeted drugs.Kai Zhao Dong Zhang Xinchi Xu Shangqian Wang Zhanpeng Liu Xiaohan Ren Xu Zhang Zhongwen Lu Shancheng Ren Chao Qin 2023Asian Journal of Urology2023,10,3:0

17SARS-CoV-2 effects in the genitourinary system and prospects of sex hormone therapy显示文摘Object:Corona virus disease 2019(COVID-19)is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which leads to acute respiratory infection symptoms.SARS-CoV-2 infection is not always limited to the respiratory tract,and renal infection and dysfunction have been shown to be specific risk factors for death.In addition,COVID-19 has a higher incidence,severity and mortality in men than women.This disparity is due to biological rather than comorbid or behavioral sex differences.Because the male reproductive system is unique,the function of sex hormones in COVID-19 infection may explain the differences between males and females.Understanding these factors will provide appropriate prevention measures and adequate triage strategies and guide the drug discovery process.Methods:An electronic search was completed in PubMed,ARXIV,MEDRXIV and BIORXIV.The most relevant articles were systematically reviewed.In addition,single cell RNA sequencing analysis of tissue samples from human cell landscape was conducted.Results:The influence of SARS-CoV-2 on the urogenital system,the possibility of urinary tract transmission and the functions of sex hormones were discussed in this review.Conclusion:Corona viruses can invade the genitourinary system,causing urological symptoms.Identifying the potential genitourinary organ impairments and protecting them from damage are necessary.Since sex hormones have potential as specific drugs,the gonadal hormones substitution therapy should be considered in both sexes in the COVID-19 pandemic.Kuangzheng Liu Xinglin Chen Xiaohan Ren Yuqing Wu Shancheng Ren Chao Qin 2021Asian Journal of Urology2021,8,3:0

19The older the better:The characteristic of localized prostate cancer in Chinese men显示文摘Objective:To assess the clinicopathological features and overall survival between two groups of Chinese patients older or younger than 70 years after retropubic radical prostatectomy.Methods:From January 2001 to February 2010,390 patients receive dretropubic radical prostatectomy.After excluding 89 patients with adjuvant or neoadjuvant hormonal therapy or radiotherapy,a total of 301 patients were included in this study.We arbitrarily divided these patients into younger age group(<70 years,140 cases,46.5%)and older age group(≥70 years,161 cases,53.5%).The differences in serum prostate specific antigen(PSA),Gleason score,clinical tumor stage,and biochemical-free survival were analyzed between the two groups.Results:There were not significant differences between the two groups in high Gleason score rate and clinical tumor stage.However,older patients had significantly lower biochemical recurrence rate than those of younger patients,and had significantly higher PSA levels.Multivariate analysis showed that older age,PSA level and clinical tumor stage were significantly associated with biochemical recurrence free survival.Conclusion:In Chinese men,older age(≥70 years)is associated with better outcome.If the physical condition permits,older age alone should not exclude patients from radical prostatectomy.Haifeng Wang Xu Gao Ziyu Fang Xin Lu Yan Wang Chunfei Ma Zhenkai Shi Bo Yang Shancheng Ren Chuanliang Xu Yinghao Sun 2015Asian Journal of Urology2015,2,3:0
