1A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice,rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines.Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge,even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases.Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu 2022Cell Research2022,32,3:5
2A highly potent and stable pan-coronavirus fusion inhibitor as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases显示文摘The development of broad-spectrum antivirals against human coronaviruses(HCoVs)is critical to combat the current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and its variants,as well as future outbreaks of emerging CoVs.We have previously identified a polyethylene glycol-conjugated(PEGylated)lipopeptide,EK1C4,with potent pan-CoV fusion inhibitory activity.However,PEG linkers in peptide or protein drugs may reduce stability or induce anti-PEG antibodies in vivo.Therefore,we herein report the design and synthesis of a series of dePEGylated lipopeptide-based pan-CoV fusion inhibitors featuring the replacement of the PEG linker with amino acids in the heptad repeat 2 C-terminal fragment(HR2-CF)of HCoV-OC43.Among these lipopeptides,EKL1C showed the most potent inhibitory activity against infection by SARS-CoV-2 and its spike(S)mutants,as well as other HCoVs and some bat SARS-related coronaviruses(SARSr-CoVs)tested.The dePEGylated lipopeptide EKL1C exhibited significantly stronger resistance to proteolytic enzymes,better metabolic stability in mouse serum,higher thermostability than the PEGylated lipopeptide EK1C4,suggesting that EKL1C could be further developed as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases.Jie Zhou Wei Xu Zezhong Liu Chao Wang Shuai Xia Qiaoshuai Lan Yanxing Cai Shan Su Jing Pu Lixiao Xing Youhua Xie Lu Lu Shibo Jiang Qian Wang 2022Acta Pharmaceutica Sinica B2022,12,4:5

4Particle motion behavior during gasiication of foam carrier in lost foam-squeeze casting of 20%TiC/ZG270-500 composites显示文摘The particle motion behavior affects the distribution of particles in the metal matrix and finally determines the mechanical properties of the particle reinforced metal-matrix composites. To obtain a uniform distribution of TiC particles and excellent strengthening effect in 20%TiC/ZG270-500 composites fabricated by lost foam-squeeze casting(LFSC), the particle motion behavior in the gas gap and the conditions of the particles getting into the molten steel were investigated. The results show that the airflow velocity(v1) and TiC particle motion velocity(uP) change little with the pouring temperature(TP), increase with an increase in metal filling velocity(vP), ratios of cross sections of in-gate/orifice(AP/A0) and orifice/mould cavity(A0/A1), but the increase trend of u P is more intense. The airflow pressure(P1) changes little with TP and A0/A1, but increases with the increasing of vP and AP/A0. Besides, there is a critical velocity(+|3yLGcosθ/ρpdp|1/2) for the particles getting into the molten steel. The higher the particle motion velocity, the easier the particles get into the molten steel and the more uniform the distribution of the particles in the steel matrix. When TP = 1,873 K, vP = 30 mm·s-1, AP/A0 =10 and A0/A1 = 0.02 in this study, the biggest TiC particle motion velocity(20.59 m·s-1) can be gained, and the steel matrix with the most uniformly distributed TiC particles and fine grains are obtained.Bin Shibo Xing Shuming Guo Lijun Bao Peiwei 2013China Foundry2013,10,6:1

6Alternating current stimulation promotes neurite outgrowth and plasticity in neurons through activation of the PI3K/AKT signaling pathway显示文摘As a commonly used physical intervention,electrical stimulation(ES)has been demonstrated to be effective in the treatment of central nervous system disorders.Currently,researchers are studying the effects of electrical stimu-lation on individual neurons and neural networks,which are dependent on factors such as stimulation intensity,duration,location,and neuronal properties.However,the exact mechanism of action of electrical stimulation remains unclear.In some cases,repeated or prolonged electrical stimulation can lead to changes in the morphology or function of the neuron.In this study,immunofluorescence staining and Sholl analysis are used to assess changes in the neurite number and axon length to determine the optimal pattern and stimulation parameters of ES for neurons.Neuronal death and plasticity are detected by TUNEL staining and microelectrode array assays,respec-tively.mRNA sequencing and bioinformatics analysis are applied to predict the key targets of the action of ES on neurons,and the identified targets are validated by western blot analysis and qRT-PCR.The effects of alternating current stimulation(ACS)on neurons are more significant than those of direct current stimulation(DCS),and the optimal parameters are 3μA and 20 min.ACS stimulation significantly increases the number of neurites,the length of axons and the spontaneous electrical activity of neurons,significantly elevates the expression of growth-asso-ciated protein-43(GAP-43)without significant changes in the expression of neurotrophic factors.Furthermore,application of PI3K/AKT-specific inhibitors significantly abolishes the beneficial effects of ACS on neurons,con-firming that the PI3K/AKT pathway is an important potential signaling pathway in the action of ACS.Hao Zhong Cong Xing Mi Zhou Zeyu Jia Song Liu Shibo Zhu Bo Li Hongjiang Yang Hongpeng Ma Liyue Wang Rusen Zhu Zhigang Qu Guangzhi Ning 2023Acta Biochimica et Biophysica Sinica2023,55,11:0
7Variability and correlation analysis of physical and mechanical indexes of loess in western Liaoning显示文摘Based on the method of probability and statistics,the authors analyzed the physical and mechanical properties of loess in specified area,and discussed its main indexes,the coefficient of variation,and the correlation among the physical and mechanical indexes.Regression equations are built among those indexes,the results show that the variability of mechanical indexes is higher than that of the physical indexes,and the physical indexes have better correlation than the relation between physical and mechanical indexes.The conclusions of this study would contribute to further research about loess in western Liaoning.YAO Kang WANG Changming XING Yudong LI Shibo YANG Lina 2012Global Geology2012,15,4:0
