1Breast cancer development and progression:Risk factors,cancer stem cells,signaling pathways,genomics,and molecular pathogenesis显示文摘As the most commonly occurring cancer in women worldwide,breast cancer poses a formidable public health challenge on a global scale.Breast cancer consists of a group of biologically and molecularly heterogeneous diseases originated from the breast.While the risk factors associated with this cancer varies with respect to other cancers,genetic predisposition,most notably mutations in BRCA1 or BRCA2 gene,is an important causative factor for this malignancy.Breast cancers can begin in different areas of the breast,such as the ducts,the lobules,or the tissue in between.Within the large group of diverse breast carcinomas,there are various denoted types of breast cancer based on their invasiveness relative to the primary tumor sites.It is important to distinguish between the various subtypes because they have different prognoses and treatment implications.As there are remarkable parallels between normal development and breast cancer progression at the molecular level,it has been postulated that breast cancer may be derived from mammary cancer stem cells.Normal breast development and mammary stem cells are regulated by several signaling pathways,such as estrogen receptors(ERs),HER2,and Wnt/b-catenin signaling pathways,which control stem cell proliferation,cell death,cell differentiation,and cell motility.Furthermore,emerging evidence indicates that epigenetic regulations and noncoding RNAs may play important roles in breast cancer development and may contribute to the heterogeneity and metastatic aspects of breast cancer,especially for triple-negative breast cancer.This review provides a comprehensive survey of the molecular,cellular and genetic aspects of breast cancer.Yixiao Feng Mia Spezia Shifeng Huang Chengfu Yuan Zongyue Zeng Linghuan Zhang Xiaojuan Ji Wei Liu Bo Huang Wenping Luo Bo Liu Yan Lei Scott Du Akhila Vuppalapati Hue H.Luu Rex C.Haydon Tong-Chuan He Guosheng Ren 2018Genes & Diseases2018,5,2:24
2Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
3Simultaneous quantification of ginsenoside Rg1 and its metabolites by HPLC–MS/MS:Rg1 excretion in rat bile, urine and feces显示文摘Ginsenoside Rg1(Rg1), the major effective component of ginseng, has been shown to have multiple bioactivities, but low oral bioavailability. The aim of this study was to develop a simple, sensitive and rapid high performance liquid chromatography–tandem mass spectrometry(LC–MS/MS) method, which could be used to validate and quantify the concentrations of Rg1 and its metabolites in Sprague-Dawley rat bile,urine, and feces after oral administration(25 mg/kg). Calibration curves offered satisfactory linearity(r40.995)within the determined ranges. Both intra-day and inter-day variances were less than 15%, and the accuracy was within 80–120%. The excretion recoveries of Rg1, ginsenoside Rh1(Rh1), and protopanaxatriol(Ppt) in bile,urine, and feces combined were all greater than 70%. The fecal excretion recoveries of Rg1, Rh1, and Ppt were40.11%, 22.19%, and 22.88%, respectively, whereas 6.88% of Rg1 and 0.09% of Rh1 were excreted in bile.Urinary excretion accounted for only 0.04% of Rg1. 5Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
6Efficacy of Lidan Tang on high-fat-diet induced gallstone in mice and possible mechanism显示文摘OBJECTIVE:To investigate efficacy of Lidan Tang(LDT)on gallstone induced by high fat diet in mice,and to study its underlying mechanism.METHODS:Mice were fed with high fat diet every day and treated with LDT(9.01 times of human clinic dosage).Mice were randomly divided into 6 groups as control group,gallstone model group(high-fat diet),positive control ursodeoxycholic acid(UDCA)group(80 mg·kg^-1·d^-1,i.g.),LDT low dose group(6 kg/d,i.g.),LDT middle dose group(12 kg/d,i.g.),and LDT high dose group(24 kg/d,i.g.).The whole experiment was lasted for 4 weeks.The levels of ALT,AST,LDH,CHO,HDL-C and LDL-C in serum were measured,the pathological sections were observed by hematoxylin-eosin staining,the activities of antioxidant enzymes were measured by kits,and the proteins related to oxidative stress and lipid transport were detected by Western blot analysis.RESULTS:LDT could significantly reduce the contents of ALT and AST in serum and improve the pathological tissue of liver.LDT could significantly reduce the content of MDA and LPO,and increase the level of GSH and GSH-PX in liver tissue.The data of Western blot showed that LDT had antioxidant effect promoting Keap1/Nrf2 pathway and regulated the process of lipid transport,which was statistically significant.In addition,LDT treatment inhibited the expression of ATP-binding cassette transports ABCG5/8 in liver,and reduced cholesterol transport from the hepatocytes to the gallbladder.CONCLUSION:LDT has protective effect on gallstones induced by high fat diet in mice,which might be based on the protective effect on liver,including enhancing the antioxidant capacity of liver and reducing the production of lipid peroxides.Gao Yan Liu Caipin Li Juntong Zhai Yingying Lin Meiyu Wu Qinglin Chu Shifeng Zhang Zhao Li Jianping Zhou Xin Li Yueting Chen Naihong 2020Journal of Traditional Chinese Medicine2020,40,4:4
7The development of a sensitive fluorescent protein-based transcript reporter for high throughput screening of negative modulators of lncRNAs显示文摘While the human genome is pervasively transcribed,<2%of the human genome is transcribed into protein-coding mRNAs,leaving most of the transcripts as noncoding RNAs,such as microRNAs and long-noncoding RNAs(lncRNAs),which are critical components of epigenetic regulation.lncRNAs are emerging as critical regulators of gene expression and genomic stability.However,it remains largely unknown about how lncRNAs are regulated.Here,we develop a highly sensitive and dynamic reporter that allows us to identify and/or monitor negative modulators of lncRNA transcript levels in a high throughput fashion.Specifically,we engineer a fluorescent fusion protein by fusing three copies of the PEST destruction domain of mouse ornithine decarboxylase(MODC)to the C-terminal end of the codon-optimized bilirubin-inducible fluorescent protein,designated as dBiFP,and show that the dBiFP protein is highly destabilized,compared with the commonly-used eGFP protein.We further demonstrate that the dBiFP signal is effectively down-regulated when the dBiFP and mouse lncRNA H19 chimeric transcript is silenced by mouse H19-specific siRNAs.Therefore,our results strongly suggest that the dBiFP fusion protein may serve as a sensitive and dynamic transcript reporter to monitor the inhibition of lncRNAs by microRNAs,synthetic regulatory RNA molecules,RNA binding proteins,and/or small molecule inhibitors so that novel and efficacious inhibitors targeting the epigenetic circuit can be discovered to treat human diseases such as cancer and other chronic disorders.Zongyue Zeng Bo Huang Shifeng Huang Ruyi Zhang Shujuan Yan Xinyi Yu Yi Shu Chen Zhao Jiayan Lei Wenwen Zhang Chao Yang Ke Wu Ying Wu Liping An Xiaojuan Ji Cheng Gong Chengfu Yuan Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Rex C.Haydon Hue H.Luu Lan Zhou Russell R.Reid Tong-Chuan He Xingye Wu 2018Genes & Diseases2018,5,1:4

9LA-ICP-MS zircon U-Pb age of rhyolitic lithic-crystal tuffs in Erdaohezi lead-zinc deposit,Inner Mongolia显示文摘Erdaohezi lead-zinc deposit belongs to the Derbugan metallogenic belt lying on the northwestern Hailaer-Genhe Mesozoic volcanic basin,located on the western slope of the Da Hinggan Mountains. The deposit is considered as one of the hypabyssal low-temprature hydrothermal lead-zinc deposits associated with volcanism. In order to lay the foundation on studying its diagenesis and mineralization ages,the detailed studies were carried out by dating the host rocks( i. e. rhyolitic lithic-crystal tuffs) using zircon LA-ICP-MS U-Pb method.The dating results show three groups of ages. The first group is the captured zircons( the weighted mean ^(206)Pb /^(238) U age as 175. 6 ± 2. 3 Ma,MSWD = 0. 70,n = 3). The second group can be regarded as the rockforming age( the weighted mean ^(206)Pb /^(238) U age as 165. 3 ± 1. 9 Ma,MSWD = 2. 40,n = 14). The third group should represent the late stage of the magmatic evolution( the weighted mean ^(206)Pb /^(238) U age as 161. 0 ± 3. 1Ma,MSWD = 0. 86,n = 4). According to the ages and the crystal form or CL image characteristics of zircons,it is determined that the diagenesis occurred in the late Middle Jurassic. Based on the regional geology and geochronological research,the acidic pyroclastic rocks are space accompaniment and time connection with the Tamulangou Formation intermediate-mafic volcanic rocks. Both of them constitute the host rocks of the deposit together. 10The synthesis, deprotection and properties of poly(γ-benzyl-L-glutamate)显示文摘With dicy- clohexylamine as initiator, the influence of monomer concentration, and reaction temperature and time on the polymerization of BLG NCA was examined. Three reagents were used for the deprotection of benzyl groups in PBLG, including hydrobromic acid/acetic acid (33 wt.%), NaOH aqueous solution and trimethylsilyl iodide (TMSI). Through examining the molecular weight of PLGA obtained using different deprotection methods, it was revealed that TMSI could minimize chain cleavage in the process of deprotection and retain the degree of polymerization. The biocompatibilities of PBLG obtained using different initiators were evaluated by a live/dead assay against L929 fibroblast cells. The in vitro cytotoxicities of PLGA obtained using different deprotecting agents were evaluated by a methyl thiazolyl tetrazolium assay. 11In-Situ Chemosynthesis of ZnO Nanoparticles to Endow Wood with Antibacterial and UV-Resistance Properties显示文摘Hybrid wood materials have attracted considerable attention because they have combined advantages of both wood and inorganic compounds. This work investigated the microstructural morphology, thermal stability, ultraviolet(UV) stability, and antibacterial property of composites made from wood/ZnO hybrid materials through a facile in-situ chemosynthesis methods. The X-ray diffraction(XRD) and thermogravimetric analysis(TGA) results indicated that the synthesized ZnO particles had an average grain size of about 10.8 nm. The scanning electron microscopy(SEM) observations showed that ZnO nanoflowers self-assembled with nanosheets were presented in wood cell lumens and increased with increasing Zn^(2+)concentrations. ZnO nanoparticles were also generated in the wood cell wall, which was confirmed by the results of energy-dispersive spectroscopy(EDS). The TGA tests also indicated that the thermal stability of wood/ZnO hybrid materials was improved after the formation of ZnO inorganic particles. 12Differential response of multiple zebrafish hepatic F-box protein genes to 17α-ethinylestradiol treatment显示文摘Estrogens are accumulating in environment and their effects on a variety of reproductive processes and tumorigenesis were reported by previous study, but the mechanism of estrogen promoting neoplasia was still not clear. F-box protein (FBP) is the component of E3 ubiquitin ligase which takes part in a variety of key biological processes. In this study, using mature male zebrafish, which are more sensitive to estrogen treatment, we examined influence of 17α-ethinylestradiol (EE2) exposure on the expression of a series of hepatic FBP genes, which take part in a variety of biological processes, including tumorigenesis. The influence of EE2 on the expression of hepatic mRNA concentrations of FBP genes were quantified based on the expression of the optimal internal control gene in male zebrafish after 7-day exposure to EE2, from a low-dose concentration (1 ng/L) to environmentally relevant concentrations (10, 100 ng/L). Our results showed that EE2 exposure reduced the expression of fbxl14a, fbxl14b, fbxo25 and β-TRCP2b, but enchanced the expression of skp2. While the alterations in fbxl2, fbxw7, fbxo9, β-TRCP2a, fbxl18 and fbxo45 mRNA levels were not observed after EE2 exposure. Thus, our results showed that the expression of hepatic FBP genes exhibited differentially in male zebrafish exposed EE2. 15Mechanical properties and microstructure characteristics of lattice-surfaced PEEK cage fabricated by high-temperature laser powder bed fusion显示文摘Porous structure design on the contact surface is crucial to promote the osseointegration of the intervertebral cage while preventing subsidence and displacement.However,the stress response will undergo significant changes for the current random porous cages,which can directly affect the mechanical properties and long-term usability.Here,this paper proposed a newly designed polyetheretherketone(PEEK)cage with the triply periodic minimal surface(TPMS)-structured lattice surfaces to provide tailored 3 D microporosity and studied the mechanical performance,stress/strain responses,and microstructure changes in depth.The lattice-surfaced PEEK cage mainly exhibits a multiple-point-plane stress transfer mechanism.The compression modulus and elastic limit can be adjusted by controlling the area of the Diamond TPMS surface while the energy absorption efficiency remains stable.The microstructure of high-strength PEEK is featured by the radial pattern morphology.Meanwhile,the double-stranded orthorhombic phase is more ordered,and the benzene plane subunit and lattice volume become more expanded.Peng Chen Jin Su Haoze Wang Lei Yang Haosong Cai Maoyuan Li Zhaoqing Li Jie Liu Shifeng Wen Yan Zhou Chunze Yan Yusheng Shi 2022Journal of Materials Science & Technology2022,,30:1
16Crystallization of Al-based Amorphous Alloys in Good Conductivity Solution显示文摘The corrosion-induced crystallization of Al94 exNixGd6(x=6 and 10, in at.%) metallic glasses as well as phase separation, oxidation and cracking in good conductivity solution has been investigated by various techniques.The transmission electronic microscopy(TEM) result reveals that crystalline intermetallics and oxides present on the electrochemically thinned hole edge, and the phase separation occurs in the matrix of the as-spun ribbons with the circumferential speed Rcof 29.3 m/s. In addition, the bending and cracking of the samples occur after corrosion.
