| 1 | KLF7 promotes preadipocyte proliferation via activation of the Akt signaling pathway by Cis-regulating CDKN3显示文摘Krüppel-like transcription factor 7(KLF7)promotes preadipocyte proliferation;however,its target gene in this process has not yet been identified.Using KLF7 ChIP-seq analysis,we previously showed that a KLF7-binding peak is present upstream of the cyclin-dependent kinase inhibitor 3 gene(CDKN3)in chicken preadipocytes.In the present study,we identify CDKN3 as a target gene of KLF7 that mediates the effects of KLF7 on preadipocyte proliferation.Furthermore,5′-truncating mutation analysis shows that the minimal promoter is located between nt–160 and nt–7(relative to the translation initiation codon ATG)of CDKN3.KLF7 overexpression increases CDKN3 promoter activity in the DF-1 and immortalized chicken preadipocyte(ICP1)cell lines.Deletion of the putative binding site of KLF7 abolishes the promotive effect of KLF7 overexpression on CDKN3 promoter activity.Moreover,CDKN3 knockdown and overexpression assays reveal that CDKN3 enhances ICP1 cell proliferation.Flow cytometry analysis shows that CDKN3 accelerates the G1/S transition.Furthermore,we find that KLF7 promotes ICP1 cell proliferation via Akt phosphorylation by regulating CDKN3.Taken together,our results suggest that KLF7 promotes preadipocyte proliferation by activating the Akt signaling pathway by cis-regulating CDKN3,thus driving the G1/S transition. | Ziqiu Jia Zhao Jin Shuli Shao Hu Xu Wen Li Mahmood Khan Weiyu Wang Weiwei Zhang Yingning Sun | 2022 | Acta Biochimica et Biophysica Sinica2022,54,10: | 2 |
| 2 | Consensus on pediatric epilepsy Che up surgery for young children:an investigation by the China Association Against Epilepsy task force on epilepsy surgery显示文摘Researchers have widely acknowledged the therapeutic value of epilepsy surgery for drug-resistant epilepsy.None-theless,there is a substantial gap in the surgical treatment for appropriate candidates owing to several factors,particularly in the population of young children.To standardize the protocols of preoperative evaluation and sur-gery of young children for epilepsy surgery,the China Association Against Epilepsy has appointed an expert task force to standardize the protocols of preoperative evaluation and surgery in pediatric epilepsy patients.It adopted the modifed Delphi method and performed two rounds of surveys through an anonymous inquiry among 75 experts from four subgroups including pediatric neurologists,epileptologists,pediatric epilepsy surgeons,and functional neurosurgeons.The survey contents contained:(1)the participants,comprising children aged≤6 years;(2)adopted DRE definition proposed by the International League Against Epilepsy in 2010;and(3)investigated epilepsy surgery,principally referring to curative epilepsy surgeries.The neuromodulation therapies were excluded because of the differences in treatment mechanisms from the above-mentioned surgeries.According to the Delphi process,a con-sensus was achieved for most aspects by incorporating two rounds of surveys including preoperative assessment,sur-gical strategies and techniques,and perioperative and long-term postoperative management,despite controversial opinions on certain items.We hope the results of this consensus will improve the level of surgical treatment and man-agement of intractable epilepsy in young children. | Lixin Cai Kai Zhang Wenjing Zhou Xiaoqiu Shao Yuguang Guan Tao Yu Ye Wu Shuhua Chen Rui Zhao Shuli Liang Xun Wu Guoming Luan Yuwu Jiang Jianguo Zhang Xiaoyan Liu | 2023 | Acta Epileptologica2023,6,3: | 1 |
| 3 | Age effect of deafening on stereotyped song maintenance in adult male bengalese finches Lonchura striata domestica显示文摘Birdsong is a complex learned vocal behavior that relies on auditory experience for development.However,it appears that among different species of close-ended songbirds,there are some variations in the necessity of auditory feedback for maintaining stereotyped adult song.In zebra finches,the deterioration of adult songs following deafness depends on the birds' age.It is unknown whether this age effect is a general rule in other avian species as well.Therefore,we chose Bengalese finches,whose songs show more complexity and have much heavier dependency on auditory feedback than that of zebra finches,to compare the degree of song degradation after hearing loss in old(over 18 months old)and young adult birds(5-6 months old).We found that both syllable sequence and syllable phonology were much less severely affected by deafening in old adults than that in young ones.Moreover,young adults almost lost their capability to sing trills over 6 months following deafening,while old birds continued to sing plenty of trills and trilled syllables after the same period of deafening.Our results suggest that age plays an important role in affecting the dependency of adult song maintenance on auditory feedback in Bengalese finches.Furthermore,the age dependency may be a general phenomenon in different species of close-ended songbirds. | Yingyu SUN Rui WANG Shuli SHAO Shaoju ZENG Mingxue ZUO | 2009 | Current Zoology2009,55,3: | 0 |
| 4 | PU.1 interacts with KLF7 to suppress differentiation and promote proliferation in chicken preadipocytes显示文摘Krüppel-like factor 7(KLF7)is a negative regulator of preadipocyte differentiation.Our previous KLF7 ChIP-seq analysis showed that the binding motif of PU.1 was found among the KLF7 binding peaks,indicating that an interaction between KLF7 and PU.1 at preadipocyte gene promoters and other regulatory elements might be common.Here,Co-IP and FRET assays are used to confirm that PU.1 can directly bind to KLF7 and enhance the transcription activity of cyclin-dependent kinase inhibitor 3(CDKN3),which is a downstream target gene of KLF7.We show that the PU.1 expression level is decreased during preadipocyte differentiation.Furthermore,PU.1 overexpression and knockdown experiments reveal that PU.1 negatively regulates chicken preadipocyte differentiation,as evidenced by appropriate changes in lipid droplet accumulation and altered expressions of PPARγ,FAS,and PLIN.In addition,PU.1 overexpression promotes preadipocyte proliferation,while knockdown of PU.1 inhibits preadipocyte proliferation.We further demonstrate that PU.1 inhibits differentiation and promotes proliferation in preadipocytes,in part by directly interacting with KLF7. | Ming Tan Hu Xu Jinwei Li Ziqiu Jia Xin Zhang Shuli Shao Weiwei Zhang Weiyu Wang Yingning Sun | 2023 | Acta Biochimica et Biophysica Sinica2023,55,1: | 0 |
