1Cardiac autonomic nerve fiber regeneration in chronic heart failure Do Akt gene-transduced mesenchymal stem cells promote repair?显示文摘BACKGROUND: Transplantation of Akt-over-expressing mesenchymal stem cells (Akt-MSCs) has been shown to repair infarcted myocardium and improve cardiac function. However, little is known about the therapeutic effects of Akt-MSCs on cardiac autonomic neuropathy in chronic heart failure (CHF). OBJECTIVE: The present study used adriamycin-induced CHF rat models to observe the effect of Akt-MSCs on cardiac autonomic nervous regeneration and the factors mediating this effect. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Central Laboratory of Basic Medical College, China Medical University, between September 2008 and April 2009. MATERIALS: Rabbit anti-choline acetyltransferase (ChAT), growth associated protein-43 (GAP-43), synaptophysin (SYN) polyclonal antibodies and the secondary antibody (goat anti-rabbit IgG) were purchased from Boster, China. Cat-A-Kit assay system was provided by Amersham, USA. METHODS: (1) Adult rat MSCs were isolated and cultured for the preparation of Akt-MSCs. (2) Forty male Wistar rats were intramyocardially administered adriamycin at 2 mg/kg over 3 days for a total of five times and once a week for additional five times thereafter to establish CHF models. At 2 weeks after final adriamycin treatment, 34 successful CHF rat models were randomized to three groups: Akt-MSCs (n = 11), simple MSCs (s-MSCs, n =11), and control (n = 12). Each group was intravenously administered Akt-MSCs (2×106 cells in 100 μL PBS), s-MSCs (2×106 cells in 100 μL PBS) or equal volume of phosphate buffered saline, once a day for a total of three times. MAIN OUTCOME MEASURES: At 4 weeks after final adriamycin treatment, myocardial norepi- nephrine (NE) content was detected using a Cat-A-Kit assay system. Myocardial ChAT, SYN and GAP-43 were performed by immunohistochemistry and Western blot analysis. Prior to, 2 and 4 weeks after adriamycin treatment, echocardiographic examination was performed and left ventricular ejection fraction (LVEF) was determined. RESULTS: Myocardial NE content, as well as SYN-positive and GAP-43-positive nerve fiber density and expression, and LVEF, was the greatest in the Akt-MSCs group, followed by the s-MSCs group, and lastly the control group (P < 0.05 or P < 0.01). ChAT expression was similar between Akt-MSCs and s-MSCs groups, but it was higher compared with the control group (P < 0.05). NE contents were negatively correlated to LVEF (r = -0.64, P = 0.015). 2Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4Associations between individual and environmental factors and habitual physical activity among older Chinese adults:A social–ecological perspective显示文摘Purpose: To examine, within a social–ecological framework, associations between multifaceted individual and environmental factors and habitual physical activity(HPA) among older Chinese adults.Methods: Through a mix of qualitative and quantitative methods, a survey instrument assessing various factors underlying 3 social–ecological dimensions of intrapersonal, interpersonal, and community and environmental resources was developed. Using a cross-sectional design, older adults(n = 1580, aged 67 ± 7 years) recruited from 10 communities in Shandong province completed the social–ecological survey of HPA. Data were analyzed using Partial Least Squares Path Modeling.Results: Factors related to intrapersonal(medical knowledge, motivation, physical function, sport skills, socioeconomic status, and education),interpersonal(social support, social activity, and social norms), and community and physical environmental resources(safety, capacity, availability of and access to physical activity facilities) were found to be significantly associated with older adults' participation in HPA.Conclusion: The findings provide an initial validation of a social–ecological approach to the study of HPA in China, suggesting that strategies aimed at promoting physical activity in older adults should address multiple levels of factors that may contribute to the likelihood of older Chinese adults being physically active.Xiangren Yi Zachary Pope Zan Gao Shumei Wang Fang Pan Jingpeng Yan Meng Liu Peipei Wu Jingjing Xu Rui Wang 2016Journal of Sport and Health Science2016,5,3:11
5Review of the fabrication and application of porous materials from silicon-rich industrial solid waste显示文摘Porous materials have promise as sound insulation, heat barrier, vibration attenuation, and catalysts. Most industrial solid wastes, such as tailings, coal gangue, and fly ash are rich in silicon. Additionally, a high silicon content waste is a potential raw material for the syn- thesis of silicon-based, multi-porous materials such as zeolites, mesoporous silica, glass-ceramics, and geopolymer foams. Representative sil- icon-rich industrial solid wastes (SRISWs) are the focus of this mini review of the processing and application of porous silicon materials with respect to the physical and chemical properties of the SRISW. The transformation methods of preparing porous materials from SRISWs are summarized, and their research status in micro-, meso-, and macro-scale porous materials are described. Possible problems in the application of SRISWs and in the preparation of functional porous materials are analyzed, and their development prospects are discussed. 6High quality reference genome of drumstick tree(Moringa oleifera Lam.), a potential perennial crop显示文摘The drumstick tree(Moringa oleifera Lam.) is a perennial crop that has gained popularity in certain developing countries for its high-nutrition content and adaptability to arid and semi-arid environments. Here we report a high-quality draft genome sequence of M. oleifera. This assembly represents 91.78% of the estimated genome size and contains 19,465 protein-coding genes. Comparative genomic analysis between M. oleifera and related woody plant genomes helps clarify the general evolution of this species, while the identification of several species-specific gene families and positively selected genes in M. oleifera may help identify genes related to M. oleifera's high protein content, fast-growth, heat and stress tolerance. 7Medical management of gastric cancer:A 2014 update显示文摘Gastric cancer represents a serious health problem on a global scale. It is the second leading cause of cancer-related death worldwide. Novel therapeutic targets are desperately needed because the meager improvement in the cure rate of about 10% realized by adjunctive treatments to surgery is unacceptable as > 50% patients with localized gastric cancer succumb to their disease. Either postoperative chemoradiotherapy(United States),pre-and post-operative chemotherapy(Europe),and adjuvant chemotherapy after a D2 resection(Asia) can all be regarded as standards of care in the localized gastric cancer management. In metastatic disease the addition of trastuzumab to chemotherapy is standard of care in Her2 positive disease. 10Long-Term Cognitive Improvement After Benfotiamine Administration in Patients with Alzheimer's Disease显示文摘To date,we still lack disease-modifying therapies for Alzheimer's disease(AD).Here,we report that long-term administration of benfotiamine improved the cognitive ability of patients with AD.Five patients with mild to moderate AD received oral benfotiamine(300 mg daily) over 18 months.All patients were examined by positron emission tomography with Pittsburgh compound B(PiB-PET) and exhibited positive imaging with pamyloid deposition,and three received PiB-PET imaging at follow-up.The five patients exhibited cognitive improvement as assayed by the Mini-Mental Status Examination(MMSE) with an average increase of 3.2 points at month18 of benfotiamine administration.The three patients who received follow-up PiB-PET had a 36.7%increase in the average standardized uptake value ratio in the brain compared with that in the first scan.Importantly,the MMSE scores of these three had an average increase of 3 points during the same period.Benfotiamine significantly improved the cognitive abilities of mild to moderate AD patients independently of brain amyloid accumulation.Our study provides new insight to the development of diseasemodifying therapy.Xiaoli Pan Zhichun Chen Guoqiang Fei Shumei Pan Weiqi Bao Shuhua Ren Yihui Guan Chunjiu Zhong 2016Neuroscience Bulletin2016,32,6:9
11Two possible hydrothermal vents in the northern Okinawa Trough显示文摘As the Okinawa Trough is a back-arc basin in early spreading, modern submarine hydrothermal activity and minerallization have many characteristics which have aroused wide attention. Up to now, three well-known hydro-thermal venting areas are all located in the middle part of the trough. During two cruise investigations to map and sample the seafloor, numbers of Calyptogena sp. shells were dredged at two sites in the northern trough with comparatively thicker crust and numerous submarine volcanoes. Based on the fact that Calyptogena sp. is only observed around the hydrothermal vents and lives on hydrothermal activities, it is predicted that there is the possibility of modern hydrothermal activities in the northern part of the trough. In this note, the shell is carefully characterized and the sample locations with possible hydrothermal activity are given. 12Bacterial Community and Function of Biological Activated Carbon Filter in Drinking Water Treatment显示文摘Objective It aims to investigate the changes in composition and structure of bacterial communities de-veloping on biological activated carbon (BAC) particles, and the bacterial functions. Method A pilot plant had been in service for 180 days, aiming to develop bacterial communities on acti-vated carbon naturally. After 180 days of operation, the bacterial communities were determined by dena-turing gradient gel electrophoresis (DGGE) analyses of PCR-amplified 16S rRNA genes. The study on community composition and the phylogenetic relationships of the organisms was complemented by a se-quence analysis of cloned PCR products from 16S rRNA genes. Gas chromatorgaphy-mass (GC-MS) mea-surement was used to determine organic chemical composition of inflow and outflow water on the 300th day. TOC and NH 4 + -N were also tested in this experiment. Results It showed that the stable bacterial structure did not develop on BAC particles until the 9th month during running time of the BAC filter. The communities were finally dominated by Pseudomonas sp., Ba-cillus sp., Nitrospira sp., and an uncultured bacterium. Stable bacterial communities played an important role in removal of NH 4 + -N and total organic carbon (TOC). Results from gas chromatorgaphy-mass (GC-MS) showed that 36 kinds of chemicals in feed water were eliminated, and concentrations of 5 kinds of chemicals decreased. These chemicals served as nutrients for the dominant bacteria. Conclusion The findings from the study suggested that the stability of microbial structure was beneficial for improving NH 4 + -N and TOC removal efficiencies. 15Basin assemblages and tectonic evolution in the Bohai Gulfand its neighbors显示文摘According to multi- disciplinary research advances in recent years, three blocks which named Jiaoliao - Bohai block, central North China block and Ordos block are divided. The differences of the tectonic association, types, styles and features in the space, and evolution from Archaean to Cenozoic in the time, between the former two blocks are discussed. Tectonic features and evolution in the four tectonic stages of Precambrian, Indo- China, Yanshanian and Cenozoic are emphasized especially here. All of them described above have inherit and neogenic property. The temporal-spatial change and interaction among basin assemblages in the above four stages are analyzed. 17Simultaneous desulfurization and denitrification of flue gas by pre-ozonation combined with ammonia absorption显示文摘A process of simultaneous desulfurization and denitrification of flue gas was conducted in this study.The flue gas containing 200 mg·m^-3NO,1000-4000 mg·m^-3SO(2,)3%-9%O(2)and 10%-20%CO(2)was first oxidized b(y)O3 and then absorbed by ammonia in a bubbling reactor.Increasing the ammonia concentration or the SO2 content in flue gas can promote the absorption of NOx and extend the effective absorption time.On the contrary,both increasing the absorbent temperature or the O(2)content shorten the effective absorption time of NO((x.))The change of solution pH had substantial influence on NOx absorption.In the presence of CO(2)the NOx removal efficiency reached 89.2%when the absorbent temperature was raised to 60℃and the effective absorption time can be maintained for 8 h,which attribute to the buffering effect in the absorbent.Besides,both the addition of Na(2)S2 O3 and urea can promote the NOx removal efficiency when the absorbent temperature is 25℃and the addition of Na(2)S2 O3 had achieved better results.The advantage of adding Na(2)S2 O3 became less evident at higher absorbent temperature and coexistence of CO(2.)In all experiments,SO(2)removal efficiency was always above 99%,and it was basically not affected by the above factors.Baowei Wang Shumei Yao Yeping Peng 2020Chinese Journal of Chemical Engineering2020,28,9:5
18Effective methods for isolation and purification of extracellular vesicles from plants显示文摘Plant extracellular vesicles(EVs)play critical roles in the cross-kingdom trafficking of molecules from hosts to interacting microbes,most notably in plant defense responses.However,the isolation of pure,intact EVs from plants remains challenging.A variety of methods have been utilized to isolate plant EVs from apoplastic washing fluid(AWF).Here,we compare published plant EV isolation methods,and provide our recommended method for the isolation and purification of plant EVs.This method includes a detailed protocol for clean AWF collection from Arabidopsis thaliana leaves,followed by EV isolation via differential centrifugation.To further separate and purify specific subclasses of EVs from heterogeneous vesicle populations,density gradient ultracentrifugation and immunoaffinity capture are then utilized.We found that immunoaffinity capture is the most precise method for specific EV subclass isolation when suitable specific EV biomarkers and their corresponding antibodies are available.Overall,this study provides a guide for the selection and optimization of EV isolation methods for desired downstream applications.Yifan Huang Shumei Wang Qiang Cai Hailing Jin 2021Journal of Integrative Plant Biology2021,63,12:5
