1RNAi:antiviral therapy against dengue virus显示文摘Dengue virus infection has become a global threat affecting around 100 countries in the world.Currently,there is no licensed antiviral agent available against dengue.Thus,there is a strong need to develop therapeutic strategies that can tackle this life threatening disease.RNA interference is an important and effective gene silencing process which degrades targeted RNA by a sequence specific process.Several studies have been conducted during the last decade to evaluate the efficiency of siRNA in inhibiting dengue virus replication.This review summarizes siRNAs as a therapeutic approach against dengue virus serotypes and concludes that siRNAs against virus and host genes can be next generation treatment of dengue virus infection.Sobia Idrees Usman A Ashfaq 2013Asian Pacific Journal of Tropical Biomedicine2013,3,3:5

3Hepatitis C virus genotype 3a infection and hepatocellular carcinoma: Pakistan experience显示文摘AIM: To assess the association between chronic hepatitis C virus (HCV) infection and hepatocellular carcinoma (HCC) in Pakistan, and the genotype distribution among these HCC patients.METHODS: One hundred and sixty-one subjects with HCC were included in this study. Liver biopsy was performed on 145 of the patients; sixteen were excluded because they failed to fulfill the inclusion criteria. Qualitative polymerase chain reaction (PCR) was performed for hepatitis B virus and HCV. Samples positive for HCV RNA were genotyped using genotype-specif ic PCR and conf irmed by HCV 5' noncoding region sequencing analysis. RESULTS: Chronic HCV infection was identified a major risk factor (63.44% of tested HCC patients) forthe development of HCC. The time from HCV infection to appearance of cancer was 10-50 years. In the HCC patient population, broader distributions of genotypes were present with genotype 3a as the predominant genotype. Using the type-specific genotyping method, we found HCV genotype 3a in 40.96%, 3b in 15.66%, 1a in 9.63%, and 1b in 2.40% of HCC tissue samples. About 28% of cases were found with mixed genotypes. Two cases were unable to be genotyped because of low viral load. Sixty-six percent of treated patients with cirrhosis had an end of treatment response, but unfortunately they relapsed quickly when the treatment was discontin-ued, and HCC developed during a median 3.8 years. 4Medicinal plants against hepatitis C virus显示文摘Hepatitis C virus(HCV)is a global health concern which is responsible for most of the liver diseases.Currently,there is no vaccine available for prevention of HCV infection due to the high degree of strain variation.The current standard of care is a combination of pegylated interferonαwith ribavirin and boceprevir/telaprevir.This treatment was partially effective and had significant side effects.Hence,there is a need to develop new antiviral agents that interfere with different stages of the HCV life cycle.Recent advances in the understanding of both the cellular and molecular mechanisms of HCV replication have provided the basis for novel therapeutic strategies.Several hundred plant species and their phyto-constituents have been isolated for screening against HCV,and some have been shown to have great medicinal value in preventing and/or ameliorating viral diseases in pre-clinical and clinical trials.This review summarizes medicinal plants and their phytochemicals which inhibit different stages of HCV life cycle and discuss their potential use in HCV therapy.Usman A Ashfaq Sobia Idrees 2014World Journal of Gastroenterology2014,20,11:0
5Discovery and design of cyclic peptides as dengue virus inhibitors through structure-based molecular docking显示文摘Objective:To find potential peptide inhibitors against the NS2B/NS3 protease of DENV which in turn,can inhibit the viral replication inside host cell.Methods:Cyclic peptides were designed having combination of positively charged amino acids using ChemSketch software and were converted to 3D structures.DENV NS3 protein structure was retrieved from Protein Data Bank(PDB)using PDB Id:2FOM.DENV NS3 and cylic peptides were docked using MOE software after structural optimization.Results:Through molecular docking it was revealed that most of the peptides bound deeply in the binding pocket of DENV NS2B/NS3 protease an had interactions with catalytic triad.Peptide 2 successfully blocked the catalytic triad of NS2B/NS3 protease.Peptide 1,,4 and 6 also had potential interactions with active residues of the NS2B/NS3 protease while all other peptides were in close contact with the active sites of NS2B/NS3 protease thus,these peptides can serve as a potential drug candidate to stop viral replication.Conclusions:Thus.it can be concluded from the study that these peptides could serve as important inhibitors to inhibit the viral replication and need further in-vitro investigations to confirm their efficacy.Sobia Idrees Usman Ali Ashfaq 2014Asian Pacific Journal of Tropical Medicine2014,7,7:0
