1Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13
2Monitoring Maize Lodging Grades via Unmanned Aerial Vehicle Multispectral Image显示文摘Lodging is one of the main factors affecting the quality and yield of crops.Timely and accurate determination of crop lodging grade is of great significance for the quantitative and objective evaluation of yield losses.The purpose of this study was to analyze the monitoring ability of a multispectral image obtained by an unmanned aerial vehicle(UAV)for determination of the maize lodging grade.A multispectral Parrot Sequoia camera is specially designed for agricultural applications and provides new information that is useful in agricultural decision-making.Indeed,a near-infrared image which cannot be seen with the naked eye can be used to make a highly precise diagnosis of the vegetation condition.The images obtained constitute a highly effective tool for analyzing plant health.Maize samples with different lodging grades were obtained by visual interpretation,and the spectral reflectance,texture feature parameters,and vegetation indices of the training samples were extracted.Different feature transformations were performed,texture features and vegetation indices were combined,and various feature images were classified by maximum likelihood classification(MLC)to extract four lodging grades.Classification accuracy was evaluated using a confusion matrix based on the verification samples,and the features suitable for monitoring the maize lodging grade were screened.The results showed that compared with a multispectral image,the principal components,texture features,and combination of texture features and vegetation indices were improved by varying degrees.The overall accuracy of the combination of texture features and vegetation indices is 86.61%,and the Kappa coefficient is 0.8327,which is higher than that of other features.Therefore,the classification result based on the feature combinations of the UAV multispectral image is useful for monitoring of maize lodging grades.Qian Sun Lin Sun Meiyan Shu Xiaohe Gu Guijun Yang Longfei Zhou 2019Plant Phenomics2019,1,1:5
3LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development,stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus.Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage,and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation.In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity,and its loss leads to nucleolar phase separation defects,ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci,and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling,translationally inert and anabolically inactive state,which is a part of previously unappreciated 2C-like transcriptional program.These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development.Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang 2022Protein & Cell2022,13,7:4

8Analysis on Static Pressure Bearing Seal Characteristics of Guide Sleeve of Electro-hydraulic Servo Cylinder显示文摘As a typical bionic walking robot, hydraulic quadruped robot has attracted much attention because of its high mobility, strong load capacity and steady motion. The electro-hydraulic servo cylinder, as its power actuator, requires low friction, good lateral load resistance and high speed motion. The electro-hydraulic servo cylinder hydrostatic bearing seal guide sleeve is taken as the research object in this paper. By using Fluent software to analyze and contrast the film characteristics of rectangular and I-shaped oil chamber of hydrostatic bearing seal guide sleeve, the relationship between piston rod moving speed, eccentricity, oil film carrying capacity, friction force and leakage volume, as well as the relationship between oil feed flow and oil film bearing capacity, friction force, inlet pressure and leakage volume were analyzed. 9Digital Twin-Assisted Knowledge Distillation Framework for Heterogeneous Federated Learning显示文摘In this paper,to deal with the heterogeneity in federated learning(FL)systems,a knowledge distillation(KD)driven training framework for FL is proposed,where each user can select its neural network model on demand and distill knowledge from a big teacher model using its own private dataset.To overcome the challenge of train the big teacher model in resource limited user devices,the digital twin(DT)is exploit in the way that the teacher model can be trained at DT located in the server with enough computing resources.Then,during model distillation,each user can update the parameters of its model at either the physical entity or the digital agent.The joint problem of model selection and training offloading and resource allocation for users is formulated as a mixed integer programming(MIP)problem.To solve the problem,Q-learning and optimization are jointly used,where Q-learning selects models for users and determines whether to train locally or on the server,and optimization is used to allocate resources for users based on the output of Q-learning.Simulation results show the proposed DT-assisted KD framework and joint optimization method can significantly improve the average accuracy of users while reducing the total delay.Xiucheng Wang Nan Cheng Longfei Ma Ruijin Sun Rong Chai Ning Lu 2023China Communications2023,20,2:0
10Microtremor-based analysis of the dynamic response characteristics of a site containing grouped earth fissures显示文摘In this study,the Beibu earth fissure site in the northeastern part of Weihe Basin,which contains four nearly parallel earth fissures,was studied.A long straight microtremor measuring line,containing 49 measuring points across four earth fissures,was established to investigate the dynamic response of this site using Fourier spectrum,response acceleration spectrum,Arias intensity,and HVSR analyses.The main results are as follows:(1)The fundamental frequencies of 44 measuring points obtained from HVSR analysis are concentrated within 1.67 Hz-2.25 Hz,and the existence of the earth fissures has little effect on the fundamental frequency changes.(2)There is an amplification effect near a single earth fissure.The dynamic responses are large at the measuring points near the earth fissure,and the values decrease with increasing distance from the earth fissure.In areas between two adjacent earth fissures,these values decrease and are even lower than those in sites without amplification effects.(3)In this earth fissure site,the general area(or less affected area)and affected areas were delineated based on the amplification effect.In engineering applications,construction design should avoid these affected areas and existing structures should be reinforced to satisfy the seismic fortification requirements.Deng Yahong Xuan You Mu Huandong Chang Jiang Cao Ge He Nainan Zhao Xunchang Sun Longfei 2023Earthquake Engineering and Engineering Vibration2023,22,3:0
11Upregulation of Zip14 correlates with induction of endoplasmic reticulum stress(ERS)in hypertrophied hearts of Dahl saltsensitive rats显示文摘Zinc is a trace element involved in maintaining cellular structure and function.Although zinc is associated with left ventricular hypertrophy(LVH),there have been few reports on this association.This study aimed to evaluate the correlation between Zip14 and expression of endoplasmic reticulum stress(ERS)associated molecules in hypertrophied hearts of rats.Dahl salt-sensitive rats were fed a high salt diet to establish a left ventricular hypertrophy(LVH)rat model.RT-PCR was used to determine Zip14,activating transcription factor(ATF4),ATF6,x-box-binding protein 1(xBP1),C/EBP homologous protein(CHOP),immunoglobulin-binding protein(BiP)mRNA expression.Western blotting was used to evaluate Zip14,BiP,CHOP,GAPDH expression.Zinc levels were measured by Inductively Coupled Plasma Optical Emission Spectroscopy.The results indicated that compared with the Control group,Zip14 mRNA and protein expression in LVH rat hearts were markedly increased(P<0.01).Zinc content in rat heart tissue was significantly increased in the LVH group compared with the Control group(P<0.05).ATF4,ATF6,xBP1 mRNA expressions were increased in LVH rat hearts compared with Control hearts(P<0.001).Compared with the Control group,CHOP and BiP mRNA and protein expression were markedly increased in LVH rat hearts(P<0.05,P<0.01).Linear regression models showed that Zip14 mRNA expressions were positively correlated with zinc concentration,ATF4 and ATF6 mRNA expressions in Control hearts(P=0.0005,P=0.0052,P=0.0026,respectively)and LVH rat hearts(P<0.0001,P=0.0119,P=0.0033,respectively).In conclusion,upregulation of Zip14 in LVH rat hearts correlated with zinc accumulation and induction of ERS.JINYONG HUANG TIANMING TENG BO BIAN YUCHEN XUE LONGFEI HUANG ZHELONG XU QING YANG YUEMIN SUN 2022BIOCELL2022,46,3:0
12Study on Oil Cavity Number of Hydrostatic Guide Sleeve of Electro-hydraulic Servo Cylinder显示文摘The electro-hydraulic servo drive hydraulic cylinder has many unique advantages, such as fast response, high load stiffness, high control power, strong anti-eccentric load ability and so on, so it has been widely used in industrial control. Based on the guide sleeve of hydrostatic seal of hydraulic cylinder, the reasonable number of oil chamber of guide sleeve is studied in this paper. ICEM CFD software and FLUENT simulation software are used to calculate and analyze the number of different oil chambers of guide sleeve of hydrostatic seal. The temperature field of piston rod with different moving speed, different initial pressure of oil chamber and oil film under different number of oil chambers is analyzed. 13Single-cell profiling reveals Müller glia coordinate retinal intercellular communication during light/dark adaptation via thyroid hormone signaling显示文摘Light adaptation enables the vertebrate visual system to operate over a wide range of ambient illumination.Regulation of phototransduction in photoreceptors is considered a major mechanism underlying light adaptation.However,various types of neurons and glial cells exist in the retina,and whether and how all retinal cells interact to adapt to light/dark conditions at the cellular and molecular levels requires systematic investigation.Therefore,we utilized single-cell RNA sequencing to dissect retinal cell-type-specific transcriptomes during light/dark adaptation in mice.The results demonstrated that,in addition to photoreceptors,other retinal cell types also showed dynamic molecular changes and specifically enriched signaling pathways under light/dark adaptation.Importantly,Müller glial cells(MGs)were identified as hub cells for intercellular interactions,displaying complex cell‒cell communication with other retinal cells.Furthermore,light increased the transcription of the deiodinase Dio2 in MGs,which converted thyroxine(T4)to active triiodothyronine(T3).Subsequently,light increased T3 levels and regulated mitochondrial respiration in retinal cells in response to light conditions.As cones specifically express the thyroid hormone receptor Thrb,they responded to the increase in T3 by adjusting light responsiveness.Loss of the expression of Dio2 specifically in MGs decreased the light responsive ability of cones.These results suggest that retinal cells display global transcriptional changes under light/dark adaptation and that MGs coordinate intercellular communication during light/dark adaptation via thyroid hormone signaling.Min Wei Yanping Sun Shouzhen Li Yunuo Chen Longfei Li Minghao Fang Ronghua Shi Dali Tong Jutao Chen Yuqian Ma Kun Qu Mei Zhang Tian Xue 2023Protein & Cell2023,14,8:0
14Human IFN-k Inhibited Respiratory RNA Virus Replication Dependent on Cell-to-Cell Interaction in the Early Phase显示文摘Background:Interferon kappa(IFN-k)is a type I interferon(IFN-I)that inhibits virus replication by evoking interferon-stimulated genes(ISGs).However,as an evolutionarily ancient interferon,IFN-k may function differently from the later emerged interferon-a and b.Methods:Conventional molecular biology methods were used to determine the localization of IFN-k and its structure and function.In addition,we employed RT-PCR,western blot,and RNA-Seq technologies to characterize the ISGs expression profile and antiviral activities exerted by IFN-k or IFN-a2.Results:Human IFN-k exists in two forms upon ectopic expression,one located on the cell membrane and the other secreted outside the cells.The membrane-anchored IFN-k showed the ability to induce ISGs and curtail RNA virus replication,whereas the secreted IFN-k failed to do so.Structural analyses indicated that 1-27aa at the N-terminus was the signal peptide,and 28-37aa was predicted as the transmembrane region.However,our data demonstrated that both of them were not associated with membrane localization of IFN-k;the former influenced the expression and secretion of IFN-k,and the latter had an impact on the induction of ISGs.In addition,prokaryotic purified soluble mature human IFN-k was also capable of inducing ISGs and inhibiting RNA virus replication.Importantly,human IFN-k induced a faster ISG response but with a lower intensity and a shorter half-life than the response of IFN-a2.In contrast,IFN-a2 started to function later but was stronger and more durable than IFN-k.Conclusions:Human IFN-k-induced ISG response and inhibited respiratory RNA virus replication dependent on cell-to-cell interactions.In addition,compared with IFN-a2,IFN-k exerted effects more rapidly in the early phase,with less intensity and a shorter half-life.Therefore,IFN-k may constitute the first line of IFN-I against respiratory virus infections.Weihui Fu Peng Sun Jun Fan Longfei Ding Songhua Yuan Guanxing Zhai Miaomiao Zhang Chenli Qiu Shuye Zhang Xiaoyan Zhang Jianqing Xu 2022Infectious Diseases & Immunity2022,2,2:0
