Comparative studies of lightweight deflectometer and Benkelman beam deflectometer in low volume roads显示文摘A comparative subgrade moduli study is carried out by static and dynamic deflection methods using lightweight deflectometer and conventional Benkelman beam deflectometer on low volume road.Field and laboratory tests are performed at 40 test locations on in-service road of 2 km stretch that contains three common types of cohesive soils(CH,CI,and CL).Pavement static and dynamic responses are estimated to ascertain static, backcalculated, and composite moduli of subgrade. The backcalculated and composite moduli of subgrade is validated at given moisture content using repeated triaxial test.Static moduli values are on lower side as compared with dynamic moduli values whereas the composite, and laboratory moduli of subgrade are approximately consistent with 2% to7% variation, respectively.Correlation analyses between static and dynamic moduli of different types of subgrade soils depict good correlation of determination(R2) varies between 0.75 and 0.91.Subsequently,validation of static moduli with California bearing ratio(CBR) related subgrade moduli shows moderate correlation of 0.67 to 0.74 whereas dynamic moduli shows good correlation of 0.74 to 0.93 for different types of soils, respectively. Therefore, the comparative analysis shows that lightweight deflectometer provides reliable subgrade moduli values, and it can be used as a quick subgrade strength evaluating tool for low volume roads.Sunny Deol Guzzarlapudi Vinod Kumar Adigopula Rakesh Kumar 2016Journal of Traffic and Transportation Engineering(English Edition)2016,3,5:2

Targeting PD-1/PD-L1 in cancer immunotherapy:An effective strategy for treatment of triple-negative breast cancer(TNBC)patients显示文摘Maintaining the balance between eliciting immune responses against foreign pro-teins and tolerating self-proteins is crucial for maintenance of homeostasis. The functions of programmed death protein 1(PD-1)and its ligand programmed death ligand 1(PD-L1)are to inhibit immune responses so that over-reacting immune cells does not cause any damage to its own body cells. However, cancer cells hijack this mechanism to attenuate immune cells functions and create an immunosuppressive environment that fuel their continuous growth and proliferation.Over the past few years’rapid development in cancer immunotherapy has opened a new avenue in cancer treatment.Blockade of PD-1 and PD-L1 has become a potential strategy that rescue the functions of immune cells to fight against cancer with high efficacy.Initially, immune checkpoint monotherapies were not very successful, making breast cancer less immunogenic.Although, recent reports support the presence of tumor infiltrating lympho-cytes(TILs)in breast cancer that make it favorable for PD-1/PD-L1 mediated immunotherapy, which is effective in PD-L1 positive patients.Recently,anti-PD-1(pembrolizumab)and anti-PD-L1(atezolizumab)gets FDA approval for breast cancer treatment and make PD-1/PD-L1 immunotherapy is meaningful for further research.Likewise, this article gathered understand-ing of PD-1 and PD-L1 in recent years, their signaling networks, interaction with other mole-cules,regulations of their expressions and functions in both normal and tumor tissue microenvironments are crucial to find and design therapeutic agents that block this pathway and improve the treatment efficacy.Additionally, authors collected and highlighted most of the important clinical trial reports on monotherapy and combination therapy.Sunny Kumar Mouli Chatterjee Pratyasha Ghosh Kirat K.Ganguly Malini Basu Mrinal K.Ghosh 2023Genes & Diseases2023,10,4:1

Numerical Investigation of Green Water Loading on Flexible Structures Using Three-Step CFD-BEM-FEM Approach显示文摘In this paper, the effect of green water impact on a flexible structure is studied based on three-step computational fluid dynamics (CFD)-boundary element method (BEM)-fmite element method (FEM) approach. The impact due to shipping of water on the deck of the vessel is computed using commercial CFD software and used as an external force in coupled BEM-FEM solver.Other hydrodynamic forces such as radiation, diffraction, and Froude-Krylov forces acting on the structure are evaluated using 3D time domain panel method. To capture the structural responses such as bending moment and shear force,1 D finite element method is developed. Moreover,a direct integration scheme based on the Newmark-Beta method is employed to get the structural velocity, displacement, etc.,at each time step.To check the effect of the green water impact on the structure,a rectangular barge without forward speed is taken for the analysis. The influence is studied in terms of bending moment, shear force, etc. Results show that the effect of green water impact on the bow region can be severe in extreme seas and lead to various structural damages.Similarly,it is also verified that vessel motion affects green water loading significantly and therefore one must consider its effect while designing a vessel.Sumit Kumar Pal Ravindra Babu Kudupudi Mohammed Rabius Sunny Ranadev Datta 2018Journal of Marine Science and Application2018,17,3:1

Natural and artificial small RNAs:a promising avenue of nucleic acid therapeutics for cancer显示文摘Since the failure of traditional therapy, gene therapy using functional DNA sequence and small RNA/DNA molecules(oligonucleotide) has become a promising avenue for cancer treatment. The discovery of RNA molecules has impelled researchers to investigate small regulatory RNA from various natural and artificial sources and determine a cogent target for controlling tumor progression. Small regulatory RNAs are used for therapeutic silencing of oncogenes and aberrant DNA repair response genes.Despite their advantages, therapies based on small RNAs exhibit limitations in terms of stability of therapeutic drugs, precisionbased delivery in tissues, precision-based intercellular and intracellular targeting, and tumor heterogeneity-based responses. In this study, we summarize the potential and drawbacks of small RNAs in nucleic acid therapeutics for cancer.Sunny Yadav Mamta Shekhawat Devashree Jahagirdar Nilesh Kumar Sharma 2017Cancer Biology & Medicine2017,14,3:0
Protection of Viola odorata L.against Neurodegenerative Diseases:Potential of the Extract and Major Phytoconstituents显示文摘Background:Neurodegenerative diseases cause cognitive impairment owing to neuronal death and deterioration.Only a few brain-protecting chemical investigations yielded neuroprotective medications.Viola odoata L.is a traditional medicinal herb that has lately been shown to have strong pharmacological effectiveness against major neurological illnesses with fewer adverse effects. Objective:The objective of the current review is to provide a thorough overview of the key biologically active components and extract of Viola odorata L.in their capacity to safeguard nerve cells. Methods:Science Direct,PubMed,Springer Nature, and Google Scholar were scoured for relevant articles using the terms“Viola odorata L.”,and in the heading and abstract or main text,“neuroprotective activity”,“neurode-generative disease”,“pharmacological potential”,and“therapeutic activity”along with“extract”or“phytocon-stituents”.Neuroprotective activity of Viola odorata L.extract or phytoconstituents were used to classify and investigate the articles that passed the screening process. Results:A total of one hundred twenty one papers were reviewed based on the collected data. The following topics have been elaborated upon in the text:pathological factors of neurodegenerative diseases;role of medic-inal plants in neurodegenerative diseases;Viola odorata L.taxonomy, pharmacological activities, toxicity and adverse effect;neuroprotective activity of extract and various active components of Viola odorata L.The primary phytocomponents of Viola odorata L.were discovered to be anthocyanins, flavonoids, melatonin, salicylic acid, cumarins and various alkaloids, which are primarily responsible for its neuroprotective action. These natural plant compounds can boost antioxidant levels, reduce harmful oxidants, alleviate nerve pain, decrease the pro-duction of pro-inflammatory cytokines such as IL-6,IL-1β,and TNF-α,lower the total amount of nitrite produced, and trigger various neuroprotective mechanisms in the nervous system. It has been also concluded in several in-vestigations that Viola odorata L.extract and some of its major molecular components can protect against toxic affronts, either directly through neuroprotective mechanisms or indirectly through antioxidant properties. Conclusion:Despite the need for further research, the presence of bioactive components in Viola odarata L.that hold the potential to become effective neuroprotective therapeutic agents shows its potential use in treating neurodegenerative diseases characterized by neuro-inflammation and neurotransmitter deficiency.Sunny Dhiman Shivali Singla Inder Kumar Priyankul Palia Pankaj Kumar Sachin Goyal 2023Clinical Complementary Medicine and Pharmacology2023,3,3:0
COVID-19 therapeutics:Clinical application of repurposed drugs and futuristic strategies for target-based drug discovery显示文摘Severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)causes the compli-cated disease COVID-19.Clinicians are continuously facing huge problems in the treatment of patients,as COVID-19-specific drugs are not available, hence the principle of drug repurpos-ing serves as a one-and-only hope.Globally, the repurposing of many drugs is underway;few of them are already approved by the regulatory bodies for their clinical use and most of them are in different phases of clinical trials. Here in this review, our main aim is to discuss in detail the up-to-date information on the target-based pharmacological classification of repurposed drugs, the potential mechanism of actions, and the current clinical trial status of various drugs which are under repurposing since early 2020.At last,we briefly proposed the probable phar-macological and therapeutic drug targets that may be preferred as a futuristic drug discovery approach in the development of effective medicines.Sunny Kumar Malini Basu Pratyasha Ghosh Uttam Pal Mrinal K.Ghosh 2023Genes & Diseases2023,10,4:0
Chaperone-assisted E3 ligase CHIP:A double agent in cancer显示文摘The carboxy-terminus of Hsp70-interacting protein(CHIP)is a ubiquitin ligase and co-chaperone belonging to Ubox family that plays a crucial role in the maintenance of cellular homeostasis by switching the equilibrium of the folding-refolding mechanism towards the proteasomal or lysosomal degradation pathway. It links molecular chaperones viz.HSC70,HSP70 and HSP90 with ubiquitin proteasome system(UPS),acting as a quality control system.CHIP contains charged domain in between N-terminal tetratricopeptide repeat(TPR)and C-terminal Ubox domain.TPR domain interacts with the aberrant client proteins via chaperones while Ubox domain facilitates the ubiquitin transfer to the client proteins for ubiquitination. Thus,CHIP is a classic molecule that executes ubiquitination for degradation of client proteins.Further,CHIP has been found to be indulged in cellular differentiation, proliferation, metastasis and tumorigenesis.Additionally,CHIP can play its dual role as a tumor suppressor as well as an oncogene in numerous malignancies, thus acting as a double agent. Here,in this review,we have reported almost all substrates of CHIP established till date and classified them according to the hallmarks of cancer. In addition,we discussed about its architectural alignment, tissue specific expression,sub-cellular localization,folding-refolding mechanisms of client proteins,E4 ligase activity, normal physiological roles,as well as involvement in various diseases and tumor biology.Further,we aim to discuss its importance in HSP90 inhibitors mediated cancer therapy. Thus, this report concludes that CHIP may be a promising and worthy drug target towards pharmaceutical industry for drug development.Sunny Kumar Malini Basu Mrinal K.Ghosh 2022Genes & Diseases2022,9,6:0
TDP-43 triggers immune response via mitochondrial DNA release显示文摘The molecular mechanisms of toxicity associated with cytoplasmic accumulation of TAR DNA binding protein-43(TDP-43),a pathological feature of many neurodegenerative diseases, are not fully understood.A recent study by Yu et al.is proposing that noxious neuroinflammatory stimulation associated with TDP-43 pathology is caused by excess TDP-43 penetration into mitochondria followed by cytosolic release of mitochondrial DNA(mtDNA)and activation of the cGAS/STING immune sensor.Sunny Kumar Jean-Pierre Julien 2021Cell Research2021,31,4:0
Computational Microuidic Channel for Separation of Escherichia coli from Blood-Cells显示文摘Microuidic channels play a vital role in separation of analytes of interest such as bacteria and platelet cells, etc.,in various biochemical diagnosis procedures including urinary tract infections(UTI)and bloodstream infections. This paper presents the multi physics computational model specifically designed to study the effects of design parameters of a microuidics channel for the separation of Escherichia coli(E.coli)from various blood constituents including red blood cells(RBC)and platelets.A standard two inlet and a two outlet microchannel of length 805µm with a channel width of 40µm is simulated. The effect of electrode potentials and the effect of electrode placement along the channel length and also the levitation of electrodes from the channel wall are studied to optimize the selective particle separation throughput.Simulated results show the efcient separation of E-coli with a mean diameter 0.68µm is achieved at low voltages(less than 20 V)when electrodes placed near to the micro channel and also noticed that the applied electric potential is inversely proportional to the number of electrodes placed along the microuidic channel. The computer aided multi physics simulations with multiple governing parameters could be advantage in design optimization of microuidics channels and support precise bioparticle separation for better diagnosis.Chinnapalli Likith Kumar A.Vimala Juliet Bandaru Ramakrishna Shubhangi Chakraborty Mazin Abed Mohammed Kalakanda Alfred Sunny 2021Computers, Materials & Continua2021,,5:0
