| 2 | IL-25 stimulates M2 macrophage polarization and thereby promotes mitochondrial respiratory capacity and lipolysis in adipose tissues against obesity显示文摘Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially macrophages.Immune molecules,including some cytokines,have a close relationship with metabolism.Interleukin(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively clarified.Human serum and liver biopsy specimens,high-fat diet-induced obesity mice and DB/DB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLD).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in vitro.IL-25 was decreased in NAFLD patients and obese mice.In addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating macrophages.IL-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated macrophages.Furthermore,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more NAD+/NADH and ATP.In conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes. | Juan Feng Lingyi Li Zhiying Ou Qiao Li Baoyong Gong Zhenxian Zhao Weiwei Qi Ti Zhou Jun Zhong Weibin Cai Xia Yang Aiping Zhao Guoquan Gao Zhonghan Yang | 2018 | Cellular & Molecular Immunology2018,15,5: | 10 |

| 5 | A PILOT TRIAL FOR SEVERE ,REFRACTORY SYSTEMIC AUTOIMMUNE DISEASE WITH STEM CELL TRANSPLANTATION显示文摘Objective To evaluate the feasibility, efficacy, and safety of high dose immunosuppressive therapy (HDIT) and autolo- gous hemopoietic stem cell transplantation (HSCT) with CD34+ cell selection in patients with severe, refractory autoim- mune diseases. Methods Twenty-six patients with persistent systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), primary Sjgren’s syndrome (pSS), or systemic sclerosis (SSc) who had been treated unsuccessfully with conventional treatment were enrolled in the trial in Peking Union Medical College Hospital from September 1999 to June 2004. The patients received HDIT with 200 mg/kg cyclophosphamide followed by an infusion of autologous stem cells that were CD34 selected. Disease acti- vity, adverse effect, hemopoietic and immune reconstitution, and time to recurrence of disease were monitored. Results Overall treatment related mortality was 7.7% (2/26) with 1 patient died of cytomegalovirus infection and an- other of severe pneumonia. Relapse occurred in 3 SLE patients (17.6%) in 37, 26, and 19 months posttransplantation respectively, and 1 RA patient in 15 months posttransplantation. SLE Disease Activity Index (SLEDAI) scores of SLE survivors decreased significantly (P < 0.01). RA patients recorded a drop of Disease Activity Score 28 (DAS 28). The pSS patient remained symptoms free up to now, more than 50 months after the transplantation. Conclusion HSCT can be performed relative safely in patients with severe autoimmune disease. Short-term effect of HSCT is promising. | 7 | Production of Embryogenic Callus and Plant Regeneration from Elite Guizhou Waxy Maize Inbred Lines显示文摘Immature embryos from three elite Guizhou waxy maize inbred lines (W21019, B7, and QCL5036) were evaluated for their ability of forming callus and regeneration into plants. Immature embryos harvested at different physiological stages were used as explants to initiate callus on N6 basal medium with 0-3.5 mg L-1 of 2,4-dichlorophenoxy acetic acid (2,4-D). The concentration of 2,4-D, physiological age of immature embryos and genotype had a significant effect (P<0.05) on the percentage of embryogenic callus formed. The optimum 2,4-D concentration for the initiation of embryogenic callus was varied among the waxy maize genotypes from 2.0 mg L-1 (B7 and QCL5036) to 3.0 mg L-1 (W21019). The shoots were generated from embryogenic callus which were transferred into the regeneration medium supplemented with 0-2.5 mg L-1 of 6-benzylaminopurine (6-BA). 6-BA in the medium significantly promoted the regeneration of embryogenic callus. Embryogenic size was also an important factor that affected regeneration capacity. 0.6-0.7 cm was proved to be the best size for regeneration from embryogenic callus and the mean number of shoots per primary callus in all genotypes achieved the highest number. The ability of the plant regeneration was also affected by genotype. W21019 had the highest number of shoots formed per primary embryogenic callus. With the optimum condition, the highest mean number of shoots per primary callus was up to 12.13, 5.73, and 3.33 in line W21019, B7, and QCL5036, respectively. | 20 | Surgical management of non-parasitic hepatic cyst with biliary communication: a case report显示文摘Non-parasitic hepatic cysts with biliary communication are rare. The clinical symptoms involved are not specific to this condition, thereby making diagnosis difficult and treatment controversial. Here, we report a case of 70-year-old woman complaining of abdominal satiety, combined with non-specific pain in the right upper quadrant. The abdominal contrast-enhanced MRI-scan revealed a large and thick-walled septus cystic lesion in the liver. During operation, the biliary fistula was confirmed in the cyst cavity. A silica gel tube was inserted via the cystic duct for cholangiography, which demonstrated communication between the cyst and biliary tract. We performed wide-scale cyst wall resection; the biliary fistula was completely repaired by the closure of communicated bile ducts. The postoperative course was uneventful, and the patient was discharged with no sign of cholangitis or any other symptoms.
