Human umbilical cord-derived mesenchymal stem cell therapy in patients with COVID-19:a phase 1 clinical trial显示文摘No effective drug treatments are available for coronavirus disease 2019(COVID-19).Host-directed therapies targeting the underlying aberrant immune responses leading to pulmonary tissue damage, death,or long-term functional disability in survivors require clinical evaluation. We performed a parallel assigned controlled,non-randomized,phase 1 clinical trial to evaluate the safety of human umbilical cord-derived mesenchymal stem cells(UC-MSCs)infusions in the treatment of patients with moderate and severe COVID-19 pulmonary disease. The study enrolled 18 hospitalized patients with COVID-19(n=9 for each group). The treatment group received three cycles of intravenous infusion of UC-MSCs(3×107 cells per infusion)on days 0,3,and 6.Both groups received standard COVID-treatment regimens.Adverse events, duration of clinical symptoms, laboratory parameters, length of hospitalization, serial chest computed tomography(CT)images, the PaO2/FiO2 ratio, dynamics of cytokines, and IgG and IgM anti-SARS-CoV-2 antibodies were analyzed. No serious UC-MSCs infusion-associated adverse events were observed. Two patients receiving UC-MSCs developed transient facial flushing and fever, and one patient developed transient hypoxia at 12 h post UC-MSCs transfusion.Mechanical ventilation was required in one patient in the treatment group compared with four in the control group. All patients recovered and were discharged. Our data show that intravenous UC-MSCs infusion in patients with moderate and severe COVID-19 is safe and well tolerated.Phase 2/3 randomized,controlled,double-blinded trials with long-term follow-up are needed to evaluate the therapeutic use of UC-MSCs to reduce deaths and improve long-term treatment outcomes in patients with serious COVID-19. | Fanping Meng Ruonan Xu Siyu Wang Zhe Xu Chao Zhang Yuanyuan Li Tao Yang Lei Shi Junliang Fu Tianjun Jiang Lei Huang Peng Zhao Xin Yuan Xing Fan Ji-Yuan Zhang Jinwen Song Dawei Zhang Yanmei Jiao Limin Liu Chunbao Zhou Markus Maeurer Alimuddin Zumla Ming Shi Fu-Sheng Wang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 16
Estimates of genetic and phenotypic parameters for weight and length in Paralichthys olivaceus(Temminck et Schlegel)显示文摘The complete diallel cross design was employed to estimate the genetic parameters of the growth of Japanese flounder (Paralichthys olivaceus). A total of 60 full-sib families were cultivated and two growth-related traits, body weight (BW) and body length (BL), were examined at average 2, 3 and 8 months of age respectively, with 1 800 individuals measured in each age group (30 per family). Based on the additive-dominance-maternal-effect genetic analysis model, the restricted maximum likelihood approach was used to estimate various (co)variance components. The results showed that narrow-sense heritability estimates of BW and BL were respectively: 0.29 ± 0.01 and 0.22 ± 0.02 at 2 months of age, 0.32 ± 0.02 and 0.30 ± 0.04 at 3 months of age, 0.48 ± 0.04 and 0.40 ± 0.05 at 8 months of age; broad-sense heritability estimates were respectively: 0.44 ± 0.02 and 0.54 ± 0.04 at 2 months of age, 0.35 ± 0.01 and 0.36 ± 0.03 at 3 months of age, 0.49 ± 0.03 and 0.45 ± 0.04. All heritabilities were statistically significant (P <0.05). Additive genetic correlations between BW and BL at 2, 3 and 8 months of age were consistently positive and highly significant (P <0.01): 0.93 ± 0.02, 0.95 ± 0.03 and 0.92 ± 0.03 respectively. Maternal effect was significant (P <0.05) only at 2 months of age, and was not detected at 3 and 8 months of age. According to the heritability estimates, the mass selection strategy should be efficient for the breeding of Japanese flounder. | TIAN Yongsheng XU Tianjun LIANG You CHEN Songlin | 2011 | Acta Oceanologica Sinica2011,30,6: | 13
Paleoclimate Modeling in China: A Review显示文摘This paper provides a review of paleoclimate modeling activities in China. Rather than attempt to cover all topics, we have chosen a few climatic intervals and events judged to be particularly informative to the international community. In historical climate simulations, changes in solar radiation and volcanic activity explain most parts of reconstructions over the last millennium prior to the industrial era, while atmospheric greenhouse gas concentrations play the most important role in the20 th century warming over China. There is a considerable model–data mismatch in the annual and boreal winter temperature change over China during the mid-Holocene [6000 years before present(ka BP)], while coupled models with an interactive ocean generally perform better than atmospheric models. For the Last Glacial Maximum(21 ka BP), climate models successfully reproduce the surface cooling trend over China but fail to reproduce its magnitude, with a better performance for coupled models. At that time, reconstructed vegetation and western Pacific sea surface temperatures could have significantly affected the East Asian climate, and environmental conditions on the Qinghai–Tibetan Plateau were most likely very different to the present day. During the late Marine Isotope Stage 3(30–40 ka BP), orbital forcing and Northern Hemisphere glaciation, as well as vegetation change in China, were likely responsible for East Asian climate change. On the tectonic scale, the Qinghai–Tibetan Plateau uplift, the Tethys Sea retreat, and the South China Sea expansion played important roles in the formation of the East Asian monsoon-dominant environment pattern during the late Cenozoic. | JIANG Dabang YU Ge ZHAO Ping CHEN Xing LIU Jian LIU Xiaodong WANG Shaowu ZHANG Zhongshi YU Yongqiang LI Yuefeng JIN Liya XU Ying JU Lixia ZHOU Tianjun YAN Xiaodong | 2015 | Advances in Atmospheric Sciences2015,32,2: | 12

High levels of circulating GM-CSF^(+) CD4^(+) T cells are predictive of poor outcomes in sepsis patients: a prospective cohort study显示文摘Granulocyte colony-stimulating factor(GM-CSF),produced by CD4^(+)T cells, has recently been implicated in the pathogenesis of inflammatory diseases, such as multiple sclerosis and juvenile arthritis. However, the role of GM-CSF-producing CD4^(+)T cells in sepsis remains unknown. This study reports peripheral changes in GM-CSF-producing CD4^(+)T cells in septic patients and the possible underlying mechanism by which GM-CSF influences the outcome of sepsis.Forty-three septic patients,20 SIRS patients, and 20 healthy controls were enrolled in this study and followed for 28 days to assess mortality. We measured the peripheral frequency of GM-CSF^(+)CD4^(+)T cells and recorded their associated relationship with disease progression. Our data demonstrated that peripheral GM-CSF-producing CD4^(+)T cells were significantly higher in septic patients than in both SIRS patients and healthy controls. These cells exhibit a memory phenotype and impaired IFN-γ-secreting capacity in sepsis patients.Using a receiver operating curve analysis with 8.01%as a cut-off point, the percentage of GM-CSF^(+)CD4^(+)T cells could predict the outcome of septic patients.Combined with the increase in GM-CSF-producing CD4^(+)T cells, inflammatory cytokines IL-1βand IL-6 were also upregulated.Using an in vitro neutrophil model,we found that GM-CSF inhibited C3aR expression, while inducing IL-8 production.Furthermore, this effect was transferrable in plasma from sepsis patients and was attenuated by inhibition of GM-CSF using an anti-GM-CSF antibody. These results indicate that GM-CSF-producing CD4^(+)T cells may serve as a marker of sepsis severity. Thus, targeting GM-CSF overproduction may benefit sepsis patients. | Huihuang Huang Siyu Wang Tianjun Jiang Rong Fan Zheng Zhang Jinsong Mu Ke Li Yonggang Wang Lei Jin Fang Lin Jie Xia Lijian Sun Biao Xu Chengcheng Ji Jing Chen Juan Chang Bo Tu Bing Song Chao Zhang Fu-Sheng Wang Ruonan Xu | 2019 | Cellular & Molecular Immunology2019,16,6: | 6
A nonlinear rheological model of backfill material for retaining roadways and the analysis of its stability显示文摘The long-term stability of backfill material is the key to retaining roadways successfully. In order to study the rheological deformation of backfill material and its long-term stability, given the visco-elastoplastic properties of this material,we introduced a softening and a hardening function for a new nonlinear rheological model with time-varying parameters.Based on this,we presented the instability condition of this model by using the principle of minimum potential energy.Combined with engineering practice,we calculated the unstable time period of backfill material. The results show that the time of instability of the backfill material relate to the initial parameters of the material, the coefficients decided by temperature and the ratio of the plastic zone of the backfill material.Based on the results of our analysis from the point of view of energy,we can quickly obtain the time of instability of this model from our graphical analysis. The time of instability of the backfill material obtained from our investigation coincides with an actual project. | Zhang Tianjun Ma Mina Wang Hongsheng Xu Hongjie | 2011 | Mining Science and Technology2011,21,4: | 5
Cloning, characterization of two female-specific AFLP markers and development of PCR-based sex identification method for the half-smooth tongue sole Cynoglossus semilaevis显示文摘Two female-specific AFLP(amplified fragment length polymorphism)markers(named CseF464 and CseF136)were isolated by using one selective primer combination(E-AGC/M-CTG)from the genomic DNA of 20 females and 20 males of the half-smooth tongue sole Cynoglossus semilaevis. Both the markers were re-amplified,recovered from the agarose gels, cloned and sequenced.Bioinformatics analysis indicated that the length of the two markers were 468 bp and 134 bp,respectively, and the sequences showed no similarity to each other,as well as to the known sequences deposited in the GenBank database using BLASTn.Two pairs of SCAR(sequence characterized amplified regions)primers were designed based on the sequences of the two female-specific markers.Furthermore,PCR-based genetic sex identification method was developed in Cynoglossus semilaevis.A specific fragment was amplified in all females but not in any males by using these SCAR primers on the initial 20 female and 20 male individuals of Cynoglossus semilaevis. The feasibility of the two SCAR primer pairs was confirmed in additional 100 individuals(50 females and 50 males). This allowed for reliable, rapid molecular identification of genetic sex of the species, genetic mapping on the sex chromosomes and better understanding of the sex determination and sex differentiation in the half-smooth tongue sole. | Hongyu MA Songlin CHEN Jing LI Jin-Zhen BI Tianjun XU | 2009 | Current Zoology2009,55,4: | 4
Long noncoding RNA AANCR modulates innate antiviral responses by blocking miR-210-dependent MITA downregulation in teleost fish,Miichthys miiuy显示文摘Viral infection induces the initiation of antiviral effectors and cytokines which are critical mediators of innate antiviral responses. The critical molecular determinants are responsible for triggering an appropriate immune response.Long noncoding RNAs(lncRNAs)have emerged as new gene modulators involved in various biological processes, while how lncRNAs operate in lower vertebrates are still unknown. Here,we discover a long noncoding RNA,termed antiviral-associated long noncoding RNA(AANCR),as a novel regulator for innate antiviral responses in teleost fish. The results indicate that fish MITA plays an essential role in host antiviral responses and inhibition of Siniperca chuatsi rhabdovirus(SCRV)production.miR-210 reduces MITA expression and suppress MITA-mediated antiviral responses, which may help viruses evade host antiviral responses.Further,AANCR functions as a competing endogenous RNA(ceRNA)for miR-210 to control protein abundance of MITA,thereby inhibiting SCRV replication and promoting antiviral responses. Our data not only shed new light on understanding the function role of lncRNA in biological processes in teleost fish, but confirmed the hypothesis that ceRNA networks exist widely in vertebrates. | Qing Chu Tianjun Xu Weiwei Zheng Renjie Chang Lei Zhang | 2021 | Science China(Life Sciences)2021,64,7: | 3
From unusual suspect to serial killer: Cyanotoxins boosted by climate change may jeopardize megafauna显示文摘The recent mass mortality event of more than 330 African elephants in Botswana has been attributed to biotoxins produced by cyanobacteria;however, scientific evidence for this is lacking. Here,by synthesizing multiple sources of data,we show that, during the past decades, the widespread hypertrophic waters in Southern Africa have entailed an extremely high risk and frequent exposure of cyanotoxins to the wildlife within this area, which functions as a hotspot of mammal species richness. The hot and dry climatic extremes have most likely acted as the primary trigger of the recent and perhaps also of prehistoric mass mortality events. As such climate extremes are projected to become more frequent in Southern Africa in the near future, there is a risk that similar tragedies may take place, rendering African megafauna species, especially those that are already endangered,in risk of extinction. Moreover, cyanotoxin poisoning amplified by climate change may have unexpected cascading effects on human societies.Seen within this perspective, the tragic mass death of the world's largest terrestrial mammal species serves as an alarming early warning signal of future environmental catastrophes in Southern Africa. We suggest that systematic, quantitative cyanotoxin risk assessments are made and precautionary actions to mitigate the risks are taken without hesitation to ensure the health and sustainability of the megafauna and human societies within the region. | Haijun Wang Chi Xu Ying Liu Erik Jeppesen Jens-Christian Svenning Jianguo Wu Wenxia Zhang Tianjun Zhou Puze Wang Shingirai Nangombe Jinge Ma Hongtao Duan Jingyun Fang Ping Xie | 2021 | The Innovation2021,2,2: | 3
Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients. We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment. We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients. The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone. The cure rate, changes of oxygen saturation and interference, and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group, and 32 patients to the control group. The cure rate in the tocilizumab group was higher than that in the control group, but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions, the hypoxia ameliorated earlier after tocilizumab treatment, and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative. For patients with bilateral pulmonary lesions and elevated IL-6 levels, tocilizumab could be recommended to improve outcome. | Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu | 2021 | Frontiers of Medicine2021,15,3: | 2
Tailoring Porosity and Titanium Species of TS-1 Zeolites via Organic Base-assisted Sequential Post-treatment显示文摘The exploration of high-efficient catalysts based on hierarchical Ti-containing zeolites with optimized active titanium species distribution is of great value in enhancing the epoxidation of bulky olefins. Herein, hierarchical TS-1(MFI) zeolite with an extra-large external surface area(210 m2/g) and highly active octahedral- coordinated Ti species was prepared via organic base-assisted sequential post-treatment. Such a catalyst afforded a high turnover number value(TON, 114) in 1-octene epoxidation reaction, which was over twice than that(53) of the untreated conventional microporous TS-1 parent. According to the detailed characterization results, we revealed the hierarchical porosity construction effect of tetrapropylamonium hydroxide(TPAOH) treatment(first step) and the octahedral-coordinated Ti species fabrication effect of ethylamine(EA) treatment(second step) under hydrothermal condition. Such a facile post-treatment strategy reported in this work may provide guidance for the rational synthesis of TS-1 zeolite with enhanced catalytic activity. | XU Wenjing LI Li ZHANG Tianjun YU Jihong | 2022 | Chemical Research in Chinese Universities2022,38,1: | 2
MicroRNA negatively regulates NF-κB-mediated immune responses by targeting NOD1 in the teleost fish Miichthys miiuy显示文摘Inflammation is a self-protection mechanism that can be triggered when innate immune cells detect infection.Eradication of pathogen infection requires appropriate immune and inflammatory responses, but excessive inflammatory responses can cause uncontrolled inflammation, autoimmune diseases,or pathogen dissemination.Mounting evidence has shown that microRNAs(miRNAs)in mammals act as important and versatile regulators of innate immunity and inflammation. However,miRNAmediated regulation networks are largely unknown in inflammatory responses in lower vertebrates.Here,miR-144 and miR-217 are identified as negative regulators in teleost inflammatory responses. We find that Vibrio harveyi and lipopolysaccharide(LPS)treatment significantly upregulate the expression of fish miR-144 and miR-217.Upregulated miR-144 and miR-217 suppress LPS-induced inflammatory cytokine expression by targeting nucleotide-binding oligomerization domain-containing protein 1(NOD1),thereby avoiding excessive inflammatory responses. In addition,miR-144 and miR-217 regulate inflammatory responses through NOD1-induced nuclear factor kappa(NF-κB)signaling pathways. These findings demonstrate that miR-144 and miR-217 play regulatory roles in inflammatory responses by modulating the NOD1-induced NF-κB signaling pathway. | Qing Chu Dekun Bi Weiwei Zheng Tianjun Xu | 2021 | Science China(Life Sciences)2021,64,5: | 2

Variability of Atlantic Meridional Overturning Circulation in FGOALS-g2显示文摘The variability of Atlantic Meridional Overturning Circulation(AMOC) in the pre-industrial control experiment of the Flexible Global Ocean–Atmosphere–Land System model, Grid-point Version 2(FGOALS-g2) was investigated using the model outputs with the most stable state in a 512-yr time window from the total 1500-yr period of the experiment. The period of AMOC in FGOALS-g2 is double peaked at 20 and 32 years according to the power spectrum, and 22 years according to an auto-correlation analysis, which shows very obvious decadal variability. Like many other coupled climate models, the decadal variability of AMOC in FGOALS-g2 is closely related to the convection that occurs in the Labrador Sea region. Deep convection in the Labrador Sea in FGOALS-g2 leads the AMOC maximum by 3–4 years. The contributions of thermal and haline effects to the variability of the convection in three different regions [the Labrador, Irminger and Greenland–Iceland– Norwegian(GIN) Seas] were analyzed for FGOALS-g2. The variability of convection in the Labrador and Irminger Seas is thermally dominant, while that in the colder GIN Seas can be mainly attributed to salinity changes due to the lower thermal expansion. By comparing the simulation results from FGOALS-g2 and 11 other models, it was found that AMOC variability can be attributed to salinity changes for longer periods(longer than 35 years) and to temperature changes for shorter periods. | HUANG Wenyu WANG Bin LI Lijuan DONG Li LIN Pengfei YU Yongqiang ZHOU Tianjun LIU Li XU Shiming XIA Kun PU Ye WANG Lu LIU Mimi SHEN Si HU Ning WANG Yong SUN Wenqi DONG Fang | 2014 | Advances in Atmospheric Sciences2014,31,1: | 1
