| 1 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 3 | Whole-exome sequencing identifies novel mutations in genes responsible for retinitis pigmentosa in 2 nonconsanguineous Chinese families显示文摘AIM: To detect the pathogenetic mutations responsible for nonsyndromic autosomal recessive retinitis pigmentosa(RP) in 2 nonconsanguineous Chinese families. METHODS: The clinical data, including detailed medical history, best corrected visual acuity(BCVA), slit-lamp biomicroscope examination, fundus photography, optical coherence tomography, static perimetry, and full field electroretinogram, were collected from the members of 2 nonconsanguineous Chinese families preliminarily diagnosed with RP. Genomic DNA was extracted from the probands and other available family members;wholeexome sequencing was conducted with the DNA samples provided by the probands, and all mutations detected by whole-exome sequencing were verified using Sanger sequencing in the probands and the other available family members. The verified novel mutations were further sequenced in 192 ethnicity matched healthy controls.RESULTS: The patients from the 2 families exhibited the typical symptoms of RP, including night blindness and progressive constriction of the visual field, and the fundus examinations showed attenuated retinal arterioles, peripheral bone spicule pigment deposits, and waxy optic discs. Whole-exome sequencing revealed a novel nonsense mutation in FAM161 A(c.943 A>T, p.Lys315*) and compound heterozygous mutations in RP1 L1(c.56 C>A, p.Pro19 His;c.5470 C>T, p.Gln1824*). The nonsense c.5470 C>T, p.Gln1824* mutation was novel. All mutations were verified by Sanger sequencing. The mutation p.Lys315* in FAM161A co-segregated with the phenotype, and all the nonsense mutations were absent from the ethnicity matched healthy controls and all available databases.CONCLUSION: We identify 2 novel mutations in genes responsible for autosomal recessive RP, and the mutation in FAM161A is reported for the first time in a Chinese population. | 5 | Impact of 5-Fu/oxaliplatin on mouse dendritic cells and synergetic effect with a colon cancer vaccine显示文摘Objective: The aim of the present study was to investigate the effects of 5-fluorouracil(5-Fu) and oxaliplatin on the function and activation pathways of mouse dendritic cells(DCs), and to clarify whether 5-Fu/oxaliplatin combined with the CD1d-MC38/α-galactosylceramide(α-GC) tumor vaccine exhibits synergistic effects on the treatment of colon cancer in mice.Methods: The combination of the Toll like receptor(TLR) ligands and/or 5-Fu/oxaliplatin was added into myeloid-derived DCs in vitro culture. DC phenotypic changes were detected by flow cytometry, and the secretion of DC cytokines was detected by cytometric bead array(CBA). A MC38 mouse colon cancer model was constructed and the DCs were isolated from the spleen, tumor tissue and lymph nodes following intraperitoneal injection of 5-Fu/oxaliplatin. The cell phenotypes were detected by flow cytometry. The tumor infiltrating leukocytes,splenocytes and lymph node cells were co-cultured with the dead MC38 tumor cells, and the secretion levels of interferon-γ(IFN-γ) were detected. 5-Fu/oxaliplatin combined with our previously developed CD1d-MC38/α-GC tumor vaccine was used to inhibit the growth of MC38 colon cancer in mice, and the tumor growth rate and survival time were recorded.Results: 5-Fu/oxaliplatin exerted no significant effect on the expression of the stimulating phenotypes of DCs in vitro, while it could reduce the expression of programmed death ligand 1/2(PD-L1/L2) and promote interleukin-12(IL-12) secretion by DCs. Furthermore 5-Fu/oxaliplatin was beneficial to the differentiation of T-helper 1(Th1) cells. 5-Fu/oxaliplatin further enhanced the stimulating phenotypic expression of DCs in tumor bearing mice, decreased PD-L1/L2 expression, and specifically activated the lymphocytes. | 9 | Elevated preoperative CA125 is associated with poor survival in patients with metastatic colorectal cancer undergoing primary tumor resection:a retrospective cohort study显示文摘Background The impact of the preoperative carbohydrate antigen 125(CA125)level on the survival of metastatic colorectal cancer(CRC)patients undergoing primary tumor resection(PTR)remains uncertain.The aim of this study was to assess the prognostic value in overall survival(OS)and cancer-specific survival(CSS)between patients with and without an elevated preoperative CA125 level.Methods All metastatic CRC patients receiving PTR between 2007 and 2017 at the Sixth Affiliated Hospital of Sun Yat-sen University(Guangzhou,China)were retrospectively included.OS and CSS rates were compared between patients with and without elevated preoperative CA125 levels.Results Among 326 patients examined,46(14.1%)exhibited elevated preoperative CA125 levels and the remaining 280(85.9%)had normal preoperative CA125 levels.Patients with elevated preoperative CA125 levels had lower body mass index,lower preoperative albumin level,lower proportion of preoperative chemotherapy,higher carcinoembryonic antigen and carbohydrate antigen 19–9(CA19–9)levels,poorer differentiation,and more malignant histopathological type than patients with normal preoperative CA125 levels.In addition,patients with elevated preoperative CA125 levels exhibited more advanced pathological T and N stages,more peritoneal metastasis,and more vessel invasion than patients with normal preoperative CA125 levels.Moreover,the primary tumor was more likely to be located at the colon rather than at the rectum in patients with elevated CA125 levels.Both OS and CSS rates in patients with elevated preoperative CA125 levels were significantly lower than those in patients with normal preoperative CA125 levels.Multivariate Cox regression analysis revealed that an elevated preoperative CA125 level was significantly associated with poor prognosis in metastatic CRC patients undergoing PTR.The hazard ratio(HR)in OS was 2.36(95%confidence interval[CI],1.67–3.33,P<0.001)and the HR in CSS was 2.50(95%CI,1.77–3.55,P<0.001).The survival analysis stratified by peritoneal metastasis also demonstrated that patients with elevated preoperative CA125 levels had lower OS and CSS rates regardless of peritoneal metastasis.Conclusion Based on an analysis of metastatic CRC patients undergoing PTR,an elevated preoperative CA125 level was associated with poor prognosis,which should be taken into consideration in clinical practice. | Jun-Hua Huang Hua-Shan Liu Tuo Hu Zong-Jin Zhang Xiao-Wen He Tai-Wei Mo Xiao-Feng Wen Ping Lan Lei Lian Xian-Rui Wu | 2022 | Gastroenterology Report2022,10,1: | 2 |
| 10 | Ferroptosis promotes T-cell activation-induced neurodegeneration in multiple sclerosis显示文摘While many drugs are effective at reducing the relapse frequency of multiple sclerosis (MS), there is an unmet need for treatments that slow neurodegeneration resulting from secondary disease progression. The mechanism of neurodegeneration in MS has not yet been established. Here, we discovered a potential pathogenetic role of ferroptosis, an iron-dependent regulated cell death mechanism, in MS. We found that critical ferroptosis proteins (acyl-CoA synthetase long-chain family member 4, ACSL4) were altered in an existing genomic database of MS patients, and biochemical features of ferroptosis, including lipid reactive oxygen species (ROS) accumulation and mitochondrial shrinkage, were observed in the experimental autoimmune encephalitis (EAE) mouse model. Targeting ferroptosis with ferroptosis inhibitors or reducing ACSL4 expression improved the behavioral phenotypes of EAE mice, reduced neuroinflammation, and prevented neuronal death. We found that ferroptosis was an early event in EAE, which may promote T-cell activation through T-cell receptor (TCR) signaling in vitro and in vivo. | 12 | TMEM16F may be a new therapeutic target for Alzheimer's disease显示文摘TMEM16F is involved in many physiological processes such as blood coagulation,cell membrane fusion and bone mineralization.Activation of TMEM16F has been studied in various central nervous system diseases.High TMEM16F level has been also found to participate in microglial phagocytosis and transformation.Microglia-mediated neuroinflammation is a key factor in promoting the progression of Alzheimer’s disease.However,few studies have examined the effects of TMEM16F on neuroinflammation in Alzheimer’s disease.In this study,we established TMEM16F-knockdown AD model in vitro and in vivo to investigate the underlying regulatory mechanism about TMEM16F-mediated neuroinflammation in AD.We performed a Morris water maze test to evaluate the spatial memory ability of animals and detected markers for the microglia M1/M2 phenotype and NLRP3 inflammasome.Our results showed that TMEM16F was elevated in 9-month-old APP/PS1 mice.After TMEM16F knockdown in mice,spatial memory ability was improved,microglia polarization to the M2 phenotype was promoted,NLRP3 inflammasome activation was inhibited,cell apoptosis and Aβplaque deposition in brain tissue were reduced,and brain injury was alleviated.We used amyloid-beta(Aβ_(25-35))to stimulate human microglia to construct microglia models of Alzheimer’s disease.The levels of TMEM16F,inducible nitric oxide synthase(iNOS),proinflammatory cytokines and NLRP3 inflammasome-associated biomarkers were higher in Aβ_(25-35) treated group compared with that in the control group.TMEM16F knockdown enhanced the expression of the M2 phenotype biomarkers Arg1 and Socs3,reduced the release of proinflammatory factors interleukin-1,interleukin-6 and tumor necrosis factor-α,and inhibited NLRP3 inflammasome activation through reducing downstream proinflammatory factors interleukin-1βand interleukin-18.This inhibitory effect of TMEM16F knockdown on M1 microglia was partially reversed by the NLRP3 agonist Nigericin.Our findings suggest that TMEM16F participates in neuroinflammation in Alzheimer’s disease through participating in polarization of microglia and activation of the NLRP3 inflammasome.These results indicate that TMEM16F inhibition may be a potential therapeutic approach for Alzheimer’s disease treatment. | Zhi-Qiang Cui Xiao-Ying Hu Tuo Yang Jing-Wei Guan Ying Gu Hui-Yuan Li Hui-Yu Zhang Qing-Huan Xiao Xiao-Hong Sun | 2023 | Neural Regeneration Research2023,18,3: | 2 |
| 13 | Efficient CO_(2) adsorption and mechanism on nitrogen-doped porous carbons显示文摘In this work,nitrogen-doped porous carbons(NACs)were fabricated as an adsorbent by urea modification and KOH activation.The CO_(2) adsorption mechanism for the NACs was then explored.The NACs are found to present a large specific surface area(1920.72-3078.99 m2·g^(-1))and high micropore percentage(61.60%-76.23%).Under a pressure of 1 bar,sample NAC-650-650 shows the highest CO_(2) adsorption capacity up to 5.96 and 3.92 mmol·g^(-1) at 0 and 25℃,respectively.In addition,the CO_(2)/N_(2) selectivity of NAC-650-650 is 79.93,much higher than the value of 49.77 obtained for the nonnitrogen-doped carbon AC-650-650.The CO_(2) adsorption capacity of the NAC-650-650 sample maintains over 97% after ten cycles.Analysis of the results show that the CO_(2) capacity of the NACs has a linear correlation(R^(2)=0.9633)with the cumulative pore volume for a pore size less than 1.02 nm.The presence of nitrogen and oxygen enhances the CO_(2)/N_(2) selectivity,and pyrrole-N and hydroxy groups contribute more to the CO_(2) adsorption.In situ Fourier transform infrared spectra analysis indicates that CO_(2) is adsorbed onto the NACs as a gas.Furthermore,the physical adsorption mechanism is confirmed by adsorption kinetic models and the isosteric heat,and it is found to be controlled by CO_(2) diffusion.The CO_(2) adsorption kinetics for NACs at room temperature and in pure CO_(2) is in accordance with the pseudo-first-order model and Avramís fractional-order kinetic model. | Yanxia Wang Xiude Hu Tuo Guo Jian Hao Chongdian Si Qingjie Guo | 2021 | Frontiers of Chemical Science and Engineering2021,15,3: | 1 |

| 15 | Enhanced Mechanical Properties of Poly(arylene sulfide sulfone)Membrane by Co-electrospinning with Poly(m-xylene adipamide)显示文摘The mechanical properties of poly(arylene sulfide sulfone)(PASS) electrospun membrane were significantly enhanced by coelectrospinning with semi-aromatic nylon poly(m-xylene adipamide)(MXD6), another engineering plastic with high thermal stability and good mechanical properties. The tensile strength of PASS membrane increased with increased incorporation of MXD6, and was tripled when 20%MXD6 was incorporated. The mechanism of the mechanical property improvement is the existence of hydrogen bonding interaction between PASS and MXD6 and between adjacent fibers at the intersections. Thermal properties of the PASS/MXD6 membranes were evaluated by differential scanning calorimetry(DSC) and thermogravimetric analysis(TGA), which showed that the membranes could be stably utilized up to180 °C without any change in appearance and without decomposition. Contact angle measurements of all the membranes showed hydrophobic character. To demonstrate the potential applications of PASS/MXD6 blend membranes, their oil absorption capacities were evaluated with three oils of different viscosities, which proved that the PASS/MXD6 membranes are better absorbents than commercial non-woven polypropylene fibers. | 20 | Effect of bicarbonate on CO_(2) electroreduction over cathode catalysts显示文摘CO_(2) electroreduction (CO_(2) ER) using renewable energy is ideal for mitigating the greenhouse effect and closing the carbon cycle. Bicarbonate (HCO_(3)−) is most commonly employed as the electrolyte anion because it is known to facilitate CO_(2) ER. However, its dynamics in the electric double layer remains obscure and requires more in-depth investigation. Herein, we investigate the refined reduction process of bicarbonate by employing in situ attenuated total reflectance surface-enhanced infrared absorption spectroscopy. By comparing the product distributions in Ar-saturated KCl and KHCO_(3) electrolytes, we confirmed CO production from HCO_(3)^(−) in the absence of an external CO_(2) source. Notably, in contrast to an electric compulsion, negatively charged HCO_(3)− anions were found to accumulate near the electrode surface. A reduction mechanism of HCO3− is proposed in that HCO3− is not adsorbed over a catalyst, but may be enriched near the electrode surface and converted to CO_(2) and react over Au and Cu electrodes. The dependence of the CO_(2) ER activity on the local HCO3− concentration was subsequently discovered, which was in turn dependent on the bulk HCO3− concentration and cathodic potential. In particular, the local HCO3− concentration was limited by the cathodic potential, leading to a plateau in the CO_(2) ER activity.
