| 2 | Genetic diversity of wild barley (Hordeum vulgare ssp. spontaneum) and its utilization for barley improvement显示文摘Wild barley(Hordeum vulgare spp.spontaneum) is the undisputed progenitor of cultivated barley and offers considerable potential as a genetic resource for barley improvement.Naturally occurring populations are distributed primarily across the Fertile Crescent,but secondary areas of diversity are also found in central Asia and Tibet.The pattern of its genetic diversity is non-random,and is structured on a global,regional and local scale.On a global scale,diversity follows ecogeography,while on a regional and local scale,it is more strongly determined by ecology.Populations growing in areas where the climate is unreliable and/or the soil is heterogeneous tend to retain the most diversity.A large number of accessions are currently conserved ex situ,but a high proportion of these have originated from the Fertile Crescent.Given the likelihood of future climate change,conserving germplasm both in situ and ex situ in the various regions where the species' currently existing is a research priority.The value of H.vulgare spp.spontaneum for the improvement of cultivated barley lies largely in its potential contribution to the levels of attainable pathogen resistance and abiotic stress tolerance. | XiaoYing Ma Chao Li AiDong Wang RuiJun Duan GenLin Jiao Eviatar Nevo GuoXiong Chen | 2012 | Research in Cold and Arid Regions2012,4,6: | 2 |

| 18 | Knockdown of sAC affects sperm hyperactivation by cAMP-signaling pathway in male rat (Rattus norvegicus)显示文摘Soluble adenylyl cyclase (sAC) plays a critical role in male fertility in mammals by regulating sperm hyperactivation. We aimed to study the mechanism of sAC in this phenomenon and to explore potential target sites for male contraception. In this study, in vivo electroporation and rete testis microinjection-mediated short hairpin (sh)RNA plasmids were adopted to silence sAC gene expression in male rats. The results showed that high transfection efficiency (shRNA717, 49.0% and shRNA4205, 65.0%) was achieved by shRNA plasmids injected directly into the rete testis. When the sAC was downregulated, the cyclic adenosine mono-phosphate (cAMP) content and protein phosphorylation level of spermatozoa both declined with a significantly lower hyperactivation rate compared with negative controls. The highest transfection efficiency occurred at 15 d and was obviously time dependent. Bioinformatic and experimental results showed that sAC and tmAC both belong to the AC family and might have analogous functions. ShRNA717 and shRNA4205 were the best targets for the sAC gene, suggesting that they could be candidates for male contraception. | 19 | Study of the Climbing Behavior of the Flagella of Calamus Simplicifolius Based on Micro-CT and Nanoindentation显示文摘Rattan is a typical tropical climbing plant that uses flagella to climb supports to grow.A comprehensive understanding of the anatomic structure and micromechanics of rattan flagella might inspire more research on biomimetic climbing materials.Here,the structure and micromechanical properties of flagella in calamus simplicifolius were examined by Micro-Computed Tomography(Micro-CT)and nanoindentation techniques,respectively.The results showed that the rachis of the flagella mainly comprised vascular bundles surrounded by basic tissues,which had a gradient density decreasing from outsides to insides.The prickles are derived from the epidermis or the epidermis and cortical tissue of the flagellum,which do not possess vascular tissue.The entire tip of the prickle was composed almost of fibrous cells.The indentation modulus of elasticity of the prickle was 17.03 GPa,which was 17.93%higher in comparison with the rachis.The hardness of the prickle was 539.27 MPa and was slightly higher than that of the rachis.The results indicated that the discrepancy of micromechanical strengths in different parts of flagella reflects on their unique roles in the process of climbing. | Fukuan Dai Ziwei Wang Tuhua Zhong Hankun Wang Genlin Tian | 2022 | Journal of Bionic Engineering2022,19,6: | 0 |
