3Genome Analysis of Dasineura jujubifolia Toursvirus 2,A Novel Ascovirus显示文摘So far,ascoviruses have only been identified from Lepidoptera host insects and their transmission vectors-endoparasitic wasps.Here,we reported the first finding of a complete novel ascovirus genome from a Diptera insect,Dasineura jujubifolia.Initially,sequence fragments with homology to ascoviruses were incidentally identified during metagenomic sequencing of the mitochondria of D.jujubifolia(Cecidomyiidae,Diptera)which is a major pest on Ziziphus jujuba.Then a full circular viral genome was assembled from the metagenomic data,which has an A+T percentage of 74%and contains 142,600 bp with 141 open reading frames(ORFs).Among the 141 ORFs,37 were conserved in all sequenced ascoviruses(core genes)including proteins predicted to participate in DNA replication,gene transcription,protein modification,virus assembly,lipid metabolism and apoptosis.Multi-gene families including those encode for baculovirus repeated open reading frames(BROs),myristylated membrane proteins,RING/U-box E3 ubiquitin ligases,and ATP-binding cassette(ABC)transporters were found in the virus genome.Phylogenetic analysis showed that the newly identified virus belongs to genus Toursvirus of Ascoviridae,and is therefore named as Dasineura jujubifolia toursvirus 2(DjTV-2a).The virus becomes the second reported species of the genus after Diadromus pulchellus toursvirus 1(DpTV-1a).The genome arrangement of DjTV-2a is quite different from that of DpTV-1a,suggesting these two viruses separated in an early time of evolution.The results suggest that the ascoviruses may infect a much broader range of hosts than our previous knowledge,and shed lights on the evolution of ascoviruses and particularly on that of the toursviruses.Jun Wang Minglu Yang Haibing Xiao Guo-Hua Huang Fei Deng Zhihong Hu 2020Virologica Sinica2020,35,2:4
4Overcoming chemotherapy resistance via simultaneous drug-efflux circumvention and mitochondrial targeting显示文摘Multidrug resistance(MDR) has been considered as a huge challenge to the effective chemotherapy. Therefore, it is necessary to develop new strategies to effectively overcome MDR. Here,based on the previous research of N-(2-hydroxypropyl)methacrylamide(HPMA) polymer–drug conjugates, we designed an effective system that combined drug-efflux circumvention and mitochondria targeting of anticancer drug doxorubicin(Dox). Briefly, Dox was modified with mitochondrial membrane penetrating peptide(MPP) and then attached to(HPMA) copolymers(P-M-Dox). Our study showed that macromolecular HPMA copolymers successfully bypassed drug efflux pumps and escorted Dox into resistant MCF-7/ADR cells via endocytic pathway. Subsequently, the mitochondria accumulation of drugs was significantly enhanced with 11.6-fold increase by MPP modification. The excellent mitochondria targeting then resulted in significant enhancement of reactive oxygen species(ROS) as well as reduction of adenosine triphosphate(ATP)production, which could further inhibit drug efflux and resistant cancer cell growth. By reversing Dox resistance, P-M-Dox achieved much better suppression in the growth of 3D MCF-7/ADR tumor spheroids compared with free Dox. 5Optimal Control of a Universal Rotating Magnetic Vector for Petal-shaped Capsule Robot in Curve Environment显示文摘Steering control of a capsule robot in curve environment by magnetic navigation is not yet solved completely.A petal-shaped capsule robot with less steering resistance based on multiple wedge effects is presented,and an optimization method with two processes for determining the orientation of a pre-applied universal magnetic spin vector is proposed.To realize quick and non-contact steering swimming,a fuzzy comprehensive evaluation method for optimizing the steering driving angle is presented based on two evaluation indexes including the average steering speed and the average steering trajectory deviation,achieving the initial optimal orientation of a universal magnetic spin vector.To further reduce robotic magnetic vibration,a main target method for optimizing its final orientation,which is used for fine adjustment,is employed under the constrains of the magnetic moments.Swimming experimental results in curve pipe verified the effectiveness of the optimization method,which can be effectively used to realize non-contact steering swimming of the petal-shaped robot and reduce its vibration.ZHANG Yongshun BAI Jianwei CHI Minglu CHENG Cunxin WANG Dianlong 2014Chinese Journal of Mechanical Engineering2014,27,5:2

11Efficient control of chronic LCMV infection by a CD4 T cell epitope-based heterologous prime-boost vaccination in a murine model显示文摘CD4^(+)T cells are essential for sustaining CD8^(+)T cell responses during a chronic infection.The adoptive transfer of virus-specific CD4^(+)T cells has been shown to efficiently rescue exhausted CD8^(+)T cells.However,the question of whether endogenous virus-specific CD4^(+)T cell responses can be enhanced by certain vaccination strategies and subsequently reinvigorate exhausted CD8^(+)T cells remains unexplored.In this study,we developed a CD4^(+)T cell epitope-based heterologous prime-boost immunization strategy and examined the efficacy of this strategy using a mouse model of chronic lymphocytic choriomeningitis virus(LCMV)infection.We primed chronically LCMV-infected mice with a Listeria monocytogenes vector that expressed the LCMV glycoprotein-specific I-Ab-restricted CD4^(+)T cell epitope GP61–80(LM-GP61)and subsequently boosted the primed mice with an influenza virus A(PR8 strain)vector that expressed the same CD4^(+)T cell epitope(IAV-GP61).This heterologous prime-boost vaccination strategy elicited strong anti-viral CD4^(+)T cell responses,which further improved both the quantity and quality of the virusspecific CD8^(+)T cells and led to better control of the viral loads.The combination of this strategy and the blockade of the programmed cell death-1(PD-1)inhibitory pathway further enhanced the anti-viral CD8^(+)T cell responses and viral clearance.Thus,a heterologous prime-boost immunization that selectively induces virus-specific CD4^(+)T cell responses in conjunction with blockade of the inhibitory pathway may represent a promising therapeutic approach to treating patients with chronic viral infections.Ran He Xinxin Yang Cheng Liu Xiangyu Chen Lin Wang Minglu Xiao Jianqiang Ye Yuzhang Wu Lilin Ye 2018Cellular & Molecular Immunology2018,15,9:1

13Design and characterization of plasticized bacterial cellulose/waterborne polyurethane composite with antibacterial function for nasal stenting显示文摘A nasal stent capable of preventing adhesions and inflammation is of great value in treating nasal diseases.In order to solve the problems of tissue adhesion and inflammation response,we prepared plasticized bacterial cellulose(BCG)and waterborne polyurethane(WPU)composite with antibacterial function used as a novel nasal stent.The gelation behavior of BCG could contribute to protecting the paranasal sinus mucosa;meanwhile,the WPU with improved mechanical property was aimed at supporting the narrow nasal cavity.The thickness,size and the supporting force of the nasal stent could be adjusted according to the specific conditions of the nasal.Thermogravimetric analysis,contact angle and water absorption test were applied to investigate the thermal,hydrophilic and water absorption properties of the composite materials.The composite materials loaded with poly(hexamethylene biguanide)hydrochloride maintained well antibacterial activity over 12days.Animal experiments further revealed that the mucosal epithelium mucosae damage of BCG-WPU composite was minor compared with that of WPU.This new type of drug-loaded nasal stent can effectively address the postoperative adhesions and infections while ensuring the health of nasal mucosal,and thus has an immense clinical application prospects in treating nasal diseases.Zhaoxuan Feng Minglu Li Xing Jin Yudong Zheng Junxiu Liu Liang Zhao Yansen Wang Hao Li Danlin Zuo 2020Regenerative Biomaterials2020,7,6:1
14A Mandatory Access Control Framework in Virtual Machine System with Respect to Multi-level Security I: Theory显示文摘At present,there are few security models which control the communication between virtual machines (VMs).Moreover,these models are not applicable to multi-level security (MLS).In order to implement mandatory access control (MAC) and MLS in virtual machine system,this paper designs Virt-BLP model,which is based on BLP model.For the distinction between virtual machine system and non-virtualized system,we build elements and security axioms of Virt-BLP model by modifying those of BLP.Moreover,comparing with BLP,the number of state transition rules of Virt-BLP is reduced accordingly and some rules can only be enforced by trusted subject.As a result,Virt-BLP model supports MAC and partial discretionary access control (DAC),well satisfying the requirement of MLS in virtual machine system.As space is limited,the implementation of our MAC framework will be shown in a continuation.LIU Qian WANG Guanhai WENG Chuliang LUO Yuan LI Minglu 2010China Communications2010,7,4:1
15Simultaneously improved actuated performance and mechanical strength of silicone elastomer by reduced graphene oxide encapsulated silicon dioxide显示文摘Herein,graphene oxide(GO)-encapsulated silica(SiO 2)hybrids(GO@SiO 2)were prepared via electrostatic self-assembly of the 3-aminopropyltriethoxysilane(APS)-modified SiO_(2) and GO.The as-prepared GO@SiO 2 was introduced into polydimethyl-siloxane(PDMS)elastomer to simultaneously increase the dielectric constant(k)and mechanical properties of PDMS.Then,the in situ thermal reduction of GO@SiO_(2)/PDMS composites was conducted at 180℃ for 2 h to increase the interfacial polariz-ability of GO@SiO_(2).As a result,the values of k at 1000 Hz are largely improved from 3.2 for PDMS to 13.3 for the reduced GO@SiO_(2)(RGO@SiO_(2))/PDMS elastomer.Meanwhile,the dielectric loss of the composites remains low(<0.2 at 1000 Hz).More importantly,the actuated strain at low electric field(5 kV/mm)obviously increases from 0.3%for pure PDMS to 2.59%for the composites with 60 phr of RGO@SiO_(2),an eightfold increase in the actuated strain.In addition,both the tensile strength and elastic modulus are obviously improved by adding 60 phr of RGO@SiO_(2),indicating a good reinforcing effect of RGO@SiO_(2) on PDMS.Our goal is to develop a simple and effective way to improve the actuated performance and mechanical strength of the PDMS dielectric elastomer for its wider application.Nanying Ning Minglu Wang Jing Zhang Liqun Zhang Ming Tian 2015International Journal of Smart and Nano Materials2015,6,4:1
16Improving performance of macro electrolyte jet machining of TC4 titanium alloy:Experimental and numerical studies显示文摘Electrolyte jet machining(EJM)is a promising method for shaping titanium alloys due to its lack of tool wear,thermal and residual stress,and cracks and burrs.Recently,macro-EJM has attracted increasing attention for its high efficiency in machining wide grooves or planes.However,macro-EJM generates large amounts of electrolytic products,thereby increasing the difficulty of rapid product removal with a standard tool and reducing the surface quality.Therefore,for enhanced product transport,a novel tool with a back inclined end face was proposed for macroEJM of TC4 titanium alloy.For comparison,also proposed were ones with a standard flat end face,a front inclined end face,and both front and back inclined end faces.The flow field distributions of all proposed tools were simulated numerically,and experiments were also conducted to validate the simulation results.The results show that one with a 5°back inclined end face can decrease the lowvelocity flow zone in the machining area and increase the high-velocity flow zone at the back end of tool,thereby promoting rapid product removal.A relatively smooth bright-white groove surface was obtained.The same tool also resulted in the highest machining depth and material removal rate among the tested ones.In addition,rapid product removal was beneficial to the subsequent processing.Because of its rapid product removal,the machining depth and material removal rate during deep groove machining using the tool with a 5°back inclined end face were respectively 7%and14%higher than those produced using a standard one.Moreover,the lowest bottom height difference of 0.027 mm can be obtained when the step-over value was 8.2 mm,and a plane with a depth of0.285 mm and a bottom height difference of 0.03 mm was fabricated using the tool with a 5°back inclined end face.Minglu WANG Ningsong QU 2022Chinese Journal of Aeronautics2022,35,8:0

18Abdominal Grey Color Variation along an Elevation Gradient in a Lizard显示文摘Color variation among populations is thought to be either due to genetic variation or phenotype plasticity.In this study,we found that an agamid lizard,Phrynocephalus theobaldi,inhabiting elevations greater than any other reptile,displayed a significantly darker spotted marking on the central ventral abdomen in higher populations than in lower populations.The grey spotted area only occurred in elevations>4200 m,and also increased with increased elevation.The ratio of the size of the dark colored area to the degree of skin luminance/reflectance was used to test whether the lizard has a larger and darker spotted area at higher elevations.This ratio was significantly positively associated with elevation rather than substrate color.The lizard did not display sexual difference in this phenotype.Temperature had a significant influence on the brightness of central abdomen,but temperature-related plasticity of this trait was much smaller than the magnitude of the difference between higher(>4200 m)and lower populations(<4200 m).Common garden experiments showed that genetic inheritance,rather than phenotype plasticity,played a dominant role in this variation.Our research addressed the darkness of a body region that is rarely analyzed because it is shielded from direct insolation.The idea that ventral color could be related to thermoregulation is shown to be worthy of further research.Zian WEI Jiasheng LI Tao WANG Minglu YANG Yuanting JIN 2023Asian Herpetological Research2023,14,2:0
19Responses of microbial interactions to elevated salinity in activated sludge microbial community显示文摘Biological treatment processes are critical for sewage purification,wherein microbial interactions are tightly associated with treatment performance.Previous studies have focused on assessing how environmental factors(such as salinity)affect the diversity and composition of the microbial community but ignore the connections among microorganisms.Here,we described the microbial interactions in response to elevated salinity in an activated sludge system by performing an association network analysis.It was found that higher salinity resulted in low microbial diversity,and small,complex,more competitive overall networks,leading to poor performance of the treatment process.Subnetworks of major phyla(Proteobacteria,Bacteroidetes,and Chloroflexi)and functional bacteria(such as AOB,NOB and denitrifiers)differed substantially under elevated salinity process.Compared with subnetworks of Nitrosomonadaceae,Nitrosomonas(AOB)made a greater contribution to nitrification under higher salinity(especially 3%)in the activated sludge system.Denitrifiers established more proportion of cooperative relationships with other bacteria to resist 3%salinity stress.Furthermore,identified keystone species playing crucial roles in maintaining process stability were dynamics and less abundant under salinity disturbance.Knowledge gleaned from this study deepened our understanding of microbial interaction in response to elevated salinity in activated sludge systems.Tao Ya Zhimin Wang Junyu Liu Minglu Zhang Lili Zhang Xiaojing Liu Yuan Li Xiaohui Wang 2023Frontiers of Environmental Science & Engineering2023,17,5:0
20Insights from metagenomic,metatranscriptomic,and molecular ecological network analyses into the effects of chromium nanoparticles on activated sludge system显示文摘The objectives of this study were to investigate the influence of chromium nanoparticles(Cr NPs)on the nitrogen and phosphorus removal performance and the bacterial structures of an activated sludge(AS)system.Also,we through molecular ecological networks(MENs)discussed the bacterial interactions.At last we researched the change of the functional genes and their expression patterns related to nitrogen and phosphorus removaT in an AS system.The results showed that long-term exposure to 1 mg/L Cr NPs significantly promoted the denitrifying process and phosphorus removal in the AS system.The relative abundance of denitrifying and phosphorus removal microorganisms,such as Denitratisoma,Thauera,Dechloromonas,and Defluviicoccus,increased significantly.Candidatus Accumulibacter,well-known as polyphosphate-accumulating organisms(PAOs),increased significantly;the relative abundance of Candidatus Competibacter,known as glycogen-accumulating organisms(GAOs),decreased significantly.Furthermore,metagenomic and metatranscriptomic analysis revealed that most of the genera related to denitrifying and phosphorus removal had greatly increased,according to the quantities of denitrifying and phosphorus genes,and the corresponding transcription likewise greatly increased.Lastly,MENs analysis showed that although the overall network became smaller and looser in the presence of Cr NPs,the microbial connections among members related to nitrogen and phosphorus removal were enhanced.The abundance increases of denitrifiers and PAOs,and their increased transcription of functional genes,together with the enhanced interactions may be associated with the promotion of the denitrifying process and phosphorus removal.Quan Zheng Minglu Zhang Tingting Zhang Xinhui Li Minghan Zhu Xiaohui Wang 2020Frontiers of Environmental Science & Engineering2020,14,4:0
