1Genome-wide analysis of the WRKY transcription factor gene family in Gossypium raimondii and the expression of orthologs in cultivated tetraploid cotton显示文摘WRKY proteins are members of a family of transcription factors in higher plants that function in plant responses to various physiological processes.We identified 120 candidate WRKY genes from Gossypium raimondii with corresponding expressed sequence tags in at least one of four cotton species,Gossypium hirsutum,Gossypium barbadense,Gossypium arboreum,and G.raimondii.These WRKY members were anchored on 13 chromosomes in G.raimondii with uneven distribution.Phylogenetic analysis showed that WRKY candidate genes can be classified into three groups,with 20 members in group I,88 in group II,and 12 in group III.The88 genes in group II were further classified into five subgroups,groups IIa–e,containing 7,16,37,15,and 13 members,respectively.We characterized diversity in amino acid residues in the WRKY domain and/or other zinc finger motif regions in the WRKY proteins.The expression patterns of WRKY genes revealed their important roles in diverse functions in cotton developmental stages of vegetative and reproductive growth and stress response.Structural and expression analyses show that WRKY proteins are a class of important regulators of growth and development and play key roles in response to stresses in cotton.Caiping Cai Erli Niu Hao Du Liang Zhao Yue Feng Wangzhen Guo 2014The Crop Journal2014,2,Z1:23
2Characterizati0n,development and exploitation of EST-derived microsatellites in Gossypium raimondii Ulbrich显示文摘Microsatellite DNA or simple sequence repeats (SSRs) can be derived from expressed se- quence tags (ESTs). These markers are important for gene mapping as well as marker-assisted selection (MAS). To develop EST-SSRs for cotton gene map- ping, we selected and characterized functional markers in Gossypium raimondii, which consisted of 58906 non-redundant EST sequences from NCBI. Among them there were 2620 microsatellite se- quences containing 2818 EST-SSRs, which amoun- ted to 4.45% of the non-redundant starting sequence population. This incidence was equivalent to one EST-SSR in every 14.8 kb of G. raimondii genetic material. Among the different motifs ranging from 1 to 6 bp, trinucleotide repeats were most abundant (38.31%), followed by dinucleotide repeats (24.09%) and mononucleotide repeats (23.35%). Among all identified motif types, A/T had the highest frequency (18.67%), followed by AT/TA (14.83%). Among the compound motifs, tandem trinucleotides occurred with the highest frequency (48.65%). In all, we identi- fied 1554 EST-SSRs primer pair sequences. 300 of them were randomly selected to screen the poly- morphisms between the mapping parents G· hirsutum acc. TM-1 and G· barbadense cv. Hai7124, to con- struct linkage groups in cultivated allotetraploid cot- ton. Among them, 129 (43%) primer pairs were found to have polymorphisms. 3Association analysis of fiber quality traits and exploration of elite alleles in Upland cotton cultivars/accessions(Gossypium hirsutum L.)显示文摘Exploring the elite alleles and germplasm accessions related to fiber quality traits will accelerate the breeding of cotton for fiber quality improvement. In this study, 99 Gossypium hirsutum L. accessions with diverse origins were used to perform association analysis of fiber quality traits using 97 polymorphic microsatellite marker primer pairs. A total of 107 significant marker-trait associations were detected for three fiber quality traits under three different environments, with 70 detected in two or three environments and 37 detected in only one environment. Among the 70 significant marker-trait associations, 52.86% were reported previously, implying that these are stable loci for target traits. Furthermore, we detected a large number of elite alleles associated simultaneously with two or three traits. These elite alleles were mainly from accessions collected in China, introduced to China from the United States, or rare alleles with a frequency of less than5%. No one cultivar contained more than half of the elite alleles, but 10 accessions were collected from China and the two introduced from the United States did contain more than half of these alleles. 4Molecular mapping of Verticillium wilt resistance QTL clustered on chromosomes D7 and D9 in upland cotton显示文摘Verticillium wilt is a destructive disease with international consequences for cotton production. Breeding broad-spectrum resistant cultivars is considered to be one of the most effective means for reducing crop losses. A resistant cotton cultivar,60182,was crossed with a susceptible cultivar,Jun-mian 1,to identify markers for Verticillium resistance genes and validate the mode of its inheritance. Genetic segregation analysis for Verticillium wilt resistance was evaluated based upon infected leaf percentage in the seedling stage using major gene-polygene mixed inheritance models and joint analysis of P1,P2,F1,B1,B2 and F2 populations obtained from the cultivar cross. We found that resistance of upland cotton cultivar 60182 to isolates BP2,VD8 and T9,and their isoconcentration mixture was controlled by two major genes with additive-dominance-epistatic effects,and the inheritance of the major gene was dominant. Furthermore,a genetic linkage map was constructed using F2 segregating population and resistance phenotypic data were obtained using F2:3 families inoculated with different isolates and detected in different developmental stages. The genetic linkage map with 139 loci was comprised of 31 linkage groups covering 1165 cM,with an average distance of 8.38 cM between two markers,or 25.89% of the cotton genome length. From 60182,we found 4 QTL on chromosome D7 and 4 QTL on D9 for BP2,5 QTL on D7 and 9 QTL on D9 for VD8,4 QTL on D7 and 5 QTL on D9 for T9 and 3 QTL on D7 and 7 QTL on D7 for mixed pathogens. The QTL mapping results revealed that QTL clusters with high contribution rates were screened simultaneously on chromosomes D9 and D7 by multiple interval mapping (CIM),whether from resistance phenotypic data from different developmental stages or for different isolates. The result is consistent with the genetic model of two major genes in 60182 and suggests broad-spectrum resistance to both defoliating isolates of V. dahliae and nondefoliating isolates. 5Identification of RAPD marker linked with fertility-restoring gene of cytoplasmic male sterile lines in upland cotton显示文摘Bulked segregant analysis was employed to construct two mixed DNA pools to screen the RAPd marker linked with the fertilityrestoring gene(\%Rf\-i)\% of upland cotton. A total of 425 arbitrary 10mer oligonucleotide primers were screened on two DNA pools, bulked male fertile and sterile DNAs isolated from BC\-3 segregating population of (06132R×Simian No. 3). Three primers produced repeatable polymorphisms between the paired bulks and their parents. DNA was extracted and amplified with these three primers for 92 plants of (Zhong 12A1×06132R)F\-2. 6Fine mapping of the dominant glandless Gene Gl_2~e in Sea-island cotton (Gossypium barbadense L.)显示文摘Gle2 is a mutant gene that controls glandless trait in cotton plants and seeds. It is an important gene resource to gossypol-free cottonseed breeding. The objective of this research was to develop SSR markers tightly linked with Gle2 by using the F2 segregating population containing 1599 plants derived from the cross of G. hirsutum genetic standard line TM-1 and G. barbadense glandless mutant line Hai-1. Genetic analysis suggested that the Gle2 was an incomplete dominant gene. Based on the backbone of genetic linkage map from G. hirsutum × G. barbadense BC1 published by our laboratory,Gle2 was lo-cated between CIR362 and NAU2251b,NAU3860b,STV033,with a genetic distance 9.27 and 0.96 cM,respectively. 7Corporate Governance, Ownership and Financial Distress of Publicly Listed Companies in China显示文摘This paper examines the effects of internal control mechanisms on the financial distress of publicly listed companies in China. Using t-test and Logistic regression analysis, we investigated the following aspects: ownership by top managers and the CEO, the number of directors, the percentage of inside directors, CEO duality, and ownership concentration. The empirical tests indicate a lower probability of financial distress when ownership by top managers and that by the CEO are higher. 8Isolation and characterization of resistance and defense gene analogs in cotton (Gossypium barbadense L.)显示文摘Plant disease resistance gene (R gene) and defense response gene encode some con-served motifs. In the present work,a PCR strategy was used to clone resistance gene analogs (RGAs) and defense gene analogs (DGAs) from Sea-island cotton variety Hai7124 using oligonucleotide primers based on the nucleotide-binding site (NBS) and serine/threonine kinase (STK) in the R-gene and pathogenesis-related proteins of class 2 (PR2) of defense response gene. 79 NBS sequences,21 STK sequences and 11 DGAs were cloned from disease-resistance cotton. Phylogenic analysis of 79 NBS-RGAs and NBS-RGAs nucleotide sequences of cotton already deposited in GenBank identified one new sub-cluster. The deduced amino acid sequences of NBS-RGAs and STK-RGAs were divided into two distinct groups respectively: Toll/Interleukin-1 receptor (TIR) group and non-TIR group,A group and B group. The expression of RGAs and DGAs having consecutive open reading frame (ORF) was also investigated and it was found that 6 NBS-RGAs and 1 STK-RGA were induced,and 1 DGA was up-regulated by infection of Verticillium dahliae strain VD8. 4 TIR-NBS-RGAs and 4 non-TIR-NBS-RGAs were arbitrarily used as probes for Southern-blotting. There existed 2-10 blotted bands. 9Fine mapping of the red plant gene R_1 in upland cotton (Gossypium hirsutum)显示文摘Sub 16 is a substitution line with G. hirsutum cv. TM-1 genetic background except that the 16th chro-mosome (Chr. 16) is replaced by the corresponding homozygous chromosome of G. barbadense cv. 3-79, and T586 is a G. hirsutum multiple gene marker line with 8 dominant mutation genes. The R1 gene for anthocyanin pigmentation was tagged in Chr. 16 in T586. The objective of this research was to screen SSR markers tightly linked with R1 by using the F2 segregating population containing 1259 plants derived from the cross of Sub 16 and T586 and the backbone genetic linkage map from G. hir-sutum×G. barbadense BC1 newly updated by our laboratory. Genetic analysis suggested that the seg-regation ratio of red plants in the F2 population fit Mendelian 1:2:1 inheritance, confirming that the red plant trait was controlled by an incomplete dominance gene. Preliminary mapping of R1 was conducted using 237 randomLy selected F2 individuals and JoinMap v3.0 software. Then, a fine map of R1 was constructed using the F2 segregating population containing 1259 plants, and R1 was located between NAU4956 and NAU6752, with only 0.49 cM to the nearest maker loci (NAU6752). 10QTL mapping in A-genome diploid Asiatic cotton and their congruence analysis with AD-genome tetraploid cotton in genus Gossypium显示文摘Asiatic cotton(Gossypium arboreum L.) is an Old World cultivated cotton species.The sinense race was planted extensively in China.Due to the advances in spinning technology during the last century,the species was replaced by the New World allotetraploid cotton G.hirsutum L.Gossypium arboreum is still grown in India and Pakistan and also used as an elite in current cotton breeding programs.In addition,G.arboreum serves as a model for genomic research in Gossypium.In the present study,we generated an A-genome diploid cotton intraspecific genetic map including 264 SSR loci with three morphological markers mapped to 13 linkage groups.The map spans 2,508.71 cM with an average distance of 9.4 cM between adjacent loci.A population containing 176 F2:3 families was used to perform quantitative trait loci(QTL) mapping for 17 phenotypes using Multiple QTL Model(MQM) of MapQTL ver 5.0.Overall,108 QTLs were detected on 13 chromosomes.Thirty-one QTLs for yield and its components were detected in the F2 population.Forty-one QTLs for yield and its components were detected in the F2:3 families with a total of 43 QTLs for fiber qualities.Two QTLs for seed cotton weight/plant and lint index and three QTLs for seed index were consistently detected both in F2 and F2:3.Most QTLs for fiber qualities and yields were located at the same interval or neighboring intervals.These results indicated that the negative correlation between fiber qualities and yield traits may result from either pleiotropic effect of one gene or linkage effects of multiple closely linked genes.Xuexia Ma Yezhang Ding Baoliang Zhou Wangzhen Guo Yanhui Lv Xiefei Zhu Tianzhen Zhang 2008Journal of Genetics and Genomics2008,35,12:5
11Surface Treatment of Crl2MoV Steel towards Long-Life Cold-Work Dies显示文摘With the rapid development of the automobile industry in China, there is an ever-increasing demand for long-life cold working dies used for punching automobile components. However, the full potential of such advanced surface engineering technologies as PVD coatings and duplex surface treatments in cold work dies has not been realized. In the present study, Crl2MoV steel has been surface engineered by single PVD Ti/TiN coating and duplex treatment combining low temperature plasma nitriding (LTPN) with PVD Ti/TiN coatings. The properties of Ti/TiN coatings in terms of surface morphology, microhardness, load bearing capacity, bonding strength and wear resistance were evaluated by microhardness, scratch and wear tests. The experimental results show that PVD Ti/TiN coatings can significantly enhance the surface load bearing capacity (especially for duplex treatments) and wear resistance of Crl2MoV steel by more than one order of magnitude. This can be mainly attributed to the hard and well-adherent PVD Ti/TiN surface coatings and strong mechanical support of the LTPN sublayer. 12Mining,characterization,and exploitation of EST-derived microsatellites in Gossypium barbadense显示文摘Simple sequence repeats(SSRs) have been widely applied as molecular markers in genetic studies.However,the number of expressed sequence tags(ESTs) and SSR markers from Gossypium barbadense is fewer than those from other cotton species.In this study,EST-SSR distribution from G.barbadense was characterized and new G.barbadense-derived EST-SSR markers were determined on the basis of the ESTs obtained by randomly sequencing 2 cDNA libraries associated with fiber development in G.barbadense.By mining 9697 non-redundant ESTs,a total of 638 SSR loci derived from 595 ESTs were observed.In G.barbadense,the frequency of ESTs containing SSRs was 6.13%,with an average of 1 SSR in every 10.4 kb of EST sequence.Furthermore,trinucleotide was found to be the most abundant repeat type among 2-6-nucleotide repeat types.It accounted for 26.6% of the total,followed by the hexanucleotide(26.0%) and pentanucleotide repeats(25.9%).Among all the repeat motifs,(AAG)n accounted for the highest proportion.EST-SSR primer pairs were developed using the Primer3 program,and the redundant primers were removed using the virtual PCR approach.As a result,380 non-redundant EST-SSR primer pairs were developed and used to detect polymorphisms between the mapping parents G.hirsutum 'TM-1' and G.barbadense 'Hai7124' for constructing linkage groups in cultivated allotetraploid cotton.Out of these,98(25.8%) primer pairs detected polymorphisms.Finally,95 polymorphic loci from 82 primer pairs were integrated into the backbone genetic map;of these,42 were mapped into the A subgenome and 53 into the D subgenome.The present work provided the foundation for constructing saturated genetic maps and conducting comparative genomic studies on different cotton species.LU YuanDa CAI CaiPing WANG Lei LIN ShaoYan ZHAO Liang TIAN LiangLiang LU JunHong ZHANG TianZhen GUO WangZhen 2010Chinese Science Bulletin2010,55,18:5
13Characteristics of the binding features of Nelin with F-actin and screening Nelin interactive proteins显示文摘The interaction of nelin, a cardiac-specific ex- pressed protein of human novel gene nelin, with F-actin was studied by both F-actin cosedimentation in vitro and colocal- ization assays. The results showed that nelin is a new F-actin binding protein and is colocolized with F-actin in cytoplasm of cells. Three new nelin binding proteins, filamin C subtype, titin N2B subtype and inter-alpha trypsin inhibitor heavy chain precursor (ITIH), were identified from human heart cDNA library using yeast two-hybrid screening. The binding activity of filamin C with nelin was confirmed by coim- munoprecipitation. Filamin C binds to nelin through its C-terminal region. 14Gene cloning and molecular breeding to improve fiber qualities in cotton显示文摘Cotton fiber is one of known natural resources comprising the highest purity cellulose. It plays an important role worldwide in the textile industry. With the acceleration of spinning speeds and the improvement of the peoples liv-ing level, the demand of improving cotton fiber qualities is getting stronger and stronger. So, making clear the develop-mental model of fiber cell and elucidating systematically the molecular mechanisms of cotton fiber development and regulation will produce a great significance to make full use of cotton gene resources, raise cotton yield and improve fiber quality, and even develop man-made fiber. In the paper, the status of the gene cloning and the molecular breeding to im-prove cotton fiber quality were reviewed, the importance and potential of gene cloning related with cotton fiber quality were put forward and the proposal and prospect on fiber quality improvement were made. Using national resources available and through the creative exploration in corre-sponding research, some international leading patents in genes or markers linked with cotton fiber development hav-ing Chinese own intellectual property should be licensed quickly. 15Inheritance of resistance to Helicoverpa armigera of 3 kinds of transgenic Bt strains available in upland cotton in China显示文摘There are 3 kinds of transgenic Bt strains, Shanxi 94-24, Zhongxin 94, and R19, in upland cotton in China. Their transgenic Bt insect-resistance cultivars or hybrids have been developed and grown by farmers. Genetic studies indicate that the resistance of the 3 transgenic Bt cotton strains to Helicoverpa armigera is controlled by one pair of non-allelic dominant genes. Linkage relationship between the resistant genes of R19 and Shanxi 94-24 transgenic Bt strains shows that they may be inserted in the same chromosome. FT hybrids crossed among the 3 strains show that high levels of protection from feeding damage are the same as that of their parents. Therefore, there is no co-suppression phenomenon in many transgenic plants. 18Genetic dissection of tetraploid cotton resistant to Verticillium wilt using interspecific chromosome segment introgression lines显示文摘Verticillium wilt(caused by the pathogen Verticillium dahliae) is of high concern for cotton producers and consumers. The major strategy for controlling this disease is the development of resistant cotton(Gossypium spp.) cultivars. We used interspecific chromosome segment introgression lines(CSILs) to identify quantitative trait loci(QTL) associated with resistance to Verticillium wilt in cotton grown in greenhouse and inoculated with three defoliating V. dahliae isolates. A total of 42 QTL, including 23 with resistance-increasing and 19 with resistancedecreasing, influenced host resistance against the three isolates. These QTL were identified and mapped on 18 chromosomes(chromosomes A1, A3, A4, A5, A7, A8, A9, A12, A13, D1, D2,D3, D4, D5, D7, D8, D11, and D12), with LOD values ranging from 3.00 to 9.29. Among the positive QTL with resistance-increasing effect, 21 conferred resistance to only one V. dahliae isolate, suggesting that resistance to V. dahliae conferred by most QTL is pathogen isolate-specific. The At subgenome of cotton had greater effect on resistance to Verticillium wilt than the Dt subgenome. 19Variation induced by DNA rearrangement in a transgenic Bt+CpTI cotton strain显示文摘In the development, of transgenic Bt + CpTI cotton cultivars, one male and female sterile mutant has been found in a homozygous T4 strain in our laboratory. The mutant plant, as well as its leaves, buds and flowers, is only 1/2-1/3 as large as that of the wild transgenic Bt + CpTI bivalant cotton plants. Cytological observation found that the chromosome number of the mutant is 2n=52; however, there are 4-8 univalents observed in meiosis I of pollen mother cells. Laboratory bioassay indicated that the mutant was highly resistant to bollworm as the wild plants. PCRamplification revealed that Bt and CpTl genes in the mutant were still intactly inserted. However, small deletion of flanked area had been observed in the mutant by Southern blotting analysis. So it is proposed that the mutant pheno-type might result from either the DNA deletion or T-DNA transferring in plant genome.
