| 1 | A high-quality genome sequence of alkaligrass provides insights into halophyte stress tolerance显示文摘Alkaligrass(Puccinellia tenuiflora) is a monocotyledonous halophytic forage grass widely distributed in Northern China. It belongs to the Gramineae family and shares a close phylogenetic relationship with the cereal crops, wheat and barley. Here, we present a high-quality chromosome-level genome sequence of alkaligrass assembled from Illumina, Pac Bio and 10× Genomics reads combined with genome-wide chromosome conformation capture(Hi-C) data. The ~1.50 Gb assembled alkaligrass genome encodes 38,387 protein-coding genes, and 54.9% of the assembly are transposable elements, with long terminal repeats being the most abundant. Comparative genomic analysis coupled with stress-treated transcriptome profiling uncovers a set of unique saline-and alkaline-responsive genes in alkaligrass. The high-quality genome assembly and the identified stress related genes in alkaligrass provide an important resource for evolutionary genomic studies in Gramineae and facilitate further understanding of molecular mechanisms underlying stress tolerance in monocotyledonous halophytes. | 2 | Creating burdock polysaccharide-oleanolic acid-ursolic acid nanoparticles to deliver enhanced anti-inflammatory effects:fabrication,structural characterization and property evaluation显示文摘This study explored the potential of polysaccharides from Actium lappa(ALPs)as natural wall materials for producing ALP-based nanoparticles to deliver poorly water-soluble oleanolic acid(OA)and ursolic acid(UA).Encapsulating OA+UA with ALPs(ALP:OA+UA,50:1;OA:UA,1:1)changed the crystalline nature to a more amorphous state through hydrogen bonding and involving O-H/C-O/O-C-O groups.ALP-OA/UA nanoparticles had a particle size and zeta potential(in water)of 199.1 nm/-7.15 mV,with a narrow unimodal size distribution,and excellent pH,salt solution,temperature and storage stability.Compared with ALPs,ALPOA/UA nanoparticles showed enhanced anti-inflammatory activity(especially at a dose of 100μg/mL)in a CuSO-induced zebrafish inflammation model via down-regulating the NF-κB signalling pathway and gene expression of associated transcription factors and cytokines(TNF-α,IL-1βand IL-8).Therefore,ALP-based nanoparticles are natural and anti-inflammatory carriers for hydrophobic bioactive molecules. | Shanshan Zhu Zhichang Qiu Xuguang Qiao Geoffrey I.N.Waterhouse Wenqing Zhu Wenting Zhao Qiuxia He Zhenjia Zheng | 2023 | Food Science and Human Wellness2023,12,2: | 2 |
| 3 | Lycium barbarum polysaccharides ameliorate canine acute liver injury by reducing oxidative stress,protecting mitochondrial function,and regulating metabolic pathways显示文摘The development of acute liver injury can result in liver cirrhosis,liver failure,and even liver cancer,yet there is currently no effective therapy for it.The purpose of this study was to investigate the protective effect and therapeutic mechanism of Lycium barbarum polysaccharides(LBPs)on acute liver injury induced by carbon tetrachloride(CCl_(4)).To create a model of acute liver injury,experimental canines received an intraperitoneal injection of 1 mL/kg of CCl_(4)solution.The experimental canines in the therapy group were then fed LBPs(20 mg/kg).CCl_(4)-induced liver structural damage,excessive fibrosis,and reduced mitochondrial density were all improved by LBPs,according to microstructure data.By suppressing Kelch-like epichlorohydrin(ECH)-associated protein 1(Keap1),promoting the production of sequestosome 1(SQSTM1)/p62,nuclear factor erythroid 2-related factor 2(Nrf2),and phaseⅡdetoxification genes and proteins downstream of Nrf2,and restoring the activity of anti-oxidant enzymes like catalase(CAT),LBPs can restore and increase the antioxidant capacity of liver.To lessen mitochondrial damage,LBPs can also enhance mitochondrial respiration,raise tissue adenosine triphosphate(ATP)levels,and reactivate the respiratory chain complexes I–V.According to serum metabolomics,the therapeutic impact of LBPs on acute liver damage is accomplished mostly by controlling the pathways to lipid metabolism.9-Hydroxyoctadecadienoic acid(9-HODE),lysophosphatidylcholine(Lyso PC/LPC),and phosphatidylethanolamine(PE)may be potential indicators of acute liver injury.This study confirmed that LBPs,an effective hepatoprotective drug,may cure acute liver injury by lowering oxidative stress,repairing mitochondrial damage,and regulating metabolic pathways. | JIANJIA HUANG YUMAN BAI WENTING XIE RONGMEI WANG WENYUE QIU SHUILIAN ZHOU ZHAOXIN TANG JIANZHAO LIAO RONGSHENG SU | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,2: | 2 |

| 6 | A Global Multiregional Proteomic Map of the Human Cerebral Cortex显示文摘The Brodmann area(BA)-based map is one of the most widely used cortical maps for studies of human brain functions and in clinical practice;however,the molecular architecture of BAs remains unknown.The present study provided a global multiregional proteomic map of the human cerebral cortex by analyzing 29 BAs.These 29 BAs were grouped into 6 clusters based on similarities in proteomic patterns:the motor and sensory cluster,vision cluster,auditory and Broca’s area cluster,Wernicke’s area cluster,cingulate cortex cluster,and heterogeneous function cluster.We identified 474 cluster-specific and 134 BA-specific signature proteins whose functions are closely associated with specialized functions and disease vulnerability of the corresponding cluster or BA.The findings of the present study could provide explanations for the functional connections between the anterior cingulate cortex and sensorimotor cortex and for anxiety-related function in the sensorimotor cortex.The brain transcriptome and proteome comparison indicates that they both could reflect the function of cerebral cortex,but show different characteristics.These proteomic data are publicly available at the Human Brain Proteome Atlas(www.brain-omics.com).Our results may enhance our understanding of the molecular basis of brain functions and provide an important resource to support human brain research. | Zhengguang Guo Chen Shao Yang Zhang Wenying Qiu Wenting Li Weimin Zhu Qian Yang Yin Huang Lili Pan Yuepan Dong Haidan Sun Xiaoping Xiao Wei Sun Chao Ma Liwei Zhang | 2022 | Genomics, Proteomics & Bioinformatics2022,20,4: | 0 |
