| 1 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 5 | TIMODAZ:A successful international cooperation project to investigate the thermal impact on the EDZ around a radioactive waste disposal in clay host rocks显示文摘Disposal of spent nuclear fuel and long lived radioactive waste in deep clay geological formations is one of the promising options worldwide. In this concept of the geological disposal system, the host clay formation is considered as a principal barrier on which the fulfillment of key safety functions rests. Between 2006 and 2010, the European Commission project TIMODAZ, which gathered 15 partners from 8 countries, has investigated the coupled thermo-hydro-mechanical (THM) effects on clay formations for geological disposal of radioactive waste, and specific attention was paid to investigating the thermal effect on the evolution of the damaged zone (DZ). Three types of potential host clay formations were investigated: the Boom Clay (Belgium), the Opalinus Clay (Switzerland) and the Callovo-Oxfordian argillite (France). Intensive experimental (laboratory and in situ in underground research laboratories) and numerical studies have been performed. Multi-scale approach was used in the course of the project. High degree of similarities between the failure modes, sealing process, stress paths, deformation, etc., observed in laboratories and in situ has been obtained, which increased the confidence in the applicability of laboratory test results and up-scaling perspective. The results of the laboratory and in situ tests obtained allowed the parameters for numerical models at various scales to be derived and provided the basis for the simplified performance assessment models that are used to assess the long-term safety of a repository. The good cooperation between the numerical modeler and experimenters has allowed an in-depth analysis of the experimental results and thus better understanding the underlying processes, and consequently increased the capabilities to model the THM effects in claystones. | 6 | Antimonene:a long-term stable two-dimensional saturable absorption material under ambient conditions for the mid-infrared spectral region显示文摘We experimentally demonstrate a long-term stable two-dimensional saturable absorption material under ambient conditions—multi-layer antimonene feasible for the mid-infrared spectral region—for the first time to our knowledge. The multi-layer antimonene material prepared using a liquid-phase exfoliation method was coated on a quartz/Ca F_2 for characterizations and an Au mirror as a reflection-type saturable absorber(SA) device. It has a modulation depth of 10.5%, a saturation peak intensity of 0.26 GW∕cm^2, and a non-saturation loss of 19.1% measured at 2868.0 nm using the typical power-dependent method. By introducing the SA device into a linear-cavity Ho^(3+)∕ Pr^(3+)-codoped fluoride fiber laser at 2865.0 nm, stable Q-switched pulses were obtained. It generated a maximum output power of 112.3 m W and pulse energy of 0.72 μJ, while the shortest pulse duration and largest repetition rate were 1.74 μs and 156.2 k Hz, respectively. The long-term stability of the SA device was also checked using the same laser setup within 28 days. | 7 | Prevalence of liver injury among patients with acquired immunodeficiency syndrome treated with highly active antiretroviral therapy in China显示文摘OBJECTIVE: To estimate the prevalence of liver injury among patients with acquired immunodeficiency syndrome(AIDS) who received highly active antiretroviral therapy(HAART) in rural Henan Province in China, and to explore whether Traditional Chinese Medicine(TCM) treatment based on HAART would increase this risk.METHODS: This was a retrospective cross-sectional study. We collected medical information on patients with AIDS from two treatment databases in2014. Criteria established by the AIDS Clinical Trials Group in 1996 were used for grading liver injury,classified based on the limit of normal(ULN) for alanine transaminase and aspartate aminotransferase:grade 1(1.25-2.5 × ULN); grade 2(2.6-5 × ULN);grade 3(5.1-10 × ULN); and grade 4(> 10 × ULN).Factors associated with liver injury were evaluated using a logistic regression model.RESULTS: A total 6953 patients with AIDS(3324 male and 3629 female patients) were enrolled into this study. The prevalence of liver injury was 22.0%(18.0% grade 1, 3.1% grade 2, 0.9% grade 3). In multivariate analysis, patients aged 34-45 years were more likely to have liver injury than patients in other age groups [adjusted odds ratio(AOR), 1.39; 95%CI, 1.01-1.91)]. Other factors associated with liver injury included male sex(AOR, 1.64; 95% CI,1.46-1.85), HIV infection via blood(AOR, 1.47; 95%CI, 1.19-1.82), hepatitis B virus antibody positive(AOR, 1.07; 95% CI, 0.85-1.36), and hepatitis C virus(HCV) antibody positive(AOR, 2.76; 95% CI,2.28-3.34).CONCLUSION: The prevalence of liver injury was relatively high among HAART-experienced patients. Several factors associated with liver injury included male sex, age 35-45 years old, HIV infection through blood, and concurrent HCV infection. There were no appreciable changes in the NE and NW wind systems and their wind fqrces that deposited dune sand, sandy loam and loess in the global cold stage since the last glacial epoch (accordingly no marked shifts of the boundaries of these aeolian facies belts took place. In the global warm stage since then, the climate in the Taklimakan desert and areas south of it became warm and dry, resulting in ablation of substantial volumes of ice and snow in their surrounding mountains and thus forming alluvial and diluvial deposits in the region. | 10 | Smartphone-powered iontophoresis-microneedle array patch for controlled transdermal delivery显示文摘The incidence rate of diabetes has been increasing every year in nearly all nations and regions.The traditional control of diabetes using transdermal insulin delivery by metal needles is generally associated with pain and potential infections.While microneedle arrays(MAs)have emerged as painless delivery techniques,the integration of MA systems with electronic devices to precisely control drug delivery has rarely been realized.In this study,we developed an iontophoresis-microneedle array patch(IMAP)powered by a portable smartphone for the active and controllable transdermal delivery of insulin.The IMAP in situ integrates iontophoresis and charged nanovesicles into one patch,achieving a one-step drug administration strategy of“penetration,diffusion and iontophoresis”.The MA of the IMAP is first pressed on the skin to create microholes and then is retracted,followed by the iontophoresis delivery of insulin-loaded nanovesicles through these microholes in an electrically controlled manner.This method has synergistically and remarkably enhanced controlled insulin delivery.The amount of insulin can be effectively regulated by the IMAP by applying different current intensities.This in vivo study has demonstrated that the IMAP effectively delivers insulin and produces robust hypoglycemic effects in a type-1 diabetic rat model,with more advanced controllability and efficiency than delivery by a pristine microneedle or iontophoresis.The IMAP system shows high potential for diabetes therapy and the capacity to provide active as well as long-term glycemic regulation without medical staff care. | Jingbo Yang Yanjun Li Rui Ye Ying Zheng Xiangling Li Yuzhen Chen Xi Xie Lelun Jiang | 2020 | Microsystems & Nanoengineering2020,6,1: | 3 |

| 14 | Soil physical properties,nutrients,and crop yield with two-year tillage rotations under a winter wheat-summer maize double cropping system显示文摘Winter wheat and summer maize were planted from 2015-2017 to study the effects of different rotational tillage patterns on soil physicochemical properties,crop yield,water content,and fertilizer utilization.The tillage treatments were designed as wheat subsoiling-maize no tillage(WS-MN),wheat rotary tillage-maize subsoiling(WR-MS),wheat subsoiling-maize subsoiling(WS-MS),and conventional wheat rotary tillage-maize no tillage(WR-MN)as a control.Among the four treatments,WS-MN and WR-MS were single-season subsoiling treatments,and WS-MS was a two-season subsoiling treatment.The average soil bulk density decreased by 7.6%in the single-and double-season subsoiling groups compared to the WR-MN group,and the total porosity and noncapillary porosity increased by 10.7%and 12.2%,respectively.Single-or double-season subsoiling treatment was not conducive to water storage in the 0-20 cm soil layer but increased the water content of the 20-140 cm soil layer,and the average soil water content of the 0-140 cm layer was increased by 11.6%in the two-growing season treatment groups compared with the WR-MN group.In WS-MS and WS-MN groups compared with the WR-MN group,the soil ammonium nitrogen content was increased by an average of 18.6%in 0-20 cm soil and 16.8%in 20-100 cm soil;soil nitrate-nitrogen content was decreased by 13.5%in 0-100 cm soil;total organic carbon and microbial carbon contents in the 15-30 cm soil were increased by 18.1%and 12.7%,respectively;and soil urease,catalase,and alkaline phosphatase activities were increased by 46.1%,15.2%,and 23.1%,respectively.Annual crop yield and water use efficiency increased by 8.9%and 15.0%,respectively,in both the single-and double-season subsoiling treatment groups.This study demonstrated the advantages of subsoiling tillage and suggested that it is suitable for crop cultivation in the Haihe Plain,China. | Baozhong Yin Pan Liu Yongwei Cui Zhaohui Hu Xiangling Li Zhihua Pan Wenchao Zhen | 2022 | International Journal of Agricultural and Biological Engineering2022,15,1: | 2 |

SOD has special physiological activity and is the primary substance for scavenging free radicals in living organisms.However, the expensive and complex extraction processes, low SOD yield, as well as difficult to store at room temperature have seriously hindered its application pace. Herein, the enzyme mimetic function of Mn-doped ZnS quantum dots(QDs) was discovered. The improved Marklund and McCord method both showed that Mn-doped ZnS QDs possess intrinsic SOD-like activity. | 16 | Circ_002059 suppresses cell proliferation and migration of gastric cancer via miR-182/MTSS1 axis显示文摘Circular RNAs(circRNAs)play either oncogenic or tumor suppressive roles in gastric cancer(GC).A previous study demonstrated that circ_002059,a typical circRNA,was downregulated in GC tissues.However,the role and mechanism of circ_002059 in GC development are still unknown.In this study,the levels of circ_002059,miR-182,and metastasis suppressor-1(MTSS1)were examined by real-time quantitative polymerase chain reaction and western blot analysis.Cell proliferation and migration were evaluated by MTT assay and Transwell migration assay,respectively.The interactions between miR-182 and circ_002059 or MTSS1 were analyzed by dual-luciferase reporter assay.A GC xenograft model was established to validate the role of circ_002059 in GC progression in vivo.Overexpression of circ_002059 significantly inhibited,whereas knockdown of circ_002059 notably facilitated,cell proliferation and migration in GC cells.MTSS1 was found to be a direct target of miR-182 and circ_002059 upregulated MTSS1 expression by competitively sponging miR-182.Transfection with miR-182 mimic and MTSS1 silencing abated the inhibitory effect of circ_002059 on GC progression.Circ_002059 inhibited GC cell xenograft tumor growth by regulating miR-182 and MTSS1 expression.Collectively,Circ_002059 inhibited GC cell proliferation and migration in vitro and xenograft tumor growth in mice,by regulating the miR-182/MTSS1 axis. | Ting Li Xiaomin Zuo Xiangling Meng | 2021 | Acta Biochimica et Biophysica Sinica2021,53,4: | 1 |
| 17 | Semi-Implantable Bioelectronics显示文摘Developing techniques to effectively and real-time monitor and regulate the interior environment of biological objects is significantly important for many biomedical engineering and scientific applications, including drug delivery, electrophysiological recording and regulation of intracellular activities. Semi-implantable bioelectronics is currently a hot spot in biomedical engineering research area, because it not only meets the increasing technical demands for precise detection or regulation of biological activities, but also provides a desirable platform for externally incorporating complex functionalities and electronic integration. Although there is less definition and summary to distinguish it from the well-reviewed non-invasive bioelectronics and fully implantable bioelectronics, semi-implantable bioelectronics have emerged as highly unique technology to boost the development of biochips and smart wearable device. Here, we reviewed the recent progress in this field and raised the concept of “Semi-implantable bioelectronics”, summarizing the principle and strategies of semi-implantable device for cell applications and in vivo applications, discussing the typical methodologies to access to intracellular environment or in vivo environment, biosafety aspects and typical applications. | 18 | Hybrid Satellite-UAV-Terrestrial Maritime Networks:Network Selection for Users on A Vessel Optimized with Transmit Power and UAV Position显示文摘The hybrid satellite-UAV-terrestrial maritime networks have shown great promise for broadband coverage at sea.The existing works focused on vessels collaboratively served by UAV-enabled aerial base station(ABSs)and terrestrial base stations(TBSs)deployed along the coast,and proved that data rate could be improved by optimizing transmit power and ABS’s position.In practice,users on a vessel can be collaboratively served by an ABS and a vesselenabled base station(VBS)in different networks.In this case,how to select the network for users on a vessel is still an open issue.In this paper,a TBS and a satellite respectively provide wireless backhaul for the ABS and the VBS.The network selection is jointly optimized with transmit power of ABS and VBS,and ABS’s position for improving data rate of all users.We solve it by finding candidates for network selection and iteratively solving transmit power and ABS’s position for each candidate.Simulation results demonstrate that data rate can be improved by collaborative coverage for users on a vessel. | Xiangling Li Wenjing Shi | 2022 | China Communications2022,19,9: | 1 |
| 19 | Differential gene expression in porcine SK6 cells infected with wild-type and SAP domain-mutant foot-and-mouth disease virus显示文摘Foot-and-mouth disease virus(FMDV) is the causative agent of a highly contagious disease in livestock. The viral proteinaseL^(rop) of FMDV is involved in pathogenicity, and mutation of theL^(rop) SAP domain reduces FMDV pathogenicity in pigs. To determine the gene expression profiles associated with decreased pathogenicity in porcine cells, we performed transcriptome analysis using next-generation sequencing technology and compared differentially expressed genes in SK6 cells infected with FMDV containingL^(rop) with either a wild-type or mutated version of the SAP domain. This analysis yielded 1,853 genes that exhibited a ≥ 2-fold change in expression and was validated by real-time quantitative PCR detection of several differentially expressed genes. Many of the differentially expressed genes correlated with antiviral responses corresponded to genes associated with transcription factors, immune regulation, cytokine production, inflammatory response, and apoptosis. Alterations in gene expression profiles may be responsible for the variations in pathogenicity observed between the two FMDV variants.
