| 1 | Genome-wide profiling of RNA editing sites in sheep显示文摘Background: The widely observed RNA-DNA differences(RDDs) have been found to be due to nucleotide alteration by RNA editing. Canonical RNA editing(i.e., A-to-I and C-to-U editing) mediated by the adenosine deaminases acting on RNA(ADAR) family and apolipoprotein B mRNA editing catalytic polypeptide-like(APOBEC)family during the transcriptional process is considered common and essential for the development of an individual.To date, an increasing number of RNA editing sites have been reported in human, rodents, and some farm animals;however, genome-wide detection of RNA editing events in sheep has not been reported. The aim of this study was to identify RNA editing events in sheep by comparing the RNA-seq and DNA-seq data from three biological replicates of the kidney and spleen tissues.Results: A total of 607 and 994 common edited sites within the three biological replicates were identified in the ovine kidney and spleen, respectively. Many of the RDDs were specific to an individual. The RNA editing-related genes identified in the present study might be evolved for specific biological functions in sheep, such as structural constituent of the cytoskeleton and microtubule-based processes. Furthermore, the edited sites found in the ovine BLCAP and NEIL1 genes are in line with those in previous reports on the porcine and human homologs, suggesting the existence of evolutionarily conserved RNA editing sites and they may play an important role in the structure and function of genes.Conclusions: Our study is the first to investigate RNA editing events in sheep. We screened out 607 and 994 RNA editing sites in three biological replicates of the ovine kidney and spleen and annotated 164 and 247 genes in the kidney and spleen, respectively. The gene function and conservation analysis of these RNA editing-related genes suggest that RNA editing is associated with important gene function in sheep. | 4 | Genome sequence of Gossypium anomalum facilitates interspecific introgression breeding显示文摘Crop wild relatives are an important reservoir of natural biodiversity. However, incorporating wild geneticdiversity into breeding programs is often hampered by reproductive barriers and a lack of accurate genomicinformation. We assembled a high-quality, accurately centromere-anchored genome of Gossypium anomalum, a stress-tolerant wild cotton species. We provided a strategy to discover and transfer agronomicallyvaluable genes from wild diploid species to tetraploid cotton cultivars. With a (Gossypium hirsutum 3 G.anomalum)2 hexaploid as a bridge parent, we developed a set of 74 diploid chromosome segment substitution lines (CSSLs) of the wild cotton species G. anomalum in the G. hirsutum background. This set of CSSLsincluded 70 homozygous substitutions and four heterozygous substitutions, and it collectively containedabout 72.22% of the G. anomalum genome. Twenty-four quantitative trait loci associated with plant height,yield, and fiber qualities were detected on 15 substitution segments. Integrating the reference genome withagronomic trait evaluation of the CSSLs enabled location and cloning of two G. anomalum genes thatencode peroxiredoxin and putative callose synthase 8, respectively, conferring drought tolerance andimproving fiber strength. | 8 | Health education improves effectively therapy of inpatients with hypertension显示文摘Objective: Low rate of medication usage and blood pressure(BP) control, bad lifestyle, poor cognition to hypertension, have been obstacles in prevention from hypertensive disease and complication in Chinese cohort. Health education appears to be an impendent measure to improve this situation. To study the effect of health education on the rate of medication usage and BP control, the lifestyle, and the incidence of complication among patients with hypertension. Methods: 284 hypertensive inpatients were divided randomly into two groups: the control group was intervened with routine medication; the education group was intervened with routine medication in combination with intensive health education according to the investigation on the risk factors of cardiocerebrovascular diseases and cognitive levels of hypertensive disease. During 12.16 ± 1.95 months of following up, the status of lifestyle, the rate of medication usage and BP control, and incidence of complications between two groups were analyzed. Results: In education group, the rate of smoking, alcohol intake, lower-salt ingesting, exercise, medication usage and BP control were improved after intervention and significantly different from those before intervention(P < 0.01). After intervention, the incidence of complications in education group and control group was 10.9% and 30.5% respectively(P < 0.01). | 9 | Recognition mechanism and sequence optimization of organophosphorus pesticides aptamers for better monitoring contaminations in food显示文摘Aptamers as a kind of biological recognition element have shown great potential in monitoring and the rapid quantification of organophosphorus pesticides(OPPs). However, molecules of OPPs are structurally similar and original aptamers selected by systematic evolution of ligands by exponential enrichment are usually long-chain bases, which hamper the further application under OPPs-aptamer recognition. The aim of the research was to develop a new strategy to design oligonucleotide sequences for binding OPPs by combination of experimental and molecular modeling methods. 3D models of aptamers binding OPPs were constructed, and binding energy and the most probable binding site for the OPPs were then determined by molecular docking, and the binding sites were further confirmed by the results of 2-AP replaced experiments. Based on the docking results, a new aptamer for detection 4 representative OPPs with only 29 bases was designed by reasonable truncation and mutation of the reported aptamer(named S4-29). The interaction between this new aptamer and OPPs were analyzed by molecular docking, microscale thermophoresis, circular dichroism and fluorometric analysis. | 11 | Highly improved cyclic stability of Ni-rich/Li batteries with succinic anhydride as electrolyte additive and underlying mechanism显示文摘Lithium-metal battery based on Ni-rich cathode provides high energy density but presents poor cyclic stability due to the unstable electrode/electrolyte interfaces on both cathode and anode.In this work,we report a new strategy to address this issue.It is found that the cyclic stability of Ni-rich/Li battery can be significantly improved by using succinic anhydride(SA) as an electrolyte additive.Specifically,the capacity retention of LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)(NCM811)/Li cell is improved from 14% to 83% after 200cycles at 1 C between 3.0 and 4.35 V by applying 5% SA.The underlying mechanism of SA contribution is understood by comparing the effects of malic anhydride(MA) and citraconic anhydride(CA), both of which share a similar molecular structure to SA but show different effects.On anode side,SA can but MA and CA cannot form a protective solid electrolyte interphase(SEI) on Li anode.On cathode side,three anhydrides can suppress the formation of hydrogen fluoride from electrolyte oxidation decomposition,but SA behaves best.Typically,MA shows adverse effects on the interface stability of Li anode and NCM811 cathode,which originates from its high acidity.Though the acidity of MA can be mitigated by substituting a methyl for one H atom at its C=C bond,the substituent CA cannot compete with SA in cyclic stability improvement of the cell,because the SEI resulting from CA is not as robust as that from SA,which is related to the binding energy of the SEI components.This understanding reveals the importance of the electrolyte acidity on the Ni-rich cathode and the robustness of the SEI on Li anode,which is helpful for rationally designing new electrolyte additives to further improve the cyclic stability of high-energydensity Ni-rich/Li batteries. | Shu Yang Guanjie Li Xiaoyan Lin Changyong Mo Xianggui Zhou Lijiao Quan Kuan Zhou Suli Li Hai Wang Weishan Li | 2023 | Journal of Energy Chemistry2023,,3: | 0 |

| 13 | Multiple Cavernous Hemangiomas of the Orbit显示文摘Purpose:Cavernous hemangioma of the orbit is often unilateral and solitary.Multiple cavernous hemangiomas of the orbit are extremely rare.The authors reported four patients who had more than two cavernous hemangiomas in one orbit.Methods:Case reports.The clinical features,imaging findings,differential diagnosis and surgical management were presented.Results:We found 4 cases of multiple cavernous hemangiomas of the orbit out of 214 records of all patients with orbital cavernous hemangiomas who were referred to us from Jan 1,1986 to Dec 31,2000.Two patients were female and two were male.The mean age was 46.5 years,ranging from 37.0 to 57.0 years.The left orbits were affected in all patients,with two,three,eight and ten tumors,respectively.The most common complaint was a painless,gradually progressive proptosis.The characteristic CT imaging features were multiple well circumscribed,round or ovoid masses that were sharply delineated from the surrounding tissue.B-scan ultrasonography showed round-shaped orbital masses with high internal reflectivity.The therapeutic outcomes of lateral or anterior orbitotomy were excellent.Conclusion:Although rare,cavernous hemangioma should be considered in the differential diagnosis of patients with multiple orbital mass lesions. | Xianggui Wang Jianhua Yan | 2011 | Eye Science2011,26,1: | 0 |
