1PKS5,a SNF1-related kinase,interacts with and phosphorylates NPR1,and modulates expression of WRKY38 and WRKY62显示文摘NPR1 (Nonexpressor of Pathogenesis-Related gene 1) is a major co-activator of plant defense. Phosphorylations of NPR1 play impor-tant roles in fine-tuning its activity, however a kinase corresponding to such modification remains uncharacterized. Here, we report that NPR1 interacts with PKS5 (SOS2-like Protein Kinase 5). The AKR (AnKyrin Repeats) motif of NPR1 is required for this interaction. PKS5 phosphorylates NPR1 at the C-terminal region. Expression of PKS5 is induced quickly by Pseudomonas syringae pv. tomato DC3000. Expression level of two NPR1 target genes, WRKY38 and WRKY62, is reduced and/or delayed in pks5 mutants. Moreover, the expression of WRKY38 and WRKY62 displays a similar pattern in npr1-1pks5-1 double mutant comparing to that in npr1-1. 2Enhanced Vitamin C Production Mediated by an ABA-Induced PTP-like Nucleotidase Improves Plant Drought Tolerance in Arabidopsis and Maize显示文摘Abscisic acid(ABA)is a key phytohormone that mediates environmental stress responses.Vitamin C,or L-ascorbic acid(AsA),is the most abundant antioxidant protecting against stress damage in plants.How the ABA and AsA signaling pathways interact in stress responses remains elusive.In this study,we characterized the role of a previously unidentified gene,PTPN(PTP-like Nucleotidase)in plant drought tolerance.In Arabidopsis,(AtPTPN was expressed in multiple tissues and upregulated by ABA and drought treatments.Loss-of-function mutants oiAtPTPN were hyposensitive to ABA but hypersensitive to drought stresses,whereas plants with enhanced expression AtPTPN showed opposite phenotypes to.Overexpression of maize PTPN(ZmPTPN)promoted,while knockdown oiZmPTPN inhibited plant drought tolerance,indicating conserved and positive roles of PTPN in plant drought tolerance.We found that both AtPTPN and ZmPTPN release Pi by hydrolyzing GDP/GMP/dGMP/IMP/dIMP,and that AtPTPN positively regulated AsA production via endogenous Pi content control.Consistently,overexpression of VTC2,the rate-limiting synthetic enzyme in AsA biosynthesis,promoted AsA production and plant drought tolerance,and these effects were largely dependent on AtPTPN activity.Furthermore,we demonstrated that the heat shock transcription factor HSFA6a directly binds the AtPTPN promoter and activates AtPTPN expression.Genetic analyses showed that AtPTPN is required for HSFA6a to regulate ABA and drought responses.Taken together,our data indicate that PTPN-mediated crosstalk between the ABA signaling and AsA biosynthesis pathways positively controls plant drought tolerance.Hui Zhang Yanli Xiang Neng He Xiangguo Liu Hongbo Liu Liping Fang Fei Zhang Xiaopeng Sun Delin Zhang Xingwang Li William Terzaghi Jianbing Yan Mingqiu Dai 2020Molecular Plant2020,13,5:7
3Structural Characteristics and their Significance for Hydrocarbon Accumulation in the Northern Slope of the Central Sichuan Paleo-uplift显示文摘Based on 2D and 3D seismic data,the latest drilling data and field outcrop data of the northern slope of the Central Sichuan paleo-uplift,the structural analysis method is used to analyze unconformity development characteristics and fault characteristics during the key structural transformation period,discussing the influence of the structural characteristics on the hydrocarbon accumulation of deep carbonate rocks.The results show that:(1)The two key unconformities of the Tongwan and Caledonian periods were primarily developed in deep carbonate rocks.Firstly,Tongwan’s unconformities are characterized by regional disconformities between the second and third members of the Dengying Formation,the top formation of the Sinian and the lower Cambrian,strips of which zigzag through the north and south sides of the study area.Secondly,the Caledonian unconformity is characterized by a regional unconformable contact between the lower Permian and the ower Paleozoic strata.From NE to SW,the age of the strata,which were subject to erosion,changes from new to old,the denudation distribution showing as a nose-shaped structure which inclines towards the ENE.(2)Boundary fault and transtensional strike-slip faults developed in the Sinian to Paleozoic strata.In profile,there are three types of structural styles:steep and erect,flower structures,’Y’and reversed’Y’type faults.In plane view,the Sinian developed extensional boundary faults extending in an almost NS direction,strike-slip faults developing and extending linearly in approximately EW,WNW and NE strikes in the Cambrian,with characteristically more in the south and less in the north.(3)The faults in the northern slope show obvious zonal deformations in transverse view as well as significant stages and stratified activity in a longitudinal direction.Among them,the activity of faults in the Sinian was the strongest,followed by the activity in the Cambrian period,the activity intensity of faults in the Permian period being the weakest.This fault activity can be divided into four periods:Sinian,Cambrian-Permian,the early Indosinian period and the late Indosinian-Himalayan period,the transtensional strikeslip faults being the products of oblique extensions of pre-existing weak zones in the Xingkai and Emei taphrogenesis,with a particular inheritance in the main faults.(4)Combined with hydrocarbon accumulation factors,it is considered that the epigenetic karstification of the Tongwan and Caledonian unconformities in the northern slope controlled the formation and distribution of carbonate karst reservoirs over a large area,also acting as a good pathway for oil and gas migration.The extensional faults developed at the margin of the NS trending rift,controlling the sag-platform sedimentary pattern in the Dengying Formation of the Sinian.Strike-slip faults in NE,WNW and ENE directions may control the microgeomorphological pattern inside the platform and intensify the differential distribution of grain beach facies.The multi-stage hereditary activity of strike-slip faults not only improved the porosity and permeability of the reservoirs,but also acted as the main channel of oil and gas migration,providing favorable conditions for the development of the current multi-layer gasbearing scenario in the northern slope of the Central Sichuan Basin.TIAN Xingwang LIU Guangdi LUO Bing YANG Yu WEN Long ZHANG Benjian CHEN Xiao SU Guiping SONG Zezhang PENG Hanlin ZHOU Gang YAN Wei YANG Dailin WANG Yunlong SUN Yiting DOU Shuang 2022Acta Geologica Sinica(English Edition)2022,96,4:2
4Texture evolution and slip mode of a Ti-5.5Mo-7.2Al-4.5Zr-2.6Sn-2.1Cr dual-phase alloy during cold rolling based on multiscale crystal plasticity finite element model显示文摘The complex micromechanical response among grains remains a persistent challenge to understand the deformation mechanism of titanium alloys during cold rolling.Therefore,in this work,a multiscale crystal plasticity finite element method of dual-phase alloy was proposed and secondarily developed based on LS-DYNA software.Afterward,the texture evolution and slip mode of a Ti-5.5Mo-7.2Al-4.5Zr-2.6Sn-2.1Cr alloy,based on the realistic 3D microstructure,during cold rolling(20%thickness reduction)were systematically investigated.The relative activity of the■slip system in theαphase gradually increased,and then served as the main slip mode at lower Schmid factor(<0.2).In contrast,the contribution of the■slip system to the overall plastic deformation was relatively limited.For theβphase,the relative activity of the<111>{110}slip system showed an upward tendency,indicating the important role of the critical resolved shear stress relationship in the relative activity evolutions.Furthermore,the abnormally high strain of very fewβgrains was found,which was attributed to their severe rotations compelled by the neighboring pre-deformedαgrains.The calculated pole figures,rotation axes,and compelled rotation behavior exhibited good agreement to the experimental results.Duoduo Wang Qunbo Fan Xingwang Cheng Yu Zhou Ran Shi Yan Qian Le Wang Xinjie Zhu Haichao Gong Kai Chen Jingjiu Yuan Liu Yang 2022Journal of Materials Science & Technology2022,,16:2
5In-Situ Synthesis of CQDs/BiOBr Material via Mechanical Ball Milling with Enhanced Photocatalytic Performances显示文摘Designing simple, efficient, and environmentally friendly methods to construct high-efficient photocatalysts is an important strategy to promote the further development of the field of photocatalysis. Herein, flower-like carbon quantum dots(CQDs)/Bi OBr composite photocatalysts have been prepared via in-situ synthesis by mechanical ball milling in the existence of ionic liquid. The CQDs/Bi OBr composites exhibit higher photo-degradation performance for tetracycline(TC) than Bi OBr monomer and the commercial Bi_(2)O_(3) under visible light irradiation. For comparison, the different Br sources and synthetic methods are chosen to prepare Bi OBr and CQDs/Bi OBr composites. Photocatalysts prepared by ball milling and ionic liquid present significantly enhanced photocatalytic performance for removing TC. In addition, the introduction of CQDs could distinctly enhance the photocatalytic performances of pure Bi OBr. The reason is that CQDs as electron acceptor effectively separate electrons and holes and inhibit their recombination. The intermediates during photocatalytic degradation were tested using liquid chromatography-mass spectrometry(LC-MS) and possible degradation pathways were given. During degradation, ·OH, O_(2)^(·-)and h^(+) were identified to be the main active species based on electron spin resonance(ESR) spectra and free radical trapping experiments. 8Exploring the genetic characteristics of 93-11 and Nipponbare recombination inbred lines based on the Golden Gate SNP assay显示文摘Understanding genetic characteristics in rice populations will facilitate exploring evolutionary mechanisms and gene cloning. Numerous molecular markers have been utilized in linkage map construction and quantitative trait locus(QTL) mappings. However, segregation-distorted markers were rarely considered, which prevented understanding genetic characteristics in many populations. In this study, we designed a 384-marker Golden Gate SNP array to genotype 283 recombination inbred lines(RILs) derived from 93-11 and Nipponbare Oryza sativa crosses. Using 294 markers that were highly polymorphic between parents, a linkage map with a total genetic distance of 1,583.2 c M was constructed, including 231 segregation-distorted markers. This linkage map was consistent with maps generated by other methods in previous studies. In total, 85 significant quantitative trait loci(QTLs) with phenotypic variation explained(PVE) values?5% were identified. Among them, 34 QTLs were overlapped with reported genes/QTLs relevant to corresponding traits, and 17 QTLs were overlapped with reported sterility-related genes/QTLs. Our study provides evidence that segregation-distorted markers can be used in linkage map construction and QTL mapping. Moreover, genetic information resulting from this study will help us to understand recombination events and segregation distortion. 9Epitaxial growth of large area ZrS_(2)2D semiconductor films on sapphire for optoelectronics显示文摘Recently,group-IVB semiconducting transition metal dichalcogenides(TMDs)of ZrS_(2) have attracted significant research interest due to its layered nature,moderate band gap,and extraordinary physical properties.Most device applications require a deposition of high quality large-area uniform ZrS_(2) single crystalline films,which has not yet been achieved.In this work,for the first time,we demonstrate the epitaxial growth of high quality large-area uniform ZrS_(2) films on c-plane sapphire substrates by chemical vapor deposition.An atomically sharp interface is observed due to the supercell matching between ZrS_(2) and sapphire,and their epitaxial relationship is found to be ZrS_(2)(0001)[1010]||Al_(2)O_(3)(0001)[1120].The epitaxial ZrS_(2) film exhibits n-type semiconductor behavior with a room temperature mobility of 2.4 cm^(2)·V^(−1)·s^(−1),and the optical phonon is the dominant scattering mechanism at room temperature or above.Furthermore,the optoelectronic applications of ZrS_(2) films are demonstrated by fabricating photodetector devices.The ZrS_(2) photodetectors exhibit the excellent comprehensive performance,such as a light on/off ratio of 106 and a specific detectivity of 2.6×10^(12) Jones,which are the highest values compared with the photodetectors based on other group-IVB two-dimensional TMDs.Yan Tian Yong Cheng Jidong Huang Siyu Zhang Hao Dong Gaokai Wang Jingren Chen Jinliang Wu Zhigang Yin Xingwang Zhang 2022Nano Research2022,15,7:1
10Direct growth of hexagonal boron nitride films on dielectric sapphire substrates by pulsed laser deposition for optoelectronic applications显示文摘Recently,hexagonal boron nitride(h-BN),an ultra-wide bandgap semiconductor,has attracted considerable atten-tion owing to its excellent properties.In thin films grown on metal catalysts,contamination and damage induced by a transfer process cannot be avoided.Therefore,synthesizing h-BN films on non-catalytic dielectric substrates is desirable for electronic applications.In this study,we demonstrate the direct growth of high-quality h-BN films with a controllable thickness on sapphire substrates by using the pulsed laser deposition(PLD)technique.The effects of the deposition conditions and laser parameters on the growth of the h-BN films are systematically investigated by evaluating their characteristic Raman peaks.Among the various growth parameters studied,the substrate temperature has the greatest influence on the crystalline quality of the h-BN films,and the optimal pres-sure varies depending on the target-substrate distance.The h-BN film grown under optimal conditions exhibits a narrow Raman line width of∼30 cm^(−1),indicating a high crystalline quality.The photodetectors fabricated from the PLD-grown h-BN films exhibit superior deep-ultraviolet detection performance with a large on/offratio of>104,high photoresponsivity,and a sharp cut-offwavelength of 220 nm.This study presents the possibility of producing high-quality h-BN films by applying PLD on dielectric substrates for optoelectronic applications.Gaokai Wang Jingren Chen Junhua Meng Zhigang Yin Ji Jiang Yan Tian Jingzhen Li Jinliang Wu Peng Jin Xingwang Zhang 2021Fundamental Research2021,1,6:1
11Mapping Multi-factor-mediated Chromatin Interactions to Assess Dysregulation of Lung Cancer-related Genes显示文摘Studies on the lung cancer genome are indispensable for developing a cure for lung cancer.Whole-genome resequencing,genome-wide association studies,and transcriptome sequencing have greatly improved our understanding of the cancer genome.However,dysregulation of longrange chromatin interactions in lung cancer remains poorly described.To better understand the three-dimensional(3D)genomic interaction features of the lung cancer genome,we used the A549 cell line as a model system and generated high-resolution chromatin interactions associated with RNA polymerase II(RNAPII),CCCTC-binding factor(CTCF),enhancer of zeste homolog 2(EZH2),and histone 3 lysine 27 trimethylation(H3K27me3)using long-read chromatin interaction analysis by paired-end tag sequencing(ChIA-PET).Analysis showed that EZH2/H3K27me3-mediated interactions further repressed target genes,either through loops or domains,and their distributions along the genome were distinct from and complementary to those associated with RNAPII.Cancer-related genes were highly enriched with chromatin interactions,and chromatin interactions specific to the A549 cell line were associated with oncogenes and tumor suppressor genes,such as additional repressive interactions on FOXO4 and promoter–promoter interactions between NF1 and RNF135.Knockout of an anchor associated with chromatin interactions reversed the dysregulation of cancer-related genes,suggesting that chromatin interactions are essential for proper expression of lung cancer-related genes.These findings demonstrate the 3D landscape and gene regulatory relationships of the lung cancer genome.Yan Zhang Jingwen Zhang Wei Zhang Mohan Wang Shuangqi Wang Yao Xu Lun Zhao Xingwang Li Guoliang Li 2023Genomics, Proteomics & Bioinformatics2023,21,3:0
12Identification and Expression of PPAR in Sinonovacula constricta and Their Potential Regulatory Effects on Δ6 Fad Transcription显示文摘Peroxisome proliferator activated receptors(PPAR)are kinds of key transcriptional factors in regulating LC-PUFA biosynthesis.Until now,little is known about PPAR in marine molluscs as well as in other invertebrates.Sinonovacula constricta is the first marine mollusc that is proved to possess the complete LC-PUFA biosynthetic pathway,and it can be a perfect representative to clarify this situation.In this study,the molecular properties of S.constricta PPAR were characterized,and corresponding potential regulatory roles in S.constrictaΔ6 fatty acyl desaturase(Fad)transcription were estimated by dual luciferase assay.Results showed that two PPAR homologs(PPAR_a and_b)were identified in S.constricta.They both contain typical features of vertebrate PPAR,suggesting highly conserved functional regions in PPAR.By phylogenetic comparison,they are clearly different with vertebrate PPAR and can be divided into two distinctive sub-groups.Moreover,they show a high expression level in gill,labial palps,mantle and intestine.Their down-regulated expressions in trochophore larva and veliger larva might be attributed to food-deprivation.Additionally,the transcriptional activity of S.constrictaΔ6 Fad promoter was significantly activated by both PPAR_a and_b,indicating that S.constricta PPAR might play a role in the regulation of LC-PUFA biosynthesis.To our knowledge,this is the first systematic analysis of PPAR in a marine mollusc.The results will provide a valuable reference for further researches on the function of marine molluscan PPAR and their underlying mechanisms in regulating LC-PUFA biosynthesis.RAN Zhaoshou KONG Fei LIAO Kai XU Jilin LIU Xingwang SHI Peng ZHANG Mengqi WU Kaibin YAN Xiaojun 2021Journal of Ocean University of China2021,20,6:0

14Alternative Splicing of OsRAD1 Defines C-Terminal Domain Essential for Protein Function in Meiosis显示文摘Alternative splicing can generate multiple mRNAs that differ in their untranslated regions or coding sequences,and these differences might affect mRNA stability or result in different protein isoforms with diverse functions and/or localizations.In this study,we isolated a sterile mutant in rice with abnormal meiosis of microspore mother cells and megaspore mother cells that carried a point mutation in OsRAD1 gene.Cloning of OsRAD1 cDNAs revealed three transcript variants,named as OsRAD1.1,OsRAD1.2 and OsRAD1.3,respectively,which were derived from alternative splicing of the last intron.Proteins derived from the three transcripts were mostly identical except the difference in the very C-terminal domain.The three transcripts exhibited similar expression patterns in various tissues,but the expression level of OsRAD1.1 was the highest.Specific knockout of OsRAD1.1 led to sterility,while knockout of OsRAD1.2 and OsRAD1.3 together did not change the plant fertility.Overexpression of OsRAD1.2 and OsRAD1.3 cDNAs in OsRAD1.1-specific mutant did not complement the plant fertility.Yeast two-hybrid assay showed that OsRAD1.1,but not OsRAD1.2 and OsRAD1.3,interacted with the three other meiosis proteins OsHUS1,OsRAD9 and OsRAD17,suggesting that the C-terminal domain of OsRAD1.1 is critical for the protein function.YUAN Shuting XU Chunjue YAN Wei CHANG Zhenyi DENG Xingwang CHEN Zhufeng WU Jianxin TANG Xiaoyan 2020Rice science2020,27,4:0
15Association between SLC17A7 gene polymorphisms and venlafaxine for major depressive disorder in a Chinese Han population:a prospective pharmacogenetic case-control study显示文摘Objective: Venlafaxine is a common antidepressant and its therapeutic effect varies among people with different genetic backgrounds. The aim of this study was to investigate whether single nucleotide polymorphisms (SNPs) in theSLC17A7 gene are associated with the treatment outcome of venlafaxine in a Chinese Han population with major depressive disorder.Methods: This prospective pharmacogenetic case-control study that involved genotyping of four SNPs ofSLC17A7 was conducted on 175 major depressive disorder patients of Chinese Han origin, aged 18 to 65 years, participated in the study from April 2005 to September 2006. Comparisons of allele and genotype frequencies of all SNPs were performed between the responder/remission group and the nonresponder/nonremission group. This study was approved by the Institutional Ethics Committee of Sichuan University (approval No. 20151112-265).Results: The allele and genotype frequencies of the four candidate SNPs inSCL17A7 showed no significant difference between responders and nonresponders. Meanwhile, no significant difference was detected in the four investigatedSLC17A7 SNPs between patients who did and did not exhibit remission. 16A one-pot process based on P_(44414)Cl-HCl aqueous biphasic system for recovering rare earth elements from NdFeB permanent magnet显示文摘Recovering critical metals from secondary resources have attracted great interest recently.In this work,a green one-pot leaching-extraction process based on tributyl(tetradecyl)phosphonium chloride (P_(44414)Cl)aqueous biphasic system (ABS) was developed to efficiently recover rare earth elements (REEs) from Nd Fe B permanent magnet.The reaction process,phase separation mechanism,and operation conditions were thoroughly investigated.It is found that the P_(44414)Cl-HCl ABS showed strong extraction ability towards Fe (>99%) whereas only a few REEs (<10%) were extracted,leading to extremely high separation selectivity between Fe and REEs.The characterization results showed that the coordination differences of Fe and Nd in HCl were the main driving forces for such highly selective separation.The phase diagram of P_(44414)Cl-Nd Cl_(3)ABS indicated that the salting-out effect of Nd Cl_(3)was stronger than common chlorides.Due to the hydrophobic property of P_(44414)[Fe Cl_(4)]and salting-out effect of Nd Cl_(3),the P_(44414)Cl could directly form ABS at room temperature after dissolving practical roasted Nd Fe B samples without any other operations and reagents.REEs and Fe could be mutually separated in just one step.Compared with traditional liquid-liquid extraction or ABS separation,this recovery process is green and facile and shows great application prospects in the field of rare-earth recovery.Chuanying Liu Qibin Yan Xingwang Zhang Lecheng Lei Chengliang Xiao 2022Chinese Chemical Letters2022,33,2:0

18Dynamic response and damage evolution of Zr-based bulk metallic glass under shock loading显示文摘Dynamic response and damage evolution of Zr_(70)Cu_(13)Ni_(9.8)Al_(3.6)Nb_(3.4)Y_(0.2) bulk metallic glass(Zr-based BMG)under impact pressure ranging from 4.03 GPa to 27.22 GPa were studied.The Hugoniot Elastic Limit(HEL)and the spalling Strength(σ_(sp))were measured as 7.09 GPa and 2.28 GPa,and the curve of impact velocity(D)and particle velocity(u)were also obtained.Under the strain rate of~10^(5)s^(-1),local crystallization phenomenon was observed.As increasing the impact pressure,the failure mode of Zr-based BMG changed from spallation to fragmentation caused by the combination of spalling cracks and longitudinal cracks.Cone-cup structures were also observed in the internal spalling zone via nano-CT characterization.When increasing the impact pressure,the thickness of Zr-based BMG increased after impact and the remelting and cladding layers were also observed on the fracture surfaces.The fragments of the specimen were welded after impact due to the high temperature remelting,which causes plastic deformation of Zr-based BMG under shock loading.Yan Li Xingwang Cheng Zhaolong Ma Xuhai Li Meng Wang 2021Journal of Materials Science & Technology2021,,34:0
19Correlation of PCSK1 with nonalcoholic fatty liver disease in a Han Chinese population:a case-control observational study显示文摘Objective: This study aimed to investigate the association between single-nucleotide polymorphisms (SNPs) ofPCSK1 (proprotein convertase subtilisin/kexin type 1) related to obesity and nonalcoholic fatty liver disease (NAFLD).Methods: In this case-control observational study, four candidate SNPs (rs6234, rs155971, rs6232, rs3811951) ofPCSK1 were genotyped in 732 NAFLD patients and 823 healthy control participants, all of whom were of ethnic Han Chinese descent. All participants came from Shanghai, China, and joined our study during 2015 to 2016. The frequencies of each allele and genotype, paired linkage disequilibrium, and haplotype were calculated on the SHEsis platform. In addition to SHEsis, five different genetic models (codominant, dominant, recessive, overdominant, and log-additive) were employed to identify the correlation between genotype frequency and NAFLD. This study was approved by the Medical Ethics Committee of Shanghai University of Traditional Chinese Medicine (approved No. 2017LCSY069).Results: In a comparison of NAFLD patients and healthy participants, none of the fourPCSK1 SNPs were significantly correlated with the occurrence of NAFLD (P>0.05), in either genotypic or allelic distribution.
