| 1 | Interaction between the gut microbiome and mucosal immune system显示文摘The gut microbiota, the largest symbiotic ecosystem with the host, has been shown to play important roles in maintaining intestinal homeostasis. Dysbiosis of the gut microbiome is caused by the imbalance between the commensal and pathogenic microbiomes. The commensal microbiome regulates the maturation of the mucosal immune system, while the pathogenic microbiome causes immunity dysfunction, resulting in disease development.The gut mucosal immune system, which consists of lymph nodes, lamina propria and epithelial cells, constitutes a protective barrier for the integrity of the intestinal tract. The composition of the gut microbiota is under the surveillance of the normal mucosal immune system. Inflammation, which is caused by abnormal immune responses,influences the balance of the gut microbiome, resulting in intestinal diseases. | 3 | Hepatitis B virus X gene induces human telomerase reverse transcriptase mRNA expression in cultured normal human cholangiocytes显示文摘AIM: To study the transcriptional regulation of human telomerase reverse transcriptase (hTERT) mRNA in normal human cholangiocytes (HBECs) after hepatitis B virus X (HBx) gene transfection and to elucidate the possible mechanism of HBV infection underlying cholangiocarcinoma.METHODS: HBECs were cultured in vitroand co-transfected with a eukaryotic expression vector containing the HBx coding region and a doning vector containing coding sequences of enhanced green fluorescent protein (EGFP) using lipidmediated gene transfer. The transfection efficiency was determined by the expression of EGFP. The expressions of hTERT mRNA and HBx protein in HBECs were detected by RT-PCR and immunocytochemical stain, respectively.RESULTS: The transfection efficiencies were about 15% for both HBx gene expression plasmid and empty vector.No hTERT mRNA was expressed in HBECs when transfected with OPTI-NEN medium and empty vector, but a dramatic increase was observed for hTERT mRNA expression in HBECs when transfected with HBx expression vector. HBx proyein was only expressed in HBECs when transfected with HBx expression vector.CONCLUSION: HBx transfection can activate thet ranscriptional expression of hTERT mRNA. | 6 | Experiment and simulation study on construction of a three-dimensional network model显示文摘The construction of a network model is one of the key techniques in organic com-bination of microscopic flow experiment and simulation. The construction method of a three-dimensional network model is presented on the basis of CT scanning images in this paper. A series of CT slice images describing microscopic pore structure and fluid distribution of actual rock is obtained with the help of the in-dustrial microfocus CT system. Based on the extraction of pore space skeleton, pore and throat information, the corresponding network model is established, and the conversion from three-dimensional CT image information to pore-throat size distribution and topological information is also achieved. The feature of this me-thod lies in the fact that complicated pore space of rock may be characterized by pores and throats with a simple shape while keeping the geometry and flow char-acteristics. It is indicated that the calculated results of porosity, permeability, rela-tive permeability curve and microscopic remaining oil distribution match very well the experimental results of water flooding and polymer flooding. The fundamental characteristic of IVIFS is that the values of its membership function and non-membership function are intervals rather than exact numbers. There are various averaging operators defined for IVIFSs. These operators are not monotone with respect to the total order of IVIFS, which is undesirable. This paper shows how such averaging operators can be represented by using additive generators of the product triangular norm, which simplifies and extends the existing constructions. Moreover, two new aggregation operators based on the ukasiewicz triangular norm are proposed, which are monotone with respect to the total order of IVIFS. | 10 | Sulindac induces apoptosis and protects against colon carcinoma in mice显示文摘AIM: To study the effect of sulindac on colon cancer induction in mice.METHODS: The chemo-preventive action of 80 ppm sulindac fed during initiation and post-initiation and 100 ppm sulindac fed during progressive stages of induction of colon carcinogenesis in mice was investigated using 1,2-dimethylhydrazine (DMH). Using the terminal deoxynucleotidyltransferase-mediated dUTP nick-end labeling (TUNEL)technique and PCNA immunohistochemical staining, we observed the apoptotic and proliferative cell density changes at different carcinogenic stages and the effect of sulindac on these two phenomena.RESULTS: Dietary sulindac significantly inhibited the incidence of colonic neoplasmas in mice. Compared with the control group, feeding sulindac during initiation and post-initiation stages inhibited the incidence by 46.7-50.4%,and feeding sulindac during progressive stages inhibited the incidence by 41.1%. Animals that were fed sulindac showed less serious pathological changes than those that were fed the control diet (P<0.01, H= 33.35). There was no difference in the density of proliferating cells among those groups which were or were not fed sulindac. In the same period, feeding sulindac resulted in a higher density of apoptotic cells than feeding control diet. CONCLUSION: Sulindac has an anti-carcinogenic function in mice. Its effect on preventing colon carcinogenesis is better than its effect on treating established tumors. By inducing apoptosis, sulindac inhibited the development of colon cancer and delayed canceration. Sulindac has no effect on proliferation. | 11 | ACBD3 is required for FAPP2 transferring glucosylceramide through maintaining the Golgi integrity显示文摘Glycosphingolipid (GSL) metabolism is involved in various physiological processes, including all major cell signaling pathways, and its dysregulation is linked to some diseases. The four-phosphate adaptor protein FAPP2-mediated glucosylceramide (GlcCer) transport for complex GSL synthesis has been studied extensively. However, the molecular machinery of FAPP2 as a GlcCer-transferring protein remains poorly defined. Here, we identify a Golgi-resident protein, acyl-coenzyme A binding domain containing 3 (ACBD3), as an interacting partner of FAPP2. We find that ACBD3 knockdown leads to dramatic Golgi fragmentation, which subsequently causes FAPP2 dispersal throughout the cytoplasm and a decreased localization at trans-Golgi network. The further quantitative Upidomic analysis indicates that ACBD3 knockdown triggers abnormal sphingolipid metabolism. Interestingly, the expression of siRNA-resistant full-length ACBD3 can rescue these defects caused by ACBD3 knockdown. | 13 | Redox-sensitive micelles for targeted intracellular delivery and combination chemotherapy of paclitaxel and all-trans-retinoid acid显示文摘The application of paclitaxel(PTX) in clinic has been restricted due to its poor solubility.Several traditional nano-medicines have been developed to improve this defect,while they are still lack of tumor targeting ability and rapid drug release. In this work,an amphiphilic polymeric micelle of hyaluronic acid(HA) – all-trans-retinoid acid(ATRA) with a disulfide bond,was developed successfully for the co-delivery of PTX and ATRA. The combination chemotherapy of PTX and ATRA can strengthen the anti-tumor activity. Along with selfassembling to micelles in water,the delivery system displayed satisfying drug loading capacities for both PTX(32.62% ± 1.39%) and ATRA,due to directly using ATRA as the hydrophobic group. Rapid drug release properties of the PTX-loaded redox-sensitive micelles(HA-SS-ATRA) in vitro were confirmed under reducing condition containing GSH. Besides,HA-CD44 mediated endocytosis promoted the uptake of HA-SS-ATRA micelles by B16 F10 cells. Due to these properties,cytotoxicity assay verified that PTX-loaded HA-SS-ATRA micelles showed concentration-dependent cytotoxicity and displayed obvious combination therapy of PTX and ATRA. Importantly,HA-SS-ATRA micelles could remarkably prolong plasma circulation time after intravenously administration. | 16 | Effects of Shear Fracture on In-depth Profile Modification of Weak Gels显示文摘Two sand packs were filled with fine glass beads and quartz sand respectively.The characteristics of crosslinked polymer flowing through the sand packs as well as the influence of shear fracture of porous media on the in-depth profile modification of the weak gel generated from the crosslinked polymer were investigated.The results indicated that under the dynamic condition crosslinking reaction happened in both sand packs,and the weak gels in these two cases became small gel particles after water flooding.The differences were:the dynamic gelation time in the quartz sand pack was longer than that in the glass bead pack.Residual resistance factor(FRR)caused by the weak gel in the quartz sand pack was smaller than that in the glass bead pack.The weak gel became gel particles after being scoured by subsequent flood water.A weak gel with uniform apparent viscosity and sealing characteristics was generated in every part of the glass bead pack,which could not only move deeply into the sand pack but also seal the high capacity channels again when it reached the deep part.The weak gel performed in-depth profile modification in the glass bead pack,while in the quartz sand pack,the weak gel was concentrated with 100 cm from the entrance of the sand pack.When propelled by the subsequent flood water,the weak gel could move towards the deep part of the sand pack but then became tiny gel particles and could not effectively seal the high capacity channels there.The in-depth profile modification of the weak gel was very weak in the quartz sand pack.It was the shear fracture of porous media that mainly affected the properties and weakened the in-depth profile modification of the weak gel. | Li Xianjie Song Xinwang Yue Xiang'an Hou Jirui Fang Lichun Zhang Huazhen | 2007 | Petroleum Science2007,4,1: | 2 |

According to the results,there were three genotypes( AA,AB and BB) of NPY gene in Donglan black-bone chicken group. Different genotypes exhibited significant effects( P < 0. 05) on the total number of eggs at 300 days of age. The total number of eggs at 300 days of age of AA genotype was significantly higher than that of BB genotype( P < 0. 05).
