| 1 | Blockade of cholecystokinin-2 receptor and cyclooxygenase-2 synergistically induces cell apoptosis, and inhibits the proliferation of human gastric cancer cells in vitro显示文摘Gastrin and cyclooxygenase-2(COX-2) playimportant roles in the carcinogenesis and progression ofgastric cancer.However,it remains unknown whether the combination of cholecystokinin-2(CCK-2) receptor antagonist plus COX-2 inhibitor exerts synergistic anti-tumor effects on human gastric cancer.Here,we demonstrated that the combination of AG-041R(a CCK-2 receptor antagonist) plus NS-398(a selective COX-2 inhibitor) treatment had synergistic effects on proliferation inhibition,apoptosis induction,down-regulation of Bcl-2 and up-regulation of Bax expression in MKN-45 cells.These results indicate that simultaneous targeting of CCK-2 receptor and COX-2 may inhibit gastric cancer development more effectively than targeting either molecule alone.(C)2008 Elsevier Ireland Ltd.All rights reserved. | Sun, W. | 4 | Polymorphisms of MTHFD,plasma homocysteine levels, and risk of gastric cancer in ahigh-risk Chinese population显示文摘Purpose:Accumulative evidence suggests that folate has a protective effect on gastric cancer. The methylenetetrahyd-rofolate dlehydrogenase(MTHFD) plays an important role in folate and homocysteine metabolisms, and polymorphisms of MTHFD may result in disturbance of the folate-mediated homocysteine pathway. The aim of this study is to test the hypothesis that genetic variants of MTHFD and plasma homocysteine levels are associated with risk of gastric cancer and modulated by genotypes of methylenetetrahydrofolate reductase(MTHFR). Experimental Design: We genotyped G1958A and T401C in MTHFD and C677T in MTHFR and detected total plasma homocysteine(tHcy) levels in a case-control study of 589 gastric cancer cases and 635 cancer-free controls in a high-risk Chinese population. Results:The variant genotypes of MTHFD 1958AA and 401CC were associated with a significantly increased risk of gastric canceradjusted odds ratio(OR), 2.05; 95% confidence interval(95% CI),1.34-3.13 for 1958AA; adjusted OR,1.43; 95% CI,1.14-1.80 for 401CC compared with 1958GG/GA and 401TT/TC genotypes, respectively. Both of the effects were more evident in the subjects carrying MTHFR 677CT/TT genotypes. The average tHcy level was significantly higher in gastric cancer cases than in controls(P < 0.01), and the upper quartile of tHcy (> 13.6 mu mol/L) was associated with an 82% significantly increased risk of gastric cancer, compared with the lowest quartile of tHcy(<= 8.0 pmol/L;adjusted OR,1.82; 95% CI,1.20-2.75). Conclusions:The strong associations between MTHFD variants and the plasma tHcy levels and gastric cancer risk suggest, for the first time, a possible gene-environment interaction between genetic variants of folate-metabolizing genes and high tHcy levels in gastric carcinogenesis. | Wang, L. | 5 | BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Thus, BubR1-mediated CENP-E phosphorylation produces a temporal switch that enables transition from lateral to end-on microtubule capture and organization of microtubules into stable midzone arrays. | Yuejia Huang Lin Lin Xing Liu Sheng Ye Phil Y. | 9 | NUMERICAL SIMULATION OF FORGING AND SUBSEQUENT HEAT TREATMENT OF A ROD BY A FINITE VOLUME METHOD显示文摘A method to simulate processes of forging and subsequent heat treatment of an axial symmetric rod is formulated in eulerian description and the feasibility is investigated. This method uses finite volume mushes for troching material deformation and an automatically refined facet surface to accurately trace the free surface of the deforming material.In the method,the deforming work piece flows through fixed finite volume meshes using eulerian formulation to describe the conservation laws,Fixed finite volume meshing is particularly suitable for large three-dimensional deformation such as forging because remeshing techniques are not required, which are commonly considered to be the main bottelencek in the ssimulations of large defromation by using the finite element method,By means of this finite volume method, an approach has been developed in the framework of 'metallo-thermo-mechanics' to simulate metallic structure, temperature and stress/strain coupled in the heat treatment process.In a first step of simulation, the heat treatment solver is limited in small deformation hypothesis,and un- coupled with forging. The material is considered as elastic-plastic and takes into account of strain, strain rate and temperature effects on the yield stress.Heat generation due to deformation,heat con- duction and thermal stress are considered.Temperature - dependent phase transformation,stress-in- duced phase transformation,latent heat,transformation stress and strain are included.These ap- proaches are implemented into the commerical commercial computer program MSC/SuperForge and a verification example with experimental date is given as comparison. | P. | 10 | Interfacial bonding mechanism in Al/coated steel dissimilar refill friction stir spot welds显示文摘Defect-free dissimilar Al/zinc coated steel and Al/AlSi coated steel welds were successfully fabricated by refill friction stir spot welding. However, Al alloy and uncoated steel could not be welded under the same welding condition. Al-Zn eutectic layer formed at the Al/zinc coated steel interface showed nonuniformity in thickness and nanoscale intermetallic(IMC) produced was discontinuous. The bonding formation between the Al-Zn layer and the surrounding materials was attributed to a liquid/solid reaction mechanism. Bonding formation at Al alloy and AlSi coated steel interface was attributed to a solid/solid reaction mechanism, as the joining process did not involve with melting of base metals or AlSi coating materials. Kissing bond formed at the weld boundary acted as a crack initiation and propagation site,and the present study showed that weld strength of Al 5754/AlSi coated steel was greatly influenced by properties of original IMC layer. | Z. | 12 | Pathological and biochemical alterations of astrocytes in ovariectomized rats injected with D-galactose:A potential contribution to Alzheimer's disease processes显示文摘Astrocytes are implicated in the pathological changes of Alzheimer's disease. Our previous studies have demonstrated that estrogen deprivation and oxidative stress act synergistically to accelerate the progress of Alzheimer's disease. Long-term D-galactose injection combined with ovariectomy may serve as a rodent model for Alzheimer's disease. To address the potential contribution of astroglia to the Alzheimer's disease pathogenesis, we investigated pathological and biochemical alterations of astrocytes under this animal model. Ovadectomized rats injected with D-galactose for 2 weeks showed extensive localization of glial fibrillary acidic protein immunoreactive astrocytes and slightly elevated glutathione levels in the hippocampus without significant impairments in the water maze test and deficits of the cholinergic analyses, compared to the saline-injected rats. Ovariectomized rats injected with D-galactose for 6 weeks, however, exhibited degeneration of astrocytes and decreased glutathione levels in the hippocampus, accompanied with severe dysfunction of behavioral test and deficiency of cholinergic terminals. Electron microscopy further confirmed the pathological changes of astrocytes, especially in the aggregated area of synapse and brain microvessels. Consistent with degeneration of perivascular astrocytic endfeet, analysis of the horseradish peroxidase demonstrated an impairment of the blood-brain barrier permeability. These findings indicate that biochemical and pathological alterations of astrocytes may partially contribute to exacerbating neuronal deficits in the course of Alzheimer's disease. | 13 | A transmission electron microscopy study of microscopic causes for localized-corrosion morphology variations in the AA7055 Al alloy显示文摘By Using(scanning) transmission electron microscopy, localized-corrosion morphology variations of the AA7055 AlZn(Cu)Mg alloy with different thermal processes and their underlying microscopic causes were investigated systematically. Our study shows that the corrosion resistance of the nanoscale precipitates varies with their structure type and Cu-content. Just like the Al-matrix, the early-stage precipitates are corrosion resistant, as compared with the η_p/η-precipitates without high Cu-content. With a high Cucontent, however, the η-precipitates become most corrosion resistant among all phases involved. Hence,tailoring the precipitate microstructure and chemistry though thermal processes may change the overall corrosion morphology and improve corrosion resistance property of the alloy. | X.B.
