2Changes in circulating Foxp3^+ regulatory T cells and interleukin-17-producing T helper cells during HBV-related acute-on-chronic liver failure显示文摘AIM:To longitudinally investigate cytokine gene expression and protein levels in Th17 and Treg cells,to observe T-cell phenotypes during hepatitis B virus(HBV)-related acute-on-chronic liver failure(ACHBLF)and to analyze changes in Th17 and Treg phenotypes during disease progression.METHODS:We measured the expression of seven Th17/Treg differentiation-related genes and serum concentrations of the corresponding cytokines in 18ACHBLF,18 chronic hepatitis B(CHB)disease controls and 10 healthy controls(HCs)by real-time quantitative pCR and enzyme linked immunosorbent assay.peripheral Th17 and Treg cell frequencies were analyzed by flow cytometry.RESULTS:From the onset of ACHBLF,patients presented with a conductive Th17 differentiation cytokine environment accompanied by high Th17 frequency and high serum IL-17 levels,which were sustained throughout the disease course.The Treg-related cytokine IL-2and Foxp3 were also up-regulated from disease onset,and Foxp3 gene expression showed a gradually increasing trend during ACHBLF.The circular phenotype of Treg and Th17 cells showed changes from the onset of ACHGLF.At disease onset,Th17 frequency increased significantly compared with both CHB and HCs,but Treg cell frequency decreased significantly compared with CHB.During the ACHBLF event,Th17 frequency remained higher compared with HCs,but decreased sharply from the peak point to the recovery point;Treg cell frequency increased gradually during the ACHBLF event.Treg and Th17 cell counts correlated with ACHBLF development;in all patients,serum IL-17 levels significantly correlated with patient serum ALT levels.In survivors,Th17 frequency at the onset point and the Treg to Th17 ratio at the peak point correlated with the patient’s model for end stage liver disease(MELD)plus sodium(MELD-Na)score.The Treg to Th17 ratio and the Th17 frequency at onset were significant predictors of patient survival.Low Treg/Th17 cell ratios at the onset predicted poor survival.Survivors exhibited an initial decrease in the circulating Treg/Th17 ratio from the onset to the peak time,and subsequently displayed a continuous increase.CONCLUSION:Treg and Th17 cells showed changes in genes,protein levels and T cell phenotypes during ACHBLF events.An increased Treg/Th17 ratio was associated with the survival of ACHBLF patients.Xue-Song Liang Cheng-Zhong Li Yin Zhou Wei Yin Ya Yun Liu Wen-Han Fan 2014World Journal of Gastroenterology2014,20,26:44
3The Relationship between Glucose Excursion and Cognitive Function in Aged Type 2 Diabetes Patients显示文摘Objective Evidence suggests that type 2 diabetes (T2DM) is associated with an increased risk of dementia and that glucose variability is an independent risk factor for diabetic complications.This study investigated the relationship between glucose excursion and cognitive function in aged T2DM patients.Methods A total of 248 aged T2DM patients wore a continuous glucose monitoring system (CGMS) for 3 days in order to evaluate glucose excursion,including mean amplitude of glycemic excursions (MAGE) and mean of daily difference (MODD).All subjects were evaluated with a number of accepted cognitive function tests,including themini‐mental status examination (MMSE).The relationship between MAGE and MODD and performance on these cognitive tests was assessed.Results The MAGE and MMSE score were negatively correlated,likewise with the correlation between MODD and MMSE.Liner multivariate regression analysis showed that MAGE and MODD were also negatively related to MMSE independent of age,sex,glycemic control,hypertension,smoking,or coronary heart disease history.Conclusion Glucose excursion is related to cognitive function in aged T2DM patients.Elevated glucose excursion decreased the MMSE score,which reflects general cognitive function.Thus,therapy aimed at controlling glucose excursion may be beneficial for maintaining cognitive function in aged T2DM patients.ZHONG Yuan ZHANG Xiao Yan MIAO Ya ZHU Jie Hua YAN Hong WANG Bei Yun JIN Jun HU Ting Jun JIA Wei Ping 2012Biomedical and Environmental Sciences2012,25,1:34

5Second harmonic generation in a high-Q lithium niobate microresonator fabricated by femtosecond laser micromachining显示文摘We report on second harmonic generation(SHG) in on-chip high-Q(>105) lithium niobate(Li Nb O3, LN) microresonators fabricated by femtosecond laser micromachining. We examine the efficiency of SHG with either a continuous-wave(CW) or an ultrashort pulsed pump laser. 8Echinococcus Granulosus 14-3-3 Protein:A Potential Vaccine Candidate Against Challenge with Echinococcus Granulosus in Mice显示文摘Objective To investigate the protective immunity against Echinococcus granulosus in mice immunized with rEg14-3-3.Methods ICR mice were subcutaneously immunized three times with rEg14-3-3,followed by the challenge with Echinococcus granulosus protoscoleces intraperitoneally and then sacrificed after six months of post-challenge to detect the proliferation of splenocytes by MTT assay,and to measure the secretion of IL-2,IL-4,IL-10,and IFN-γ by ELISA.The rate of reduced hydatid cyst and the levels of IgE,IgG and IgG subclasses in sera were examined.Results Mice vaccinated with rEg14-3-3 and challenged with protoscoleces revealed significant protective immunity of 84.47%.ELISA analysis indicated that the immunized mice generated specific high levels of IgG and the prevailing isotypes of IgG were IgG1 and IgG2a.Splenocytes from mice immunized with rEg14-3-3 showed a significant proliferation response.The secretion of IFN-γ and IL-2 increased significantly in the vaccinated mice whereas there was no significant difference in IL-4 and IL-10 levels between vaccinated and control mice.Conclusion The results indicate that the rEg14-3-3 vaccine could induce a high level of protective immunity as a promising vaccine candidate to prevent cystic echinococcosis.LI Zong Ji WANG Ya Na WANG Qi ZHAO Wei 2012Biomedical and Environmental Sciences2012,25,3:18
9Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 10Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. 12Late Pleistocene glaciation of the Hulifang Massif of Gongwang mountains in Yunnan Province显示文摘Late Pleistocene glaciation was restricted to only a few high mountains in eastern China. The Gongwang mountainsconstitute one ofthe typicalplacesonce glaciated.Geomorphic mapping of the area and the TL dating provides evidence for at least four distinct glaciations. YJT-I glacial advance occurred about100 ka BP and two TL absolute ages(101,100 ±7780 a BP;104,000±8300 a BP)indicate thisadvance happened during the Penultimale Glaciation.The early stage glacialadvance (YJT-II advance) during the last glaciation occurred about 40,920±3400 a BP. The last glacial maximum advance (YJT-IIIadvance)about18-25 ka BP,which sustained by two TL ages(18,230 ± 1420 a BP;25,420 ±2110 a BP).The Penultimale and the early stage glaciationswere more extensive and the lastglacialmaximum (LGM )and the late-glacialperiod (YJT-IV advance,10 ka BP) were progressively less extensive. 14Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 15Hyperinsulinemia,Insulin Resistance and Cognitive Decline in Older Cohort显示文摘Objective Type 2 diabetes has been recently recognized as an important risk factor for cognitive decline of patients with Alzheimer's disease (AD).But the roles of hyperinsulinemia (HI) and insulin resistance (IR) in the development of AD are still controversial.This study was designed to evaluate whether HI or IR influenced the cognitive functions of older cohort.Methods The cognitive functions of 328 consecutive elderly patients were evaluated with a battery of cognitive rating scales.Their fasting blood glucose (FBG) and fasting insulin (FINS) were analyzed and IR was calculated with modified‐Homa.The cognitive scores in different groups and the correlation of cognitive functions with HI or IR were analyzed.Results In our study,there were 180 participants with HI and 148 without HI,and 192 with IR and 136 without IR.The participants with HI showed worse cognitive functions than those without HI in MMSE,MOCA,CDR,orientation,delayed memory,and attention/calculation domains.Similarly,the elderly with IR had lower cognitive scores than those without IR in MMSE,MOCA,CDR,GDS,orientation,delayed memory,and attention/calculation domains.The insulin levels and Homa IR had negative correlation with the scores of MMSE and delayed memory,not only in the model 1 adjusted for FBG and diabetes history,but also in the model 2 adjusted for all nine demographic characteristics.Conclusion HI and IR are important risk factors for cognitive decline of the elderly,especially for the dysfunctions in delayed memory domains.ZHONG Yuan MIAO Ya JIA Wei Ping YAN Hong WANG Bei Yun JIN Jun 2012Biomedical and Environmental Sciences2012,25,1:11
16TNF-α and IL-1RA Polymorphisms and Silicosis Susceptibility in Chinese Workers Exposed to Silica Particles:A Case-Control Study显示文摘Objective To assess the association of TNF- α and IL-1RA SNPs with the risk of silicosis in Chinese workers exposed to silica particles. Methods Case-control study design was used to enroll 68 silicotic patients induced by silica particles and 68 healthy workers matched for length of silica particle exposure as controls. Both cases and controls were from the same company in southwest China, and each of them was requested to complete a questionnaire. Blood samples were drawn for genomic DNA extraction from each participant. The genotyping of TNF-α (-238 and -308) and IL-1RA (+2018) was performed using polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP) and SYBR green-based quantitative polymerase chain reaction (qPCR), respectively. Unconditional logistic regression model was used to estimate odds ratios (ORs) and their 95% confidential intervals (CI) for SNPs. Results No significant differences were found between cases and controls in particles exposure length, body mass index (BMI), and status of smoking and alcohol consumption except for age (P=0.001) and blood type (P=0.042). The frequencies of TNF-α (-238) and IL-1RA (+2018) genotypes in cases were significantly different from those in controls, (P=0.001 and P=0.002, respectively), while a borderline significant difference was found in the frequencies of TNF-α (-308) between cases and controls (P=0.063). The variants of three SNPs increased the risk of silicosis in the Chinese workers exposed to silica particles. The adjusted ORs of TNF-α (-308), TNF-α (-238) and IL-1RA (+2018) were 2.8 (95% CI: 1.1-7.5), 20.9 (95% CI: 1.8-236.4) and 4.0 (95% CI: 1.6-10.1), respectively. Conclusion It is suggested that cytokine polymorphisms of TNF- α (-238, -308) and IL-1RA (+2018) are associated with the risk of silicosis in the Chinese workers exposed to silica particles. 17Identification and Fine Mapping of a Gene Related to Pale Green Leaf Phenotype near the Centromere Region in Rice(Oryza sativa)显示文摘A thermo-insensitive pale green leaf mutant (pgl2) was isolated from T-DNA inserted transgenic lines of rice (Oryza sativa L. subsp. japonica cv. Nipponbare). Genetic analysis indicated that the phenotype was caused by a recessive mutation in a single nuclear-encoded gene. To map the PGL2 gene, an F2 population was constructed by crossing the mutant with Longtefu (Oryza sativa L. subsp. indica). The PGL2 locus was roughly linked to SSR marker RM331 on chromosome 8. To finely map the gene, 14 new InDel markers were developed around the marker, and PGL2 was further mapped to a 2.37 Mb centromeric region. Analysis on chlorophyll contents of leaves showed that there was no obvious difference between the mutant and the wild type in total chlorophyll (Chl) content, while the ratio of Chl a / Chl b in the mutant was only about 1, which was distinctly lower than that in the wild type, suggesting that the PGL2 gene was related to the conversion between Chl a and Chl b. Its performances were compared with that prepared by the traditional liquid phase reaction method. The results demonstrate that the electrocatalytic activity of Pt/C catalyst with solid phase reaction method for methanol oxidation is higher than that with liquid phase reaction method. 20Dendrobium officinale Kimura et Migo and American ginseng mixture: A Chinese herbal formulation for gut microbiota modulation显示文摘Dendrobium officinale Kimura et Migo(D. officinale) is a famous traditional Chinese medicine(TCM). A mixture of D. officinale and American ginseng has been shown to enhance cell-mediated immunity, humoral immunity, and monocyte/macrophage functions in mice. Here, the effects of a D. officinale and American ginseng mixture on the structure of gut microbial community in dogs were examined using high-throughput 16S rRNA gene amplicon sequencing. The data revealed that while the mixture did not change the diversity of gut microbial community significantly, differences among individuals were significantly reduced. Furthermore, the mixture-responsive operational taxonomic units(OTUs) exhibited a phase-dependent expression pattern. Fifty-five OTUs were found to exhibit a mixture-induced expression pattern, among which one third were short-chain fatty acid(SCFA)-producing genera and the others were probiotic genera included Lactobacillus spp., Sutterella, Alistipes, Anaerovorax, Bilophila, Coprococcus, Gordonibacter, Oscillibacter, among others. By contrast, 36% of the OTUs exhibiting a mixture-repressed expression pattern were disease-associated microorganisms, and six genera, namely Actinomyces, Escherichia/Shigella, Fusobacterium, Slackia, Streptococcus and Solobacterium, were associated with cancer. In addition, five genera were closely associated with diabetes, namely Collinsella,Rothia, Howardella, Slackia and Intestinibacter.
