| 1 | Discovery of mitochondrial chimeric-gene associated with cytoplasmic male sterility of HL-rice显示文摘The mitochondrial genome libraries of HL-type sterile line(A) and maintainer line(B) have been constructed. Mitochondrial gene, atp6, was used to screen libraries, due to the different Southern and Northern blot results between sterile and maintainer line. Sequencing analysis of positive clones proved that there were two copies of atp6 gene in sterile line and only one in maintainer line. One copy of atpt6 in sterile line was same to that in maintainer line; the other showed different flanking sequence from the 49th nucleotide downstream of the termination codon of atp6 gene. A new chimeric gene, orfH79, was found in the region. | 4 | Evaluation and optimization of blasting approaches to reducing oversize boulders and toes in open-pit mine显示文摘A new blasting approach of combined blastholes with different diameters is proposed to solve the problems of oversize boulders and rock toes in open-pit mine. A non-ideal detonation model and a statistical damage constitutive model are implemented in dynamic finite element analysis to investigate the formation mechanism of oversize boulders and toes. The damage distribution and evolution process of rock blasting fragmentation is simulated, and the scheme is further optimized. Numerical analysis results showed that pocket charges and satellite blastholes can only improve bench top fragmentation, but they cannot reduce the oversize in the middle and bottom of bench as well as the toe problem. | 5 | Is a low dose of hepatitis B vaccine enough for a rapid vaccination scheme?显示文摘AIM: To determine whether or not a low dose of HB vaccine can be effectively used in the rapid vaccination.METHODS: Rapid vaccination (0, 1, 2 months) with low dose (5 μg) or routine dose (10 μg) HB vaccine was studied in 250 subjects (130 school children and 120 university students). Serum from all the participants was tested for HBsAg, anti-HBs and anti-HBc at 1, 3 and 7 months after the first dose of vaccination and all subjects were serum HBV marks negative before the vaccination. Non-responders to a complete initial vaccination from university students were given an additional vaccination with 10 μg of HB vaccine and their serum anti-HBs was tested again one month later.RESULTS: One month after the third dose of vaccination (third month) sero-conversion rates and geometric mean titer (GMTs) were significantly (P<0.01) higher in the routine dose (resp. 89 % and 106.8) than in the low dose group (resp. 72 % and 59.5). Sero-conversion rates and GMTs were maintained stable for another 4 months in both groups.After an additional vaccination to non-responders with 10 μg HB vaccine, 17/23 subjects (13/15 from those vaccinated with 5 μg vaccine and 4/8 from those vaccinated with 10 μg vaccine) became anti-HBs positive, yielding similar seroconversion rates for both dose groups.CONCLUSION: Higher sero-conversion rates and GMTs were reached in those vaccinated with 10 μg HB vaccine than in those vaccinated with 5 μg HB vaccine after a comp^te vaccination with a 0, 1, 2 month scheme. | 6 | Detection of bcl-2 and bax expression and bcl-2/JH fusion gene in intrahepatic cholangiocarcinoma显示文摘AIM: To investigate the relationship between bcl-2 gene and its related protein bax and intrahepatic cholangiocellular carcinoma (CCC). METHODS: Semi-nested in situ PCR (SNISPCR) and immunohistochemistry were performed to detect bcl-2/JH fusion gene and bcl-2, bax protein expression in 29 cases of CCC. RESULTS: No bcl-2/JH fusion gene was found in all cases of CCC, 72.4% of 29 cases expressed bcl-2 protein. Bcl-2 protein expression was related to histopathological grades (P<0.05). There was no corresponding relationship between bcl-2/JH fusion gene formation and bcl-2 protein expression in CCC (P<0.05). Bax was expressed in 10.3% of 29 cases. The ratio of bcl-2 to bax in normal liver tissues (3.5 to 1) was different from that in tumor tissues (7.0 to 1). CONCLUSION: It is suggested that bcl-2/JH fusion gene formation is not a frequent event and may not play an important role in the pathogenesis of CCC. However, aberrant ratio of bcl-2 to bax protein expression may be involved in the course of tumorigenesis of CCC. | 7 | Activation of transcription factors NF-kappaB and AP-1 and their relations with apoptosis-associated proteins in hepatocellular carcinoma显示文摘AIM: To study the distribution pattern of transcription factors NF-κB and AP-1 and their relations with the expression of apoptosis associated-proteins Fas/FasL and ICH-1L/S in human hepatocellular carcinoma (HCC).METHODS: We performed in situ hybridization and immunohistochemical techniques for NF-κB, AP-1,Fas/FasL and ICH-1 in 40 cases of human HCC along with corresponding nontumoral tissues and 7 cases of normal liver tissues.RESULTS: Twenty-two (55%) and 25 (62.5%) of 40 cases for NF-κB and AP-1 were presented for nuclear or both nuclear and cytoplastic staining respectively, while less cases were presented for only cytoplastic staining for NF-κB (18%) and AP-1 (10%) in adjacent nontumoral tissues and negative staining in normal liver tissues. There was no statistically significant difference of NF-κB or AP-1activation between well differentiated tumors and poorly differentiated tumors (P>0.05). NF-κB activity is positively corresponded to AP-1 activation. The expression of ICH-1L/S was associated with the activation of NF-κB and AP-1 (P<0.05), but no significant relationship was found between Fas/FasL and NF-κB or AP-1(P>0.05).CONCLUSION: Activation of both NF-κB and AP-1 may be required for ICH-1L/S-induced apoptosis in HCC, but not for Fas/FasL-mediated apoptosis. | 8 | Effect of vector-expressed siRNA on HBV replication in hepatoblastoma cells显示文摘AIM: To study the effect of siRNA expressed from DNA vector on HBV replication.METHODS: Human U6 promoter was amplified from genomic DNA and cloned into plasmid pUC18 to construct a mammalian siRNA expression vector pUC18U6. Then oligonucleotides coding for a short hairpin RNA against HBV were cloned into pUC18U6 to form pUC18U6HBVsir which was introduced into 2.2.15 cells by using liposome-mediated transfection.2.2.15 cells transfected by pUC18U6 and pUC18U6GFPsir which expressed siRNA against green fluorescent protein and mock-transfected 2.2.15 cells were used as controls.Concentration of HBsAg in the supernatant of the transfected cells was measured by using solid-phase radioimmunoassay.RESULTS: A mammalian siRNA expression vector pUC18U6 was constructed successfully. | 9 | The editing sites in transcripts of functional genes of rice mitochondria显示文摘RNA editing exists extensively in the higher plant mitochondria, and is a required step for forming functional proteins. There may be some relationship between RNA editing and cytoplasmic male sterility (CMS), a kind of phenomenon that is attributed to mitochondrial genome mutations. The research materials used are the gametophytic male sterility line (A), maintainer line (B) and F1 hybrid (F1) of HL-type CMS rice. cDNAs and DNAs of atp6 and coxII have been obtained from A, B and F1 by PCR and RT-PCR. Comparing sequences of cDNAs and DNAs, 18 and 15 editing sites were found respectively in the transcripts of atp6 and coxll. A, B and F1 shared the same editing sites. | 10 | Dam foundation excavation techniques in China:A review显示文摘A protective layer(PL) is commonly reserved above foundation surface to protect the underlying rock mass during dam foundation excavation. In China, the PL of dam foundation is conventionally subdivided into two or three thin layers and excavated with the shallow-hole blasting method, even by pneumatic pick method in case of soft rock mass. The aforementioned layered excavation of the PL delays the construction of the whole project. After nearly 30-year practices, several safe and effcient methods for the PL excavation of dam foundation are gradually developed. They include shallow-hole bench blasting with cushion material(SBC) at the bottom of the hole, and horizontal smooth blasting(HSB). The PL is even cancelled on the condition that horizontal pre-split technique is employed during dam foundation excavation. This paper introduces the aforementioned two PL excavation methods(shallow-hole blasting and bench blasting) and horizontal pre-split technique of dam foundation without protective layer(HPP). The basic principles of blasting method, blasting geometry, charge structure, drill-and-blast parameters of typical projects are examined. Meanwhile, the merits and limitations of each method are compared. Engineering practices in China show that HSB is basically the optimal method for dam foundation PL excavation in terms of foundation damage control and rapid construction. | 11 | Effect of vector-expressed shRNAs on hTERT expression显示文摘AIM: To study the effect of short hairpin RNAs (shRNAs)expressed from DNA vector on hTERT expression.METHODS: Oligonucleotides coding for four shRNAs against hTERT were cloned into a mammalian shRNA expressionvector pUC18U6 to form pUC18U6ht1-4, which were thenintroduced into HepG2 cells by using liposome-mediated transfection. HepG2 cells transfected by pUC18U6 and pUC18U6GFPsir, which expressed shRNA against green fluorescent protein (GFP), were used as controls. hTERT mRNA in the transfected cells were quantified by using real-time fluorescent RT-PCR.RESULTS: Among the four shRNAs against hTERT, two decreased the hTERT mRNA level. | 14 | Unveiling the Effects of Interchain Hydrogen Bonds on Solution Gelation and Mechanical Properties of Diarylfluorene-Based Semiconductor Polymers显示文摘The intrinsically rigid and limited strain of most conjugated polymers has encouraged us to optimize the extensible properties of conjugated polymers.Herein,learning from the hydrogen bonds in glucose,which were facilitated to the toughness enhancement of cellulose,we introduced interchain hydrogen bonds to polydiarylfluorene by amide-containing side chains.Through tuning the copolymerization ratio,we systematically investigated their influence on the hierarchical condensed structures,rheology behavior,tensile performances,and optoelectronic properties of conjugated polymers.Compared to the reference copolymers with a low ratio of amide units,copolymers with 30%and 40%amide units present a feature of the shearthinning process that resembled the non-Newtonian fluid,which was enabled by the interchain dynamic hydrogen bonds.Besides,we developed a practical and universal method for measuring the intrinsic mechanical properties of conjugated polymers.We demonstrated the significant impact of hydrogen bonds in solution gelation,material crystallization,and thin film stretchability.Impressively,the breaking elongation for P4 was even up to~30%,which confirmed the partially enhanced film ductility and toughness due to the increased amide groups.Furthermore,polymer light-emitting devices(PLEDs)based on these copolymers presented comparable performances and stable electroluminescence(EL).Thin films of these copolymers also exhibited random laser emission with the threshold as low as 0.52μJ/cm^(2),suggesting the wide prospective application in the field of flexible optoelectronic devices. | Lubing Bai Yamin Han Chen Sun Xiang An Chuanxin Wei Wei Liu Man Xu Lili Sun Ning Sun Mengna Yu He Zhang Qi Wei Chunxiang Xu Yingguo Yang Tianshi Qin Linghai Xie Jinyi Lin Wei Huang | 2020 | Research2020,,1: | 2 |
