1Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. According to the substitution rate,we concluded that the approximate divergence time between Tibetan Mastiff and grey wolf was 58,000 years before the present (YBP),and the approximate divergence time between other domestic dogs and grey wolf was 42,000 YBP,demonstrating that the time of origin of the Tibetan Mastiff was earlier than that of the other domestic dogs.Qifa Li Zhenshan Liu Yinxia Li Xingbo Zhao Liyan Dong Zengxiang Pan Yuanrong Sun Ning Li Yinxue Xu Zhuang Xie 2008Journal of Genetics and Genomics2008,35,6:15

3Prepartum body conditions affect insulin signaling pathways in postpartum adipose tissues in transition dairy cows显示文摘Background: Overconditioned dairy cows are susceptible to excessive lipolysis and increased insulin resistance during the transition period.The associations among body fat reserve,insulin resistance,and lipolysis in adipose tissues(AT) remain to be elucidated.Therefore,this study aimed to investigate whether excessive fat reserves influence the insulin signaling pathway in AT postpartum.Results: Twenty multiparous dairy cows were selected and assigned to one of two groups,according to prepartum body condition score(BCS): Control group(BCS = 3.0–3.5;n = 10) and Overconditioned group(BCS ≥ 4.0;n = 10).Blood samples were collected on days-14,-7,-4,-2,-1,0,1,2,4,7,and 14 relative to parturition.Subcutaneous AT were collected on day 2 following parturition for quantitative real-time polymerase chain reaction and western blot analyses.No differences were observed between the two groups in serum glucose,non-esterified fatty acids,β-hydroxybutyric acid,tumor necrosis factor(TNF)α,insulin,or leptin concentrations during the experimental period.Compared with the control cows,the overconditioned cows had lower serum triglyceride levels and higher adiponectin concentrations.In the AT postpartum,insulin receptor mRNA and protein levels were lower in the overconditioned cows than in the control cows,and no differences were found in glucose transporter 4 mRNA.Compared with the control cows,the overconditioned cows had lower mRNA levels of TNFα and higher mRNA levels of peroxisome proliferator-activated receptor gamma(PPARγ) in AT postpartum.The phosphorylated protein kinase B(AKT) content and phosphorylation rate of AKT were increased in the overconditioned cows compared with the control cows,which suggested that the downstream insulin signaling in AT was affected.Conclusions: In the present study,transition dairy cows with higher BCS did not show more fat mobilization.The changes of insulin signaling pathway in AT postpartum of overconditioned cows may be partly related to the expression of PPARγ and TNFα,and the secretion of adiponectin.Fanjian Zhang Dan Li Qiong Wu Jian Sun Wenyi Guan Yinxu Hou Yaohong Zhu Jiufeng Wang 2019Journal of Animal Science and Biotechnology2019,10,3:2
4Dental stem cell-derived extracellular vesicles as promising therapeutic agents in the treatment of diseases显示文摘Dental stem cells(DSCs), an important source of mesenchymal stem cells(MSCs), can be easily obtained by minimally invasive procedures and have been used for the treatment of various diseases. Classic paradigm attributed the mechanism of their therapeutic action to direct cell differentiation after targeted migration, while contemporary insights into indirect paracrine effect opened new avenues for the mystery of their actual low engraftment and differentiation ability in vivo. As critical paracrine effectors, DSC-derived extracellular vesicles(DSC-EVs) are being increasingly linked to the positive effects of DSCs by an evolving body of in vivo studies. Carrying bioactive contents and presenting therapeutic potential in certain diseases, DSC-EVs have been introduced as promising treatments. Here, we systematically review the latest in vivo evidence that supports the therapeutic effects of DSC-EVs with mechanistic studies. In addition, current challenges and future directions for the clinical translation of DSC-EVs are also highlighted to call for more attentions to the(Ⅰ) distinguishing features of DSC-EVs compared with other types of MSC-EVs,(Ⅱ)heterogeneity among different subtypes of DSC-derived EVs,(Ⅲ) action modes of DSC-EVs,(Ⅳ) standardization for eligible DSC-EVs and(Ⅴ) safety guarantee for the clinical application of DSC-EVs. 7Steady-state weights solution to affine projection algorithm显示文摘A new expression of the weights update equation for the affine projection algorithm (APA) is proposed that improves the convergence rate of an adaptive filter, particularly for highly colored input signals, and yields greater details of the internal structure. The steady-state weights solution to the APA algorithm is calculated in different step-sizes, which is significantly different from the iteration method. The weights error in steady-state is proved to be zero as the number of the input direction vector increases to infinity, ensuring that the estimated weights of the APA algorithm in steady-state are unbiased and consistent. The sensitivity of the step-size parameter for the steady-state weights is also analyzed. 9High performance removal of sulfamethoxazole using large specific area of biochar derived from corncob xylose residue显示文摘To remove antibiotics from waste water,an alkali active porous biochar,850BC,was prepared from corncob xylose residue.In preparation,NaOH dipping was used for silicon removal and KOH activation was operated at 850℃.Further characterization containing BET,SEM,and FTIR were confirmed.850BC possessed a huge specific surface area of 3043 m^(2)·g^(−1),developed pore structure and abundant oxygen functional groups.The adsorption performance of sulfamethoxazole on 850BC was quick and efficient,and the adsorption capacity reached 1429 mg·g^(−1),which was significantly higher than other adsorbents reported previously.While pseudo-second-order kinetic model and Langmuir model could better describe the adsorption,chemisorp-tion dominated the SMX adsorption onto 850BC.In virtue of pore-filling andπ-πinteraction as major mechanism,a large surface area and rich oxygen-containing functional groups led to an excellent adsorption performance.Thus,this preparation method provided a biochar-based adsorbent with enhanced specific surface for efficient removal of antibiotic pollutants.Yinxue Li Hongru Shang Yongna Cao Chunhui Yang Yujie Feng Yanling Yu 2022Biochar2022,4,1:1
10Monitoring of Avian Influenza Virus and Newcastle Disease Virus in Suichuan Region of Jiangxi Province显示文摘[Objective] The research aimed to investigate the epidemiological characteristics of Avian influenza virus( AIV) and Newcastle disease virus( NDV)of wild Ardeid birds in Suichuan region of Jiangxi Province. [Method] A total of 110 nasopharyngeal swabs and 110 cloacal swabs of Ardeid birds were collected from Suichuan region of Jiangxi Province in October of 2014. The swabs were conducted the virus isolation using SPF chicken embryos,suspected positive samples were screened by hemagglutination( HA) test and identified by Reverse Transcription-Polymerase Chain Reaction( RT-PCR). [Results] HA titre of all 110 test samples was 0 except four samples. Detection on AIV and NDV of the test samples by RT-PCR showed that all the samples were negative,which indicated that the infection risk of avian influenza( AI) and newcastle disease( ND) of Ardeid birds in Suichuan region in Jiangxi Province in the summer of 2014 was low.
