1The altered expression of inflammation-related microRNAs with microRNA-155 expression correlates with Th17 differentiation in patients with acute coronary syndrome显示文摘MicroRNAs(miRNAs)are a novel class of small,non-coding RNAs that play a significant role in both inflammatory and cardiovascular diseases.Immune cells,especially T helper(Th)cells,are critical in the development of atherosclerosis and the onset of acute coronary syndrome(ACS).To assess whether inflammation-related miRNAs(such as miR-155,146a,21,125a-5p,125b,31)are involved in the imbalance of Th cell subsets in patients with ACS,we measured the expression of related miRNAs in patients with acute myocardial infarction(AMI),unstable angina(UA),stable angina(SA)and chest pain syndrome(CPS);analyzed the relationship between miRNA expression and the frequency of Th cell subsets;and observed the co-expression of miR-155 and IL-17A in peripheral blood mononuclear cells(PBMCs)of patients with ACS.The results showed that the expression of miR-155 in the PBMCs of patients with ACS was decreased by approximately 60%,while the expression of both miR-21 and miR-146a was increased by approximately twofold.The expression patterns of miRNAs in plasma correlated with those in PBMCs,except for miR-21,which was increased by approximately sixfold in the AMI group and showed no significant difference between the UA group and the CPS group.We also found that the expression of miR-155 inversely correlated with the frequency of Th17 cells(r520.896,P,0.01)and that miR-155 was co-expressed with IL-17A in patients with ACS.In conclusion,our study revealed the expression patterns of inflammation-related miRNAs in patients with ACS and found that miR-155 may be associated with Th17 cell differentiation.Rui Yao Yulan Ma Youyou Du Mengyang Liao Huanhuan Li Wei Liang Jing Yuan Zhijun Ma Xian Yu Hong Xiao Yuhua Liao 2011Cellular & Molecular Immunology2011,8,6:32
2Artemisinin, the Magic Drug Discovered from Traditional Chinese Medicine显示文摘Artemisinin and its derivatives represent the most important and influential class of drugs in the fight against malaria. Since the discovery of artemisinin in the early 1970s, the global community has made great strides in characterizing and understanding this remarkable phytochemical and its unique chemical and pharmacological properties. Today, even as artemisinin continues to serve as the foundation for antimalarial therapy, numerous challenges have surfaced in the continued application and development of this family of drugs. These challenges include the emergence of delayed treatment responses to artemisinins in malaria and efforts to apply artemisinins for non-malarial indications. Here, we provide an overview of the story of artemisinin in terms of its past, present, and future. 4Enhanced high-temperature performance of Li-rich layered oxide via surface heterophase coating显示文摘Li-rich layered oxides have become one of the most concerned cathode materials for high-energy lithiumion batteries, but they still suffer from poor cycling stability and detrimental voltage decay, especially at elevated temperature. Herein, we proposed a surface heterophase coating engineering based on amorphous/crystalline Li3 PO4 to address these issues for Li-rich layered oxides via a facile wet chemical method. The heterophase coating layer combines the advantages of physical barrier effect achieved by amorphous Li3 PO4 with facilitated Li+diffusion stemmed from crystalline Li3 PO4. Consequently, the modified Li(1.2) Ni(0.2) Mn(0.6) O2 delivers higher initial coulombic efficiency of 92% with enhanced cycling stability at 55 °C(192.9 mAh/g after 100 cycles at 1 C). More importantly, the intrinsic voltage decay has been inhibited as well, i.e. the average potential drop per cycle decreases from 5.96 mV to 2.99 mV. 5Niacin alters the ruminal microbial composition of cattle under high-concentrate condition显示文摘To understand the effects of niacin on the ruminal microbial ecology of cattle under high-concentrate diet condition, Illumina MiSeq sequencing technology was used. Three cattle with rumen cannula were used in a 3 x 3 Latin-square design trial. Three diets were fed to these cattle during 3 periods for 3 days, respectively: high-forage diet(HF; forage-to-concentrate ratio = 80:20), high-concentrate diet(HC;forage-to-concentrate ratio=20:80), and HC supplemented with 800 mg/kg niacin(HCN). Ruminal pH was measured before feeding and every 2 h after initiating feeding. Ruminal fluid was sampled at the end of each period for microbial DNA extraction. Overall, our findings revealed that subacute ruminal acidosis(SARA) was induced and the a-diversity of ruminal bacterial community decreased in the cattle of HC group. Adding niacin in HC could relieve the symptoms of SARA in the cattle but the ruminal pH value and the Shannon index of ruminal bacterial community of HCN group were still lower than those of HF group. Whatever the diet was, the ruminal bacterial community of cattle was dominated by Bacteroidetes,Firmicutes and Proteobacteria. High-concentrate diet significantly increased the abundance of Prevotella,and decreased the abundance of Paraprevotella, Sporobacter, Ruminococcus and Treponema than HF.Compared with HC, HCN had a trend to decrease the percentage of Prevotella, and to increase the abundance of Succiniclasticum, Acetivibrio and Treponema. Increasing concentrate ratio could decrease ruminal pH value, and change the ruminal microbial composition. 6The mechanosensitive lncRNA Neat1 promotes osteoblast function through paraspeckle-dependent Smurf1 mRNA retention显示文摘Mechanical stimulation plays an important role in bone remodeling. Exercise-induced mechanical loading enhances bone strength,whereas mechanical unloading leads to bone loss. Increasing evidence has demonstrated that long noncoding RNAs(lnc RNAs) play key roles in diverse biological, physiological and pathological contexts. However, the roles of lnc RNAs in mechanotransduction and their relationships with bone formation remain unknown. In this study, we screened mechanosensing lnc RNAs in osteoblasts and identified Neat1, the most clearly decreased lnc RNA under simulated microgravity. Of note, not only Neat1 expression but also the specific paraspeckle structure formed by Neat1 was sensitive to different mechanical stimulations, which were closely associated with osteoblast function. Paraspeckles exhibited small punctate aggregates under simulated microgravity and elongated prolate or larger irregular structures under mechanical loading. Neat1 knockout mice displayed disrupted bone formation, impaired bone structure and strength, and reduced bone mass. Neat1 deficiency in osteoblasts reduced the response of osteoblasts to mechanical stimulation. In vivo, Neat1 knockout in mice weakened the bone phenotypes in response to mechanical loading and hindlimb unloading stimulation. Mechanistically, paraspeckles promoted nuclear retention of E3 ubiquitin ligase Smurf1 m RNA and downregulation of their translation, thus inhibiting ubiquitination-mediated degradation of the osteoblast master transcription factor Runx2, a Smurf1 target. 7EGFR tyrosine kinase inhibitor HS-10182 increases radiation sensitivity in non-small cell lung cancers with EGFR T790M mutation显示文摘Objective:To investigate the potential of HS-10182,a second-generation epidermal growth factor receptor tyrosine kinase inhibitor(EGFR-TKI),as a radiosensitizer in non-small cell lung cancer(NSCLC).Methods:Two cell lines of NSCLCs,A549 that possesses wild-type(WT)EGFRs and H1975 that possesses EGFR L858R/T790M double mutations,were treated with HS-10182 at various concentrations,and cell viabilities were determined using the MTS assay.The cells were tested by clonogenic survival assays to identify the radiosensitivity of both groups.Western blot was performed to analyze the expression of phosphorylated EGFR,AKT,DNA-dependent protein kinase,and catalytic subunit(DNA-PKcs)proteins.Immunofluorescence analyses were performed to examine the formation and changes in nuclearγ-H2AX foci.Cell apoptosis was examined by flow cytometry and Western blots for cleaved caspase-3,-8,-9,and cleaved poly ADP-ribose polymerase(PARP).Furthermore,we established xenograft models in mice and the effects of different treatments on tumor growth were then assessed.Results:Clonogenic survival assays revealed that HS-10182 significantly enhanced the radiosensitivity of H1975 cells but not A549cells[dose enhancement ratios(DERs)=2.36(P<0.05)vs.1.43(P>0.05)].Western blot results showed that HS-10182 increased the levels of cleaved caspase-3,-8,-9,and cleaved PARP in H1975 cells but not in A549 cells.In addition,flow cytometry analysis showed that HS-10182 enhanced irradiation-induced apoptosis in H1975.Immunofluorescence results found that HS-10182increased the average number ofγ-H2AX foci after irradiation in H1975 cells,but not in A549 cells.Combined radiation and HS-10182 treatment increased the expression of DNA-PKcs but this increase was more significant in H1975 cells than in A549 cells.Moreover,HS-10182 suppressed the increased expression of Rad50 in H1975 cells in response to irradiation.In vivo experiments found that the combined therapy significantly inhibited tumor growth.Conclusions:HS-10182 enhances the radiosensitivity of H1975 cells which is possibly because that HS-10182 could enhance irradiation-induced apoptosis,increase irradiation-induced DNA damage,and cause a delay in DNA damage repair.Our findings suggest that radiotherapy combined HS-10182 is a novel treatment for lung cancer cells which have acquired the T790M mutation.HS-10182 could be brought to the clinic as a radiosensitizer in NSCLCs with the EGFR T790M mutation.Yang Chen Youyou Wang Lujun Zhao Ping Wang Jifeng Sun Rudi Bao Chenghai Li Ningbo Liu 2018Cancer Biology & Medicine2018,15,1:2

9Large-scale and high-quality III-nitride membranes through microcavity-assisted crack propagation by engineering tensile-stressed Ni layers显示文摘Epitaxially grown III-nitride alloys are tightly bonded materials with mixed covalent-ionic bonds.This tight bonding presents tremendous challenges in developing III-nitride membranes,even though semiconductor membranes can provide numerous advantages by removing thick,inflexible,and costly substrates.Herein,cavities with various sizes were introduced by overgrowing target layers,such as undoped GaN and green LEDs,on nanoporous templates prepared by electrochemical etching of n-type GaN.The large primary interfacial toughness was effectively reduced according to the design of the cavity density,and the overgrown target layers were then conveniently exfoliated by engineering tensile-stressed Ni layers.The resulting III-nitride membranes maintained high crystal quality even after exfoliation due to the use of GaN-based nanoporous templates with the same lattice constant.The microcavity-assisted crack propagation process developed for the current III-nitride membranes forms a universal process for developing various kinds of large-scale and high-quality semiconductor membranes.Jung-Hong Min Kwangjae Lee Tae-Hoon Chung Jung-Wook Min Kuang-Hui Li Chun Hong Kang Hoe-Min Kwak Tae-Hyeon Kim Youyou Yuan Kyoung-Kook Kim Dong-Seon Lee Tien Khee Ng Boon S.Ooi 2022Opto-Electronic Science2022,1,10:1

11Interfacial Degradation and Optimization of Li-rich Cathode Materials显示文摘High-energy and safe lithium ion batteries(LIBs)are in increasing need as the rapid development of electronic devices,electric vehicles,as well as energy storage station.Li-rich oxides have attracted a lot of attention due to their high capacity and low cost as cathode material for LIBs.However,they still suffer from the vulnerable cathode/electrolyte interface,which presents the huge challenges of surface degradation and gas release,particularly at high state of charge.Some issues of Li-rich cathode materials,such as moderate cycle stability and voltage decay,are in tight connection with electrode-electrolyte interfacial side reactions.Research in the area of interfacial degradation mechanism and optimization strategies is of great significance as for Li-rich cathode,and extensive efforts have been poured.This review aims to understand the degradation mechanism of Li-rich cathode materials,and summarize the corresponding valuable and effective optimization strategies.Based on these considerations,we also have discussed the remaining challenges and the future research direction.Yuefeng Su Jiayu Zhao Lai Chen Ning Li Yun Lu Jinyang Dong Youyou Fang Shi Chen Feng Wu 2021Chinese Journal of Chemistry2021,39,2:1
