Highly expressed BMP9/GDF2 in postnatal mouse liver and lungs may account for its pleiotropic effects on stem cell differentiation, angiogenesis, tumor growth and metabolism显示文摘Bone morphogenetic protein 9(BMP9)(or GDF2)was originally identified from fetal mouse liver cDNA libraries.Emerging evidence indicates BMP9 exerts diverse and pleiotropic functions during postnatal development and in maintaining tissue homeostasis. However, the expression landscape of BMP9 signaling during development and/or in adult tissues remains to be analyzed. Here,we conducted a comprehensive analysis of the expression landscape of BMP9 and its signaling mediators in postnatal mice.By analyzing mouse ENCODE transcriptome datasets we found Bmp9 was highly expressed in the liver and detectable in embryonic brain, adult lungs and adult placenta. We next conducted a comprehensive qPCR analysis of RNAs isolated from major mouse tissues/organs at various ages.We found that Bmp9 was highly expressed in the liver and lung tissues of young adult mice, but decreased in older mice.Interestingly,Bmp9 was only expressed at low to modest levels in developing bones.BMP9-associated TGFβ/BMPR type I receptor Alk1 was highly expressed in the adult lungs.Furthermore, the feedback inhibitor Smads Smad6 and Smad7 were widely expressed in mouse postnatal tissues. However, the BMP signaling antagonist noggin was highly expressed in fat and heart in the older age groups,as well as in kidney, liver and lungs in a biphasic fashion. Thus, our findings indicate that the circulating BMP9 produced in liver and lungs may account for its pleiotropic effects on postnatal tissues/organs although possible roles of BMP9 signaling in liver and lungs remain to be fully understood. | Wei Liu Zhongliang Deng Zongyue Zeng Jiaming Fan Yixiao Feng Xi Wang Daigui Cao Bo Zhang Lijuan Yang Bin Liu Mikhail Pakvasa William Wagstaff Xiaoxing Wu Huaxiu Luo Jing Zhang Meng Zhang Fang He Yukun Mao Huiming Ding Yongtao Zhang Changchun Niu Rex C.Haydon Hue H.Luu Jennifer Moriatis Wolf Michael J.Lee Wei Huang Tong-Chuan He Yulong Zou | 2020 | Genes & Diseases2020,7,2: | 9
Fluxes of methane, carbon dioxide and nitrous oxide in an alpine wetland and an alpine grassland of the Tianshan Mountains,China显示文摘Methane(CH4), carbon dioxide(CO2) and nitrous oxide(N2O) are known to be major greenhouse gases that contribute to global warming. To identify the flux dynamics of these greenhouse gases is, therefore, of great significance. In this paper, we conducted a comparative study on an alpine grassland and alpine wetland at the Bayinbuluk Grassland Eco-system Research Station, Chinese Academy of Sciences. By using opaque, static, manual stainless steel chambers and gas chromatography, we measured the fluxes of CH4, N2O and CO2 from the grassland and wetland through an in situ monitoring study from May 2010 to October 2012. The mean flux rates of CH4, N2O and CO2 for the experimental alpine wetland in the growing season(from May to October) were estimated at 322.4 μg/(m2?h), 16.7 μg/(m2?h) and 76.7 mg/(m2?h), respectively; and the values for the alpine grassland were –88.2 μg/(m2?h), 12.7 μg/(m2?h), 57.3 mg/(m2?h), respectively. The gas fluxes showed large seasonal and annual variations, suggesting weak fluxes in the non-growing season. The relationships between these gas fluxes and environmental factors were analyzed for the two alpine ecosystems. The results showed that air temperature, precipitation, soil temperature and soil moisture can greatly influence the fluxes of CH4, N2O and CO2, but the alpine grassland and alpine wetland showed different feedback mechanisms under the same climate and environmental conditions. | GuiXiang HE KaiHui LI XueJun LIU YanMing GONG YuKun HU | 2014 | Journal of Arid Land2014,6,6: | 7
Resveratrol compares with melatonin in improving in vitro porcine oocyte maturation under heat stress显示文摘Background: Resveratrol, an important phyto-antioxidant commonly found in grapes, mulberry, and other plants, has a variety of functions including anti-aging, anti-cancer and anti-inflammatory activities. In the current study, we investigated the beneficial effects of resveratrol on in vitro porcine oocyte maturation under heat stress(HS). The effect of resveratrol, melatonin and their combination on alleviating HS was compared according to the maturation rate of oocytes and the development competence of embryos after parthenogenetic activation(PA).Results: Supplementation with resveratrol(2.0 μmol/L) not only improved the nuclear maturation but also raised the blastocyst rate of porcine embryos' PA from oocytes that underwent HS by increasing their glutathione(GSH)level, reducing reactive oxygen species(ROS) and up-regulating the expression of Sirtuin 1(SIRT1). It was also found that melatonin(10^(-7)mol/L) and the combination of resveratrol(2.0 μmol/L) plus melatonin(10^(-7)mol/L) exhibited more potent effects than resveratrol alone regarding their protective activities on oocyte maturation under HS.Conclusions: This study compared the efficiencies of resveratrol, melatonin and their combination for protecting porcine oocytes from heat stress. The mechanisms are attributed to the fact that each treatment may have different ability to regulate the synthesis of steroid hormones and the expression of mature related genes. | Yu Li Jing Wang Zhenzhen Zhang Jinyun Yi Changjiu He Feng Wang Xiuzhi Tian Minghui Yang Yukun Song Pingli He Guoshi Liu | 2017 | Journal of Animal Science and Biotechnology2017,8,1: | 7
Effects of Yishengukang decoction on expression of bone-specific alkaline phosphatase, carboxyterminal propeptide of typeⅠprocollagen, and carboxyterminal cross-linked telepeptide of typeⅠcollagen in malignant tumor patients with bone metastasis显示文摘OBJECTIVE: To investigate the effect of Yishengukang decoction on the expression of the metabolic bone markers, bone-specific alkaline phosphatase(BAP), carboxyterminal propeptide of type Ⅰ procollagen(PICP), and carboxyterminal cross-linked telepeptide of type Ⅰ collagen(ICTP), in cancer patients with bone metastasis.METHODS: Patients(n = 180) were divided into three groups:(a) bone metastasis patients treated with Yishengukang and pamidronate disodium injection(treatment group, n = 60);(b) bone metastasis patients treated with pamidronate disodium injection alone(control group, n = 60);(c) cancer patients without metastatic bone lesion(non-bone metastasis group, n = 60). Serum levels of the metabolic markers BAP, PICP, and ICTP were detected by enzyme-linked immunosorbent assay pre- and post-therapy.RESULTS: A significant decrease in serum BAP level was observed in the treatment group compared with the control group. However there were no significant differences in serum levels of PICP and ICTP before or after treatment compared with the control group.CONCLUSION: Yishengukang decoction combined with pamidronate disodium injection reduced serum BAP level to a greater extent that pamidronate disodium injection alone. Furthermore, the combined therapy was more beneficial in regulating imbalanced bone metabolism after bone metastasis, and may represent the molecular mechanism underpinning the effects of Yishengukang decoction. | Yin Yukun Feng Li Zhou Lei Li Jie Gao Yin Wang Ningjun Yu Jianhua Jiang Zhenglong He Shengqi Lu Dianrong Wang Fang Du Yanlin | 2017 | Journal of Traditional Chinese Medicine2017,37,1: | 6
Combination of ablative fractional carbon dioxide laser and platelet-rich plasma treatment to improve hypertrophic scars:a retrospective clinical observational study显示文摘Background:Hypertrophic scars are one of the main complications that affect the quality of life of patients after burns. Many methods have been shown to be effective in the treatment of hypertrophic scars, such as ablative fractional CO_(2) laser(AFCL)and platelet-rich plasma(PRP).However, there are few studies on the effect of the combined application of these measures. The purpose of this study was to explore the therapeutic effect of AFCL combined with PRP on hypertrophic burn scars. Methods:A retrospective clinical observation study was conducted on 50 patients with hypertrophic burn scars. The AFCL+PRP group included 31 patients who received AFCL combined with PRP treatment;the AFCL group included 19 patients who received AFCL treatment only. The University of North Carolina 4P Scar Scale(UNC4P)and the Vancouver Scar Scale(VSS)scores that were collected before each treatment were used as indicators of the effectiveness of the previous treatment. The scores recorded at the second, fourth and seventh months were analysed. Results:The demographic data of the 2 groups were not significantly different. Before treatment, therewas no difference in the UNC4P and VSS scores between the 2 groups.Therewas a significant decline in the UNC4P and VSS total scores over 6 months in both groups(p<0.05)and scores in the 2 groups were comparable after 3 and 6 months(p<0.05).UNC4P scores in the AFCL+PRP group decreased from a mean of 8.26 to 2.61(p<0.05)with a concomitant drop in VSS scores from a mean of 11.74 to 6.06(p<0.01).In the AFCL group UNC4P and VSS scores decreased from 7.68 to 4.63(p<0.05)and from 10.89 to 8.16(p<0.05),respectively.The sub-items of these 2 assessments were analysed and the results suggest that AFCL combined with PRP can comprehensively improve scarring. Conclusions:This study shows that PRP is an effective adjunct for AFCL in the treatment of hypertrophic burn scars and that the combination of PRP and AFCL proved to be more useful than AFCL alone. This combination may be a new and effective clinical practice for the treatment of scars. However, larger and higher-level clinical studies are still needed to determine its efficacy and possible mechanisms. | Zhanzhan Dai Xiaozhen Lou Tuo Shen Yu Sun Yongqiang Xiao Xingfeng Zheng Xuexin Wang Yu Peng Yukun Guo Yibin Guo Jiannan Wen He Fang Bing Ma Zhaofan Xia | 2021 | Burns & Trauma2021,9,1: | 4

Calibration procedures for the CHASE/HIS science data显示文摘The Hα line is an important optical line in solar observations containing the information from the photosphere to the chromosphere. To study the mechanisms of solar eruptions and the plasma dynamics in the lower atmosphere, the Chinese Hα Solar Explorer(CHASE) was launched into a Sun-synchronous orbit on October 14, 2021. The scientific payload of the CHASE satellite is the Hα Imaging Spectrograph(HIS). The CHASE/HIS acquires, for the first time, seeing-free Hα spectroscopic observations with high spectral and temporal resolutions. It consists of two observational modes. The raster scanning mode provides full-Sun or region-of-interest spectra at Hα(6559.7-6565.9 ?) and Fe I(6567.8-6570.6 ?) wavebands. The continuum imaging mode obtains full-Sun photospheric images at around 6689 ?. In this paper, we present detailed calibration procedures for the CHASE/HIS science data, including the dark-field and flat-field correction, slit image curvature correction, wavelength and intensity calibration, and coordinate transformation. | 8 | Progress in octahedral spherical hohlraum study显示文摘In this paper,we give a review of our theoretical and experimental progress in octahedral spherical hohlraum study. From our theoretical study, the octahedral spherical hohlraums with 6 Laser Entrance Holes(LEHs)of octahedral symmetry have robust high symmetry during the capsule implosion at hohlraum-to-capsule radius ratio larger than 3.7.In addition, the octahedral spherical hohlraums also have potential superiority on low backscattering without supplementary technology. We studied the laser arrangement and constraints of the octahedral spherical hohlraums, and gave a design on the laser arrangement for ignition octahedral hohlraums. As a result, the injection angle of laser beams of 50°-60°was proposed as the optimum candidate range for the octahedral spherical hohlraums. We proposed a novel octahedral spherical hohlraum with cylindrical LEHs and LEH shields,in order to increase the laser coupling efficiency and improve the capsule symmetry and to mitigate the influence of the wall blowoff on laser transport. We studied on the sensitivity of the octahedral spherical hohlraums to random errors and compared the sensitivity among the octahedral spherical hohlraums, the rugby hohlraums and the cylindrical hohlraums, and the results show that the octahedral spherical hohlraums are robust to these random errors while the cylindrical hohlraums are the most sensitive.Up till to now,we have carried out three experiments on the spherical hohlraum with 2 LEHs on Shenguang(SG)laser facilities, including demonstration of improving laser transport by using the cylindrical LEHs in the spherical hohlraums, spherical hohlraum energetics on the SGIII prototype laser facility, and comparisons of laser plasma instabilities between the spherical hohlraums and the cylindrical hohlraums on the SGIII laser facility. | Ke Lan Jie Liu Zhichao Li Xufei Xie Wenyi Huo Yaohua Chen Guoli Ren Chunyang Zheng Dong Yang Sanwei Li Zhiwen Yang Liang Guo Shu Li Mingyu Zhang Xiaoying Han Chuanlei Zhai Lifei Hou Yukun Li Keli Deng Zheng Yuan Xiayu Zhan Feng Wang Guanghui Yuan Haijun Zhang Bobin Jiang Lizhen Huang Wei Zhang Kai Du Runchang Zhao Ping Li Wei Wang Jingqin Su Xuewei Deng Dongxia Hu Wei Zhou Huaiting Jia Yongkun Ding Wanguo Zheng Xiantu He | 2016 | Matter and Radiation at Extremes2016,1,1: | 2
Kupffer cell restoration after partial hepatectomy is mainly driven by local cell proliferation in IL-6-dependent autocrine and paracrine manners显示文摘Kupffer cells(KCs),which are liver-resident macrophages, originate from the fetal yolk sac and represent one of the largest macrophage populations in the body. However, the current data on the origin of the cells that restore macrophages during liver injury and regeneration remain controversial. Here,we address the question of whether liver macrophage restoration results from circulating monocyte infiltration or local KC proliferation in regenerating livers after partial hepatectomy(PHx)and uncover the underlying mechanisms.By using several strains of genetically modified mice and performing immunohistochemical analyses,we demonstrated that local KC proliferation mainly contributed to the restoration of liver macrophages after PHx.Peak KC proliferation was impaired in Il6-knockout(KO)mice and restored after the administration of IL-6 protein, whereas KC proliferation was not affected in Il4-KO or Csf2-KO mice. The source of IL-6 was identified using hepatocyte-and myeloid-specific Il6-KO mice and the results revealed that both hepatocytes and myeloid cells contribute to IL-6 production after PHx.Moreover, peak KC proliferation was also impaired in myeloid-specific Il6 receptor-KO mice after PHx,suggesting that IL-6 signaling directly promotes KC proliferation.Studies using several inhibitors to block the IL-6 signaling pathway revealed that sirtuin 1(SIRT1)contributed to IL-6-mediated KC proliferation in vitro.Genetic deletion of the Sirt1 gene in myeloid cells, including KCs,impaired KC proliferation after PHx.In conclusion, our data suggest that KC repopulation after PHx is mainly driven by local KC proliferation, which is dependent on IL-6 and SIRT1 activation in KCs. | Yeni Ait Ahmed Yaojie Fu Robim M.Rodrigues Yong He Yukun Guan Adrien Guillot Ruixue Ren Dechun Feng Juan Hidalgo Cynthia Ju Fouad Lafdil Bin Gao | 2021 | Cellular & Molecular Immunology2021,18,9: | 2

Essential role of Msx1 in regulating anterior-posterior patterning of the secondary palate in mice显示文摘Development of the secondary palate displays molecular heterogeneity along the anterior-posterior axis;however, the underlying molecular mechanism remains largely unknown. MSX1 is an anteriorly expressed transcription repressor required for palate development. Here, we investigate the role of Msx1 in regional patterning of the secondary palate. The Wnt1-Cre-mediated expression of Msx1(Rosa Msx1^(Wnt1-Cre))throughout the palatal mesenchyme leads to cleft palate in mice, associated with aberrant cell proliferation and cell death. Osteogenic patterning of the hard palate in Rosa Msx1^(Wnt1-Cre)mice is severely impaired, as revealed by a marked reduction in palatine bone formation and decreased expression of the osteogenic regulator Sp7. Overexpression and knockout of Msx1 in mice show that the transcription repressor promotes the expression of the anterior palate-specific Alx1 but represses the expression of the medialposterior palate genes Barx1, Meox2, and Tbx22. Furthermore, Tbx22 constitutes a direct Msx1 target gene in the secondary palate, suggesting that Msx1 can directly repress the expression of medial-posterior specific genes. Finally, we determine that Sp7 is downstream of Tbx22 in palatal mesenchymal cells, suggesting that a Msx1/Tbx22/Sp7 axis participates in the regulation of palate development. Our findings unveil a novel role for Msx1 in regulating the anterior-posterior growth and patterning of the secondary palate. | Shicheng Zhu Hanjing Song Liangjun Zhong Suman Huo Yukun Fang Wanxin Zhao Xueqin Yang Zhong-Min Dai Rui He Mengsheng Qiu Zunyi Zhang Xiao-Jing Zhu | 2022 | Journal of Genetics and Genomics2022,49,1: | 2

Effects of grain boundary diffusion process on magnetic properties enhancement and microstructure evolution of hot-deformed Nd-Fe-B magnets显示文摘Grain boundary diffusion(GBD)process is an important approach for producing Nd-Fe-B magnets with high coercivity and high thermal stability. The GBD for hot-deformed Nd-Fe-B magnets with nanocrystalline micro structure is more complicated compared to sintered magnets. Here,we investigated the effects of different GBD methods,i.e.,intergranular addition(in-situ GBD 1#),in-situ GBD from magnet surface during hot pressing and hot deformation(in-situ GBD 2#),and conventional GBD,on the magnetic properties and microstructure of hot deformed magnets. After the treatment by these three GBD approaches using 2 wt%Pr_(40)Tb_(30)Cu_(30)diffusion source, the coercivity of the hot-deformed magnet increases from 1281 to 1567,1412 and 2022 kA/m,respectively.The coercivity enhancement is attributed to the formation of local(Nd,Tb)2Fe14B phase with stro ng magnetic anisotropy.Reduced grain orientation is found in both in-situ GBD 1#and conventional GBD treated samples mainly due to the local stress state variation and the rotation of platelet grains.Interestingly, the in-situ GBD 2#processed sample has a high orientation at diffusion surface, which may be caused by the modified surface state of the magnet by the diffusion source.Compared with the in-situ GBD processes, the conventional GBD exhibits a higher utilization efficiency of Tb.Since the in-situ GBD is effective to treat thick hot-deformed magnets, further effort should be aimed at enhancing its diffusion efficiency. | Jiayi He Yukun Liu Zhigao Yu Jiali Cao Hongya Yu Zhongwu Liu | 2023 | Journal of Rare Earths2023,41,5: | 1
Advanced characterizations and measurements for sodium-ion batteries with NASICON-type cathode materials显示文摘NASICON (Na superionic conductor)-type cathode materials for sodium-ion batteries (SIBs) have attractedextensive attention due to their mechanically robust three-dimensional (3D) framework, which has sufficient openchannels for fast Na^(+) transportation. However, they usually suffer from inferior electronic conductivity and lowcapacity, which severely limit their practical applications. To solve these issues, we need to deeply understand thestructural evolution, redox mechanisms, and electrode/electrolyte interface reactions during cycling. Recently, rapid developments in synchrotron X-ray techniques, neutron-based resources, magnetic resonance, as well asoptical and electron microscopy have brought numerous opportunities to gain deep insights into the Na-storagebehaviors of NASICON cathodes. In this review, we summarize the detection principles of advanced characterization techniques used with typical NASICON-structured cathode materials for SIBs. The special focus is on bothoperando and ex situ techniques, which help to investigate the relationships among phase, composition, andvalence variations within electrochemical responses. Fresh electrochemical measurements and theoretical computations are also included to reveal the kinetics and energy-storage mechanisms of electrodes upon charge/discharge. Finally, we describe potential new developments in NASICON-cathodes with optimized SIB systems, foreseeing a bright future for them, achievable through the rational application of advanced diagnostic methods. | Yukun Liu Jie Li Qiuyu Shen Jian Zhang Pingge He Xuanhui Qu Yongchang Liu | 2022 | eScience2022,2,1: | 1
Comprehensive Humoral and Cellular Immune Responses to SARS-CoV-2 Variants in Diverse Chinese Population显示文摘The SARS-CoV-2 variants have been emerging and have made great challenges to current vaccine and pandemic control strategies. It is urgent to understand the current immune status of various Chinese populations given that the preexisting immunity has been established by national vaccination or ex posure to past variants.Using sera from 85 individuals(including 21 convalescents of natural infection,15 cases which suffered a breakthrough infection after being fully vaccinated, and 49 healthy vaccinees),we showed signifcantly enhanced neutralizing activities against SRAS-CoV-2 variants in convalescent sera, especially those who had been fully vaccinated. The neut ralizing antibodies against Omicron were detectable in 75%of convalescents and 4.9%of healthy vaccinees(p=0.006),with a GMT of 289.5,180.9-463.3,and 42.6,31.3-59,respetively.However,the neutralizing activities were weaker in young convalescents(aged<18y),with a detectable rate of 50%and a GMT of 46.4 against Omicron. We also examined and found no pan.sarbecovirus neutralizing activities in vaccinated SARS-CoV-I survivors.A booster dose could further increase the breadth and magnitude of neutralization against WT and variants of concern(VOCs)to different degrees. In addition,we showed that COVID-19-inactivated vaccines can elicit Omicron-specific T-cell responses. The positive rates of ELISpot reactions were 26.7%(4/15)and 43.8%(7/16)in the full vaccination group and the booster vaccination group, respectively, although without statistically significant difference. The neutralizing antibody titers declined while T-cell responses remain consistent over 6 months. These findings will inform the optimization of public health vaccination and intervention strategies to protect diverse populations against SARS-CoV-2 variants.Advances.Breakthrough infection significantly boosted neutralizing activities against SARS-CoV-2 variants a compared to booster immunization with inaclivated vaccine.Vaccine-induced vir us-specific T-cell immunity,on the other hand, may compensate for the shortall.Furthermore, the public health system should target the most vulnerable group due to a poorer protective serological response in both infected and vaccinated adolescents. | Jiwei Li Jing Wu Qiuyue Long Yan'an Wu Xiaoyi Hu Yukun He Mingzheng Jiang Jia Li Lili Zhao Shuoqi Yang Xiaoyong Chen Minghui Wang Jianshi Zheng Fangfang Wu Ruiliang Wu Lihong Ren Liang Bu Houzhao Wang Ke Li Lijuan Fu Guojun Zhang Yali Zheng Zhancheng Gao | 2022 | Research2022,,2: | 1
Modeling colorectal tumorigenesis using the organoids derived from conditionally immortalized mouse intestinal crypt cells (ciMICs)显示文摘Intestinal cancers are developed from intestinal epithelial stem cells(ISCs)in intestinal crypts through a multi-step process involved in genetic mutations of oncogenes and tumor suppressor genes.ISCs play a key role in maintaining the homeostasis of gut epithelium. In 2009,Sato et al established a three-dimensional culture system, which mimicked the niche microenvironment by employing the niche factors, and successfully grew crypt ISCs into organoids or Mini-guts in vitro.Since then, the intestinal organoid technology has been used to delineate cellular signaling in ISC biology. However, the cultured organoids consist of heterogeneous cell populations, and it was technically challenging to introduce genomic changes into three-dimensional organoids. Thus, there was a technical necessity to develop a twodimensional ISC culture system for effective genomic manipulations. In this study,we established a conditionally immortalized mouse intestinal crypt(ciMIC)cell line by using a piggyBac transposon-based SV40 T antigen expression system. We showed that the ciMICs maintained long-term proliferative activity under two-dimensional niche factor-containing culture condition, retained the biological characteristics of intestinal epithelial stem cells, and could form intestinal organoids in three-dimensional culture. While in vivo cell implantation tests indicated that the ciMICs were non-tumorigenic,the ciMICs overexpressing oncogenic b-catenin and/or KRAS exhibited high proliferative activity and developed intestinal adenoma-like pathological features in vivo.Collectively, these findings strongly suggested that the engineered ciMICs should be used as a valuable tool cell line to dissect the genetic and/or epigenetic underpinnings of intestinal tumorigenesis. | Xiaoxing Wu Zhaoxia Li Hongyu Zhang Fang He Min Qiao Huaxiu Luo Jing Zhang Meng Zhang Yukun Mao William Wagstaff Yongtao Zhang Changchun Niu Xia Zhao Hao Wang Linjuan Huang Deyao Shi Qing Liu Na Ni Kai Fu Rex C.Haydon Russell R.Reid Hue H.Luu Tong-Chuan He Ziwei Wang Houjie Liang Bing-Qiang Zhang Ning Wang | 2021 | Genes & Diseases2021,8,6: | 1
Argonaute(AGO)proteins play an essential role in mediating BMP9-induced osteogenic signaling in mesenchymal stem cells(MSCs)显示文摘As multipotent progenitor cells, mesenchymal stem cells(MSCs)can renew themselves and give rise to multiple lineages including osteoblastic, chondrogenic and adipogenic lineages. It’s previously shown that BMP9 is the most potent BMP and induces osteogenic and adipogenic differentiation of MSCs.However, the molecular mechanism through which BMP9 regulates MSC differentiation remains poorly understood.Emerging evidence indicates that noncoding RNAs,especially microRNAs,may play important roles in regulating MSC differentiation and bone formation. As highly conserved RNA binding proteins,Argonaute(AGO)proteins are essential components of the multi-protein RNA-induced silencing complexes(RISCs),which are critical for small RNA biogenesis. Here,we investigate possible roles of AGO proteins in BMP9-induced lineage-specific differentiation of MSCs.We first found that BMP9 upregulated the expression of Ago1,Ago2 and Ago3 in MSCs.By engineering multiplex siRNA vectors that express multiple siRNAs targeting individual Ago genes or all four Ago genes,we found that silencing individual Ago expression led to a decrease in BMP9-induced early osteogenic marker alkaline phosphatase(ALP)activity in MSCs.Furthermore,we demonstrated that simultaneously silencing all four Ago genes significantly diminished BMP9-induced osteogenic and adipogenic differentiation of MSCs and matrix mineralization, and ectopic bone formation.Collectively, our findings strongly indicate that AGO proteins and associated small RNA biogenesis pathway play an essential role in mediating BMP9-induced osteogenic differentiation of MSCs. | Yukun Mao Na Ni Linjuan Huang Jiaming Fan Hao Wang Fang He Qing Liu Deyao Shi Kai Fu Mikhail Pakvasa William Wagstaff Andrew Blake Tucker Connie Chen Russell R.Reid Rex C.Haydon Sherwin H.Ho Michael J.Lee Tong-Chuan He Jian Yang Le Shen Lin Cai Hue H.Luu | 2021 | Genes & Diseases2021,8,6: | 1
A new approach for high fidelity seismic data recovery by fractal interpolation显示文摘Recovering accurate data is important for both earthquake and exploration seismology studies, when data are sparsely sampled or partially missing. We present a method that allows for precise and accurate recovery of seismic data using a localized fractal recovery method. This method requires that the data are self-similar on local and global spatial scales. We present examples that show that the intrinsic structure associated with seismic data can be easily and accurately recovered by using this approach. This result, in turn, indicates that seismic data are indeed self-similar on local and global scales. This method is applicable not only for seismic studies, but also for any field studies that require accurate recovery of data from sparsely sampled datasets with partially missing data. Our ability to recover the missing data with high fidelity and accuracy will qualitatively improve the images of seismic tomography. | Hongyan Liu Tongjiang He Yukun Chen Xinfu Li | 2012 | Earthquake Science2012,25,4: | 0
Development of a simplified and inexpensive RNA depletion method for plasmid DNA purification using size selection magnetic beads(SSMBs)显示文摘Plasmid DNA(pDNA)isolation from bacterial cells is one of the most common and critical steps in molecular cloning and biomedical research.Almost all pDNA purification in-volves disruption of bacteria, removal of membrane lipids, proteins and genomic DNA,purifi-cation of pDNA from bulk lysate, and concentration of pDNA for downstream applications. While many liquid-phase and solid-phase pDNA purification methods are used, the final pDNA preparations are usually contaminated with varied degrees of host RNA,which cannot be completely digested by RNase A.To develop a simple,cost-effective,and yet effective method for RNA depletion,we investigated whether commercially available size selection magnetic beads(SSMBs),such as Mag-Bind®TotalPure NGS Kit(or Mag-Bind),can completely deplete bacterial RNA in pDNA preparations. In this proof-of-principle study,we demonstrated that, compared with RNase A digestion and two commercial plasmid affinity purification kits, the SSMB method was highly efficient in depleting contaminating RNA from pDNA minipreps.Gene transfection and bacterial colony formation assays revealed that pDNA purified from SSMB method had superior quality and integrity to pDNA samples cleaned up by RNase A digestion and/or commercial plasmid purification kits.We further demonstrated that the SSMB method completely depleted contaminating RNA in large-scale pDNA samples.Furthermore, the Mag-bind-based SSMB method costs only 5-10%of most commercial plasmid purification kits on a per sample basis. Thus, the reported SSMB method can be a valuable and inexpensive tool for the removal of bacterial RNA for routine pDNA preparations. | Xi Wang Ling Zhao Xiaoxing Wu Huaxiu Luo Di Wu Meng Zhan Jing Zhang Mikhail Pakvasa William Wagstaff Fang He Yukun Mao Yongtao Zhang Changchun Niu Meng Wu Xia Zhao Hao Wang Linjuan Huang Deyao Shi Qing Liu Na Ni Kai Fu Kelly Hynes Jason Strelzow Mostafa El Dafrawy Tong-Chuan He Hongbo Qi Zongyue Zeng | 2021 | Genes & Diseases2021,8,3: | 0
First experimental comparisons of laser-plasma interactions between spherical and cylindrical hohlraums at SGⅢ laser facility显示文摘We present our recent laser-plasmas instability(LPI)comparison experiment at the SGIII laser facility between the spherical and cylindrical hohlraums.Three kinds of filling are considered:vacuum,gas-filling with or without a capsule inside.A spherical hohlraum of 3.6 mm in diameter, and a cylindrical hohlraum of 2.4 mm?4.3 mm are used. The capsule diameter is 0.96 mm.A flat-top laser pulse with 3 ns duration and up to 92.73 kJ energy is used. The experiment has shown that the LPI level in the spherical hohlraum is close to that of the outer beam in the cylindrical hohlraum, while much lower than that of the inner beam. The experiment is further simulated by using our 2-dimensional radiation hydrodynamic code LARED-Integration,and the laser back-scattering fraction and the stimulated Raman scatter(SRS)spectrum are post-processed by the high efficiency code of laser interaction with plasmas HLIP.According to the simulation, the plasma waves are strongly damped and the SRS is mainly developed at the plasma conditions of electron density from 0.08 n_(c) to 0.1 n_(c) and electron temperature from 1.5 keV to 2.0 keV inside the hohlraums. However, obvious differences between the simulation and experiment are found, such as that the SRS back-scattering is underestimated, and the numerical SRS spectrum peaks at a larger wavelength and at a later time than the data. These dif-ferences indicate that the development of a 3D radiation hydrodynamic code, with more accurate physics models,is mandatory for spherical hohlraum study. | Yaohua Chen Zhichao Li Xufei Xie Chunyang Zheng Chuanlei Zhai Liang Hao Dong Yang Wenyi Huo Guoli Ren Jie Liu Xiaoshi Peng Tao Xu Yulong Li Sanwei Li Zhiwen Yang Liang Guo Lifei Hou Yonggang Liu Huiyue Wei Xiangming Liu Weiyi Cha Yukun Li Keli Deng Zheng Yuan Xiayu Zhan Haijun Zhang Baibin Jiang Wei Zhang Kai Du Xuewei Deng Yongkun Ding Xiaofeng Wei Wanguo Zheng Xiaodong Chen Xiantu He Ke Lan | 2017 | Matter and Radiation at Extremes2017,2,2: | 0
