2Influences of Seasonal Freezing and Thawing on Soil Water-stable Aggregates in Orchard in High Cold Region,Northeast China显示文摘Soil aggregate stability,as an important indicator of soil functions,may be affected by seasonal freezing and thawing(SFT)and land use in high cold and wet regions.Therefore,comprehensive understanding the effects of SFT on aggregate stability in orchards during winter and spring is crucial to develop appropriate management strategies that can effectively alleviate the degradation of soil quality to ensure sustainable development of orchard ecosystems.To determine the mechanism of degradation in orchard soil quality,the effects of SFT on the stability of water-stable aggregates were examined in apple-pear orchards(Pyrus ussuriensis var.ovoidea)of four different ages(11,25,40,and 63 yr)on 0 to 5%slopes before freezing and after thawing from October 2015 to June 2016 in Longjing City,Yanbian Prefecture,Northeast China,involving a comparison of planted versus adjacent uncultivated lands(control).Soil samples were collected to investigate water-stable aggregate stability in three incremental soil layers(0–20,20–40 and 40–60 cm).In the same samples,iron oxide,organic matter,and clay contents of the soil were also determined.Results showed that the destructive influences of SFT on water-stable aggregates were more pronounced with the increased orchards ages,and SFT exerted severe effects on water-stable aggregates of older orchards(40 and 63 yr)than juvenile orchards.Undergoing SFT,the soil instability index and the percentage of aggregate destruction increased by mean 0.15 mm and 1.86%,the degree of aggregation decreased by mean 1.32%,and the erosion resistance weakened,which consequently led to aggregate stability decreased.In addition,soil free,amorphous,and crystalline iron oxide as well as soil organic matter and clay contents are all important factors affecting the stability of water-stable aggregates,and their changes in their contents were consistent with those in the stability of water-stable aggregates.The results of this study suggest that long-term planting fruit trees can exacerbate the damaging effects of SFT on aggregate stability and further soil erosion increases and nutrient losses in an orchard,which hider sustainable use of soil and the productivity orchards.LIANG Yunjiang DENG Xu SONG Tao CHEN Guoshuang WANG Yuemei ZHANG Qing LU Xinrui 2021Chinese Geographical Science2021,31,2:3
3Luteolin prevents uric acid-induced pancreatic β-cell dysfunction显示文摘Elevated uric acid causes direct injury to pancreaticβ-cells.In this study,we examined the effects of luteolin,an important antioxidant,on uric acid-inducedβ-cell dysfunction.We first evaluated the effect of luteolin on nitric oxide(NO)formation in uric acid-stimulated Min6 cells using the Griess method.Next,we performed transient transfection and reporter assays to measure transcriptional activity of nuclear factor(NF)-κB.Western blotting assays were also performed to assess the effect of luteolin on the expression of MafA and inducible NO synthase(iNOS)in uric acid-treated cells.Finally,we evaluated the effect of luteolin on uric acid-induced inhibition of glucose-stimulated insulin secretion(GSIS)in Min6 cells and freshly isolated mouse pancreatic islets.We found that luteolin significantly inhibited uric acid-induced NO production,which was well correlated with reduced expression of iNOS mRNA and protein.Furthermore,decreased activity of NF-κB was implicated in inhibition by luteolin of increased iNOS expression induced by uric acid.Besides,luteolin significantly increased MafA expression in Min6cells exposed to uric acid,which was reversed by overexpression of iNOS.Moreover,luteolin prevented uric acidinduced inhibition of GSIS in both Min6 cells and mouse islets.In conclusion,luteolin protects pancreaticβ-cells from uric acid-induced dysfunction and may confer benefit on the protection of pancreaticβ-cells in hyperuricemiaassociated diabetes.Ying Ding Xuhui Shi Xuanyu Shuai Yuemei Xu Yun Liu Xiubin Liang Dong Wei Dongming Su 2014The Journal of Biomedical Research2014,28,4:3
4Studies on the isolation and culture of protoplasts from Kappaphycus alvarezii显示文摘In this study, protoplasts were successfully isolated from Kappaphycus alvarezii using snail enzymes, abalone enzymes and cellulase. The optimum enzymic ratio was fixed to be 20% of abalone enzyme, 12% of cellulase and the osmotic stabilizer was 2.0 mol/L glucose. The optimum enzymic hydrolysis conditions were found to be dark enzymolysis at 30°C continuing for 4.0 h. The resultant density and yield of protoplasts achieved 32.60×104 mL-1, 65.20×104 g-1 tissue for Kappaphycus alvarezii. 7Postnatal development of NADPH-diaphorase expression in the visual cortex of the golden hamster显示文摘Nitric oxide is an important neuromodulator in the brain and is involved in the development of visual system. But it is not clear how nitric oxide and nitric oxide synthase (NOS) are involved in the developing visual cortex of rodents. Thus we examined the expression of NOS activity in the postnatal developing visual cortex of the golden hamster by using histochemical technique for NADPH-diaphorase (NADPH-d). A heavily stained NADPH-d band was observed in the neuropil of the visual cortex. This NADPH-d band initially appeared in the cortical plate from the day of birth (P0) to postnatal day 4 (P4). From P7 to P21, this band was confined to area 17 and migrated to the deeper layers III-IV and V-VI before it eventually disappeared at P28. Such developmental trends of the band correlated well with the process of formation and establishment of the geniculo-cortical projection patterns. Thus, the areal specific development of the band suggests that NOS is closely related to the cortical differentiation and synaptic formation of the primary visual cortex. On the other hand, monocular eye enucleation on P1 could not alter the appearance of this NADPH-d positive band, indicating a non-activity dependant role of NOS. 8Control of an Intermittent Outbreak Caused by an Emerging Pandrug-Resistant Acinetobacter baumannii Clone ST457 in a Tertiary Hospital显示文摘Acinetobacter baumannii is a frequent cause of nosocomial infections.Here we report an intermittent outbreak caused by the emerging carbapenem-resistant A.baumannii(CRAB)clone ST457 and assess the effectiveness of patient screening for outbreak control.CRAB isolates were collected from 74 patients admitted to the respiratory intensive care unit of a tertiary hospital between May 2018 and March 2019.Fourteen CRAB isolates were assigned to ST457,and 13 belonged to an outbreak clone,as determined by phylogenomic analysis.Strict patient screening was started at the respiratory intensive care unit on May 2019,through which a ST457-positive patient transferred from the intensive care unit was detected.No positive patients were detected within 3months onwards.The results of this study highlight the importance of rigorous infection prevention and control measures,combined with patient screening,in controlling the CRAB outbreak.Chuqiu Zhang Tingting Xu Yang Ji Wenping Zheng Jingsong Wu Yuemei Lu Huiping Li Rongchang Chen Chen Qiu Kai Zhou 2021Infectious Microbes & Diseases2021,3,4:1

10Transfer matrices of dipoles with bending radius variation显示文摘With the increasing demand of high brightness in light source, the uniform dipole can not meet the needs of low emittance, and thus the dipole with bending radius variation is introduced in this paper. The transfer matrix of a non-uniform dipole whose bending radius is linearly changed is chosen as an example and a very simple calculation formula of non-uniform dipole transfer matrices is given. The transfer matrices of some common profile non-uniform dipoles are also listed. The comparison of these transfer matrices and the matrices calculated with slices method verifies the numerical accuracy of this formula. 11Intact regulation of G1/S transition renders esophageal squamous cell carcinoma sensitive to PI3Kαinhibitors显示文摘Phosphatidylinositol 3-kinase alpha(PI3Kα)inhibitors are currently evaluated for the therapy of esophageal squamous cell carcinoma(ESCC).It is of great importance to identify potential biomarkers to predict or monitor the efficacy of PI3Kαinhibitors in an aim to improve the clinical responsive rate in ESCC.Here,ESCC PDXs with CCND1 amplification were found to be more sensitive to CYH33,a novel PI3Kα-selective inhibitor currently in clinical trials for the treatment of advanced solid tumors including ESCC.Elevated level of cyclin D1,p21 and Rb was found in CYH33-sensitive ESCC cells compared to those in resistant cells.CYH33 significantly arrested sensitive cells but not resistant cells at G1 phase,which was associated with accumulation of p21 and suppression of Rb phosphorylation by CDK4/6 and CDK2.Hypo-phosphorylation of Rb attenuated the transcriptional activation of SKP2 by E2F1,which in turn hindered SKP2-mediated degradation of p21 and reinforced accumulation of p21.Moreover,CDK4/6 inhibitors sensitized resistant ESCC cells and PDXs to CYH33.These findings provided mechanistic rationale to evaluate PI3Kαinhibitors in ESCC patients harboring amplified CCND1 and the combined regimen with CDK4/6 inhibitors in ESCC with proficient Rb.Xu Zhang Yuxiang Wang Xi Zhang Yanyan Shen Kang Yang Qingyang Ma Yuemei Qiao Jiajie Shi Yi Wang Lan Xu Biyu Yang Gaoxiang Ge Landian Hu Xiangyin Kong Chunhao Yang Yi Chen Jian Ding Linghua Meng 2023Signal Transduction and Targeted Therapy2023,8,5:0
