1Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 2Mechanochemical preparation of sulfur-doped nanosized TiO_2 and its photocatalytic activity under visible light显示文摘In order to make nanosized TiO2 photocatalyst responsive to visible light and effectively utilize solar energy, we have, for the first time, prepared S-doped nanosized TiO2 by a mechanochemical method with hydrolysis of TiCl4. The as-prepared S-doped nanosized TiO2 possesses strong ab-sorption for visible light of 400―650 nm and shows high photocatalytic activity for decomposition of methylene blue under irradiation of visible light. The oxidation states of the S atoms incorporated into TiO2 were determined to be S6+ and S4+. 4A Dormant Multi-Controller Model for Software Defined Networking显示文摘In order to improve the scalability and reliability of Software Defined Networking(SDN),many studies use multiple controllers to constitute logically centralized control plane to provide load balancing and fail over.In this paper,we develop a flexible dormant multi-controller model based on the centralized multi-controller architecture.The dormant multi-controller model allows part of controllers to enter the dormant state under light traffic condition for saving system cost.Meanwhile,through queueing analysis,various performance measures of the system can be obtained.Moreover,we analyze the real traffic of China Education Network and use the results as the parameters of computer simulation and verify the effects of parameters on the system characteristics.Finally,a total expected cost function is established,and genetic algorithm is employed to find the optimal values of various parameters to minimize system cost for the deployment decision making.FU Yonghong BI Jun WU Jianping CHEN Ze WANG Ke LUO Min 2014China Communications2014,11,3:11
5Cytological and Molecular Characteristics of Pollen Abortion in Lily with Dysplastic Tapetum显示文摘Lily was grown worldwide as a fresh cutting flower because of its colorful petals, but its anther contained a large number of pollen grains that cause serious pollen contamination, however, pollen abortion can effectively reduce the level of pollen pollution. Our analysis aims to use cytological observation to detect the critical stage when pollen abortion occurs and to provide comprehensive gene expression information at the transcriptional level. The result showed that pollen abortion in ‘Little Kiss’ began at the mononuclear stage and the callose that covers the microspores failed to degenerate when young pollens were released from the tetrads. In addition, compared with the normally developed one,the tapetum of ‘Little Kiss’ degraded in advance while the degradation of callose was delayed. Furthermore, 103 differentially expressed genes(DEGs) related to the advance degeneration of tapetum cells and callose were found in the expression levels, including 22 transcription factors(TFs). In particular, two β-glucanase genes(endo-1,3(4)-β-glucanase, exo-β-glucanase) responsible for callose degeneration were significantly down-regulated. These results suggested that pollen abortion may occur at mononuclear stage and that early degeneration of tapetum cells resulted in a significant down-regulation of β-glucanase genes. 8Molecular diversity analysis of Tetradium ruticarpum(Wu Zhu Yu) in China based on inter-primer binding site(iPBS) markers and inter-simple sequence repeat(ISSR) markers显示文摘'Wu zhu yu', which is obtained from the dried unripe fruits of Tetradium ruticarpum(A. Jussieu) T. G. Hartley, has been used as a traditional Chinese medicine for treatment of headaches, abdominal colic, and hypertension for thousands of years. The present study was designed to assess the molecular genetic diversity among 25 collected accessions of T. ruticarpum(Wu zhu yu in Chinese) from different areas of China, based on inter-primer binding site(i PBS) markers and inter-simple sequence repeat(ISSR) markers. Thirteen ISSR primers generated 151 amplification bands, of which 130 were polymorphic. Out of 165 bands that were amplified using 10 iPBS primers, 152 were polymorphic. The iPBS markers displayed a higher proportion of polymorphic loci(PPL = 92.5%) than the ISSR markers(PPL = 84.9%). The results showed that T. ruticarpum possessed high loci polymorphism and genetic differentiation occurred in this plant. The combined data of iPBS and ISSR markers scored on 25 accessions produced five clusters that approximately matched the geographic distribution of the species. 9Variability of Soil Salinity at Multiple Spatio-Temporal Scales and the Related Driving Factors in the Oasis Areas of Xinjiang,China显示文摘Located in the inland arid area of central Asia, salt-affected farmlands take up one third of the total irrigated land area in Xinjiang of Northwest China. Spatio-temporal variability of soil salinity and the underlying mechanism are fundamental problems challenging the sustainability of oasis agriculture in China. In this study, the data of total dissolved solids(TDS) measured for soil samples collected from 27 representative study areas in the oasis areas of Xinjiang were analyzed and the coefficient of variation(CV) and stratification ratio(SR) of TDS were used to describe the lateral and vertical soil salinity variations, respectively. Weekly, monthly,and annual changes in soil salinity were also summarized. Results showed that the top(0–20 cm) soil salinity was highly variable(CV> 75%) for most studied areas. Lateral variation of soil salinity was significantly correlated with the sampling interval; as a result, a maximum sampling interval of 0.9 m was found for reducing evaluation uncertainty. The top 0–20 cm soil salt accounted for about25.2% of the total salt in the 0–100 cm soil profile. The stratification ratio values(the ratio of TDS at the 20–40 cm depth to that at the 0–20 cm depth) were mostly smaller than 1 and on average 0.92, illustrating that the top 0–20 cm soil contained slightly more salt and a considerable amount of salt still existed in subsurface and deep horizons. Irrigation reduced top soil salinity by 0.52 g kg-1, or14.6%, within the first week. On average, the relative range of soil salinity, calculated to indicate monthly changes in soil salinity, was58.2% from May to September. A 27-year experiment indicated that cultivation increased soil salinity by 44.4% at a rate of 0.14 g kg-1year-1. At small spatio-temporal scales, soil salinity variation was mainly affected by anthropogenic factors, such as irrigation and land use. However, natural factors, including groundwater, topography, and climate conditions, mainly influenced soil salinity variation at large spatio-temporal scales. This study displayed the highly variable nature of soil salinity in space and time. 10Global crustal movement and tectonic plate boundary deformation constrained by the ITRF2008显示文摘On the basis of the newly released International Terrestrial Reference Frame(ITRF2008) by the International Earth Rotation Service(IERS),a new global plate model ITRF2008 plate for the major plates is established.This ITRF2008-derived model is analyzed in comparison with NNR-NUVEL1A model,which is mainly based on geological and geophysical data.The Eurasia and Pacific plates display obvious differences in terms of the velocity fields derived from the two plate motion models.Plate acceleration is also introduced to characterize the differences of the two velocity fields which obtained from ITRF2008-plate and NNR-NUVEL1A models for major individual plates.The results show that the Africa,South America and Eurasia plates are undergoing acceleration,while the North America and Australia plates are in the state of deceleration motion.Zhu Ze Meng Guojie Su Xiaoning Wu Jicang Li Jean Xiaojing 2012Geodesy and Geodynamics2012,3,3:4

13Epsilon-near-zero photonics:infinite potentials显示文摘With its unique and exclusive linear and nonlinear optical characteristics,epsilon-near-zero(ENZ)photonics has drawn a tremendous amount of attention in the recent decade in the fields of nanophotonics,nonlinear optics,plasmonics,light-matter interactions,material science,applied optical science,etc.The extraordinary optical properties,relatively high tuning flexibility,and CMOS compatibility of ENZ materials make them popular and competitive candidates for nanophotonic devices and on-chip integration in all-optical and electro-optical platforms.With exclusive features and high performance,ENZ photonics can play a big role in optical communications and optical data processing.In this review,we give a focused discussion on recent advances of the theoretical and experimental studies on ENZ photonics,especially in the regime of nonlinear ENZ nanophotonics and its applications.First,we overview the basics of the ENZ concepts,mechanisms,and nonlinear ENZ nanophotonics.Then the new advancements in theoretical and experimental optical physics are reviewed.For nanophotonic applications,the recent decades saw rapid developments in various kinds of different ENZ-based devices and systems,which are discussed and analyzed in detail.Finally,we give our perspectives on where future endeavors can be made.JIAYE WU ZE TAO XIE YANHUA SHA H.Y.FU QIAN LI 2021Photonics Research2021,9,8:4
14Exosomal PD-L1 induces osteogenic differentiation and promotes fracture healing by acting as an immunosuppressant显示文摘A moderate inflammatory response at the early stages of fracture healing is necessary for callus formation.Over-active and continuous inflammation,however,impairs fracture healing and leads to excessive tissue damage.Adequate fracture healing could be promoted through suppression of local over-active immune cells in the fracture site.In the present study,we achieved an enriched concentration of PD-L1 from exosomes(Exos)of a genetically engineered Human Umbilical Vein Endothelial Cell(HUVECs),and demonstrated that exosomes overexpressing PD-L1 specifically bind to PD-1 on the T cell surface,suppressing the activation of T cells.Furthermore,exosomal PD-L1 induced Mesenchymal Stem Cells(MSCs)towards osteogenic differentiation when pre-cultured with T cells.Moreover,embedding of Exos into an injectable hydrogel allowed Exos delivery to the surrounding microenvironment in a time-released manner.Additionally,exosomal PD-L1,embedded in a hydrogel,markedly promoted callus formation and fracture healing in a murine model at the early over-active inflammation phase.Importantly,our results suggested that activation of T cells in the peripheral lymphatic tissues was inhibited after local administration of PD-L1-enriched Exos to the fracture sites,while T cells in distant immune organs such as the spleen were not affected.In summary,this study provides the first example of using PD-L1-enriched Exos for bone fracture repair,and highlights the potential of Hydrogel@Exos systems for bone fracture therapy through immune inhibitory effects.Ze Lin Yuan Xiong Weilin Meng Yiqiang Hu Lili Chen Lang Chen Hang Xue Adriana CPanayi Wu Zhou Yun Sun Faqi Cao Guodong Liu Liangcong Hu Chenchen Yan Xudong Xie Chuanchuan Lin Kaiyong Cai Qian Feng Bobin Mi Guohui Liu 2022Bioactive Materials2022,7,7:4
15Enhanced photocatalytic properties of molybdenum-doped BiVO4 prepared by sol–gel method显示文摘Bismuth vanadate(BiVO_4) has been investigated intensively and extensively due to its potential applications in the photocatalytic treatment of organiccontaining wastewater.In this study,Mo-BiVO_4 samples were synthesized by a facile sol-gel process.These samples were characterized by X-ray diffraction(XRD),transmission electron microscopy(TEM),and ultravioletvisible(UV-Vis) diffuse reflectance spectroscopy.It is found that the phase structure of Mo-BiVO_4 photocatalyst is monoclinic scheelite.The photocatalytic activities of the obtained BiVO_4 samples were investigated by means of the decolorization efficiency of methyl orange(MO).The BiVO_4 and Mo-BiVO_4 photocatalysts show the response on the visible light.The doped BiVO_4 photocatalysts show higher decolorization efficiency than that of bare BiVO_4.Gui-Lin Wang Lian-Wei Shan Ze Wu Li-Min Dong 2017Rare Metals2017,36,2:3
16Comparison of in vitro digestion methods for determining bioaccessibility of Hg in rice of China显示文摘Rice consumption can be a major pathway for mercury(Hg) exposure, which can cause health hazards to Chinese residents. To accurately assess the potential health risks, the bioaccessibility of Hg in rice samples collected at background and Hg-contamination sites was examined using four in vitro methods. The results showed that < 50% of the total mercury(THg) was bioaccessible in the studied rice samples. The THg bioaccessibility in the rice samples collected at the Hg-contaminated site was higher than that observed at the background area. The bioaccessibility of arsenic(As) and cadmium(Cd) was also evaluated for comparison of the four in vitro methods used in this study. The Physiologically Based Extraction Test was found to be the most accurate method based on the consistency of the results compared to those reported in previous studies. The estimated daily intakes of THg via rice consumption using the bioaccessibility data were found to meet the recommendation value set by the JECFA and the WHO in both sites. 17Dynamic Airy imaging through high_efficiency broadband phase microelements by femtosecond laser direct writing显示文摘Manipulating Airy beams to symmetric Airy beams(SABs)with abruptly autofocusing and self accelerating properties has attracted much attention.With such a particular propagation dynamic,SABs may provide great potential in dynamic signal imaging.On the other hand,the generation of SABs by spatial light modulators suffers from the limitations of phase gradient accuracy,low optical efficiency(<40%),and a bulky footprint.Therefore,exploring imaging applications and optimal generation methods of these Airy-type beams deserves further research.Here,based on the coordinate transformation of SAB,an asymmetric Airy beam(AAB)is realized.Symmetric/asymmetric cubic phase microplates(SCPPs/ACPPs)are designed and fabricated for generating SAB/AAB.The SCPP/ACPP demonstrates superior performance:compact construction(60μm × 60μm × 1.1μm),continuous variation of phase,high efficiency(~81%at 532 nm),and broadband operation from 405 to 780 nm.Dynamic imaging under monochromatic and polychromatic lights is realized by the SAB/AAB,indicating various results at different propagation distances with a certain initial signal.Further investigation reveals that the SCPP on a soft substrate maintains its physical dimensions and optical properties unchanged during stretching.Our work enables wide potential applications in integrated optics,beam manipulation,and imaging.ZE CAI XINBO QI DENG PAN SHENGYUN JI JINCHENG NI ZHAOXIN LAO CHEN XIN JIAWEN LI YANLEI HU DONG Wu JIARU CHI 2020Photonics Research2020,8,6:3
18First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 20Influence of Dust Accumulation on the Solar Reflectivity of a Linear Fresnel Reflector显示文摘Solar reflecting and heat collecting systems are generally operated outdoors year-round,and the optical performance of the reflector is reduced by dust accumulation on the surface.In this study,the dust accumulation on a linear Fresnel reflector was investigated.The relative reflectivity of the mirror before and after dust accumulation and the physical and chemical characteristics of the dust were measured using an ultraviolet spectrophotometer,scanning electron microscope,and X-ray diffractometer.The results showed that the dust density on the mirror increased,and the relative reflectivity decreased with an increase in the dust accumulation time.During 48 days of dust accumulation,the average relative reflectivity decreased 9.4%for a 1 g/m^(2)increase in the dust density.Additionally,the dust density on the mirror increased while the relative reflectivity decreased with a decrease in the mirror tilt angle.The rate of decrease of the relative reflectivity was higher for the aluminum mirror than that of the silver mirror after dust accumulation.The main component of the dust particles in the test area was SiO_(2),and the particle size range of the dust was 0.9 um to 87µm.According to the physical and chemical properties of the dust and the shielding effect of the dust on the mirror,a model to predict the influence of natural dust accumulation on the relative reflectivity of the linear Fresnel reflector was proposed.The predicted results deviated about 1%from the test results.ZHAO Xiaoyan CHEN Zhuang YAN Suying MING Tingzhen WU Ze MA Rui 2021Journal of Thermal Science2021,30,5:3
