| 3 | Antitumor effects of an engineered and energized fusion protein consisting of an anti-CD20 scFv fragment and lidamycin显示文摘Antibody-based fusion proteins are the next generation of antibody therapies for cancer and other diseases.CD20 antigen,which is overexpressed on cell membranes in nearly 95% of cases of B-cell Non-Hodgkin's Lymphoma,is an attractive target for the therapy of B-lymphoid malignancies.Lidamycin (LDM) is a potent enediyne-containing antitumor antibiotic that now has entered phase II clinical trials.In this study,we prepared an engineered fusion protein,scFv-LDP,consisting of an anti-CD20 scFv fragment and the apoprotein LDP of LDM using DNA recombination.After purification and refolding,scFv-LDP was found to bind specifically to CD20-positive lymphoma cells using ELISA and indirect immunofluorescent cytochemical staining assays.The energized fusion protein scFv-LDP-AE was obtained using molecular reconstitution of the active chromophore AE of LDM and scFv-LDP.MTT assay revealed potent cytotoxicity of scFv-LDP-AE to CD20-positive Raji and Daudi cells,with IC 50 values of 1.21×10-11 and 6.24×10-11 mol L-1,respectively.An in vivo subcutaneous xenograft model of CD20-positive B cell lymphoma in BALB/c (nu/nu) mice was also utilized.Drugs were given intravenously on day 14 and 21 after tumor transplantation.In terms of maximal tolerated doses,scFv-LDP-AE at 0.3 mg kg-1 suppressed tumor growth by 79.3%,and LDM at 0.05 mg kg-1 by 68.6% (P<0.05).Results suggested scFv-LDP-AE could be a potential candidate for tumor-targeting therapy. | FANG Hong MIAO QingFang ZHANG ShengHua CHENG Xin XIONG DongSheng ZHEN YongSu | 2011 | Science China(Life Sciences)2011,54,3: | 6 |
| 4 | Establishment and evaluation of a peanut association panel and analysis of key nutritional traits显示文摘Breeding programs aim to improve the yield and quality of peanut(Arachis hypogaea L.); using association mapping to identify genetic markers linked to these quantitative traits could facilitate selection efficiency. A peanut association panel was established consisting of 268 lines with extensive phenotypic and genetic variation, meeting the requirements for association analysis. These lines were grown over 3 years and the key agronomic traits, including protein and oil content were examined. Population structure(Q) analysis showed two subpopulations and clustering analysis was consistent with Q-based membership assignment and closely related to botanical type. Relative Kinship(K) indicated that most of the panel members have no or weak familial relatedness, with 52.78% of lines showing K ? 0. Linkagedisequilibrium(LD) analysis showed a high level of LD occurs in the panel. Model comparisons indicated false positives can be effectively controlled by taking Q and K into consideration and more false positives were generated by K than Q. A preliminary association analysis using a Q t K model found markers significantly associated with oil, protein, oleic acid, and linoleic acid, and identified a set of alleles with positive and negative effects. | 5 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 9 | Effect of Soil Drying Intensity During an Experimental Drying-Rewetting Event on Nutrient Transformation and Microbial Community Composition显示文摘Soil drying-rewetting(DRW) events affect nutrient transformation and microbial community composition; however, little is known about the influence of drying intensity during the DRW events. Therefore, we analyzed soil nutrient composition and microbial communities with exposure to various drying intensities during an experimental drying-rewetting event, using a silt loam from a grassland of northern China, where the semi-arid climate exposes soils to a wide range of moisture conditions, and grasslands account for over 40% of the nation's land area. We also conducted a sterilization experiment to examine the contribution of soil microbes to nutrient pulses. Soil drying-rewetting decreased carbon(C) mineralization by 9%–27%. Both monosaccharide and mineral nitrogen(N) contents increased with higher drying intensities(drying to ≤ 10% gravimetric water content), with the increases being 204% and 110% with the highest drying intensity(drying to 2% gravimetric water content), respectively, whereas labile phosphorus(P)only increased(by 105%) with the highest drying intensity. Moreover, levels of microbial biomass C and N and dissolved organic N decreased with increasing drying intensity and were correlated with increases in dissolved organic C and mineral N, respectively,whereas the increases in labile P were not consistent with reductions in microbial biomass P. The sterilization experiment results indicated that microbes were primarily responsible for the C and N pulses, whereas non-microbial factors were the main contributors to the labile P pulses. Phospholipid fatty acid analysis indicated that soil microbes were highly resistant to drying-rewetting events and that drought-resistant groups were probably responsible for nutrient transformation. The results show that crust_mantle structures in the investigated region obviously display layered characteristics and velocity structures and tectonic features have larger distinction in different geological structure blocks. The boundary interface C between the upper and lower crust and Moho fluctuate greatly. The shallowest depths of C (30 0km) and Moho (45 5km) under Jiashi deepen sharply from Jiashi to the western Kunlun mountain areas, where the depths of C and Moho are 44 0km and 70 0km, respectively. The higher velocity structures in the Tarim massif determine its relatively “stable” characteristics in crust tectonics. | 19 | Ammonia Volatilization from Soils Fertilized with Different Nitrogen Type and Application Method in Germination and Early Seedling Stages from the Radish Field显示文摘Ammonia volatilization(AV) from basal fertilizer with different nitrogen(N) types and application methods was investigated by the ventilation method in germination and early seedling stages during radish growth seasons in 2014. Four N fertilizer types, urea(U), ammonium bicarbonate(AB), ammonia sulfate(AS), and controlled urea formaldehyde(CUF) were applied through 5 cm depth placement(I) and 10 cm depth placement(II). The results showed that the N fertilizer type was the main factor that caused AV loss in germination and early seedling stages from the radish field. The highest and the lowest cumulative AV losses in germination and early seedling stages from the radish fields were 33.23 and 11.21 N kg/hm^2 for the treatments of AB+I and CUF+II, respectively, accounting for 60.40 and 26.40% of the N application for each treatment. The 10 cm deep placement of N reduced AV rates and lagged the AV process, and CUF significantly reduced ammonia volatilization. | 20 | Enhancement of terbium efficiency by gallium and copper co-doping in(Pr,Nd)-Fe-B sintered magnets显示文摘It is well known that Tb substitution for(Pr,Nd)in(Pr,Nd)-Fe-B based sintered magnetic materials is an effective way to increase intrinsic coercivity,but it is not quite clear whether the increment depends on the different matrix phases with various doping ingredient or not,which is essential to develop high quality magnets with high coercivity more efficiently and effectively with economic consumption of expensive Tb and other costly heavy rare earths.In this paper,we investigated the efficiency of Tb substitution for magnetic property in(Pr,Nd)-Fe-B sintered permanent magnets by co-doping Ga and Cu elements.It is shown that Ga and Cu co-doping can effectively improve the efficiency of Tb substitution to increase the thermal stability and the coercivity.The intrinsic coercivity increases up to 549 and 987 kA/m respectively by 1.5 wt%and 3.0 wt%Tb substitution in Ga and Cu co-doped magnets while the intrinsic coercivity increases up to only 334 and 613 kA/m respectively by the same amounts of Tb substitution in non-Ga and low-Cu magnets.In other words,it demonstrates that there is about 329-366 kA/m linear equivalent enhancement of intrinsic coercivity by 1.0 wt%Tb substitution for(Pr,Nd)in Ga and Cu co-doped magnets.The temperature coefficients of both intrinsic coercivityβand remanenceαat 20-150℃by 3.0 wt%Tb substitution for the magnets with Ga and Cu co-doping are-0.47%/K and-0.109%/K respectively,and in contrast those values are-0.52%/K and 0.116%/K respectively for the non-Ga and low-Cu magnets.It is the principal reason for more efficient enhancement of magnetic property by Tb substitution in the Ga and Cu co-doped magnets in which Tb atoms are expelled from triple junction phases(TJPs)to penetrate into the grain boundary phases(GB phases)and thus modify the grain boundary.It is prospected that the efficiency of Tb substitution would rely on different matrix phases with various doping constituents. | Qingfang Huang Qingzheng Jiang Yao Shi Sajjad Ur Rehman Xing Wei Ruoshu Wang Yongjian Zhang Deqin Xu Dakun Chen Zhenchen Zhong | 2023 | Journal of Rare Earths2023,41,4: | 0 |
