| 1 | Constitutive STAT3 phosphorylation and IL-6/IL-10 co-expression are associated with impaired T-cell function in tuberculosis patients显示文摘T-cells critically contribute to protection against Mycobacterium tuberculosis infection,and impaired T-cell responses can lead to disease progression.Pro-inflammatory and immunosuppressive cytokines affect T-cells,and fine-tuned regulation of cytokine signaling via the Jak/STAT signaling pathways is crucial for appropriate T-cell function.Constitutive STAT3 phosphorylation as a consequence of aberrant cytokine signaling has been described to occur in pathognomonic T-cell responses in inflammatory and autoimmune diseases.We characterized blood samples from tuberculosis patients(n=28)and healthy contacts(n=28)from Ghana for M.tuberculosis-specific T-cell responses,constitutive cytokine production,and SOCS3 and pSTAT3 expression.Lentiviral modulation of primary CD4+T-cells was performed to determine the effects of SOCS3 on T-cell functions.T-cells from tuberculosis patients expressed higher levels of IL-10 and IL-6 and lower levels of T helper type(TH)17 cytokines after M.tuberculosis-specific stimulation compared to healthy contacts.In addition,tuberculosis patients had higher IL-10 and IL-6 levels in the supernatants of non-stimulated immune cells and plasma samples compared to healthy contacts.Notably,aberrant cytokine expression was accompanied by high constitutive pSTAT3 levels and SOCS3 expression in T-cells.Multivariate analysis identified an IL-6/IL-10 co-expression-based principal component in tuberculosis patients that correlated with high pSTAT3 levels.SOCS3 contributed to a regulatory component,and tuberculosis patients with high SOCS3 expression showed decreased TH1 cytokine expression and impaired IL-2-induced STAT5 phosphorylation.SOCS3 over-expression in primary CD4+T-cells confirmed the SOCS3 inhibitory function on IL-2-induced STAT5 phosphorylation.We conclude that constitutive pSTAT3 and high SOCS3 expression are influential factors that indicate impaired T-cell functions in tuberculosis patients. | Kirstin Harling Ernest Adankwah Alptekin Güler Anthony Afum-Adjei Awuah Louis Adu-Amoah Ertan Mayatepek Ellis Owusu-Dabo Norman Nausch Marc Jacobsen | 2019 | Cellular & Molecular Immunology2019,16,3: | 15 |
| 2 | Repeat endoscopic retrograde cholangiopancreaticography after failed initial precut sphincterotomy for biliary cannulation显示文摘AIM: To investigate the outcome of repeating endoscopic retrograde cholangiopancreaticography(ERCP) after initially failed precut sphincterotomy to achieve biliary cannulation.METHODS: In this retrospective study, consecutive ERCPs performed between January 2009 and September 2012 were included. Data from our endoscopy and radiology reporting databases were analysed for use of precut sphincterotomy, biliary access rate, repeat ERCP rate and complications. Patients with initially failed precut sphincterotomy were identified.RESULTS: From 1839 consecutive ERCPs, 187(10%) patients underwent a precut sphincterotomy during the initial ERCP in attempts to cannulate a native papilla. The initial precut was successful in 79/187(42%). ERCP was repeated in 89/108(82%) of patients with failed initial precut sphincterotomy after a median interval of 4 d, leading to successful biliary cannulation in 69/89(78%). In 5 patients a third ERCP was attempted(successful in 4 cases). Overall, repeat ERCP after failed precut at the index ERCP was successful in 73/89 patients(82%). | 3 | Biloma: An unusual complication in a patient with pancreatic cancer显示文摘The term biloma describes an encapsulated collection of bile within the abdomen, usually secondary to bile duct disruption. The commonest causes reported in the literature are iatrogenic (secondary to hepatobiliary surgery), trauma or complications due to choledocholithiasis. A few cases have been reported as complications of cholangiocarcinoma or acute cholecystitis. We report the case of a 64-year-old man initially diagnosed with a non-obstructive malignancy of the pancreas, who developed a spontaneous intrahepatic biloma 8 mo later. This was identifi ed following a 1-wk history of fever, rigors and icterus. The biloma was identifi ed on computed tomography and subsequently drained under ultrasound guidance. Forty-eight hours later, a stent was inserted endoscopically into his common bile duct and he made an uneventful in-hospital recovery. | 14 | The Effect of Various Polynaphthalene Sulfonate Based Superplasticizers on the Workability of Reactive Powder Concrete显示文摘A superplasticizer is a type of chemical admixture used to alter the workability(viscosity)of fresh concrete.The workability of fresh concrete is often of particular importance when the water-to-cement(w/c)ratio is low and a particular workability is desired.Reactive Powder Concrete(RPC)is a high-strength concrete formulated to provide compressive strengths exceeding 130MPa and made of primarily powders.RPC materials typically have a very low w/c,which requires the use of a chemical admixture in order to create a material that is easier to place,handle and consolidate.Superplasticizer are commonly used for this purpose.Superplasticizers are developed from different formulations,the most common being Polycarboxylate Ether(PCE),Polymelamine Sulfonate(PMS),and Polynaphthalene Sulfonate(PNS).This study investigates the effect of various PNS based superplasticizers on the rheological performance and mechanical(compressive strength)performance of a RPC mixture.Six distinctive types of PNS based superplasticizers were used;three of various compositional strengths(high,medium,low range)from a local provider,and three of the same compositional strengths(high,medium,low)from a leading manufacturer.The properties investigated were the individual superplasticizers’viscosity,the concrete workability,determined through a mortar spread test,the concrete viscosity,and the compressive strength of the hardened RPC mixtures measured at 7,14,and 28 days.Two separate RPC mixtures were prepared,which contained two different water-to-cementitious ratios,which consequently increases the dosage of superplasticizer needed,from 34.8L/m3 to 44.7L/m3.The results show that the name brand high range composition produced the overall highest spread,lowest viscosity,and a highest compressive performance.However,the local provider outperformed the name brand in the mid and low range compositions.Lastly,the rheology assessment also confirmed that the name brand high range,and RPC fabricated with the name brand high range,developed the lowest viscosities. | Anthony Torres Federico Aguayo Srinivas Allena Michael Ellis | 2020 | Journal of Building Material Science2020,2,1: | 0 |
