2Endostatin enhances antitumor effect of tumor antigen-pulsed dendritic cell therapy in mouse xenograft model of lung carcinoma显示文摘Objective: To investigate the antitumor effect of endostatin combined with tumor antigen-pulsed dendritic cell(DC)-T cell therapy on lung cancer.Methods: Transplanted Lewis lung cancer(LLC) models of C57BL/6 mice were established by subcutaneous injection of LLC cells in left extremity axillary. Tumor antigen-pulsed DC-T cells from spleen cells and bone of mice were cultured in vitro. Tumor-bearing mice were randomly divided into three groups, including DCT+endostatin group, DC-T group, and phosphate-buffered saline(PBS) control group. Microvessel density(MVD) of tumor tissue in tumor-bearing mice was determined by immunohistochemistry(IHC). The expressions of vascular endothelial growth factor(VEGF) and hypoxia-inducible factor-1α(HIF-1α) were determined by Western blotting and IHC staining. The proportions of CD8+ T cells, mature dendritic cells(m DC), tumor-associated macrophages [TAM(M1/M2)], and myeloid-derived suppressor cells(MDSC) in suspended cells of tumor tissue were determined by flow cytometry. The expressions of interleukin(IL)-6, IL-10, IL-17, transforming growth factor-β(TGF-β) and interferon-γ(IFN-γ) in suspended cells of tumor tissue were detected by enzyme-linked immune sorbent assay(ELISA).Results: DC-T cells combined with endostatin remarkably suppressed tumor growth. MVD of mice in DCT+endostatin group was significantly lower than that of the control group and DC-T monotherapy group. The expressions of VEGF, IL-6 and IL-17 in tumors were markedly decreased, but IFN-γ and HIF-1α increased after treating with DC-T cells combined with endostatin, compared to control group and DC-T group. In the DCT+endostatin group, the proportions of MDSC and TAM(M2 type) were significantly decreased, m DC and TAM(M1 type) were up-regulated, and CD8+ T cells were recruited to infiltrate tumors, in contrast to PBS control and DC-T monotherapy. 3FGF1^(△HBS) prevents diabetic cardiomyopathy by maintaining mitochondrial homeostasis and reducing oxidative stress via AMPK/Nur77 suppression显示文摘As a classically known mitogen,fibroblast growth factor 1(FGF1)has been found to exert other pleiotropic functions such as metabolic regulation and myocardial protection.Here,we show that serum levels of FGF1 were decreased and positively correlated with fraction shortening in diabetic cardiomyopathy(DCM)patients,indicating that FGF1 is a potential therapeutic target for DCM.We found that treatment with a FGF1 variant(FGF1^(△HBS))with reduced proliferative potency prevented diabetes-induced cardiac injury and remodeling and restored cardiac function.RNA-Seq results obtained from the cardiac tissues of db/db mice showed significant increase in the expression levels of anti-oxidative genes and decrease of Nur77 by FGF1AHBS treatment.Both in vivo and in vitro studies indicate that FGF1^(△HBS) exerted these beneficial effects by markedly reducing mitochondrial fragmentation,reactive oxygen species(ROS)generation and cytochrome c leakage and enhancing mitochondrial respiration rate and β-oxidation in a 5;AMP-activated protein kinase(AMPK)/Nur77-dependent manner,all of which were not observed in the AMPK null mice.The favorable metabolic activity and reduced proliferative properties of FGF1^(△HBS) testify to its promising potential for use in the treatment of DCM and other metabolic disorders.Dezhong Wang Yuan Yin Shuyi Wang Tianyang Zhao Fanghua Gong Yushuo Zhao Beibei Wang Yuli Huang Zizhao Cheng Guanghui Zhu Zengshou Wang Yang Wang Jun Ren Guang Liang Xiaokun Li Zhifeng Huang 2021Signal Transduction and Targeted Therapy2021,6,4:6

5Electrochemiluminescence from tris(2,2′-bipyridyl) ruthenium (Ⅱ) in the presence of aminocarboxylic acid co-reactants显示文摘Due to the highly sensitive electrochemiluminescence (ECL), tris(2,2′-bipyridyl) ruthenium(II) (Ru(bpy)32+) is often used in the field of bioarrays with the help of co-reactants. However, the generally used co-reactant, tripropylamine (TPA), is toxic, corrosive and volatile. Therefore, the search for safe, sensitive and economical co-reactants is critical. Herein, three aminocarboxylic acids, ethylenediamine-tetraacetic acid (EDTA), nitrilotriacetic acid (NTA), and 2-hydroxyethylethylene diaminetriacetic acid (HEDTA), have been investigated as potential co-reactants for promoting Ru(bpy)32+ ECL behaviour. A possible ECL mechanism is also presented. The experimental results suggested that the co-reactants have a different ECL behaviour compared to TPA, such as different pH- and surfactant-responses. The detection limits of Ru(bpy)32+ using NTA, EDTA and HEDTA as co-reactants are 1, 60 and 680 fmol·L-1, respectively. The results indicate that NTA has a much higher efficiency than TPA to excite Ru(bpy)3 2+ ECL under their own optimal conditions. NTA could be widely used in many fields because it is less toxic, corrosive and volatile than TPA. Moreover, using Ru(bpy)3 2+ ECL, a sensitive method for the detection of aminocarboxylic acids is also developed. 7Novel dual inhibitor for targeting PIM1 and FGFR1 kinases inhibits colorectal cancer growth in vitro and patient-derived xenografts in vivo显示文摘Colorectal cancer(CRC) is the second most common cause of cancer-related death in the world. The pro-viral integration site for Moloney murine leukemia virus 1(PIM1) is a proto-oncogene and belongs to the serine/threonine kinase family, which are involved in cell proliferation, migration,and apoptosis. Fibroblast growth factor receptor 1(FGFR1) is a tyrosine kinase that has been implicated in cell proliferation, differentiation and migration. Small molecule HCI-48 is a derivative of chalcone, a class of compounds known to possess anti-tumor, anti-inflammatory and antibacterial effects. However,the underlying mechanism of chalcones against colorectal cancer remains unclear. This study reports that HCI-48 mainly targets PIM1 and FGFR1 kinases, thereby eliciting antitumor effects on colorectal cancer growth in vitro and in vivo. HCI-48 inhibited the activity of both PIM1 and FGFR1 kinases in an ATPdependent manner, as revealed by computational docking models. Cell-based assays showed that HCI-48inhibited cell proliferation in CRC cells(HCT-15, DLD1, HCT-116 and SW620), and induced cell cycle arrest in the G2/M phase through modulation of cyclin A2. HCI-48 also induced cellular apoptosis, as evidenced by an increase in the expression of apoptosis biomarkers such as cleaved PARP, cleaved caspase 3 and cleaved caspase 7. Moreover, HCI-48 attenuated the activation of downstream components of the PIM1 and FGFR1 signaling pathways. Using patient-derived xenograft(PDX) murine tumor models,we found that treatment with HCI-48 diminished the PDX tumor growth of implanted CRC tissue expressing high protein levels of PIM1 and FGFR1. This study suggests that the inhibitory effect of HCI-48 on colorectal tumor growth is mainly mediated through the dual-targeting of PIM1 and FGFR1kinases. 11Prediction of high-density polyethylene pyrolysis using kinetic parameters based on thermogravimetric and artificial neural networks显示文摘Pyrolysis is considered an attractive option and a promising way to dispose waste plastics.The thermogravimetric experiments of high-density polyethylene(HDPE)were conducted from 105℃ to 900℃ at different heating rates(10℃/min,20℃/min,and 30℃/min)to investigate their thermal pyrolysis behavior.We investigated four methods including three model-free methods and one modelfitting method to estimate dynamic parameters.Additionally,an artificial neural network model was developed by providing the heating rates and temperatures to predict the weight loss(wt.%)of HDPE,and optimized via assessing mean squared error and determination coefficient on the test set.The optimal MSE(2.6297×10^(−2))and R^(2) value(R^(2)>0.999)were obtained.Activation energy and preexponential factor obtained from four different models achieves the acceptable value between experimental and predicted results.The relative error of the model increased from 2.4%to 6.8%when the sampling frequency changed from 50 s to 60 s,but showed no significant difference when the sampling frequency was below 50 s.This result provides a promising approach to simplify the further modelling work and to reduce the required data storage space.This study revealed the possibility of simulating the HDPE pyrolysis process via machine learning with no significant accuracy loss of the kinetic parameters.It is hoped that this work could potentially benefit to the development of pyrolysis process modelling of HDPE and the other plastics.Xiaoxiao Yin Junyu Tao Guanyi Chen Xilei Yao Pengpeng Luan Zhanjun Cheng Ning Li Zhongyue Zhou Beibei Yan 2023Frontiers of Environmental Science & Engineering2023,17,1:0
