| 3 | PI3K/Akt/mTOR signaling orchestrates the phenotypic transition and chemo-resistance of small cell lung cancer显示文摘Small cell lung cancer(SCLC)is a phenotypically heterogeneous disease with an extremely poor prognosis,which is mainly attributed to the rapid development of resistance to chemotherapy.However,the relation between the growth phenotypes and chemo-resistance of SCLC remains largely unclear.Through comprehensive bioinformatic analyses,we found that the heterogeneity of SCLC phenotype was significantly associated with different sensitivity to chemotherapy.Adherent or semiadherent SCLC cells were enriched with activation of the PI3K/Akt/mTOR pathway and were highly chemoresistant.Mechanistically,activation of the PI3K/Akt/mTOR pathway promotes the phenotypic transition from suspension to adhesion growth pattern and confers SCLC cells with chemo-resistance.Such chemo-resistance could be largely overcome by combining chemotherapy with PI3K/Akt/mTOR pathway inhibitors.Our findings support that the PI3K/Akt/mTOR pathway plays an important role in SCLC phenotype transition and chemo-resistance,which holds important clinical implications for improving SCLC treatment. | Xuefeng Li Cheng Li Chenchen Guo Qiqi Zhao Jiayu Cao Hsin-Yi Huang Meiting Yue Yun Xue Yujuan Jin Liang Hu Hongbin Ji | 2021 | Journal of Genetics and Genomics2021,48,7: | 5 |
| 4 | Sedimentary filling and sequence structure dominated by T-R cycles of the Nenjiang Formation in the Songliao Basin显示文摘The Nenjiang Formation in the Songliao Basin is a symmetrical sequence of progradation-retrogradation,and is formed in the transgression cycle and regression cycle(T-R cycles)of the base level of deposition.We analyzed the drilling,well logging,core data,and seismic profiles of basin level,and by using the sequence stratigraphy,identified one secondary sequence boundary,two third-order sequence boundaries,and eight fourth-order sequence boundaries in the Nenjiang Formation.These eleven sequence boundaries can be divided into structural unconformity,depositional unconformity,flooding surface,and forced regression surface.Therefore the Nenjiang Formation can be subdivided into one secondary sequence,three third-order sequences,and ten fourth-order sequences.We have restored the sedimentary filling evolution within the stratigraphic framework of fourth-order sequences in the Nenjiang Formation.The sedimentary period of the first member of the Nenjiang Formation was corresponded to the global transgression period,which is also the development period of transgression cycle(T cycle),when the lake basin had the largest scope and deepest sedimentary water,the SB07(the maximum flooding surface)was formed on the top of strata during this period;covering above the SB07,there developed a set of condensation layer-oil shale,which is distributed in the whole basin and is the important source bed and regional cover.Therefore,a retrogradation sequence was formed in the T cycle of the first member of the Nenjiang Formation,characterized by the retrograding delta at a low angle.The delta has a giant front,a small plain,and many underwater distributary channels.Meanwhile,large gravity flow channels and sublacustrine fans are developed in the front of the delta.During the depositional period of the second member of the Nenjiang Formation,the R cycle began to develop due to the compression of the pacific tectonic domain;the source direction rotated 90°along with the eastern uplifting of the basin,and formed a series of east-west prograded and forced retrogradations.The prograding delta at high angel was developed in the interior of the sequence;the delta had a small front,a giant plain,and fewer underwater distributary channels,with the collapse at the foreslope,forming a series of slump fans.The slump fans can be divided into three types:discrete type,superimposed type,and fluid type.We built a whole'triad model'of the slump fan.Pointed out that the sequence of forced retrogradation formed by R cycle is a good structural mark of basin optimization,and rejected the viewpoint of'transgression'in the Nenjiang Formation of the Songliao Basin. | ZHANG ChenChen ZHANG Shun WEI Wei WU ChaoDong LIANG JiangPing NIU Wen DU JinXia FU XiuLi CUI KunNing WANG Chao WANG Hui | 2014 | Science China Earth Sciences2014,57,2: | 5 |

| 9 | Generation and application of replication-competent Venus-expressing H5N1,H7N9,and H9N2 influenza A viruses显示文摘The generation and application of replication-competent influenza A virus(IAV) expressing a reporter gene represent a valuable tool to elucidate the mechanism of viral pathogenesis and establish new countermeasures to combat the threat of influenza. Here, replication-competent IAVs with a neuraminidase(NA) segment harboring a fluorescent reporter protein, Venus, were generated in the background of H5N1, H7N9, and H9N2 influenza viruses, the three subtypes of viruses with imminent pandemic potential. All three reporter viruses maintained virion morphology, replicated with similar or slightly reduced titers relative to their parental viruses, and stably expressed the fluorescent signal for at least two passages in embryonated chicken eggs. As a proof of concept, we demonstrated that these reporter viruses,used in combination with a high-content imaging system, can serve as a convenient and rapid tool for the screening of antivirals and host factors involved in the virus life cycle. | 12 | The effect of volatile components in oil on evolutionary character- istics of diamondoids during oil thermal pyrolysis显示文摘On the basis of the results of simulation experiments, now we better understand the contribution of high carbon number hydrocarbons to diamondoid generation during thermal pyrolysis of crude oil and its sub-fractions(saturated, aromatic, resin, and asphalene fractions). However, little is known about the effect of volatile components in oil on diamondoid generation and diamondoid indices due to the lack of attention to these components in experiments. In this study, the effect of volatile components in oil on diamondoid generation and maturity indices was investigated by the pyrolysis simulation experiments on a normal crude oil from the HD23 well of the Tarim Basin and its residual oil after artificial volatilization, combined with quantitative analysis of diamondoids. The results indicate that the volatile components(≤n C12) in oil have an obvious contribution to the generation of adamantanes, which occurs mainly in the early stage of oil cracking(Easy Ro<1.0%), and influences the variations in maturity indices of adamantanes; but they have no obvious effect on the generation and maturity indices of diamantanes. | 20 | Biocatalytic Buoyancy‑Driven Nanobots for Autonomous Cell Recognition and Enrichment显示文摘Autonomously self-propelled nanoswimmers represent the nextgeneration nano-devices for bio-and environmental technology.However,current nanoswimmers generate limited energy output and can only move in short distances and duration,thus are struggling to be applied in practical challenges,such as living cell transportation.Here,we describe the construction of biodegradable metal-organic framework based nanobots with chemically driven buoyancy to achieve highly efficient,long-distance,directional vertical motion to“find-and-fetch”target cells.Nanobots surface-functionalized with antibodies against the cell surface marker carcinoembryonic antigen are exploited to impart the nanobots with specific cell targeting capacity to recognize and separate cancer cells.We demonstrate that the self-propelled motility of the nanobots can sufficiently transport the recognized cells autonomously,and the separated cells can be easily collected with a customized glass column,and finally regain their full metabolic potential after the separation.The utilization of nanobots with easy synthetic pathway shows considerable promise in cell recognition,separation,and enrichment. | Ziyi Guo Chenchen Zhuang Yihang Song Joel Yong Yi Li Zhong Guo Biao Kong John MWhitelock Joseph Wang Kang Liang | 2023 | Nano-Micro Letters2023,15,12: | 0 |
