3Establishment and application of distant hybridization technology in fish显示文摘Hybridization is widely used. However, for a long time, systematic theories and technologies related to hybridization in fish have been lacking. In this study, through long-term systematic research, we investigated and obtained the main rules regarding inheritance and reproduction related to fish distant hybridization. Furthermore, we established one-step and multistep breeding technologies that were suitable for interspecific hybridization and intraspecific hybridization. Simultaneously, we used these two breeding technologies to produce a batch of diploid fish lineages and tetraploid fish lineages and improved fishes. In addition, we widely discuss the methods, technologies and results of hybridization breeding, referring to the domestic and foreign literature on fish hybridization. 5HMGB1 from Astrocytes Promotes EAE by Influencing the Immune Cell Infiltration-Associated Functions of BMECs in Mice显示文摘High mobility group box 1(HMGB1)has been reported to play an important role in experimental autoimmune encephalomyelitis(EAE).Astrocytes are important components of neurovascular units and tightly appose the endothelial cells of microvessels by their perivascular endfeet and directly regulate the functions of the blood-brain barrier.Astrocytes express more HMGB1 during EAE while the exact roles of astrocytic HMGB1 in EAE have not been well elucidated.Here,using conditional-knockout mice,we found that astrocytic HMGB1 depletion decreased morbidity,delayed the onset time,and reduced the disease score and demyelination of EAE.Meanwhile,there were fewer immune cells,especially pathogenic T cells infiltration in the central nervous system of astrocytic HMGB1 conditional-knockout EAE mice,accompanied by up-regulated expression of the tight-junction protein Claudin5 and down-regulated expression of the cell adhesion molecules ICAM1 and VCAM1 in vivo.In vitro,HMGB1 released from astrocytes decreased Claudin5 while increased ICAM1 and VCAM1 expressed by brain microvascular endothelial cells(BMECs)through TLR4 or RAGE.Taken together,our results demonstrate that HMGB1 derived from astrocytes aggravates EAE by directly influencing the immune cell infiltration-associated functions of BMECs.Junyu Shi Yifan Xiao Na Zhang Mengya Jiao Xuhuan Tang Chan Dai Chenchen Wang Yong Xu Zheng Tan Feili Gong Fang Zheng 2022Neuroscience Bulletin2022,38,11:3
6Association of systemic inflammation and body mass index with survival in patients with resectable gastric or gastroesophageal junction adenocarcinomas显示文摘Objective:The systemic inflammation index and body mass index(BMI)are easily accessible markers that can predict mortality.However,the prognostic value of the combined use of these two markers remains unclear.The goal of this study was therefore to evaluate the association of these markers with outcomes based on a large cohort of patients with gastric cancer.Methods:A total of 2,542 consecutive patients undergoing radical surgery for gastric or gastroesophageal junction adenocarcinoma between 2009 and 2014 were included.Systemic inflammation was quantified by the preoperative neutrophil-to-lymphocyte ratio(NLR).High systemic inflammation was defined as NLR≥3,and underweight was defined as BMI<18.5 kg/m2.Results:Among 2,542 patients,NLR≥3 and underweight were common[627(25%)and 349(14%),respectively].In the entire cohort,NLR≥3 or underweight independently predicted overall survival(OS)[hazard ratio(HR):1.236,95%confidence interval(95%CI):1.069–1.430;and HR:1.600,95%CI:1.350–1.897,respectively]and recurrence-free survival(RFS)(HR:1.230,95%CI:1.054–1.434;and HR:1.658,95%CI:1.389–1.979,respectively).Patients with both NLR≥3 and underweight(vs.neither)had much worse OS(HR:2.445,95%CI:1.853–3.225)and RFS(HR:2.405,95%CI:1.802–3.209).Furthermore,we observed similar results in subgroup analyses according to pathological stage,age,and postoperative chemotherapy.Conclusions:Our results showed that preoperative elevated NLR and decreased BMI had a significant negative effect on survival.Underweight combined with severe inflammation could enhance prognostication.Taking active therapeutic measures to reduce inflammation and increase nutrition may help improve outcomes.Xianchun Gao Yanan Pan Weili Han Caie Hu Chenchen Wang Ling Chen Yong Guo Yupeng Shi Yan Pan Huahong Xie Liping Yao Jianjun Yang Jianyong Zheng Xiaohua Li Xiaonan Liu Liu Hong Jipeng Li Mengbin Li Gang Ji Zengshan Li Jielai Xia Qingchuan Zhao Daiming Fan Kaichun Wu Yongzhan Nie 2021Cancer Biology & Medicine2021,18,1:2

8Houttuynia cordata polysaccharides alleviate ulcerative colitis by restoring intestinal homeostasis显示文摘Houttuynia cordata is traditionally used as phytoantibiotics for treating lung disease in China.Houttuynia cordata polysaccharides(HCPs)have been reported to alleviate influenza virus-induced intestinal and lung immune injury by regulating the gutlung axis.The present study aims to investigate the effects and mechanisms of HCPs on ulcerative colitis(UC).Male C57BL/6 mice were induced by dextran sodium sulfate(DSS)to establish the UC animal model.Our results showed that HCPs significantly reduced the weight loss and the shortening of colon length in colitis mice,and relieved the pathological damage of colon mucosa and inhibited the expression of pro-inflammatory cytokines such as TNF-α,IL-1β,IL-6,etc.It was suggested that HCPs could significantly improve DSS-induced colitis in mice.HCPs directly protected intestinal epithelial cells,ameliorated epithelial barrier dysfunction and cell apoptosis,which was also proved in H2O2 stimulated cell apoptosis model.HCPs inhibited inflammation in the colon,which was related to suppressing the infiltration of macrophages,inhibiting the expression of pro-inflammatory cytokines and proteins(TLR4,NF-κB),and restoring the dysfunction of Th17 and Treg cells.HCPs also restored the alteration of intestinal flora induced by DSS,increased the abundance of Firmicutes and Bacteroides,and reduced the abundance of Proteobacteria.This study confirmed the protective effect of Houttuynia cordata polysaccharide extracted from traditional Chinese medicine on ulcerative colitis,of which the mechanism was closely related to the maintenance of intestinal homeostasis(intestinal barrier,immune cells,and intestinal bacteria).CEN Lifeng YI Tong HAO Yuanzhen SHI Chenchen SHI Xunlong LU Yan CHEN Daofeng ZHU Haiyan 2022Chinese Journal of Natural Medicines2022,20,12:1
9Enhanced tissue regeneration through immunomodulation of angiogenesis and osteogenesis with a multifaceted nanohybrid modified bioactive scaffold显示文摘Major traumatic tissue defects are common clinical problems often complicated by infection and local vascular dysfunction, processes which hinder the healing process. Although local application of growth factors or stem cells through various tissue engineering techniques are promising methods for the repair of tissue defects, limitations in their clinical application exist. Herein, we synthesized multifaceted nanohybrids composed of Quaternized chitosan (QCS), Graphene oxide (GO), and Polydopamine (PDA;QCS-GO-PDA). Covalent grafting of QCS and GO at a mass ratio of 5:1 (5QCS-1GO) displayed excellent biocompatibility and enhanced osteogenic ability, while addition of PDA (5QCS-1GO-PDA) reduced the level of reactive oxygen species (ROS). 5QCS-1GO-PDA was able to achieve wound tissue regeneration by reducing the inflammatory response and enhancing angiogenesis. Furthermore, Polylactic acid/hydroxyapatite (PLA/HA) composite scaffolds were printed using Selective Laser Sintering (SLS) and the hybrid nanomaterial (5QCS-1GO-PDA) was used to coat the PLA/HA scaffold (5QCS-1GO-PDA@PLA/HA) to be used for rapid bone regeneration. 5QCS-1GO-PDA not only improved angiogenesis and osteogenic differentiation, but also induced M2-type polarization of macrophages and promoted bone regeneration via the BMP2/BMPRs/Smads/Runx2 signaling pathway. 10Perivascular localized cells commit erythropoiesis in PDGF-B-expressing solid tumors显示文摘Background:Tumors possess incessant growth features,and expansion of their masses demands sufficient oxygen supply by red blood cells(RBCs).In adult mammals,the bone marrow(BM)is the main organ regulating hematopoiesis with dedicated manners.Other than BM,extramedullary hematopoiesis is discovered in various pathophysiological settings.However,whether tumors can contribute to hematopoiesis is completely unknown.Accumulating evidence shows that,in the tumor microenvironment(TME),perivascular localized cells retain progenitor cell properties and can differentiate into other cells.Here,we sought to better understand whether and how perivascular localized pericytes in tumors manipulate hematopoiesis.Methods:To test if vascular cells can differentiate into RBCs,genome-wide expression profiling was performed using mouse-derived pericytes.Genetic tracing of perivascular localized cells employing NG2-CreERT2:R26R-tdTomato mouse strain was used to validate the findings in vivo.Fluorescence-activated cell sorting(FACS),single-cell sequencing,and colony formation assays were applied for biological studies.The production of erythroid differentiationspecific cytokine,erythropoietin(EPO),in TME was checked using quantitative polymerase chain reaction(qPCR),enzyme-linked immunosorbent assay(ELISA,magnetic-activated cell sorting and immunohistochemistry.To investigate BM function in tumor erythropoiesis,BM transplantation mouse models were employed.Results:Genome-wide expression profiling showed that in response to plateletderived growth factor subunit B(PDGF-B),neural/glial antigen 2(NG2)+perivascular localized cells exhibited hematopoietic stem and progenitor-like features and underwent differentiation towards the erythroid lineage.PDGF-B simultaneously targeted cancer-associated fibroblasts to produce high levels of EPO,a crucial hormone that necessitates erythropoiesis.FACS analysis using genetic tracing of NG2+cells in tumors defined the perivascular localized cell-derived subpopulation of hematopoietic cells.Single-cell sequencing and colony formation assays validated the fact that,upon PDGF-B stimulation,NG2+cells isolated from tumors acted as erythroblast progenitor cells,which were distinctive from the canonical BM hematopoietic stem cells.Conclusions:Our data provide a new concept of hematopoiesis within tumor tissues and novel mechanistic insights into perivascular localized cell-derived erythroid cells within TME.Targeting tumor hematopoiesis is a novel therapeutic concept for treating various cancers that may have profound impacts on cancer therapy.Kayoko Hosaka Chenchen Wang Shiyue Zhang Xue Lv Takahiro Seki Yin Zhang Xu Jing Jieyu Wu Qiqiao Du Xingkang He Yulong Fan Xuan Li Makoto Kondo Masahito Yoshihara Hong Qian Lihong Shi Ping Zhu Yuanfu Xu Yunlong Yang Tao Cheng Yihai Cao 2023Cancer Communications2023,43,6:1

12Dynamic evaluation of land productivity in China显示文摘For making rational and scientific land-use planning,an inventory indicator of land productivity subject to biophysical limitations and human disturbance is required.This study introduces agricultural land productivity(ALP)as the dynamic indicator to measure land productivity,involving factors that affect agricultural production,such as characteristics of climate,soil,and terrain.As a case study of this dynamic indicator,ALP was estimated at county level and aggregated into provincial level in the whole of China.The result shows that ALP increased during 2000-2010 in China,and the ratios of ALP to the actual grain yield were 65.40%and 66.38%in the years 2000 and 2005,respectively.The ALP is an effective dynamic indicator to estimate the land productivity and to represent the basic conditions about the supply of water,energy,nutrients,and the physical basis for plant growth,as well as the local input.ALP estimation could provide supporting information for policy-making about land production planning and land-use management.Rui Yu Xiangzheng Deng Zhe Yan Chenchen Shi 2013Chinese Journal of Population,Resources and Environment2013,11,3:0
13An improved hybrid bream derived from a hybrid lineage of Megalobrama amblycephala(♀)×Culter alburnus(♂)显示文摘Distant hybridization is an important technique in fish genetic breeding.In this study,based on the establishment of an allodiploid fish lineage(BT,2n=48,F_(1)–F_(6))derived from distant hybridization between female Megalobrama amblycephala(BSB,2n=48)and male Culter alburnus(TC,2n=48),and the backcross progeny(BTB,2n=48)derived by backcrossing female F_(1) of BT to male BSB,an improved hybrid bream(BTBB,2n=48)was obtained by backcrossing BTB(♀)to BSB(♂).Moreover,the morphological and genetic characteristics of BTBB individuals were investigated;BTBB was similar to BSB in appearance but had a higher body height than BSB.The study results regarding chromosome numbers and DNA content indicated that BTBB is a diploid hybrid fish.The 5S rDNA and Hox gene of BTBB were inherited from the original parents.Gonadal development in BTBB was normal.On the other hand,BTBB had a faster growth rate,higher muscle protein level,and lower muscle carbohydrate level than BSB.Hence,bisexual fertile BTBB is promoted and can be applied as a high-quality fish,and it can also be used as a new fish germplasm resource to develop high-quality fish further.Thus,this study is of great significance for fish genetic breeding.Dingbin Gong Min Tao Lihui Xu Fangzhou Hu Zehong Wei Shi Wang Yude Wang Qingfeng Liu Chang Wu Kaikun Luo Chenchen Tang Rong Zhou Chun Zhang Yuequn Wang Shaojun Liu 2022Science China(Life Sciences)2022,65,6:0

15Chemical Synthesis of diSUMO Photoaffinity Probes for the Identification of PolySUMO Chain-Specific Interacting Proteins显示文摘Small ubiquitin-like modifiers(SUMOs)are protein modifiers that can form polymeric chains.They are important signals in cellular processes,and their study and profiling require the development of molecular tools.Herein,the authors have reported an efficient chemical protein synthesis approach for the generation of dimeric SUMO-2-based photoaffinity probes through the ligation of four readily synthesizable peptides.Proteomic studies using this diSUMO-2 probe on HeLa cell nuclear lysate found it to capture a significantly different selection of proteins compared with its monoSUMO counterparts.This resulted in the identification of several previously unknown SUMO chain-specific interacting proteins such as 40S ribosomal protein S3,which showed a significantly higher affinity for polySUMO chains than monomeric SUMO.Collectively,these results emphasize the need to develop SUMO chain-based probes in other species,and to shed light on the important role of polySUMOylation in diseases.Yu Wang Chenchen Chen Xianbin Meng Jian Fan Man Pan Jingnan Chen Haiteng Deng Jing Shi Lei Liu Yi-Ming Li 2021CCS Chemistry2021,3,4:0
16HASM quantum machine learning显示文摘The miniaturization of transistors led to advances in computers mainly to speed up their computation.Such miniaturization has approached its fundamental limits.However,many practices require better computational resources than the capabilities of existing computers.Fortunately,the development of quantum computing brings light to solve this problem.We briefly review the history of quantum computing and highlight some of its advanced achievements.Based on current studies,the Quantum Computing Advantage(QCA)seems indisputable.The challenge is how to actualize the practical quantum advantage(PQA).It is clear that machine learning can help with this task.The method used for high accuracy surface modelling(HASM)incorporates reinforced machine learning.It can be transformed into a large sparse linear system and combined with the Harrow-Hassidim-Lloyd(HHL)quantum algorithm to support quantum machine learning.HASM has been successfully used with classical computers to conduct spatial interpolation,upscaling,downscaling,data fusion and model-data assimilation of ecoenvironmental surfaces.Furthermore,a training experiment on a supercomputer indicates that our HASM-HHL quantum computing approach has a similar accuracy to classical HASM and can realize exponential acceleration over the classical algorithms.A universal platform for hybrid classical-quantum computing would be an obvious next step along with further work to improve the approach because of the many known limitations of the HHL algorithm.In addition,HASM quantum machine learning might be improved by:(1)considerably reducing the number of gates required for operating HASM-HHL;(2)evaluating cost and benchmark problems of quantum machine learning;(3)comparing the performance of the quantum and classical algorithms to clarify their advantages and disadvantages in terms of accuracy and computational speed;and(4)the algorithms would be added to a cloud platform to support applications and gather active feedback from users of the algorithms.Tianxiang YUE Chenchen WU Yi LIU Zhengping DU Na ZHAO Yimeng JIAO Zhe XU Wenjiao SHI 2023Science China Earth Sciences2023,66,9:0
17Intranasal booster using an Omicron vaccine confers broad mucosal and systemic immunity against SARS-CoV-2 variants显示文摘The highly contagious SARS-CoV-2 Omicron subvariants severely attenuated the effectiveness of currently licensed SARS-CoV-2 vaccines based on ancestral strains administered via intramuscular injection.In this study,we generated a recombinant,replication-incompetent human adenovirus type 5,Ad5-S-Omicron,that expresses Omicron BA.1 spike.Intranasal,but not intramuscular vaccination,elicited spike-specific respiratory mucosal IgA and residential T cell immune responses,in addition to systemic neutralizing antibodies and T cell immune responses against most Omicron subvariants.We tested intranasal Ad5-S-Omicron as a heterologous booster in mice that previously received intramuscular injection of inactivated ancestral vaccine.In addition to inducing serum broadly neutralizing antibodies,there was a significant induction of respiratory mucosal IgA and neutralizing activities against Omicron subvariants BA.1,BA.2,BA.5,BA.2.75,BF.7 as well as pre-Omicron strains Wildtype,Beta,and Delta.Serum and mucosal neutralizing activities against recently emerged XBB,BQ.1,and BQ.1.1 could also be detected but were much lower.Nasal lavage fluids from intranasal vaccination contained multimeric IgA that can bind to at least 10 spike proteins,including Omicron subvariants and pre-Omicron strains,and possessed broadly neutralizing activities.Intranasal vaccination using Ad5-S-Omicron or instillation of intranasal vaccinee’s nasal lavage fluids in mouse nostrils protected mice against Omicron challenge.Taken together,intranasal Ad5-S-Omicron booster on the basis of ancestral vaccines can establish effective mucosal and systemic immunity against Omicron subvariants and multiple SARS-CoV-2 variants.This candidate vaccine warrants further development as a safe,effective,and user-friendly infection and transmission-blocking vaccine.Qian Wang Chenchen Yang Li Yin Jing Sun Wei Wang Hengchun Li Zhengyuan Zhang Si Chen Bo Liu Zijian Liu Linjing Shi Xiaolin Liu Suhua Guan Chunhua Wang Linbing Qu Ying Feng Xuefeng Niu Liqiang Feng Jincun Zhao Pingchao Li Ling Chen Nanshan Zhong 2023Signal Transduction and Targeted Therapy2023,8,5:0
