| 3 | The mechanosensitive lncRNA Neat1 promotes osteoblast function through paraspeckle-dependent Smurf1 mRNA retention显示文摘Mechanical stimulation plays an important role in bone remodeling. Exercise-induced mechanical loading enhances bone strength,whereas mechanical unloading leads to bone loss. Increasing evidence has demonstrated that long noncoding RNAs(lnc RNAs) play key roles in diverse biological, physiological and pathological contexts. However, the roles of lnc RNAs in mechanotransduction and their relationships with bone formation remain unknown. In this study, we screened mechanosensing lnc RNAs in osteoblasts and identified Neat1, the most clearly decreased lnc RNA under simulated microgravity. Of note, not only Neat1 expression but also the specific paraspeckle structure formed by Neat1 was sensitive to different mechanical stimulations, which were closely associated with osteoblast function. Paraspeckles exhibited small punctate aggregates under simulated microgravity and elongated prolate or larger irregular structures under mechanical loading. Neat1 knockout mice displayed disrupted bone formation, impaired bone structure and strength, and reduced bone mass. Neat1 deficiency in osteoblasts reduced the response of osteoblasts to mechanical stimulation. In vivo, Neat1 knockout in mice weakened the bone phenotypes in response to mechanical loading and hindlimb unloading stimulation. Mechanistically, paraspeckles promoted nuclear retention of E3 ubiquitin ligase Smurf1 m RNA and downregulation of their translation, thus inhibiting ubiquitination-mediated degradation of the osteoblast master transcription factor Runx2, a Smurf1 target. | 4 | Weighted Correlation Network Analysis(WGCNA) of Japanese Flounder(Paralichthys olivaceus) Embryo Transcriptome Provides Crucial Gene Sets for Understanding Haploid Syndrome and Rescue by Diploidization显示文摘Artificial gynogenesis is of great research value in fish genetics and breeding technology. However, existing studies did not explain the mechanism of some interesting phenomena. Severe developmental defects in gynogenetic haploids can lead to death during hatching. After diploidization of chromosomes, gynogenetic diploids may dispense from the remarkable malformation and restore the viability, although the development time is longer and the survival rate is lower compared with normal diploids. The aim of this study was to reveal key mechanism in haploid syndrome of Japanese flounder, a commercially important marine teleost in East Asia. We measured genome-scale gene expression of flounder haploid, gynogenetic diploid and normal diploid embryos using RNA-Seq, constructed a module-centric co-expression network based on weighted correlation network analysis(WGCNA) and analyzed the biological functions of correlated modules. Module gene content analysis revealed that the formation of gynogenetic haploids was closely related to the abnormality of plasma proteins, and the up-regulation of p53 signaling pathway might rescue gynogenetic embryos from haploid syndrome via regulating cell cycle arrest, apoptosis and DNA repair. Moreover, normal diploid has more robust nervous system. | 8 | Integrated photonic RF self-interference cancellation on a silicon platform for full-duplex communication显示文摘In-band full-duplex(IBFD) technology can double the spectrum utilization efficiency for wireless communications,and increase the data transmission rate of B5G and 6G networks and satellite communications. RF self-interference is the major challenge for the application of IBFD technology, which must be resolved. Compared with the conventional electronic method, the photonic self-interference cancellation(PSIC) technique has the advantages of wide bandwidth, high amplitude and time delay tuning precision, and immunity to electromagnetic interference.Integrating the PSIC system on chip can effectively reduce the size, weight, and power consumption and meet the application requirement, especially for mobile terminals and small satellite payloads. In this paper, the silicon integrated PSIC chip is presented first and demonstrated for IBFD communication. The integrated PSIC chip comprises function units including phase modulation, time delay and amplitude tuning, sideband filtering, and photodetection, which complete the matching conditions for RF self-interference cancellation. Over the wide frequency range of C, X, Ku, and K bands, from 5 GHz to 25 GHz, a cancellation depth of more than 20 dB is achieved with the narrowest bandwidth of 140 MHz. A maximum bandwidth of 630 MHz is obtained at a center frequency of10 GHz. The full-duplex communication experiment at Ku-band by using the PSIC chip is carried out. Cancellation depths of 24.9 dB and 26.6 dB are measured for a bandwidth of 100 MHz at central frequencies of 12.4 GHz and14.2 GHz, respectively, and the signal of interest(SOI) with 16-quadrature amplitude modulation is recovered successfully. The factors affecting the cancellation depth and maximum interference to the SOI ratio are investigated in detail. Herein, we report an effective strategy to improve the electrochemical performance of Ni-MOF for supercapacitor by introducing a secondary Co ion. The Co substitution of Ni in Ni-MOF can improve the intrinsic reactivity and stability. As a result, the bimetallic Co/Ni-MOF-1:15 with an optimal Co/Ni ratio delivers high specific capacitance(359 F/g at 0.5 A/g), good rate performance(81.5% retention at 5 A/g) and cycling stability(81% retention after 5000 cycles).
