| 1 | Molecular subtyping and genomic profiling expand precision medicine in refractory metastatic triple-negative breast cancer:the FUTURE trial显示文摘Triple-negative breast cancer(TNBC)is a highly heterogeneous disease,and molecular subtyping may result in improved diagnostic precision and targeted therapies.Our previous study classified TNBCs into four subtypes with putative therapeutic targets.Here,we conducted the FUTURE trial(ClinicalTrials.gov identifer:NCT03805399),a phaseⅠb/Ⅱsubtyping-based and genomic biomarker-guided umbrella trial,to evaluate the efficacy of these targets.Patients with refractory metastatic TNBC were enrolled and stratifed by TNBC subtypes and genomic biomarkers,and assigned to one of these seven arms:(A)pyrotinib with capecitabine,(B)androgen receptor inhibitor with CDK4/6 inhibitor,(C)anti PD-1 with nab-paclitaxel,(D)PARP inhibitor included,(E)and(F)anti-VEGFR included,or(G)mTOR inhibitor with nab paclitaxel.The primary end point was the objective response rate(ORR).We enrolled 69 refractory metastatic TNBC patients with a median of three previous lines of therapy(range,1-8).Objective response was achieved in 20(29.0%,95%confidence interval(CI):18.7%-41.2%)of the 69 intention-to-treat(ITT)patients.Our results showed that immunotherapy(arm C),in particular,achieved the highest ORR(52.6%,95%CI:28.9%-75.6%)in the ITT population.Arm E demonstrated favorable ORR(26.1%,95%C:10.2%-48.4%in the ITT population)but with more high grade(23)adverse events.Somatic mutations of TOP2A and CD8 immunohistochemical score may have the potential to predict immunotherapy response in the immunomodulatory subtype of TNBC.In conclusion,the phaseⅠb/ⅡFUTURE trial suggested a new concept for TNBC treatment,demonstrating the clinical beneft of subtyping-based targeted therapy for refractory metastatic TNBC. | Yi-Zhou Jiang Yin Liu Yi Xiao Xin Hu Lin Jiang Wen-ia Zuo Ding Ma Jiahan Ding Xiaoyu Zhu Jianjun Zou Claire Verschraegen Daniel G.Stover Virginia Kaklamani Zhong-Hua Wang Zhi-Ming Shao | 2021 | Cell Research2021,31,2: | 37 |
| 2 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 5 | Surface and tropospheric ozone trends in the Southern Hemisphere since 1990: possible linkages to poleward expansion of the Hadley circulation显示文摘Increases in free tropospheric ozone over the past two decades are mainly in the Northern Hemisphere that have been widely documented,while ozone trends in the Southern Hemisphere(SH)remain largely unexplained.Here we first show that in-situ and satellite observations document increases of tropospheric ozone in the SH over 1990–2015.We then use a global chemical transport model to diagnose drivers of these trends.We find that increases of anthropogenic emissions(including methane)are not the most significant contributors.Instead,we explain the trend as due to changes in meteorology,and particularly in transport patterns.We propose a possible linkage of the ozone increases to meridional transport pattern shifts driven by poleward expansion of the SH Hadley circulation(SHHC).The SHHC poleward expansion allows more downward transport of ozone from the stratosphere to the troposphere at higher latitudes,and also enhances tropospheric ozone production through stronger lifting of tropical ozone precursors to the upper troposphere.These together may lead to increasing tropospheric ozone in the extratropical SH,particularly in the middle/upper troposphere and in austral autumn.Poleward expansion of the Hadley circulation is partly driven by greenhouse warming,and the associated increase in tropospheric ozone potentially provides a positive climate feedback amplifying the warming that merits further quantification. | Xiao Lu Lin Zhang Yuanhong Zhao Daniel J.Jacob Yongyun Hu Lu Hu Meng Gao Xiong Liu Irina Petropavlovskikh Audra McClure-Begley Richard Querel | 2019 | Science Bulletin2019,64,6: | 7 |

| 10 | Periodic Orbits in Rotating Second Degree and Order Gravity Fields显示文摘Periodic orbits in an arbitrary 2nd degree and order uniformly rotating gravity field are studied. We investigate the four equilibrium points in this gravity field. We see that close relation exists between the stability of these equilibria and the existence and stability of their nearby periodic orbits. We check the periodic orbits with non-zero periods. In our searching procedure for these periodic orbits, we remove the two unity eigenvalues from the state transition matrix to find a robust, non-singular linear map to solve for the periodic orbits. The algorithm converges well, especially for stable periodic orbits. Using the searching procedure, which is relatively automatic, we find five basic families of periodic orbits in the rotating second degree and order gravity field for planar motion, and discuss their existence and stability at different central body rotation rates. | Wei-Duo Hu Daniel J. | 18 | Genetic Evidence of Middle East Respiratory Syndrome Coronavirus(MERS-Cov) and Widespread Seroprevalence among Camels in Kenya显示文摘We describe the first genome isolation of Middle East respiratory syndrome coronavirus(MERS-CoV) in Kenya. This fatal zoonotic pathogen was first described in the Kingdom of Saudi Arabia in 2012. Epidemiological and molecular evidence revealed zoonotic transmission from camels to humans and between humans. Currently, MERS-CoV is classified by the WHO as having high pandemic potential requiring greater surveillance. Previous studies of MERS-CoV in Kenya mainly focused on site-specific and archived camel and human serum samples for antibodies. We conducted active nationwide cross-sectional surveillance of camels and humans in Kenya, targeting both nasal swabs and plasma samples from 1,163 camels and 486 humans collected from January 2016 to June 2018. A total of 792 camel plasma samples were positive by ELISA. Seroprevalence increased with age, and the highest prevalence was observed in adult camels(82.37%, 95%confidence interval(CI) 79.50–84.91). More female camels were significantly seropositive(74.28%, 95% CI 71.14–77.19)than male camels(P \ 0.001)(53.74%, 95% CI 48.48–58.90). Only 11 camel nasal swabs were positive for MERS-CoV by reverse transcription-quantitative PCR. Phylogenetic analysis of whole genome sequences showed that Kenyan MERSCoV clustered within sub-clade C2, which is associated with the African clade, but did not contain signature deletions of orf4 b in African viruses. None of the human plasma screened contained neutralizing antibodies against MERS-CoV. This study confirms the geographically widespread occurrence of MERS-CoV in Kenyan camels.
